A Multi-Functional Prebiotic Strategy: Crosslinked 2'-Fucosyllactose-Potato Protein Hydrolysate Conjugates Encapsulating Resveratrol for Co-Delivery to Beneficial Gut Bacteria.

Prebiotics are predominantly indigestible carbohydrate-based substrates selectively-utilized by beneficial gut-microbes to support host-health. We previously developed protein-containing prebiotics that co-deliver carbohydrate and protein substrates to the colon, where gut-microbes compete for the limited nitrogen availability, thereby enhancing microbial growth, metabolic activity, and host health compared with conventional carbohydrate prebiotics. Resveratrol is a grape-derived polyphenol with antioxidant, anti-inflammatory, and emerging prebiotic activity. Here, we developed a multifunction

The Influence of Peptidases in Intestinal Brush Border Membranes on the Absorption of Oligopeptides from Whey Protein Hydrolysate: An Ex Vivo Study Using an Ussing Chamber.

For many years, it was believed that only amino acids, dipeptides, and tripeptides could be absorbed and thus reach the bloodstream. Nowadays, the bioavailability of oligopeptides is also considered possible, leading to new research. This pilot study investigates the activity of brush border enzymes on undigested whey protein hydrolysate (WPH) and on simulated intestinal digested (ID) whey hydrolysate and the subsequent absorption of resultant peptides through the proximal jejunum of a 7-week old piglet setup in an Ussing chamber model. Amongst all samples taken, 884 oligopeptides were identif

Mechanism of galacturonic acid-rich Schisandra Chinensis polysaccharide in improving emulsifying properties of soy protein hydrolysate: Insights from interfacial behavior and molecular interactions.

In this study, Schisandra Chinensis polysaccharide (PSCP, 85.19% purity) was identified as an acidic heteropolysaccharide featuring excellent stability and favorable rheological properties. For the first time, we systematically elucidated the structure of PSCP through multiple analytical techniques, identifying a characteristic fragment: [→4)-α-D-GalpA-(1 → 4)-α-D-GalpA-(1 → 4)-β-D-Glcp-(1→]. PSCP significantly enhanced the emulsifying properties of soy protein hydrolysate (SPH, 6% hydrolysis degree) emulsions. The optimal emulsifying properties of SPH emulsion were achieved when PSCP concentr

From in silico screening to experimental verification: Identification of novel umami peptides from tamarind (Tamarindus indica L.) seed proteins and their validation in enzymatic hydrolysates.

Umami peptides are key contributors to food flavor enhancement. In this study, novel umami peptides from tamarind seed proteins were identified using an integrated strategy combining in silico screening and experimental validation. A virtual peptide library was screened based on physicochemical properties and bioactivity, yielding six candidates (VSDY, DGIDF, AKNVSDYL, YNDPHC, VPPD, and NDPHCQY). Molecular docking and dynamics simulations revealed stable binding to the umami receptor T1R1/T1R3 via hydrogen bonding and hydrophobic interactions, with binding affinities of -7.9 to -8.8 kcal/mol.

Deep eutectic solvent extraction of Camellia oleifera seed meal protein: Functional properties, antioxidant hydrolysate production and molecular mechanism compared with alkaline extraction.

Conventional alkaline extraction of Camellia oleifera seed meal (COSM) protein is not satisfactory in extraction effects and co-extraction of antinutritional saponins. This study compared five choline chloride-based deep eutectic solvents (DESs). The choline chloride/1,4-butanediol system (DES-5) showed the most favorable balance. In detail, it increased protein content from 33.91% to 50.84% while maintaining a comparable extraction rate (33.12% vs. 30.11%) and reduced saponin-equivalent levels versus alkaline extraction. DES-5-extracted protein (DES5-P) showed higher α-helix content, lower fr

Low crude protein diets supplemented with casein hydrolysate enhance the intestinal barrier function and decrease the pro-inflammatory cytokine expression in the small intestine of pigs.

To reduce nitrogen excretion and lower feeding costs, low crude protein (CP) diets are sometimes proposed, however, a great reduction of dietary CP concentration (>4% reduction vs. recommended concentration), even supplemented with essential and nonessential amino acids (AA) can detrimentally affect small intestinal barrier function and immunity, possibly due to the excessive lack of peptides. Here we hypothesize that with an extremely low CP concentration diet, protein-derived peptides, rather than AA supplementation, can improve intestinal barrier development and health. To test this hypothe

Identification and 3D modeling of bioactive peptides from Lactobacillus brevis RAMULAB49 protein hydrolysate with in silico ERK1 phosphorylation inhibition activity targeting diabetic nephropathy.

Diabetic nephropathy (DN) poses a significant health challenge, necessitating novel therapeutic approaches. In this study, we isolated proteins from cell-free supernatant (CFS) from the culture of the lactic acid bacteria Lactobacillus brevis RAMULAB49 strain. The proteins were subjected to simulated in vitro gastrointestinal digestion using gut enzymes - pepsin, pancreatin, and trypsin. The hydrolysates were filtered using 3kDa threshold ultra-centrifugal filters and were desalted using C18 disks. This was followed by nLC-ESI MS/MS tandem mass spectrometry-based identification of peptides, le

Effect of Nano- and Microparticulate Fermented Fish Protein Hydrolysate Diets on Growth, Digestive Function, Intestinal Development, and Gene Expression in Common Carp (Cyprinus carpio) Larvae.

The early larval stage is a major bottleneck in freshwater aquaculture because conventional live feeds are not always available for optimum nutrient availability and digestibility to satisfy the nutritional requirements of developing larvae. This study evaluated the effects of micro- and nano-particulate fermented and fish protein hydrolysate-based diets on growth, digestive enzyme activity, intestinal morphology, and immune-related gene expression in common carp (Cyprinus carpio) larvae. Four experimental diets, namely fish hydrolysate-based micro-feed (FHMF), fermented micro-feed (FMF), fish

Protective effects of thornback ray muscle protein hydrolysate against dyslipidemia, oxidative stress and reduced fertility induced by high cholesterol diet in adult male rats.

Enzymatic thornback ray (Raja clavata) muscle hydrolysates have been shown to have antioxidant and antihypertensive activities in vitro. The Neutrase hydrolysate exhibited the highest activities, so it was investigated along with the undigested muscle to test their hypolipidemic, antioxidative and fertility effects in rats fed with a high-cholesterol diet (HCD). Animals were allocated into four groups of 5 rats each: a normal diet group (control), a HCD group, and two groups of HCD with a daily dose of undigested muscle (Und) or the hydrolysate (MH) at 0.7 g kg-1 of body weight. Identification of Bioactive Peptides from a Laminaria digitata Protein Hydrolysate Using In Silico and In Vitro Methods to Identify Angiotensin-1-Converting Enzyme (ACE-1) Inhibitory Peptides.

Bioactive peptides range in size from 2-30 amino acids and may be derived from any protein-containing biomass using hydrolysis, fermentation or high-pressure processing. Pro-peptides or cryptides result in shorter peptide sequences following digestion and may have enhanced bioactivity. Previously, we identified a protein hydrolysate generated from Laminaria digitata that inhibited ACE-1 in vitro and had an ACE-1 IC50 value of 590 µg/mL compared to an ACE-1 IC50 value of 500 µg/mL (~2.3 µM) observed for the anti-hypertensive drug Captopril©.
