Exercise promotes host anti-infection activity through CNDP2 metabolite-mediated inflammasome hyperactivation.

Exercise can enhance the body's ability to resist infections, but its molecular mechanisms are still unclear. Here, we found that exercise can induce a hyperactivation-like state of the NLRP3 inflammasome during infection, which enhances the body's ability to resist infection by continuously secreting inflammatory cytokines and reducing immune cell pyroptosis. At the molecular level, exercise enhances the binding between CNDP2 and GSDMD, promoting the production of γ-Glu-Cys. γ-Glu-Cys binds to CypD and inhibits the binding between CypD and GSDMD, thereby avoiding further damage to the mitocho

Exerkine-Mediated Regulation of the NLRP3 Inflammasome in Neuroprotection: Mechanistic Insights and the Role of Exercise.

Neurodegeneration is a leading cause of long-term disability and cognitive impairment, and the aberrant activation of the NOD-like receptor protein 3 (NLRP3) inflammasome is closely implicated in its pathogenesis. The NLRP3 inflammasome, as a central mediator of inflammatory cascades, can, when excessively activated, promote neuroinflammation and glial polarization, induce neuronal death, disrupt the blood-brain barrier, suppress angiogenesis and neurogenesis, impair synaptic plasticity, and induce inflammaging, ultimately leading to neurodegeneration. Exerkines, including neurotrophic factors

Mechanism of effect and therapeutic potential of NLRP3 inflammasome in spinal cord injury.

Spinal cord injury (SCI) is a serious and disabling central nervous system injury that can trigger various neuropathological conditions, resulting in neuronal damage and release of various pro-inflammatory mediators, leading to neurological dysfunction. Currently, surgical decompression, drugs and rehabilitation are primarily used to relieve symptoms and improve endogenous repair mechanisms; however, they cannot directly promote nerve regeneration and functional recovery. SCI can be divided into primary and secondary injuries. Secondary injury is key to determining the severity of injury, wher

CircMLH3 induces mononuclear macrophage pyroptosis in sepsis by sponging miR-590-3p to regulate TAK1 expression.

Sepsis is a fatal syndrome characterized by uncontrolled systemic inflammatory responses. Circular RNAs (circRNAs) are involved in the modulation of various pathophysiological processes, but their potential role in sepsis has largely been unexplored. In this study, we observed differential expression of circMLH3 between healthy volunteers and septic patients, and revealed the value of circMLH3 for sepsis diagnosis and prognostic prediction. Interestingly, we discovered a correlation between the expression level of circMLH3 and the degree of pyroptosis, a critical mechanism contributing to unco

Recent advances in the treatment of gout with NLRP3 inflammasome inhibitors.

Gout is an autoinflammatory disorder characterized by the accumulation of monosodium urate crystals in joints and other tissues, representing the predominant type of inflammatory arthritis with a notable prevalence and propensity for severe outcomes. The NLRP3 inflammasome, a member of the pyrin domain-containing NOD-like receptor family, exerts a substantial impact on both innate and adaptive immune responses and serves as a pivotal factor in the pathogenesis of gout. In recent years, there has been significant academic and industrial interest in the development of NLRP3-targeted small molecu

Leukadherin-1 inhibits NLRP3 inflammasome by blocking inflammasome assembly.

Aberrant activation of the NLRP3 inflammasome has been implicated in the occurrence and development of many inflammatory diseases, and thus potent inhibitors of the NLRP3 inflammasome should be explored. An antitumor agent, Leukadherin-1 (LA-1), tested in phase 1/2 clinical trials, has been reported to exert anti-inflammatory properties by blocking the NF-κB pathway. However, the effects of LA-1 on the NLRP3 inflammasome have not been conclusively determined. In this study, we found that at lower doses (below 1 μM) ex vivo, LA-1 blocked NLRP3 inflammasome activation without affecting NF-κB sig

NLRP3-Induced NETosis: A Potential Therapeutic Target for Ischemic Thrombotic Diseases?

Ischemic thrombotic disease, characterized by the formation of obstructive blood clots within arteries or veins, is a condition associated with life-threatening events, such as stroke, myocardial infarction, deep vein thrombosis, and pulmonary embolism. The conventional therapeutic strategy relies on treatments with anticoagulants that unfortunately pose an inherent risk of bleeding complications. These anticoagulants primarily target clotting factors, often overlooking upstream events, including the release of neutrophil extracellular traps (NETs). Neutrophils are integral components of the i

The role of NLRP3 inflammasome in sepsis: A potential therapeutic target.

Sepsis is the host immune imbalance following infection and leads to organ dysfunction, with highly complicated pathophysiology. To date, sepsis still lacks effective therapies with high mortality rates. Recently, numerous studies have highlighted the potential of NLRP3 inflammasome as a therapeutic target during sepsis. NLRP3 inflammasome is a protein complex that could induce the activation of caspase-1 and the following release of pro-inflammatory cytokines such as IL-1β and IL-18. It was demonstrated that NLRP3 inflammasome was involved in the development and progression of sepsis. In cont

Skin damage induced by zinc oxide nanoparticles combined with UVB is mediated by activating cell pyroptosis via the NLRP3 inflammasome-autophagy-exosomal pathway.

BACKGROUND: Zinc oxide nanoparticles (ZnONPs) are widely used nanomaterial in personal cosmetics, such as skin creams and sunscreens, due to their whitening properties and strong UV light absorption. However, the safety issues and the hazards of ZnONPs, which can be taken up by the skin and cause skin toxicity, are still unclear. From a chemoprevention point of view, pterostilbene (PT) has been reported to prevent skin damage effectively by its anti-inflammatory and autophagy inducer effect. This study aims to determine the skin toxicity and the potential mechanisms of UVB and ZnONPs exposure

Migraine and neuroinflammation: the inflammasome perspective.

BACKGROUND: Neuroinflammation has an important role in the pathophysiology of migraine, which is a complex neuro-glio-vascular disorder. The main aim of this review is to highlight findings of cortical spreading depolarization (CSD)-induced neuroinflammatory signaling in brain parenchyma from the inflammasome perspective. In addition, we discuss the limited data of the contribution of inflammasomes to other aspects of migraine pathophysiology, foremost the activation of the trigeminovascular system and thereby the generation of migraine pain. MAIN BODY: Inflammasomes are signaling multiprotei

The Interplay between Autophagy and NLRP3 Inflammasome in Ischemia/Reperfusion Injury.

Ischemia/reperfusion (I/R) injury is characterized by a limited blood supply to organs, followed by the restoration of blood flow and reoxygenation. In addition to ischemia, blood flow recovery can also lead to very harmful injury, especially inflammatory injury. Autophagy refers to the transport of cellular materials to the lysosomes for degradation, leading to the conversion of cellular components and offering energy and macromolecular precursors. It can maintain the balance of synthesis, decomposition and reuse of the intracellular components, and participate in many physiological processes

El Tor Biotype Vibrio cholerae Activates the Caspase-11-Independent Canonical Nlrp3 and Pyrin Inflammasomes.

Vibrio cholerae is a Gram-negative enteropathogen causing potentially life-threatening cholera disease outbreaks, for which the World Health Organization currently registers 2-4 million cases and ~100.000 cholera-associated deaths annually worldwide. Genomic Vibrio cholerae research revealed that the strains causing this ongoing cholera pandemic are members of the El Tor biotype, which fully replaced the Classical biotype that caused former cholera pandemics. While both of these biotypes express the characteristic Cholera Toxin (CT), the El Tor biotype additionally expresses the accessory toxi

Inflammasome, clonal stability and the thrombotic circuit in myeloproliferative neoplasms: an antecedent framework.

El-Sehrawy and colleagues recently proposed a model of clonal thrombo-inflammation in myeloproliferative neoplasms organised around the inflammasome, clonal stability and a multi-compartmental thrombotic niche. Each of these elements was addressed in two reviews published by our group in 2021, neither of which is cited. The first set out the pathogenetic role of the NLRP3 and AIM2 inflammasomes and their link to hypercoagulable states; the second formalised the vascular consequence as the Circulating Wound, a self-sustaining lesion in which tissue-factor-rich clonal platelets, thrombin generat

Photoaged Polyvinyl Chloride Microplastics Aggravate Airway Epithelial Barrier Dysfunction via TXNIP-NLRP3-Mediated Inflammatory Amplification.

Microplastics are ubiquitous in the atmosphere and may be inhaled into the respiratory system to cause adverse health effects. Photoaging of microplastics in the atmosphere is inevitable, which changes their toxic effects greatly, yet the impacts and underlying mechanisms remain poorly understood. In this study, poly(vinyl chloride) microplastics (PVC-MPs), commonly detected in the atmosphere, were selected to investigate the effects of photoaging on pulmonary toxicological responses at 7 days after a single intratracheal instillation in a mouse model. Photoaging enhanced the abundance of oxyg

Role of inflammasomes in acute respiratory distress syndrome.

Acute respiratory distress syndrome (ARDS) is present in >10% of all people admitted to critical care and is associated with severe morbidity and mortality. Despite more than half a century since its first description, no efficacious pharmacological therapies have been developed, and little progress has been made in improving clinical outcomes. Neutrophils are the principal drivers of ARDS, with their priming and subsequent aberrant downstream functions, including interleukin (IL) 1β and IL-18 secretion, central to the disease pathogenesis. The dominant pathways through which IL-1β and IL-18 a
