Differential glycosylation in mutant vitamin D-binding protein decimates the binding stability of vitamin D.

Vitamin D (VD) is produced by the skin upon exposure to sunlight or is obtained from dietary sources. Several risk factors are associated with VD deficiency including mutations and post-translational modifications in its transport protein known as vitamin D binding protein (VDBP) or GC-globulin. The two common single nucleotide polymorphisms rs7041 and rs4588 create three major isoforms of VDBP, including GC-1F also called wild type, GC1S, and GC-2. The 3D models for both GC-1F and GC-2 were constructed in their glycosylated states to decipher the effect of these mutations on the overall confo

Self-oligomerization regulates stability of survival motor neuron protein isoforms by sequestering an SCFSlmb degron.

Spinal muscular atrophy (SMA) is caused by homozygous mutations in human SMN1 Expression of a duplicate gene (SMN2) primarily results in skipping of exon 7 and production of an unstable protein isoform, SMNΔ7. Although SMN2 exon skipping is the principal contributor to SMA severity, mechanisms governing stability of survival motor neuron (SMN) isoforms are poorly understood. We used a Drosophila model system and label-free proteomics to identify the SCFSlmb ubiquitin E3 ligase complex as a novel SMN binding partner. SCFSlmb interacts with a phosphor degron embedded within the human and fruitfl

Structural Rearrangement upon Fragmentation of the Stability Core of the ALS-Linked Protein TDP-43.

Amyotrophic lateral sclerosis (ALS) is the most common adult degenerative motor neuron disease. Experimental evidence indicates a direct role of transactive-response DNA-binding protein 43 (TDP-43) in the pathology of ALS and other neurodegenerative diseases. TDP-43 has been identified as a major component of cytoplasmic inclusions in patients with sporadic ALS; however, the molecular basis of the disease mechanism is not yet fully understood. Fragmentation within the second RNA recognition motif (RRM2) of TDP-43 has been observed in patient tissues and may play a role in the formation of aggr

Electrochemical sensing of tumor suppressor protein p53-deoxyribonucleic acid complex stability at an electrified interface.

Electrochemical biosensors have the unique ability to convert biological events directly into electrical signals suitable for parallel analysis. Here we utilize specific properties of constant current chronopotentiometric stripping (CPS) in the analysis of protein and DNA-protein complex nanolayers. Rapid potential changes at high negative current intensities (Istr) in CPS are utilized in the analysis of DNA-protein interactions at thiol-modified mercury electrodes. P53 core domain (p53CD) sequence-specific binding to DNA results in a striking decrease in the electrocatalytic signal of free p5

Reductive methylation and mutation of an anthrax toxin fusion protein modulates its stability and cytotoxicity.

We characterized an anti-cancer fusion protein consisting of anthrax lethal factor (LF) and the catalytic domain of Pseudomonas exotoxin A by (i) mutating the N-terminal amino acids and by (ii) reductive methylation to dimethylate all lysines. Dimethylation of lysines was achieved quantitatively and specifically without affecting binding of the fusion protein to PA or decreasing the enzymatic activity of the catalytic moiety. Ubiquitination in vitro was drastically decreased for both the N-terminally mutated and dimethylated variants, and both appeared to be slightly more stable in the cytosol

An Alba-domain protein contributes to the stage-regulated stability of amastin transcripts in Leishmania.

Leishmania infantum promastigotes differentiate into amastigote forms within the phagolysosome of mammalian macrophages causing visceral leishmaniasis. Delta-amastins belong to a multigenic surface protein family of potential virulence factors that are specifically expressed in the amastigote life cycle stage through distinct regulatory elements in the 3' UTR controlling either mRNA stability or translation. Here, we provide novel insights on trans-acting factors regulating amastin developmental gene expression. Using RNA affinity chromatography with a 300 nt regulatory region within the amast

Stabilizing the subtilisin BPN' pro-domain by phage display selection: how restrictive is the amino acid code for maximum protein stability?

We have devised a procedure using monovalent phage display to select for stable mutants in the pro-domain of the serine protease, subtilisin BPN'. In complex with subtilisin, the pro-domain assumes a compact structure with a four-stranded antiparallel beta-sheet and two three-turn alpha-helices. When isolated, however, the pro-domain is 97% unfolded. These experiments use combinatorial mutagenesis to select for stabilizing amino acid combinations at a particular structural locus and determine how many combinations are close to the maximum protein stability. The selection for stability is based

RNA-binding protein Epiplakin 1 promotes ovarian cancer immune escape and distal metastasis by enhancing the RNA stability of yes-associated protein.

This study investigated the mechanism of the RNA-binding protein Epiplakin 1 (EPPK1) in ovarian cancer (OC). EPPK1 expression in OC tissues was analyzed using gene expression profiling interactive analysis (GEPIA). The association between EPPK1 expression and patient overall survival (OS) and progression-free survival (PFS) was evaluated through Kaplan-Meier analysis. Reverse transcription quantitative polymerase chain reaction (RT-qPCR) and Western blot were performed to quantify EPPK1 levels in 25 clinical OC specimens. An EPPK1-knockdown xenograft model was established to examine tumor grow

Integrating click chemistry into protein expression for efficient preparation of immobilized protein bio-surface with enhanced stability and reproducibility in affinity chromatography.

BACKGROUND: Protein immobilization method play a pivotal role in developing affinity chromatographic assays for both basic and applied research, and site-specific, covalent attachment serves as the most desirable strategy. Traditional strategy often gives rise to inconsistent results due to the use of heterobifunctional cross-linker or a similar two-step protocol. Introducing unnatural amino acids into the immobilization is possible to address the issue, however, it needs purification of the protein whereas suffers from loss of the protein activity. Herein, this work developed a method by inte

COPS5 (Jab1) protein increases β site processing of amyloid precursor protein and amyloid β peptide generation by stabilizing RanBP9 protein levels.

Increased processing of amyloid precursor protein (APP) and accumulation of neurotoxic amyloid β peptide (Aβ) in the brain is central to the pathogenesis of Alzheimer's disease (AD). Therefore, the identification of molecules that regulate Aβ generation is crucial for future therapeutic approaches for AD. We demonstrated previously that RanBP9 regulates Aβ generation in a number of cell lines and primary neuronal cultures by forming tripartite protein complexes with APP, low-density lipoprotein-related protein, and BACE1, consequently leading to increased amyloid plaque burden in the brain. Interplay Between Motor Proteins and Microtubule Stability: Involvement of Tubulin PTMs and MAPs.

Microtubules and motor proteins are components of a cytoskeleton that influence each other to maintain cellular functions. Microtubules act as tracks for motor protein-driven intracellular transport of organelles, vesicles, and macromolecules; therefore, their structural organization and stability are important. Kinesin and dynein motor proteins function not only as cargo transporters but also as contributors to the regulation of microtubule organization, stability, and interactions. The interaction of motor proteins and microtubules is essential for cellular homeostasis, and dysregulations ca

The ENL-USP7 complex regulates HIV latency through BRD4 protein stabilization.

HIV persistence in CD4+ T cells and brain microglia is maintained by host mechanisms that enforce viral latency. We identify ENL and the deubiquitinase USP7 as a host complex that sustains HIV latency. Pharmacologic inhibition or genetic loss of ENL reactivated latent HIV. Proteomic analyses revealed an ENL-USP7-BRD4 regulatory axis associated with latency maintenance, where USP7 maintained BRD4 protein stability. Targeted USP7 degradation using PROTAC induced latency reversal and destabilized BRD4. Chromatin immunoprecipitation studies showed that ENL was required for USP7 and BRD4 occupancy

A temperature-triggered phase separation system for stabilizing bioactive protein structure and function.

Protein-based pharmaceuticals such as vaccines are susceptible to temperature fluctuation-induced molecular aggregation and denaturation during transport or storage. This loss of structural integrity directly compromises their biological function. This study developed a reversible water-in-water (W/W) protein shielding system triggered by heat shock and featuring a tunable activation temperature to protect against cold chain dysfunction. The aqueous system remains homogeneous until the preset cloud point of 10 °C is surpassed, disrupting the hydrogen bond network and dehydrating polypropylene

Protein-Directed Nucleation and Stabilization of Ultrasmall Silver Nanoparticles Within BSA Hydrogels.

Biocompatible nanocomposite hydrogels are emerging as versatile platforms in nanomedicine, particularly when natural proteins are used as both structural and chemical components. In this work, we report a green, simple, and rapid in situ synthesis of ultrasmall silver nanoparticles (uAgNPs) within a bovine serum albumin (BSA) hydrogel, in which albumin simultaneously acts as the reducing agent and three-dimensional scaffold. The confined reaction environment generated uniformly dispersed Ag nanostructures with diameters in the 4-40 nm range, as confirmed by DLS and TEM. High-resolution TEM rev

LRRC71 stabilizes SEPT4 protein to ensure sperm annulus integrity and male fertility.

The structural integrity of the sperm flagellum is essential for male fertility, and its impairment is associated with reduced sperm motility. The sperm annulus is a septin-based fibrous ring that demarcates the midpiece and the principal piece. Notably, defects in sperm annulus formation frequently co-occur with abnormalities in flagellar structure; however, the underlying molecular mechanisms remain poorly understood. Herein, we identify an evolutionarily conserved leucine-rich repeat-containing protein, LRRC71, and show that its deficiency results in spermatozoa with a shortened mitochondri

Are stabilizers, located on the surface of PLGA nanoparticles, able to modify the protein adsorption pattern?

Poly(lactic-co-glycolic acid) (PLGA) is an FDA-approved, biodegradable, and biocompatible polymer, which makes it a promising starting material for the development of nanoparticles. However, in vivo studies have revealed a short biological half-life due to recognition and consequently internalization of these nanoparticles by cells of the mononuclear phagocyte system, resulting in their accumulation in the liver and spleen. In this study, we analyzed the adsorption pattern of proteins on PLGA nanoparticles after incubation with human plasma and human serum. For this analysis, different nanopar

Bioorthogonal Photocatalytic Protein Labeling and Cross-Linking Enabled by Stabilized Ketyl Radicals.

Radical reactions offer transformative potential in biological contexts but remain constrained by poor selectivity and off-target reactivity. We address these limitations through visible-light photocatalytic generation of diaryl ketyl radicals from benzophenones. This strategy circumvents traditional UV excitation pathways by suppressing triplet diradical formation─which drives nonspecific [2 + 2] cycloadditions and H atom abstraction─in favor of bioorthogonal radical-radical coupling. Our platform enables precise live-cell protein labeling with minimal cytotoxicity, including in sensitive pri

Boosting antigen-specific T cell activation with lipid-stabilized protein nanoaggregates.

Vaccines based on protein antigens have numerous advantages over inactivated pathogens, including easier manufacturing and improved safety. However, purified antigens are weakly immunogenic, as they lack the spatial organization and the associated 'danger signals' of the pathogen. Formulating vaccines as nanoparticles enhances the recognition by antigen presenting cells, boosting the cell-mediated immune response. This study describes a nano-precipitation method to obtain stable protein nanoaggregates with uniform size distribution without using covalent cross-linkers. Nanoaggregates were form

Metal cofactor stabilization by a partner protein is a widespread strategy employed for amidase activation.

Construction and remodeling of the bacterial peptidoglycan (PG) cell wall must be carefully coordinated with cell growth and division. Central to cell wall construction are hydrolases that cleave bonds in peptidoglycan. These enzymes also represent potential new antibiotic targets. One such hydrolase, the amidase LytH in Staphylococcus aureus, acts to remove stem peptides from PG, controlling where substrates are available for insertion of new PG strands and consequently regulating cell size. When it is absent, cells grow excessively large and have division defects. For activity, LytH requires

Capillary-Mediated Vitrification: A Novel Approach for Improving Thermal Stability of Enzymes and Proteins.

Capillary-mediated vitrification (CMV) is a novel method for stabilizing biological molecules and complexes. CMV leverages capillary evaporation to enable rapid desiccation of aqueous solutions while avoiding both freezing and boiling. In the CMV process, an aqueous solution containing the biological material of interest and common excipients is applied to a solid, porous support, referred to as the scaffold, and desiccated under vacuum. The pores within the scaffold accelerate drying by increasing surface area while preventing boiling through the interaction of the vapor pressure, capillary f
