Transparent Graphene Interfaces for Capacitive Recordings from hiPSC-Derived Cardiomyocyte Monolayers: A Proof-of-Concept Study.

Transparent conductive interfaces can enable optical pre-assessment of cardiac cell layers while remaining compatible with label-free electrophysiological recording. Here, we evaluated the integration of a monolayer graphene electrode into a capacitive recording platform for the analysis of human-induced pluripotent stem cell-derived cardiomyocyte (hiPSC-CM) monolayers. hiPSC-CMs cultured on graphene sensors formed confluent, synchronously beating monolayers that could be assessed by light microscopy prior to recording. Capacitive current transients could be recorded from spontaneously beating

Evaluating a Novel Cell-Free Preservation Solution for Human Cardiomyocyte Protection: A Proof-of-Concept Study.

BACKGROUND: Heart transplantation remains the definitive therapy for end-stage heart failure; however, its clinical impact is constrained by donor availability and preservation-associated injury. Static cold storage (SCS) with cardioplegic solutions such as Custodiol is the current clinical standard of care, yet extended cold ischemia markedly increases the risk of primary graft dysfunction. There is therefore a critical need for advanced preservation strategies and solutions that maintain graft viability under both cold and normothermic conditions. AIM: This study is aimed at comparing the e

Using a monoplane stereoscopic videoradiography configuration to accurately measure skeletal positions: a proof-of-concept study.

Biplanar videoradiography is a validated method for measuring skeletal kinematics. However, using two x-ray source-detector pairs introduces geometric constraints that can compromise image quality. The spatial configuration of the two pairs often requires a larger object-to-image distance (OID), leading to greater magnification and geometric blur. Consequently, increased magnification can limit the effective field of view, restricting multi-joint analyses. Additionally, geometric blur degrades image quality, potentially compromising subsequent bone registrations. To address this limitation, th

A morphology-driven proof-of-concept study linking interphase nuclear abnormalities to dicentric-mediated genomic instability.

Genomic instability is a major driver of clinical heterogeneity and treatment resistance in B-cell malignancies. In chronic lymphocytic leukemia (CLL) complex karyotype (CK) and disruption of the TP53 pathway are key markers of poor prognosis. Here we investigated whether interphase nuclear abnormalities - nucleoplasmic bridges (NPB) and nuclear buds (NBUD) - visible on routine blood smears could serve as cytological surrogates of dicentric chromosomes and structural genomic instability. We analyzed 29 patients with indolent B-cell malignancies: CLL (n = 15) and splenic marginal zone lymphoma

A physics-informed decomposition network for carbon ion radiotherapy dose monitoring: a proof-of-concept study.

Objective.In-beam positron emission tomography (PET) provides a promising strategy for dose monitoring in carbon ion radiotherapy (CIRT), but accurate dose prediction remains difficult due to the complex, nonlinear relationship between positron-emitter activity and physical dose deposition. This proof-of-concept study aimed to improve activity-to-dose mapping by developing decomposition-based deep learning frameworks with auxiliary physical supervision.Approach.Idealized Monte Carlo (MC) simulations were conducted on computed tomography (CT) phantoms from 18 non-small cell lung cancer patients

Clock Proteins Have the Potential to Improve Term Delivery Date Prediction: A Proof-of-Concept Study.

Our ability to accurately predict the delivery date of term pregnancies is limited by shortcomings of modern-day clinical tools and due date estimation methods. The pregnancy clock is a series of coordinated and harmonized signals between mother, fetus, and placenta that regulate the length of gestation. Clock proteins are thought to be important mediators of these signals, yet few studies have investigated their potential utility as predictors of term delivery date. In this study, we performed a cross-sectional proteome analysis of 2648 serum samples collected between 18 and 28 weeks of gesta

Needle-free vascular access port for hemodialysis: Proof-of-concept validation in an experimental animal study.

Vascular access remains a critical challenge in kidney replacement therapy. Although arteriovenous fistulas are the reference standard, they are often compromised by maturation failure, complications, and repeated needling, which causes pain and distress for patients. A needle-free alternative could enhance safety, simplify use, and improve the patient experience, particularly in self-care and home dialysis. We evaluated the Safe Hemodialysis Implantable Vascular Access Technology arteriovenous port, a patented titanium device incorporating a twist-lock system anastomosed to a vascular graft p

Disposable Face Masks for Noninvasive Drug Detection: A Proof-of-Concept Study with Cough Syrup Constituents.

Current drug detection methods, such as blood and urine analysis, are often invasive and raise ethical and privacy concerns. This study demonstrates that breathing through typical polypropylene-based meltblown cloth face masks is an efficient and user-friendly method for collecting drugs from exhaled breath for analysis. By using codeine, ephedrine, guaifenesin, and chlorpheniramine found in cough syrup as model compounds, we found that these face masks achieved a collection efficiency exceeding 92% for the tested drugs. The analysis yielded pharmacokinetic parameters─such as half-life (t1/2),

Albumin versus saline infusion for sepsis-related peripheral tissue hypoperfusion: a proof-of-concept prospective study.

BACKGROUND: Albumin has potential endothelial protective effects through antioxidant and anti-inflammatory properties. However, the effect of albumin on peripheral tissue perfusion in human sepsis remains poorly known. METHODS: Bi-centric prospective study included patients with sepsis with or without shock and prolonged CRT > 3 s despite initial resuscitation. Clinicians in charge of the patients were free to infuse either saline 500 mL or human serum albumin 20% 100 mL over 15 min. Global hemodynamic parameters as well as peripheral tissue perfusion were analyzed after 1 (H1) and 4 h (H4).

Anterior occipital condyle screw placement through the endonasal corridor: proof of concept study with cadaveric analysis.

PURPOSE: Odontoidectomy for ventral compressive pathology may result in O-C1 and/or C1-2 instability. Same-stage endonasal C1-2 spinal fusion has been advocated to eliminate risks associated with separate-stage posterior approaches. While endonasal methods for C1 instrumentation and C1-2 trans-articular stabilization exist, no hypothetical construct for endonasal occipital instrumentation has been validated. We provide an anatomic description of anterior occipital condyle (AOC) screw endonasal placement as proof-of-concept for endonasal craniocervical stabilization. METHODS: Eight adult, inje

Predictive models in SMA II natural history trajectories using machine learning: A proof of concept study.

It is known from previous literature that type II Spinal Muscular Atrophy (SMA) patients generally, after the age of 5 years, presents a steep deterioration until puberty followed by a relative stability, as most abilities have been lost. Although it is possible to identify points of slope indicating early improvement, steep decline and relative stabilizations, there is still a lot of variability within each age group and it's not always possible to predict individual trajectories of progression from age only. The aim of the study was to develop a predictive model based on machine learning usi

A new device to treat mitral valve regurgitation: a proof of concept in bench test study.

Mitral valve repair is typically performed by implanting a ring-like device at the valve annulus to reshape the annulus and to improve leaflet coaptation. In most cases, some additional procedures are needed, including leaflet resection and artificial chordae implantation. However, artificial chordae implantation could be technically challenging and postoperative left ventricular remodeling could increase the risk of recurrent mitral regurgitation. We propose an innovative annular device made of chromo-cobalt, finalized not only to reshape the annulus but also to enable anchoring of leaflets t
