Mining predicted essential genes of Brugia malayi for nematode drug targets.

We report results from the first genome-wide application of a rational drug target selection methodology to a metazoan pathogen genome, the completed draft sequence of Brugia malayi, a parasitic nematode responsible for human lymphatic filariasis. More than 1.5 billion people worldwide are at risk of contracting lymphatic filariasis and onchocerciasis, a related filarial disease. Drug treatments for filariasis have not changed significantly in over 20 years, and with the risk of resistance rising, there is an urgent need for the development of new anti-filarial drug therapies. The recent publi

Fundamental study of expression-library immunization against Taenia taeniaeformis in rats.

Expression-library immunization has been proposed as an effective means to screen a large number of genes of the pathogen as candidate protective molecules. In this study, we examined the efficacy of expression-library immunization using a T. taeniaeformis rat model system. Total RNAs were isolated from the last 15 segments of adult T. taeniaeformis and poly A RNA was purified. cDNA library was produced using SuperScript Plasmid System, which contains a mammalian expression vector, pCMV*SPORT6. From about 3,500 clones examined, more than 800 clones were found to contain DNA fragments. About 20

A cytidine deaminase expressed in the post-infective L3 stage of the filarial nematode, Brugia pahangi, has a novel RNA-binding activity.

A number of genes have been identified that are highly expressed in the post-infective L3 stage of the filarial parasite, Brugia pahangi. Amongst these was a cDNA with homology to the cytidine deaminase (CDD) gene family. Phylogenetic analysis of the various cytosine nucleoside deaminases suggest that Brugia pahangi CDD evolved with significant divergence from the RNA editing family. In order to characterize its function, we have expressed Brugia pahangi CDD in bacteria as a chimera with maltose-binding protein (MBP). Biochemical analysis demonstrates the MBP-CDD fusion protein functions as an

Detection and pathological effects of intestinal parasites in Spotted Scat Scatophagus argus: Filisoma spp. and Cryptosporidium spp. as infective agents and their roles in fish inflammatory response.

OBJECTIVE: The Spotted Scat Scatophagus argus, particularly in aquaculture, is commonly affected by intestinal parasitic infections that impact its health and growth. Filisoma spp., an acanthocephalan parasite, and Cryptosporidium spp. cause intestinal inflammation in various marine fish; however, infections with Cryptosporidium spp. or co-infection with these parasites have not been identified in Spotted Scat. METHODS: Host-parasite interactions in fish, including the inflammatory responses of the intestine, were investigated in 32 cultured Spotted Scat from Songkhla, Thailand. The study use

Molecular identification of Trichinella spiralis and Trichinella britovi by diagnostic multiprimer large mitochondrial rRNA amplification.

Trichinella parasites with different epidemiological features still occur in Europe and four species of genus Trichinella have been identified: T. spiralis, T. britovi, T. nativa and T. pseudospiralis. Until now, two of them, T. spiralis and T. britovi, have been identified in Poland. In our studies we selected sequence coding for large mitochondrial rRNA (mt LrDNA) as a genetic marker and developed a sensitive LrDNA multiprimer PCR assay allowing for rapid identification of T. spiralis and T. britovi, parasites present in wild and domestic animals in Poland.

Molecular and immunological characterization of encoding gene and 14-3-3 protein 1 in Fasciola gigantica.

A cDNA encoding Fg14-3-3 protein 1 was cloned by immunoscreening of an adult-stage Fasciola gigantica cDNA library using a rabbit antiserum against tegumental antigens of the parasite. The protein has a deduced amino acid sequence of 252 residues and a calculated molecular weight of 28.7 kDa. It shows sequence identity values between 57.6 and 58.1% to the human 14-3-3 beta, zeta, theta, and eta proteins and is in a phylogenetic cluster with the 14-3-3 protein 1 of Schistosoma spp. Nucleic acid analyses indicate that the Fg14-3-3 protein 1 is encoded by a single copy gene and that this gene is

Eosinophil Extracellular Traps and Inflammatory Pathologies-Untangling the Web!

Eosinophils are an enigmatic white blood cell, whose immune functions are still under intense investigation. Classically, the eosinophil was considered to fulfill a protective role against parasitic infections, primarily large multicellular helminths. Although eosinophils are predominantly associated with parasite infections, evidence of a role for eosinophils in mediating immunity against bacterial, viral, and fungal infections has been recently reported. Among the mechanisms by which eosinophils are proposed to exert their protective effects is the production of DNA-based extracellular traps

Unveiling the Silent Regulators: Noncoding RNAs in Parasitic Disease Diagnosis and Therapy.

Parasitic diseases remain a major global health burden, particularly in tropical and low-resource regions, where limitations in early diagnosis and effective therapeutics contribute to high morbidity and mortality. Emerging evidence highlights noncoding RNAs (ncRNAs) as critical regulators of host-parasite interactions, offering novel opportunities for diagnosis, prognosis, and targeted therapy. However, most of these findings are derived from in vitro experiments and preclinical models with limited validation in clinical settings. This review comprehensively examines the roles of microRNAs (m

Morphology, genetic characterization and phylogeny of Moniliformis tupaia n. sp. (Acanthocephala: Moniliformidae) from the northern tree shrew Tupaia belangeri chinensis Anderson (Mammalia: Scandentia).

A new species of Moniliformis, M. tupaia n. sp. is described using integrated morphological methods (light and scanning electron microscopy) and molecular techniques (sequencing and analysing the nuclear 18S, ITS, 28S regions and mitochondrial cox1 and cox2 genes), based on specimens collected from the intestine of the northern tree shrew Tupaia belangeri chinensis Anderson (Scandentia: Tupaiidae) in China. Phylogenetic analyses show that M. tupaia n. sp. is a sister to M. moniliformis in the genus Moniliformis, and also challenge the systematic status of Nephridiacanthus major. Moniliformis t

Notocotylus ikutai n. sp. (Digenea: Notocotylidae) from lymnaeid snails and anatid birds in Hokkaido, Japan.

An unknown species of the genus Notocotylus (Digenea: Notocotylidae) was found as the larval stage from the lymnaeid snail, Radix auricularia, in a static water area of the Chubetsu River, Hokkaido, the northernmost island of Japan. A DNA barcoding identification system was applied to detect the adult stage. Through the inspection of anatid game birds in Hokkaido, Anas crecca, Anas platyrhynchos, Anas zonorhyncha, and Mareca penelope were demonstrated to serve as the definitive hosts. The detailed morphological features of the species were characterized using adults raised experimentally in im

Crossing barriers of zoogeographical regions: Molecular evidence of vicariance of the only cestode parasite of loaches (Cobitidae) in the Indomalayan region, Paracaryophyllaeus lepidocephali (Cestoda: Caryophyllidea), and its redescription.

Tapeworms of the genus Paracaryophyllaeus Kulakovskaya, 1961 (Cestoda: Caryophyllidea) are specific parasites of loaches (Cypriniformes: Cobitoidea) and occur almost exclusively in the Palaearctic region. The only exception and example of vicariance over the borders of two zoogeographical regions is Paracaryophyllaeus lepidocephali (Kundu, 1985), an insufficiently known species described from the Indomalayan region, with uncertain generic allocation. In the present paper, the species is redescribed based on new material collected from the type host, Lepidocephalichthys guntea, in West Bengal,

New dicrocoeliid digeneans from mammals in Ecuador including a highly genetically divergent new genus from an ancient marsupial lineage.

The Dicrocoeliidae is a highly diverse and broadly distributed family of digeneans typically parasitic in the gall bladder and liver of their tetrapod hosts. So far, no study has reported dicrocoeliids, or any digeneans, from the ancient marsupial family Caenolestidae. Herein, we describe a new genus of dicrocoeliids (Otongatrema n. gen.) from Tate's shrew opossum Caenolestes fuliginosus and a new species of Metadelphis (Metadelphis cesartapiai n. sp.) from a phyllostomid bat Anoura peruana collected in Ecuador. Otongatrema can be readily distinguished from the morphologically closest dicrocoe

Morphological and genetic characterization of green-banded broodsacs of Leucochloridium (Leucochloridiidae: Trematoda) sporocysts detected in Succinea lauta in Hokkaido, Japan.

Green-banded broodsacs of Leucochloridium sporocysts were obtained from land snails, Succinea lauta, collected in Esashi, Esashi District, Hokkaido, Japan. The broodsacs were similar to those of L. paradoxum, which have never been found on the Japanese archipelago. Here, we compare morphological and molecular genetic characteristics of the green-banded broodsacs to those of L. paradoxum. The broodsacs described in this study and those of L. paradoxum showed three equal parts; however, band characteristics of the second parts differed. Moreover, the sequences of nuclear 18S and 28S rRNA genes o

Analysis of Oesophagostomum dentatum excretory-secretory molecules and extracellular vesicles uncovers a repertoire of microRNAs shared with A. Suum and T. Suis and ES containing Paz and Piwi domain proteins implicated in RNAi signaling.

Soil-transmitted helminths (STH) are prevalent in pigs, with the nodular worm, Oesophagostomum dentatum, causing a chronic infection that affects animal welfare and productivity. Unlike the two other dominant soil-transmitted helminths of pigs, Ascaris suum and Trichuris suis, O. dentatum promotes immune responses that deviate from the normal parasite-induced type 2 helper (Th2) response causing long-term infection with minimally acquired immunity. O. dentatum induces a proinflammatory Type 1 helper(Th1)-like response followed by a delayed non-protective Th2 response. The mechanism underlying

Uncovering functional INDELs responsible for splice sites in sheep with different immune profiles naturally exposed to gastrointestinal nematode infection.

Gastrointestinal nematodes (GINs) are responsible for high morbidity and mortality rates in the sheep industry. This study aimed to identify functional INDELs responsible for splice sites using RNA-Sequencing from the liver tissue of sheep with different innate immune responses after being naturally exposed to GIN infection. Two GIN-naïve groups of lambs with divergent cortisol levels (high stress responders - HSR and medium stress responders - MSR) were naturally exposed to GIN during grazing. The animals were euthanized, and six HSR and six MSR, in addition to four immunological control anim

Cloning and characterisation of the Anisakis simplex allergen Ani s 4 as a cysteine-protease inhibitor.

Anisakis simplex is a nematode that can parasitise humans who eat raw or undercooked fish containing live L3s. Larvae invading the gastrointestinal mucosa excrete/secrete proteins implicated in the pathogenesis of anisakiasis that can induce IgE mediated symptoms. Misdiagnosis of anisakiasis, due to cross-reactivity, makes it necessary to develop new diagnostic tools. Recombinant allergens have proved to be useful for diagnosis of other parasitoses. Among the Anisakis allergens, Ani s 4 was considered to be a good potential diagnostic protein because of its heat resistance and its importance i

Systematic and phylogenetic insights into Neocalceostomatidae (Dactylogyridea): A new species, the first neotropical record of species of Neocalceostomoides and Neocalceostoma, and the redescription of Calceostomella herzbergii.

Neocalceostomatidae comprises a small group of monogenoids parasitizing marine catfishes (Siluriformes, Ariidae), with species reported exclusively from the Australian and Oriental regions. Despite their taxonomic relevance within Dactylogyridea, the evolutionary relationships within the group remain uncertain. In this study, we describe a new Neocalceostomoides species from the gills of Cathorops spixii using an integrative taxonomic approach that combines morphological and molecular evidence. Additionally, Calceostomella herzbergii Zambrano, Dezón & León, 2004 is redescribed and transferred

The diversity of Khawia tapeworms (Cestoda: Caryophyllidea) of barbels (Cyprinidae: Barbinae) in the western Palearctic: the existence of vicariant lineages and the description of a new species from the Iberian Peninsula.

The caryophyllidean tapeworm Khawia armeniaca has long been regarded as an exceptionally widespread species within its genus, notable for its significant morphological variability. However, with the accumulation of molecular data from different fish hosts, K. armeniaca was suspected to represent a species complex. To clarify the true identity of these parasites, a comprehensive morphological and molecular study (using 18S, 28S and ITS2 ribosomal regions) of K. armeniaca tapeworms from barbels (Barbinae) across the Iberian Peninsula and the Middle East has been conducted. The results revealed t

Phylogenetic evidence of a possible Trichuris globulosa species complex in Arabian camels from Kuwait.

During a 1-year study, Trichuris adults were obtained after necropsy of Arabian camels (Camelus dromedarius) from a slaughterhouse in Kuwait. Morphological and molecular identification was performed to confirm the identity of the Trichuris specimens obtained from C. dromedarius. Fifteen male Trichuris specimens were selected, and molecular identification was performed using mitochondrial cytochrome c oxidase subunit I, 12S ribosomal RNA, 16S ribosomal RNA genes and the nuclear internal transcribed spacer 2 (ITS2) region. Through phylogenetic analysis, 2 distinct groups were obtained using the

Patent human infections with the whipworm, Trichuris trichiura, are not associated with alterations in the faecal microbiota.

BACKGROUND: The soil-transmitted helminth (STH), Trichuris trichiura colonises the human large intestine where it may modify inflammatory responses, an effect possibly mediated through alterations in the intestinal microbiota. We hypothesised that patent T. trichiura infections would be associated with altered faecal microbiota and that anthelmintic treatment would induce a microbiota resembling more closely that observed in uninfected individuals. MATERIALS AND METHODS: School children in Ecuador were screened for STH infections and allocated to 3 groups: uninfected, T. trichiura only, and m
