Engineered Prx-LCA2 fusion protein restores oxidative skin damage via enhanced intracellular peroxidase delivery.

This study developed a novel antioxidant fusion protein Prx-LCA2 by conjugating peroxidase Prx with the LCA2 carrier derived from Escherichia coli heat-labile enterotoxin, aiming to achieve efficient intracellular delivery for oxidative damage remediation. The fusion protein Prx-LCA2 was successfully expressed in E. coli and purified. Fluorescence labeling demonstrated efficient cellular internalization of the fusion protein. In vitro, H2O2-induced oxidative stress in A431 cells was alleviated by Prx-LCA2 treatment, as evidenced by increased cell viability, reduced ROS levels, enhanced antioxi

Genetically engineered long-acting Esculentin-2CHa(1-30) fusion protein with potential applicability for the treatment of NAFLD.

Esculentin-2CHa(1-30) (‟ESC") has been reported as a potent anti-diabetic peptide with little toxicity. However, its very short plasma residence time severely limits the therapeutic efficacy. To address this issue, we genetically engineered a fusion protein of tandem trimeric ESC with an albumin binding domain (ABD) and a fusion partner, SUMO (named ‟SUMO-3×ESC-ABD"). The SUMO-3×ESC-ABD, successfully produced from E. coli, showed low cellular and hemolytic toxicity while displaying potent activities for the amelioration of hyperglycemia as well as non-alcoholic fatty liver disease (NAFLD) in v

Engineering a multi epitope vaccine against SARS-CoV-2 by exploiting its non structural and structural proteins.

SARS-CoV-2, the causative agent behind the ongoing pandemic exhibits an enhanced potential for infection when compared to its related family members- the SARS-CoV and MERS-CoV; which have caused similar disease outbreaks in the past. The severity of the global health burden, increasing mortality rate and the emergent economic crisis urgently demands the development of next generation vaccines. Amongst such emergent next generation vaccines are the multi-epitope subunit vaccines, which hold promise in combating deadly pathogens. In this study we have exploited immunoinformatics applications to

Engineered human myocardium with local release of angiogenic proteins improves vascularization and cardiac function in injured rat hearts.

Heart regeneration after myocardial infarction requires new cardiomyocytes and a supportive vascular network. Here, we evaluate the efficacy of localized delivery of angiogenic factors from biomaterials within the implanted muscle tissue to guide growth of a more dense, organized, and perfused vascular supply into implanted engineered human cardiac tissue on an ischemia/reperfusion injured rat heart. We use large, aligned 3-dimensional engineered tissue with cardiomyocytes derived from human induced pluripotent stem cells in a collagen matrix that contains dispersed alginate microspheres as lo

Phosphomimetic cardiac myosin-binding protein C partially rescues a cardiomyopathy phenotype in murine engineered heart tissue.

Phosphorylation of cardiac myosin-binding protein C (cMyBP-C), encoded by MYBPC3, increases the availability of myosin heads for interaction with actin thus enhancing contraction. cMyBP-C phosphorylation level is lower in septal myectomies of patients with hypertrophic cardiomyopathy (HCM) than in non-failing hearts. Here we compared the effect of phosphomimetic (D282) and wild-type (S282) cMyBP-C gene transfer on the HCM phenotype of engineered heart tissues (EHTs) generated from a mouse model carrying a Mybpc3 mutation (KI). KI EHTs showed lower levels of mutant Mybpc3 mRNA and protein, and

Characterization of two engineered dimeric Zika virus envelope proteins as immunogens for neutralizing antibody selection and vaccine design.

The envelope protein of Zika virus (ZIKV) exists as a dimer on the mature viral surface and is an attractive antiviral target because it mediates viral entry. However, recombinant soluble wild-type ZIKV envelope (wtZE) might preferentially exist as monomer (monZE). Recently, it has been shown that the A264C substitution could promote formation of dimeric ZIKV envelope protein (ZEA264C), requiring further characterization of purified ZEA264C for its potential applications in vaccine development. We also noted that ZEA264C, connected by disulfide bond, might be different from the noncovalent nat

Green fluorescent proteins engineered for cartilage-targeted drug delivery: Insights for transport into highly charged avascular tissues.

Osteoarthritis (OA), the most common form of arthritis, is a multi-factorial disease that primarily affects cartilage as well as other joint tissues such as subchondral bone. The lack of effective drug delivery, due to the avascular nature of cartilage and the rapid clearance of intra-articularly delivered drugs via the synovium, remains a major challenge in the development of disease modifying drugs for OA. Cationic delivery carriers can significantly enhance the uptake, penetration and retention of drugs in cartilage by interacting with negatively charged matrix proteoglycans. In this study,

The selective IGF-1 fusion protein scp776 targets apoptotic cells and promotes recovery of ischemia/reperfusion injured tissues: Engineering and preclinical validation.

Acute myocardial infarction (AMI) and acute ischemic stroke (AIS) continue to be substantial burdens on global health, despite recent advances in care. Current treatment strategies focus on blockage removal and establishment of reperfusion, but there are no approved treatments to protect cells from apoptosis resulting from hypoxia or reperfusion injury. Insulin-like growth factor-1 (IGF-1) is a pleiotropic modulator of mitogenic and anti-apoptotic pathways across multiple cell types. Administration of IGF-1 reduces cell death and promotes recovery following ischemic injury. However, the develo

A novel chassis for high-quality recombinant protein expression: apoptosis pathway engineering via multi-gene knockout and efficacy improvement of CHO cells.

BACKGROUND: Chinese hamster ovary (CHO) cells are pivotal in biopharmaceutical production, yet balancing high recombinant protein yield with cell survival remains challenging. While previous studies have targeted single apoptotic regulators, the synergistic effects of multi-gene ablation on protein stability are unknown. RESULTS: This study presents a multi-knockout strategy for CHO cells. Herewithin, CHO-K1 knockout lines were engineered via CRISPR-Cas9 targeting of key triple (TKO, BAK/BAX/CASP3), double (DKO, BAK/BAX), and single (SKO, CASP3) apoptotic nodes. Subsequently, comprehensive ana

Innovative multi-epitope vaccine engineering for dual protection against Salmonella and Shigella leveraging FliC flagellin protein adjuvant properties: an immunoinformatics-based approach.

BACKGROUND: Shigellosis and salmonellosis are prominent food-borne illnesses associated with high mortality rates and morbidity, for which, as of now, no licensed vaccine is available. Multiple epitope vaccines offer promising prospects as alternatives to conventional vaccines by combining extremely immunogenic as well as conserved epitopes to overcome weaknesses related to weak antigens as well as serotype specificity. METHODS: For the first time, this study focused on developing a new protective multiepitope vaccine for dual protection against Salmonella and Shigella by integrating six cons

Engineering Protein-Polymer Hybrid Multilayer Microreactors as Antioxidant and Anti-Inflammatory Therapeutics: Effect of Single Enzyme Nanogel Localization Within the Multilayer Systems.

Proteins have become increasingly prominent compared with conventional small molecule-based medicines in the realm of therapeutics. However, they are often prone to denaturation, instability, and aggregation, which limit their transferability to clinics. To exploit their extraordinary characteristics, emerging strategies have aimed at incorporating them into microcarriers. In this work, two complementary approaches are combined to protect catalase (CAT) from degradation and develop multilayer microreactors: the synthesis of single enzyme nanogels (SENs) and the layer-by-layer (LbL) methodology

Exploring the Structure-Function Relationships in a 5-Aminolevulinic Acid Synthase and the Use of Protein Engineering to Expand its Substrate Range.

5-Aminolevulinate synthase (ALAS) is a PLP-dependent enzyme that catalyzes the production of 5-aminolevulinate from succinyl-CoA and glycine. Its ability to catalyze the essentially irreversible C-C bond formation has significant potential in chemoenzymatic synthesis of α-amino ketones. Native ALAS, unfortunately, is extremely substrate-selective, and this seriously limits its synthetic utility. Here, we have used three different protein engineering strategies to overcome this problem for the acyl-CoA substrate. By combining previously reported mutation results and structural analysis, a serie

Deciphering the Nodamura virus Protein A Function in Schizosaccharomyces pombe and Engineering a Novel Self-Amplifying RNA (saRNA) Vector NovaVec for Vaccine Development.

Background/Objectives: Self-amplifying RNA (saRNA) vectors enable high-level transgene expression from minimal initial doses. While alphavirus-based saRNA systems are widely used, they suffer from limitations, including large genome size, complex replicase machinery, and cellular toxicity. Nodamura virus (NoV) offers a promising alternative due to its compact genome (3.2 kb) and low cytotoxicity. This study aimed to elucidate NoV RNA1 replication mechanisms and develop a novel NoV-based saRNA vector platform. Methods: We established a Schizosaccharomyces pombe system to investigate NoV RNA1 re

Engineering of the Caspase-3 Gene in Recombinant CHO Cells Caused More Apoptosis Resistance and enhanced Recombinant Protein Production Than the BAX Gene.

BACKGROUND: BAX and caspase-3 are essential genes in the apoptotic pathway of cells, promoting the apoptotic cascade through different mechanisms. Inhibition of these genes can increase the longevity of cells in cell culture. This study aimed to compare the effects of clustered regularly interspaced short palindromic repeats-CRISPR-associated protein 9 (CRISPR-Cas9)-mediated knockdown of BAX and caspase-3 genes on apoptosis inhibition, cell lifespan, and erythropoietin (EPO) production in CHO cell lines. METHODS: The BAX and caspase-3 gene expression was evaluated in the recombinant Chinese h

Sequence engineering at non-motif modulator residues yields a peptide that effectively targets a single PDZ protein in a disease-relevant cellular context.

PDZ interaction networks are finely-tuned products of evolution. These widespread binding domains recognize short linear motifs (SLiMs), usually at the C-terminus of their interacting partners, and are involved in trafficking and signaling pathways, the formation of tight junctions, and scaffolding of the post-synaptic density of neurons, amongst other roles. Typically, a single PDZ domain binds multiple targets; conversely, each PDZ-binding protein engages several PDZ domains, dependent on cellular conditions. Historical PDZ binding motifs rely on two key positions for binding. However, previ

Engineering a novel immunogenic chimera protein utilizing bacterial infections associated with atherosclerosis to induce a deviation in adaptive immune responses via Immunoinformatics approaches.

Recent studies have established the role of bacteria including Streptococcus pneumoniae, Helicobacter pylori, Chlamydia pneumonia, Mycobacterium tuberculosis, and Porphyromonas gingivalis in the development of atherosclerosis. These bacteria contribute to plaque formation via promoting Th1 immune responses and speeding up ox-LDL formation. Hence, we employed computational reverse vaccinology (RV) approaches to deviate immune response toward Th2 via engineering a novel immunogenic chimera protein. Prominent atherogenic antigens from related bacteria were identified. Then, machine learning-based

Gene-activated tissue grafts for sustained bone morphogenetic protein-2 delivery and bone engineering: Is muscle with fascia superior to muscle and fat?

Previously, we have presented an expedited strategy for sustained delivery of bone morphogenetic protein-2 (BMP-2) to bone lesions based on the implantation of gene-activated fat and muscle fragments. The aim of the present in vitro experiments was to evaluate the potential of muscle with fascia as a BMP-2 delivering osteo-regenerative implant in comparison to fat tissue and muscle alone. Subcutaneous fat, muscle, and muscle with fascia were harvested from Fischer 344 rats. The tissues were cut into small pieces and cultured for up to 90 days after direct transduction with adenoviral BMP-2 or
