WH2 and proline-rich domains of WASP-family proteins collaborate to accelerate actin filament elongation.

WASP-family proteins are known to promote assembly of branched actin networks by stimulating the filament-nucleating activity of the Arp2/3 complex. Here, we show that WASP-family proteins also function as polymerases that accelerate elongation of uncapped actin filaments. When clustered on a surface, WASP-family proteins can drive branched actin networks to grow much faster than they could by direct incorporation of soluble monomers. This polymerase activity arises from the coordinated action of two regulatory sequences: (i) a WASP homology 2 (WH2) domain that binds actin, and (ii) a proline-

Tau phosphorylation regulates the interaction between BIN1's SH3 domain and Tau's proline-rich domain.

INTRODUCTION: The application of high-throughput genomic approaches has revealed 24 novel risk loci for Alzheimer's disease (AD). We recently reported that the bridging integrator 1 (BIN1) risk gene is linked to Tau pathology. RESULTS: We used glutathione S-transferase pull-down assays and nuclear magnetic resonance (NMR) experiments to demonstrate that BIN1 and Tau proteins interact directly and then map the interaction between BIN1's SH3 domain and Tau's proline-rich domain (PRD) . Our NMR data showed that Tau phosphorylation at Thr231 weakens the SH3-PRD interaction. Using primary neurons,

Intrabodies binding the proline-rich domains of mutant huntingtin increase its turnover and reduce neurotoxicity.

Although expanded polyglutamine (polyQ) repeats are inherently toxic, causing at least nine neurodegenerative diseases, the protein context determines which neurons are affected. The polyQ expansion that causes Huntington's disease (HD) is in the first exon (HDx-1) of huntingtin (Htt). However, other parts of the protein, including the 17 N-terminal amino acids and two proline (polyP) repeat domains, regulate the toxicity of mutant Htt. The role of the P-rich domain that is flanked by the polyP domains has not been explored. Using highly specific intracellular antibodies (intrabodies), we test

Molecular recognition of proline tRNA by prolyl-tRNA synthetase from hyperthermophilic archaeon, Aeropyrum pernix K1.

To investigate the recognition mechanism of tRNA(Pro) by prolyl-tRNA synthetase from hyperthermophilic archaeon, Aeropyrum pernix K1, various tRNA(Pro) transcripts were prepared by in vitro transcription system. These transcripts were aminoacylated with proline by overexpressed A. pernix prolyl-tRNA synthetase. From prolylation experiments, recognition elements of A. pernix tRNA(Pro) were determined to be G35 and G36 of anticodon, discriminator base A73, and G1-C72 base pair at acceptor stem end.

Mutation of the GABAA receptor M1 transmembrane proline increases GABA affinity and reduces barbiturate enhancement.

All GABA(A) receptor (GABAR) subunits include an invariant proline in a consensus motif in the first transmembrane segment (M1). In receptors containing bovine alpha1, beta1 and gamma2 subunits, we analyzed the effect of mutating this M1 proline to alanine in the alpha1 or beta1 subunit using 3 different expression systems. The beta1 subunit mutant, beta1(P228A), reduced the EC(50) for GABA about 10-fold in whole cell recordings in HEK293 cells and L929 fibroblasts. The corresponding alpha1 subunit mutant (alpha1(P233A)) also reduced the GABA EC(50) when expressed in Xenopus oocytes; alpha1(P2

Novel proline substitution mutations in keratin 16 in two cases of pachyonychia congenita type 1.

Pachyonychia congenita (PC) is a group of inherited ectodermal dysplasias, the characteristic phenotype being hypertrophic nail dystrophy. Two main clinical subtypes, PC-1 and PC-2, are inherited as autosomal dominant disorders, but other less well characterized clinical forms also exist. The PC-1 phenotype may be distinguished by the absence of the epidermal cysts found in PC-2, and it has been shown to be caused by mutations in either keratin K16 or its expression partner, the K6a isoform of K6. Mutations in K16 have also been shown to cause a milder related phenotype, focal non-epidermolyti

Enantiomeric separation of basic drugs using N-benzyloxycarbonylglyclyl-L-proline as counter ion in methanol.

Direct separation of enantiomeric amines using mainly N-benzyloxycarbonylglycyl-L-proline (L-ZGP) but also N-benzyloxycarbonylglyclglcyl-L-proline (L-ZGGP) as the chiral counter ion in methanol is described. The solid phase was Hypercarb porous graphitic carbon. Several amines of pharmacological interest (e.g., alprenolol, sotalol, terbutaline, promethazine and trimipramine) were separated with high enantioselectivity (alpha = 1.16-1.98) using L-ZGP and L-ZGGP as chiral selectors. In accordance with ion-pair chromatography, the retention of the enantiomeric amines was found to increase with in

Collagen-producing mesothelial cells in adriamycin-induced pleuritis in rat. Microautoradiographic study utilizing tritiated proline.

By the microautoradiographic method using 3H-proline, collagen production by mesothelial cells was investigated in adriamycin-induced pleuritis in rats. In subpleural granulation tissue formed at 4 to 7 days after the intrapleural injection of adriamycin, proliferating fibroblasts and primitive mesenchymal cells were most intensely labeled, and abundant deposition of collagen and acid mucopolysaccharides were demonstrated about these cells. It is thus concluded that these subpleural mesenchymal cells are mainly responsible for the fibrosing process. Labeling was also observed in some reactive

Studies of the toxicological potential of tripeptides (L-valyl-L-prolyl-L-proline and L-isoleucyl-L-prolyl-L-proline): V. A 13-week toxicity study of tripeptides-containing casein hydrolysate in male and female rats.

The objective of this multiple-dose toxicity study was to assess the toxicological potential of two tripeptides, L-valyl-L-prolyl-L-proline (VPP) and L-isoleucyl-L-prolyl-L-proline (IPP), when administered once daily for 91 consecutive days to rats. The test article, powdered casein hydrolysate (CH) known to contain 0.6% VPP plus IPP, was prepared using Aspergillus oryzae protease. Prior to administration to the rats by oral gavage, the test article was suspended in sterile water. Groups of 12 male and 12 female Charles River rats were administered once daily doses of 0, 40, 200, or 1000 mg of

Exogenous nitrogen mitigation of cadmium stress in Pinellia ternata is associated with the regulation of arginine and proline metabolism.

Cadmium (Cd) contamination is a major constraint limiting the yield and quality of medicinal plants. In this study, the effects of exogenous nitrogen on alleviating Cd toxicity in Pinellia ternata seedlings were investigated, and the underlying mechanisms were explored using integrated physiological, transcriptomic, untargeted metabolomic, and rhizosphere microbial analyses. Nitrogen supplementation (150 mg N kg-¹ supplied as urea) mitigated Cd-induced growth inhibition, increasing root length by approximately 31% on days 6 and 9 and partially improving leaf area by day 9. Nitrogen supplementa

Antifreeze and self-healing hydrogels with hyaluronic acid functional cellulose nanocrystals and proline for flexible sensors.

In recent years, hydrogels have emerged as viable candidates for soft conductors in wearable flexible electronics, due to their distinctive properties including high water content, excellent biocompatibility, and adjustable mechanical properties. However, the crystallization of H2O molecules within hydrogels at low temperatures leads to significant deterioration in their toughness and mechanical strength, severely restricting their practical applications in cold environments. Cellulose nanocrystals (CNCs), rod-like nanomaterials with high crystallinity extracted from natural cellulose, have at

Mechanisms by which proline reductase of Clostridioides difficile promotes efficient metabolism and disease progression in vivo.

UNLABELLED: Clostridioides difficile is a toxin-producing pathogen that opportunistically infects those with a depleted gut microbiome, often triggered by antibiotic use. C. difficile preferentially utilizes Stickland amino acids, including proline, to promote energy generation and growth for colonization. We evaluated host outcomes from infection with wild-type and ΔprdB mouse-infective C. difficile ATCC 43255 strains to investigate how proline metabolism modulates C. difficile's pathogenesis and interactions with commensals in an enriched gut nutrient environment. While gnotobiotic mice infe

A proof-of-concept study: investigating the impact of COMT genotype and proline on negative symptoms in Alzheimer's disease.

BACKGROUND: Negative neuropsychiatric symptoms, such as apathy, are a core feature of Alzheimer's disease. Previous studies have shown that levels of fasting plasma proline and differential activity of the catechol-O-methyltransferase (COMT) enzyme, which metabolises dopamine, influence negative symptoms in patients with severe psychiatric illness and those at risk for psychosis. For patients with the COMT high activity enzyme (as assessed via the COMT Val158Met polymorphism), high plasma proline was associated with less severe negative symptoms. Conversely, high proline was associated with mo

The relationship between COMT, proline, and negative symptoms in clinical high risk and recent psychosis onset.

Individuals at clinical high-risk (CHR) of developing psychosis, as well as patients with recent psychosis onset (RO), experience significant negative symptoms that detrimentally impact daily-life functioning and are associated with poor outcomes, even in those who do not convert to psychosis. Targeting negative symptoms may thus hold promise for the treatment of CHR and RO patients. Building from previous findings we examined whether the catechol-O-methyltransferase (COMT) Val158Met functional polymorphism and fasting peripheral proline concentration predicts the severity of negative symptoms

Essential role of proline synthesis and the one-carbon metabolism pathways for systemic virulence of Streptococcus pneumoniae.

Virulence screens have indicated potential roles during Streptococcus pneumoniae infection for the one-carbon metabolism pathway component Fhs and proline synthesis mediated by ProABC. To define how these metabolic pathways affect S. pneumoniae virulence, we have investigated the phenotypes, transcription, and metabolic profiles of Δfhs and ΔproABC mutants. S. pneumoniae capsular serotype 6B BHN418 Δfhs and ΔproABC mutant strains had strongly reduced virulence in mouse sepsis and pneumonia models but could colonize the nasopharynx. Both mutant strains grew normally in complete media but had ma

The role of pH in corrosion inhibition of tin using the proline amino acid: theoretical and experimental investigations.

Herein, the effect of a mediums' pH on the interfacial interactions between proline (Pro) amino acid and tin metal was studied. In this work, acidic and near-neutral pH values were considered. DFT-based calculations and Monte Carlo simulations were carried out in the aqueous phase to understand the role of pH in Pro/tin interactions. To confirm this role in terms of inhibition efficiency, the CC and AC electrochemical techniques were used. Based on the calculated parameters, it was outlined that the partially protonated Pro form exhibits significant affinity to interact with tin metal in near-

The docking of chiral analytes on proline-based chiral stationary phases: A molecular dynamics study of selectivity.

Molecular dynamics simulations are employed to examine the selectivity of four proline-based chiral stationary phases in two solvent environments, a relatively apolar n-hexane/2-propanol solvent and a polar water/methanol solvent. The four chiral surfaces are based on a BOC-terminated diproline, a TMA-terminated diproline, a TMA-terminated triproline and a TMA-terminated hexaproline. This range of chiral selectors allows an analysis of the impact of oligomer length and terminal group on selectivity while the two solvent environments indicate the impact of solvent hydrogen bonding and polarity.

Vitamin B6 generated by obligate symbionts is critical for maintaining proline homeostasis and fecundity in tsetse flies.

The viviparous tsetse fly utilizes proline as a hemolymph-borne energy source. In tsetse, biosynthesis of proline from alanine involves the enzyme alanine-glyoxylate aminotransferase (AGAT), which requires pyridoxal phosphate (vitamin B6) as a cofactor. This vitamin can be synthesized by tsetse's obligate symbiont, Wigglesworthia glossinidia. In this study, we examined the role of Wigglesworthia-produced vitamin B6 for maintenance of proline homeostasis, specifically during the energetically expensive lactation period of the tsetse's reproductive cycle. We found that expression of agat, as wel

Insights into the roles of exogenous glutamate and proline in improving streptolydigin production of Streptomyces lydicus with metabolomic analysis.

The addition of precursors was one strategy to improve antibiotic production. The exogenous proline and glutamate, as precursors of streptolydigin, could significantly improve the streptolydigin production, but their underlying molecular mechanisms remain unknown. Herein, metabolomic analysis was carried out to explore the metabolic responses of Streptomyces lydicus to the additions of proline and glutamine. The significant differences in the quantified 53 metabolites after adding the exogenous proline and glutamate were enunciated by gas chromatography coupled to time-of-flight mass spectrome

Can zinc(II) ions be doped into the crystal structure of L-proline cadmium chloride monohydrate?

The bivalent metals Cd(II) and Zn(II) exhibit different stereochemical requirements for the set of chloride and L-proline ligands, which precludes the doping of Zn(II) ions into the crystal structure of dichloro(l-proline)cadmium(II) hydrate also referred to as L-proline cadmium chloride monohydrate (L-PCCM).
