Suboptimal choice behavior by pigeons.

Contrary to the law of effect and optimal foraging theory, pigeons show suboptimal choice behavior by choosing an alternative that provides 20% reinforcement over another that provides 50% reinforcement. They choose the 20% reinforcement alternative--in which 20% of the time, that choice results in a stimulus that always predicts reinforcement, and 80% of the time, it results in another stimulus that predicts its absence--rather than the 50% reinforcement alternative, which results in one of two stimuli, each of which predicts reinforcement 50% of the time. This choice behavior may be related

Functional analysis and treatment of rumination using fixed-time delivery of a flavor spray.

A functional analysis suggested that rumination exhibited by an adult with autism was maintained by automatic reinforcement. Next, a preference assessment with three flavor sprays (i.e., flavored sprays used by dieters) showed that apple pie spray was most preferred. Finally, the effects of fixed-time delivery of the apple pie spray on levels of rumination were evaluated. The spray reduced rumination, and the participant was taught to self-administer the spray.

Differences in corticosterone level due to inter-food interval length: implications for schedule-induced polydipsia.

The purpose of this study was to determine the effects of different food-reinforcement schedules on plasma corticosterone (CORT), and its possible involvement in the acquisition and maintenance of schedule-induced polydipsia (SIP). In Experiment 1, three groups of rats were submitted to two different fixed-interval (FI) schedules with inter-food intervals of 30 and 120 s, and to a massed-feeding presentation for 40 days until SIP was well stabilized. In Experiment 2, six groups of rats were exposed to the same schedules, FI 30s and FI 120s, and to the massed-feeding condition, but no water bot

Naltrexone alters alcohol self-administration behaviors and hypothalamic-pituitary-adrenal axis activity in a sex-dependent manner in rats.

BACKGROUND: The mu-opioid antagonist, naltrexone (NTX), is a FDA-approved treatment for alcohol use disorder (AUD); however, the data on whether it differentially affects males vs. females are mixed. NTX increases hypothalamic-pituitary-adrenal (HPA) axis activity that associates with subjective responses to alcohol and craving in individuals with AUD. The present study tested for sex differences in the ability of NTX to decrease appetitive and consummatory behaviors in rats in operant alcohol self-administration. Because the opioid system and HPA axis are sexually dimorphic, we examined NTX's

Attenuated behavioural responses to acute and chronic cocaine in GASP-1-deficient mice.

G protein-coupled receptor (GPCR) associated sorting protein 1 (GASP-1) interacts with GPCRs and is implicated in their postendocytic sorting. Recently, GASP-1 has been shown to regulate dopamine (D(2)) and cannabinoid (CB1) receptor signalling, suggesting that preventing GASP-1 interaction with GPCRs might provide a means to limit the decrease in receptor signalling upon sustained agonist treatment. In order to test this hypothesis, we have generated and behaviourally characterized GASP-1 knockout (KO) mice and have examined the consequences of the absence of GASP-1 on chronic cocaine treatme

Suppression by the cannabinoid CB1 receptor antagonist, rimonabant, of the reinforcing and motivational properties of a chocolate-flavoured beverage in rats.

Pharmacological blockade of the cannabinoid CB1 receptor has been repeatedly reported to suppress intake of food, including highly palatable foods, in laboratory animals. This study was designed to investigate whether treatment with the cannabinoid CB1 receptor antagonist, rimonabant, would reduce the reinforcing and motivational properties of a chocolate-flavoured beverage [containing 5% (w/v) chocolate powder] in nonfood-deprived and nonwater-deprived Wistar rats trained to self-administer this beverage under an operant conditioning procedure. This study was also aimed at assessing to what d

Schedule-induced stereotypy.

The phenomena of the induction and entrainment of adjunctive behaviours was investigated in eight people with severe or profound mental retardation who exhibited stereotypic behaviours. The occurrence of collateral behaviours was investigated under extinction and massed reinforcer baselines and a variety of periodic or intermittent schedules of reinforcement for the performance of a simple experimental task. Seven of the eight subjects demonstrated evidence of schedule-induced stereotypic behaviour in the experimental conditions: five of the eight subjects also showed evidence of the entrainme

Female rats express habitual behavior earlier in operant training than males.

Habitual behavior can be advantageous by increasing the availability of cognitive resources for use in other tasks. However, habitual behaviors are problematic when they are coopted to prolong the maladaptive responding present in several psychopathologies such as substance abuse, dysregulated fear responding in posttraumatic stress disorder, and obsessive-compulsive disorder. Although sex differences exist in the occurrence or progression of these psychopathologies, there are no studies that compare the development of habitual behavior systematically in male and female animals. In the present

High reinforcing efficacy of nicotine in non-human primates.

Although tobacco appears highly addictive in humans, there has been persistent controversy about the ability of its psychoactive ingredient nicotine to induce self-administration behavior in laboratory animals, bringing into question nicotine's role in reinforcing tobacco smoking. Because of ethical difficulties in inducing nicotine dependence in naïve human subjects, we explored reinforcing effects of nicotine in experimentally-naive non-human primates given access to nicotine for periods of time up to two years. Five squirrel monkeys with no experimental history were allowed to intravenously

Intermittent reinforcement of a continuous response.

Six rats were trained with food deliveries contingent upon their pressing a lever and holding it down for either fixed or variable cumulative durations. Fixed-hold requirements ranged from 15 s to 90 s over experimental conditions; variable-hold requirements ranged from 15 s to 120 s. At most long and intermediate values, variable-hold requirements maintained more lever holding than fixed requirements. At the longest hold requirements studied, more lever holding was maintained by variable requirements than by fixed requirements of equivalent mean length for each rat. Postreinforcement-pause du

Validation and optimisation of a touchscreen progressive ratio test of motivation in male rats.

RATIONALE: Across species, effort-related motivation can be assessed by testing behaviour under a progressive ratio (PR) schedule of reinforcement. However, to date, PR tasks for rodents have been available using traditional operant response systems only. OBJECTIVES: Touchscreen operant response systems allow the assessment of behaviour in laboratory rodents, using tasks that share high face validity with the computerised assessments used in humans. Here, we sought to optimise a rat touchscreen variant of PR and validate it by assessing the effects of a number of manipulations known to affect

Attenuation of cocaine-seeking behaviour by the AMPA/kainate receptor antagonist CNQX in rats.

RATIONALE: It has been suggested previously that conditioned effects on drug-seeking behaviour are in part mediated through glutamatergic neurotransmission. OBJECTIVES: To optimise a second-order schedule of IV cocaine reinforcement in Wistar rats and investigate the effects of the systemic AMPA/kainate receptor antagonist CNQX on cocaine-seeking behaviour under this schedule. METHODS: Free-feeding Wistar rats were trained to respond for an IV cocaine infusion (0.25 mg/infusion) under a FI15 min(FR7:S) schedule, whereby the completion of FR7 responses led to the presentation of a conditioned

Reducing stereotyped behaviour: a comparison of two methods of programming differential reinforcement.

Differential Reinforcement of Other behaviours (DRO) has proven to be successful in reducing challenging behaviour, although the need for staff vigilance can limit its applicability in applied settings. The present study compared the effectiveness of whole-interval DRO with momentary DRO, a less intensive schedule. In a multiple-baseline design, two participants who exhibited high-rate stereotyped behaviours were presented with the two schedules. Momentary DRO was more effective than whole-interval DRO in reducing the targeted stereotypies of one participant, and as effective with the second.

Applying a DRO schedule and compliance training to reduce aggressive and self-injurious behavior in an autistic man: a case report.

An autistic man who displayed aggressive, self-injurious, and destructive behavior in a psychiatric hospital ward despite administration of psychotropic medications and repeated application of physical restraints was treated with a differential reinforcement of other behavior (DRO) schedule and compliance training. These were employed in conjunction with ongoing medical interventions. After introduction of behavioral procedures, the frequency of applying restraints for violent behavior declined from an average of more than once per day to less than once per month. Data taken while the program

Effects of the kappa-opioid receptor antagonist nor-binaltorphimine on methamphetamine-vs-food choice in male rhesus monkeys.

BACKGROUND: Kappa-opioid receptors (KOR) are hypothesized to be involved in mediating ongoing methamphetamine self-administration. Previous rat studies have demonstrated that treatment with the KOR antagonist nor-binaltorphimine (nor-BNI) decreases methamphetamine self-administration. However, KOR antagonist effects on methamphetamine self-administration in nonhuman primates are unknown. METHODS: Operant behavior was maintained under a concurrent schedule of banana-flavored pellets (fixed-ratio 100 schedule) and intravenous methamphetamine injections (0-0.32mg/kg/injection, fixed-ratio 10 sch

Effects of chlordiazepoxide, ripazepam and d-amphetamine on conditioned acceleration of timing behaviour in rats.

The lever-pressing behaviour of three rats was maintained by a schedule in which food reinforcement was obtained by any response which was emitted at least 15 s after the previous response (DRL 15s). When performance on this schedule had stabilised, the animals were presented intermittently with 1-min periods of a white noise stimulus, the termination of which was accompanied by the delivery of a mild electric footshock. This procedure let to reliable increases in response rates furing the stimulus although responding at other times continued to be appropriate to the DRL 15-s schedule. Adminis

Effects of lick-contingent timeout on schedule-induced polydipsia.

Rats bar pressing on a 1-min fixed-interval schedule for 45-mg food pellets became polydipsic when water was concurrently available. They were then exposed to conditions in which each lick on the drinking tube produced a timeout period during which the food-schedule lever was retracted and the fixed-interval timer either did or did not continue to operate. Licks occurring within a timeout period extended its duration. As the duration of the lick-initiated timeout period was increased logarithmically through four values from 10 sec to 80 sec, lick rates as well as water intake rates generally d

Activation of G protein-coupled estradiol receptor 1 in the dorsolateral striatum enhances motivation for cocaine and drug-induced reinstatement in female but not male rats.

BACKGROUND: Estradiol potentiates drug-taking behaviors, including motivation to self-administer cocaine and reinstatement of drug-seeking after extinction in females, but not males. The dorsolateral stratum (DLS) is a region of the brain implicated in mediating drug-seeking behaviors and, more specifically, is a target brain area to study how estradiol regulates these behaviors. The estradiol receptors α, β, and G protein-coupled estradiol receptor 1 (GPER1) are all present in the DLS. In this study, the effects of activating GPER1 in the DLS on drug-seeking are investigated. METHODS: Gonad-

Predictors of new-onset depression after mild traumatic brain injury.

Methamphetamine is a drug that is often consumed at dance parties or nightclubs where the ambient temperature is high. The present study determined whether such high ambient temperatures alter intravenous methamphetamine self-administration in the rat. Male Hooded Wistar rats were trained to self-administer intravenous methamphetamine (0.1 mg/kg/infusion) under a fixed ratio 1 (FR1) or progressive ratio (PR) schedule of reinforcement at an ambient temperature of 23 +/- 1 degrees C. They were then given their daily self-administration session at a raised ambient temperature of 30 +/- 1 degrees

Culinary-Medicinal Lion's Mane Mushroom Hericium erinaceus (Agaricomycetes) Mycelium Mitigates Ketamine Abuse Potential.

Ketamine therapy is effective in alleviating symptoms of psychiatric disorders such as depression and anxiety; however, its increasing recreational use raises concerns about potential abuse. Hericium erinaceus mycelium, which contains the bioactive compound erinacine A, has shown promise in promoting brain health. This study investigated whether H. erinaceus mycelium and erinacine A could reduce ketamine's abuse potential using a rat self-administration model.
