Aberrant inhibition of B-cell receptor (BCR)-induced programmed cell death pathways is frequently associated with the development of human auto-reactive B-cell lymphomas. Here, we integrated loss-of-function, genomic, and bioinformatics approaches for the identification of oncogenic mechanisms linked to the inhibition of BCR-induced clonal deletion pathways in human B-cell lymphomas. Lentiviral (HIV)-based RNA interference screen identified MCL1 as a key survival molecule linked to BCR signaling. Loss of MCL1 by RNA interference rendered human B-cell lymphomas sensitive to BCR-induced programm

Premature fusion of one or more of the cranial sutures (craniosynostosis) in humans causes over 100 skeletal diseases, which occur in 1 of approximately 2,500 live births. Among them is Apert syndrome, one of the most severe forms of craniosynostosis, primarily caused by missense mutations leading to amino acid changes S252W or P253R in fibroblast growth factor receptor 2 (FGFR2). Here we show that a small hairpin RNA targeting the dominant mutant form of Fgfr2 (Fgfr2(S252W)) completely prevents Apert-like syndrome in mice. Restoration of normal FGFR2 signaling is manifested by an alteration o

Enteroviral diseases are widespread and impose significant importance in medicine. Although the outcome of diseases that are associated with enteroviruses such as myocarditis, pancreatitis, hepatitis, or encephalomyelitis might be fatal, no specific antiviral therapy is yet available. We and others have shown that RNA interference (RNAi) effectively limits picornaviral replication and cytopathogenicity and improves survival in susceptible mice. However, little is known about the dependence of short interfering RNA (siRNA) efficacy on target region selection and emergence of viral escape mutant

A single nucleotide polymorphism (SNP) in the sickle beta-globin gene (beta(S)) leads to sickle cell anemia. Sickling increases sharply with deoxy sickle Hb concentration and decreases with increasing fetal gamma-globin concentration. Measures that decrease sickle Hb concentration should have an antisickling effect. RNA interference (RNAi) uses small interfering (si)RNAs for sequence-specific gene silencing. A beta(S) siRNA with position 10 of the guide strand designed to align with the targeted beta(S) SNP specifically silences beta(S) gene expression without affecting the expression of the g

Three genome-wide RNA interference screens were performed in Drosophila S2 cells to dissect the contribution of host processes to Listeria monocytogenes entry, vacuolar escape, and intracellular growth. Among the 116 genes identified, several host pathways previously unrecognized as playing a role in listerial pathogenesis were identified: knockdowns affecting vacuolar trafficking to and from the multivesicular body bypassed the requirement for the essential pore-forming toxin listeriolysin O in mediating escape from phagocytic vacuoles and knockdowns affecting either subunit of serine palmito

RNA interference (RNAi) is a valuable tool for functional genomics, yet its effectiveness varies across species and developmental stages. While RNAi-mediated gene silencing is highly effective in spiny lobster (Panulirus ornatus) juveniles, silencing is inconsistent during the larval phyllosoma stages, which limits the scope of effective aquaculture biotechnologies in this species. To identify the mechanisms underlying this stage-dependent variability, we examined double-stranded RNA (dsRNA) stability, candidate uptake associated receptors, and associated immune responses in different life stag

Cotesia chilonis, a dominant endoparasitoid of the striped stem borer, Chilo suppressalis, plays a crucial role in regulating the population of C. suppressalis in the field. RNA interference (RNAi) in C. chilonis is challenging due to the unique biological traits of endoparasitoids. In this study, dsRNA was delivered to C. chilonis by microinjection of pupae within cocoons (delivery outside the host) and nanomaterial-mediated immersion, which delivered dsRNA to larvae inside the host. Both systems were effective in silencing the target gene, Cccaspase-1, with microinjection achieving 41.34% RN

The Ustilaginoidea virens effector SCRE2 has been previously identified to be an essential virulence factor. To elucidate molecular mechanisms underlying SCRE2 virulence function, we identify that SCRE2 interacts with Oryza sativa S-like ribonuclease family member OsRNS4. As a catalytically inactive RNase, OsRNS4 positively regulates rice immunity, depending on its RNA binding ability. OsRNS4 is unconventionally secreted through tetraspanin-positive extracellular vesicles (EVs). OsRNS4 binds to certain groups of rice miRNAs, whereas SCRE2 prevents the binding of OsRNS4 to miRNAs. Especially, O

The citrus root weevil, Diaprepes abbreviatus, is an economically important pest of citrus and ornamental crops whose subterranean larval feeding damages roots and predisposes plants to secondary pathogen infection. Development of efficient RNA interference (RNAi) delivery methods for early larval stages is essential for functional genomics studies and the evaluation of RNAi-based pest management strategies. In this study, we developed a droplet-based feeding assay for oral delivery of double-stranded RNA (dsRNA) to neonates of D. abbreviatus using chitin synthase 2 (DaCHS2) as a model RNAi ta

Zeugodacus tau, a highly destructive agricultural quarantine pest causing severe economic losses to global fruit crops, urgently requires the development of male fertility-based control strategies. Here, we identified and characterized the testis-specific serine/threonine protein kinase 1 gene (ZtTSSK1) in Z. tau. The encoded protein of ZtTSSK1 is highly conserved among dipteran species. Spatiotemporal expression analysis revealed predominant expression in adult males, with specific localization to the testes. In situ hybridization further localized ZtTSSK1 transcripts to the transformation zo

BACKGROUND: Osteosarcoma is one of the most common malignant tumors in children and adolescents. It occurs in the metaphysis of long bones and is a type of aggressive malignant tumor. Although there are treatment methods such as surgery and chemotherapy, the mortality and disability of osteosarcoma patients are still high. With the emergence of more and more chemotherapy resistance, it is necessary to find new therapies to improve the chemotherapy sensitivity of osteosarcoma.
METHODS: Drug-resistant MG-63 and U2OS cell strain was established in vitro by continuous exposure of human osteosarco

The brown planthopper (Nilaparvata lugens) is a major pest threatening global rice production, significantly reducing yields annually. As N. lugens increasingly develops resistance to conventional control methods, such as chemical pesticides, there is an urgent need for innovative and sustainable pest management strategies. Cleavage and Polyadenylation Specificity Factor 30 (CPSF30) is a key protein involved in mRNA 3' end processing, yet its function in N. lugens remains poorly understood. This study aims to elucidate the role of CPSF30 in the growth and development of N. lugens and evaluate

BACKGROUND: Tomato chlorosis virus (ToCV) is mainly vectored by Bemisia tabaci in China, which has a worldwide distribution, and greatly reduces the yields of tomato and other vegetables. At present, control of ToCV has been focused mainly by the use of insecticides to control whitefly populations. Transcriptome sequencing showed high expression of the B. tabaci Bta11975 gene, an α-glucosidase (AGLU) during ToCV acquisition by whitefly Mediterranean (MED) species. To investigate the role of Bta11975 gene in ToCV acquisition and transmission by B. tabaci MED, we used RNA interference (RNAi) to

Rheumatoid arthritis (RA) is the most common type of autoimmune arthritis. Hypoxia-inducible factor-1α (HIF-1α) as a transcription factor in response to hypoxia suggests that it could be a potential therapeutic target for the treatment of RA. In this study, we assessed whether the HIF pathway blockade attenuates the manifestations of RA in the collagen-induced arthritis (CIA) rat model. We constructed a short hairpin RNA (shRNA) lentiviral expression vector targeting HIF-1α (pLVX-shRNA-HIF-1α) and to achieve HIF-1α RNA interference. Quantitative RT-PCR, immunofluorescence staining, and western

DNA vaccines present the aquaculture industry with an effective and economically viable method of controlling viral pathogens that drastically affect productivity. Since specific immune response is rudimentary in invertebrates, the presence of RNA interference (RNAi) pathway in shrimps provides a promising new approach to vaccination. Plasmid DNA vaccines that express short or long double stranded RNA in vivo have shown protection against viral diseases. The design, construction and considerations for preparing such vaccines are discussed.

Lipogenesis is crucial during neuronal development. Abnormal lipid metabolism causes neurological disorders such as Refsum disease and contributes to tumor formation. Long-chain acyl-CoA synthetase (Acsl) ligates coenzyme A to fatty acids, thereby activating the lipid metabolism pathway. Here, we designed a specific small interference RNA (siRNA) against mouse Acsl6, pU6-487i and pU6-586i, and investigated the function of Acsl6 in neuron differentiation. Expression of mAcsl6 mRNA and protein was markedly decreased by pU6-487i and pU6-586i in NB41A3 mouse neuroblastoma cells. We established two

Respiratory syncytial virus (RSV) is a major cause of morbidity in infants, young children, and the elderly worldwide. Currently, there is no effective vaccine, and antiviral drugs to control infection are limited. RNA interference is a powerful tool amenable to development of antiviral drugs. Using small interfering RNA (siRNA) targeting the RSV P gene (siRNA-P), RSV replication can be silenced both in vitro and in a BALB/c model of RSV infection. In this study, we examine the effect of siRNA prophylaxis on the primary and memory immune response to RSV infection in mice. We show that mice pro

BACKGROUND: The discovery of water channel aquaporins (AQPs) has greatly expanded the understanding of the regulation of the water permeability of biological membranes. Aquaporin-1 (AQP1) may be involved in fluid transport in numerous pathological conditions. The objective of the present study was to examine whether AQP1 is present in cultured rat pleural mesothelial cells (PMCs) and to investigate the specific inhibitory effect of RNA interference (RNAi) on AQP1 expression in PMCs, which may provide a new method for the further studies on the relation between expression of AQP1 in PMCs and pl
