[Physiological manifestations of action of a gene regulating predisposition to a pendulum-like movements in rodents].

Physiological effects of the gene determining predisposition to a stereotypical hyperkinesis in the form of pedulum movements (PM) are manifested in some behavioral peculiarities (total motor activity, emotionality, intensity of the startle reflex, sensitivity of the 5-HT2 receptors, predisposition to catalepsy). High frequency of the PM occurring in Wistar rat stock suggests that the gene determining the PM expression is not a pathological one but controls some adaptive properties of the nervous system. Relationships between the PM expression and the degree of predisposition to catalepsy are

Abnormal interaction between vestibular and voluntary head control in patients with spasmodic torticollis.

The functional status of vestibulo-collic reflexes in the sternocleidomastoid (SCM) muscles was investigated in 24 patients with spasmodic torticollis using small, abrupt 'drops' of the head. None had been treated with botulinum toxin injections during at least 4 months preceding the study. Eight of the patients, four of whom had been studied before surgery, were also studied after selective peripheral denervation of neck muscles. The reflex was of normal latency and duration in the 'passive drop' condition, in which subjects were instructed not to oppose the fall of the head. To study volunta

Dynamics of defensive reactivity in patients with panic disorder and agoraphobia: implications for the etiology of panic disorder.

BACKGROUND: The learning perspective of panic disorder distinguishes between acute panic and anxious apprehension as distinct emotional states. Following animal models, these clinical entities reflect different stages of defensive reactivity depending upon the imminence of interoceptive or exteroceptive threat cues. The current study tested this model by investigating the dynamics of defensive reactivity in a large group of patients with panic disorder and agoraphobia (PD/AG). METHODS: Three hundred forty-five PD/AG patients participated in a standardized behavioral avoidance test (being entr

Fearful imagery in social phobia: generalization, comorbidity, and physiological reactivity.

BACKGROUND: Social phobia has been characterized as a disorder of exaggerated fear of social threat and heightened sensitivity to imagery of social failure. METHODS: To assess the physiological basis of this description, social phobia patients (n=75) and demographically matched control participants (n=75) imagined neutral and fearful events while acoustic startle probes were occasionally presented and eye-blink responses (orbicularis occuli) recorded. Changes in heart rate, skin conductance level, and facial expressivity were also indexed. In addition to comparing control participants and soc

The Impact of Event Scale--Revised: evaluation of the subscales and correlations to psychophysiological startle response patterns in survivors of a life-threatening cardiac event: an analysis of 129 patients with an implanted cardioverter defibrillator.

BACKGROUND: The aim of the present study was to examine the reliability and validity of the Impact of Event Scale-Revised (IES-R) with special emphasis on the evaluation of the hyperarousal subscale against a standardized psychophysiological measurement. METHODS: A total of 129 survivors of a life-threatening cardiac event underwent a psychodiagnostic evaluation and a psychophysiological acoustic startle reflex (ASR) paradigm. The ASR assessed the magnitude and habituation of electromyogram (EMG) and skin conductance responses (SCR) in response to the delivery of 15 acoustic startle trials. P

Dopaminergic stimulation disrupts sensorimotor gating in the rat.

Prepulse inhibition is a cross-species phenomenon in which reflex responses to discrete sensory events are modified by weak prestimulation. In experiments designed to investigate the neuropharmacological mechanism of this form of information processing, and its relevance to schizophrenic psychopathology, apomorphine (0.125-4.0 mg/kg) and d-amphetamine (0.5-4.0 mg/kg) were administered to rats in an attempt to modify prepulse inhibition of the acoustic startle response. Rats were presented with 40 ms, 118 dB[A] acoustic pulses which were intermittently preceded by a weak 80 dB[A] acoustic prepu

Transdiagnostic physiological correlates of unpredictable threat anticipation.

Responses to unpredictable threat have been proposed as a candidate mechanism in anxiety-related psychopathology, but clinical findings remain heterogeneous and often rely on single physiological outcomes. The present study examined whether physiological responses during threat anticipation map onto categorical and dimensional conceptualizations of psychopathology across the anxiety and obsessive-compulsive spectrum. In a transdiagnostic sample of N = 163 participants, patients with specific phobia, social anxiety disorder (SAD), and obsessive-compulsive disorder (OCD), as well as unaffected c

Missing what's right under your nose: Failed appetitive and aversive audio-olfactory conditioning in humans.

The comparison of physiological mechanisms underlying appetitive and aversive conditioning is often challenging due to the involvement of stimuli from different modalities with potentially disparate effective mechanisms (e.g., pain stimuli versus monetary rewards). The olfactory system offers a unique opportunity to examine both types of conditioning in humans, as isointense odors can serve as comparably pleasant and unpleasant stimuli. To study physiological and behavioral responses during appetitive and aversive learning, we employed odors as unconditioned stimuli (US) in a within-subjects d

Gap pre-pulse inhibition of the cortical auditory evoked potentials as a possible objective tinnitus assessment tool.

OBJECTIVES: The objective assessment tests overcome the variability of subjective methods. Cortical recordings with gap pre-pulse inhibition of the acoustic startle reflex stimulus have been used as objective tinnitus assessments in humans. This study aims to investigate this possible objective tinnitus test and compare gap-induced inhibition in different stimulus parameters and brain regions. MATERIALS & METHODS: Twenty People (18-50 years old) without hearing loss and tinnitus were included. The sound stimuli consisted of continuous background noise with a loud startle tone preceded by a si

Effects of prenatal exposure to chronic mild stress and toluene in rats.

The aim of the present study was to elucidate whether prenatal chronic stress, in combination with exposure to a developmental neurotoxicant, would increase effects in the offspring compared with the effects of either exposure alone. Development and neurobehavioral effects were investigated in female offspring of pregnant rats (Mol:WIST) exposed to chronic mild stress (CMS) during gestational days (GD) 9-20, or 1500 ppm toluene, 6 h/day during gestational days 7-20, or a combination of the two. Prenatal CMS was associated with decreased thymic weight and increased auditory startle response. Th

Effects of acoustic startle, simulated temporary threshold shift, and simulated tinnitus on sound localization and word recognition performance: A pilot study.

An unexpected loud sound can elicit an acoustic startle reflex and potentially cause a temporary threshold shift (TTS) for hearing and noise-induced tinnitus for a short period of time. This occurs, for example, with the high-amplitude noise burst that accompanies a bright light and impulsive force when police or military use a "flashbang" grenade to temporarily disable or confuse an adversary. This pilot study evaluated the effects of an unexpected loud acoustic impulse sound on human subject performance in terms of (1) word recognition in noise, and (2) sound localization, both examined in a

A diminished rate of "physiological decay" at noise offset contributes to age-related changes in temporal acuity in the CBA mouse model of presbycusis.

The persistence of afferent activity at stimulus offset was examined in mice as a function of age (experiment 1) and noise level (experiment 2), using a procedure devised by von Bekesy to study the physiological decay of afferent activity. Noise offset was presented from 1 to 10 ms (the temporal gap) prior to an acoustic startle stimulus, with an abrupt edge or a linear ramp having the duration of the gap. Noise offset inhibited the startle reflex, this effect declining with age. For young (2-3 months old) and "young-old" mice (18-19 months old), the abrupt offset was always more inhibitory th

Relationship between Toxoplasma gondii seropositivity and acoustic startle response in an inner-city population.

Toxoplasma gondii (TOXO) is a neuroinvasive protozoan parasite that induces the formation of persistent cysts in mammalian brains. It infects approximately 1.1million people in the United States annually. Latent TOXO infection is implicated in the etiology of psychiatric disorders, especially schizophrenia (SCZ), and has been correlated with modestly impaired cognition. The acoustic startle response (ASR) is a reflex seen in all mammals. It is mediated by a simple subcortical circuit, and provides an indicator of neural function. We previously reported the association of TOXO with slowed acous

Behavioral Assessment of Zwicker Tone Percepts in Gerbils.

The Zwicker tone illusion - an auditory phantom percept after hearing a notched noise stimulus - can serve as an interesting model for acute tinnitus. Recent mechanistic models suggest that the underlying neural mechanisms of both percepts are similar. To date it is not clear if animals do perceive the Zwicker tone, as up to now no behavioral paradigms are available to objectively assess the presence of this phantom percept. Here we introduce, for the first time, a modified version of the gap pre-pulse inhibition of the acoustic startle reflex (GPIAS) paradigm to test if it is possible to indu

Morphological and sensorimotor phenotypes in a zebrafish CHARGE syndrome model are domain-dependent.

CHARGE syndrome is a heterogeneous disorder characterized by a spectrum of defects affecting multiple tissues and behavioral difficulties such as autism, attention-deficit/hyperactivity disorder, obsessive-compulsive disorder, anxiety, and sensory deficits. Most CHARGE cases arise from de novo, loss-of-function mutations in chromodomain-helicase-DNA-binding-protein-7 (CHD7). CHD7 is required for processes such as neuronal differentiation and neural crest cell migration, but how CHD7 affects neural circuit function to regulate behavior is unclear. To investigate the pathophysiology of behaviora

Fear-potentiated startle response as an endophenotype: Evaluating metrics and methods for genetic applications.

The modulation of the startle response (SR) by threatening stimuli (fear-potentiated startle; FPS) is a proposed endophenotype for disorders of the fearful-fearlessness spectrum. FPS has failed to show evidence of heritability, raising concerns. However, metrics used to index FPS-and, importantly, other conditional phenotypes that are dependent on a baseline-may not be suitable for the approaches used in genetic epidemiology studies. Here, we evaluated multiple metrics of FPS in a population-based sample of preadolescent twins (N = 569 from 320 twin pairs, Mage = 11.4) who completed a fear-con

Does anxiety sensitivity correlate with startle habituation? An examination in two independent samples.

Individuals with anxiety disorders have previously demonstrated abnormal habituation to aversiveness over time. As anxiety sensitivity (AS), or an individuals' propensity to fear of anxiety-related sensations, has been shown to be a risk factor for anxiety disorders (particularly panic disorder), the present study examined whether AS was also associated with abnormal habituation. This association was examined in two independent samples of undergraduates (Ntotal=178). Habituation was operationalised as the reduction in startle response to multiple startle probes presented over 2.5 minutes and t

Familial hyperekplexia and refractory status epilepticus: a new autosomal recessive syndrome.

Hyperekplexia is a rare disorder characterized by an exaggerated startle response to noise and handling and by neonatal hypertonia. It is predominantly an autosomal dominant disease; however, atypical cases with additional variable manifestations have been reported. We report a hitherto undescribed association of hyperekplexia and refractory status epilepticus in two siblings. Both children were born after an uneventful pregnancy to healthy unrelated Ashkenazi Jews. Both had increased startle and tone from birth and later became hypotonic. A metabolic evaluation, including a muscle biopsy, was

Basic sensory risk markers characteristic of schizophrenia in autism spectrum disorder: A systematic review and meta-analysis.

AIMS: Neurophysiological impairments in basic sensory processes are a hallmark feature in schizophrenia (SZ) and are commonly reported in individuals at-risk of developing SZ. Autism spectrum disorder (ASD) is linked to a higher incidence of later psychotic disorders, including SZ. However, it remains unclear whether basic sensory risk markers for SZ are equally characteristic of ASD. METHODS: We performed a systematic review and subsequent meta-analyses of basic sensory risk markers for SZ in ASD. Following PRISMA guidelines, we included 44 studies, investigating prepulse inhibition of the s
