Halophilic bacteria and archaea in salinity-resilient agriculture: mechanisms and multi-omics perspectives.

Soil salinization due to anthropogenic activity and climate change is a bottleneck to the global agricultural yield and food security. Conventional approaches for mitigating salt stress, including utilization of chemical fertilizers, have shown limited success, and the use of genetically engineered microbes as bioinoculants or salt-tolerant crop varieties against abiotic salinity stress often faces regulatory challenges with extended timelines. Contrary to the existing approaches, employment of halophilic bacteria and archaea offers a promising platform for eco-friendly and sustainable crop cu

Plant-associated halotolerant bacteria improving growth of Vicia faba L. Mariout-2 under salinity conditions.

In this comprehensive investigation, we successfully isolated and characterized 40 distinct plant-associated halotolerant bacteria strains obtained from three halophytic plant species: Tamarix nilotica, Suaeda pruinosa, and Arthrocnemum macrostachyum. From this diverse pool of isolates, we meticulously selected five exceptional plant-associated halotolerant bacteria strains through a judiciously designed seed biopriming experiment and then identified molecularly. Bacillus amyloliquefaciens DW6 was isolated from A. macrostachyum. Three bacteria (Providencia rettgeri DW3, Bacillus licheniformis

Proteomic analysis reveals the protective role of exogenous hydrogen sulfide against salt stress in rice seedlings.

Hydrogen sulfide (H2S) is an important gaseous signal molecule which participates in various abiotic stress responses. However, the underlying mechanism of H2S associated salt tolerance remains elusive. In this study, sodium hydrosulfide (NaHS, donor of H2S) was used to investigate the protective role of H2S against salt stress at the biochemical and proteomic levels. Antioxidant activity and differentially expressed proteins (DEPs) of rice seedlings treated by NaCl or/and exogenous H2S were investigated by the methods of biochemical approaches and comparative proteomic analysis. The protein-p

Physiological and transcriptomic responses of Ginkgo biloba to salt stress and exogenous melatonin.

INTRODUCTION: Soil salinization constrains plant growth, and enhancing salt tolerance is of great significance. METHODS: Here, Ginkgo biloba seedlings were treated with control (CK), salt stress (S), or salt stress + melatonin (S+M). Integrating physiology, transcriptomics, and weighted gene co-expression network analysis (WGCNA), we explored how melatonin enhances salt tolerance. RESULTS AND DISCUSSION: Salt stress reduced chlorophyll content, suggesting potential impairment of photosynthetic capacity, appeared to increase membrane lipid peroxidation, and led to substantial proline accumula

Genome-wide characterization of soybean alcohol dehydrogenase (ADH) genes identifies GmADH13 as a positive regulator of the salt stress response.

Alcohol dehydrogenase (ADH) is a zinc-binding enzyme responsible for catalyzing the interconversion between ethanol and acetaldehyde, as well as other alcohol and aldehyde pairs, within the ethanol fermentation pathway. This enzyme plays a crucial role in plant adaptation to environmental stressors. However, knowledge regarding the ADH gene family in soybean remains limited. Here, a genome-wide analysis was conducted, leading to the identification of 22 ADH genes in soybean and their classification into five subfamilies based upon phylogenetic relationships. Heat Shock Factor TaHSFA6e Interacts With TaNAC019 to Activate TaHSP70 Expression and Positively Contributes to Salt Stress in Wheat.

Soil salinization, exacerbated by seawater intrusion and climate change, poses a major constraint on crop production. To cope with salt stress, plants have evolved complex mechanisms involving numerous tolerance-related genes. We identified that heat shock protein TaHSP70 acted as a positive regulator in response to salt stress in wheat. Further analysis revealed that heat shock transcription factor TaHSFA6e and NAC transcription factor TaNAC019 physically interact and function additively to activate TaHSP70 expression. Whereas a single-nucleotide T insertion within the coding sequence of TaNA

RNA-sequencing transcriptome analysis of Avicennia marina (Forsk.) Vierh. leaf epidermis defines tissue-specific transcriptional response to salinity treatment.

Avicennia marina (Forsk.) Vierh. is a typical mangrove plant. Its epidermis contains salt glands, which can secrete excess salts onto the leaf surfaces, improving the salt tolerance of the plants. However, knowledge on the epidermis-specific transcriptional responses of A. marina to salinity treatment is lacking. Thus, physiological and transcriptomic techniques were applied to unravel the salt tolerance mechanism of A. marina. Our results showed that 400 mM NaCl significantly reduced the plant height, leaf area, leaf biomass and photosynthesis of A. marina. In addition, 1565 differentially ex

Impact of salinity stress during reproductive stage on physiological responses and yield of landrace rice genotypes.

Salinity significantly reduces rice yield, particularly when stress occurs during the reproductive stage. This study aimed to evaluate the physiological and yield responses of rice genotypes subjected to salinity stress during this critical growth phase. The experiment followed a 4 × 10 factorial arrangements in a randomized complete block design (RCBD), comprising ten rice genotypes and four salinity levels (control, 6, 12, and 16 dS/m). Data were collected for electrolyte leakage, relative water content (RWC), proline content, SPAD chlorophyll meter reading (SCMR), total phenolic content, gr

QTL mapping and haplotype analysis reveal candidate genes for seedling-stage salinity tolerance in rice.

INTRODUCTION: Soil salinization is a major constraint on rice production, and severe salinity levels lead to pronounced yield losses. Therefore, elucidating the genetic architecture of salinity tolerance and identifying candidate genes underlying this trait are crucial for molecular design breeding. METHODS: In this study, a population of 253 recombinant inbred lines (RILs) derived from the salt-sensitive japonica cultivar Jileng 1 (JL1) and the salt-tolerant indica cultivar Milyang 23 (MY23) was evaluated for seedling-stage salinity tolerance under 0.9% NaCl stress. RESULTS: Using multiple

Impact of Salinity Duration on Growth, Photosynthesis, Water Relations, and Recovery Capacity Reveals Adaptive Responses in Hordeum maritimum.

Although the physiological mechanisms underlying salt tolerance in wild barley (Hordeum maritimum) are relatively well understood, its recovery after salt stress remains poorly characterized. In this pot experiment, plants were exposed to 200 mM NaCl for 0, 15, 30, 45, or 60 days to evaluate changes in growth and water status. Wild barley exhibited high tolerance during the early stages of salt exposure, maintaining normal growth and water balance for up to 30 days. However, longer exposure reduced plant growth and leaf water content, indicating that prolonged salinity eventually compromises p

Alfalfa Transcription Factor MsNAC2a Orchestrates the Homeostasis of Salt Stress Responses via Reactive Oxygen Species Accumulation and Hydrogen Sulphide Depletion.

Soil salinisation poses a significant threat to alfalfa (Medicago sativa L.) growth and development, limiting its productivity and hindering its widespread cultivation. Hydrogen sulphide (H2S) serves as an important gaseous signalling molecule in plants, mediating a myriad of physiological processes like salt tolerance. However, the molecular mechanisms underlying H2S accumulation and its regulation under salinity stress in alfalfa are still not fully elucidated. In this study, we demonstrated that MsNAC2a, a NAC transcription factor, is a negative modulator of salt stress resistance in alfalf

Edaphic characterization and plant zonation in the Qaidam Basin, Tibetan Plateau.

This paper presents a study of edaphic characteristics and their relationship with plant distribution in the Qaidam Basin, Tibetan Plateau, and establishes a distribution model for plants in sandy gravel Gobi to dry salt lake areas. All of the communities in the study area were dominated by plants with strong saline-alkaline tolerance. In this area, salts appeared to migrate to the surface; the surface soil was striped, and the salt distribution varied from sandy gravel Gobi to dry salt lake areas. The salt composition mainly consisted of NaCl in the surface crust. In the subsurface layers, th

OsVDE, a xanthophyll cycle key enzyme, mediates abscisic acid biosynthesis and negatively regulates salinity tolerance in rice.

OsVDE, a lipocalin-like protein in chloroplasts, negatively regulated the ABA biosynthesis and stomatal closure under salt stress in rice seedlings. Violaxanthin de-epoxidase (VDE) is a key enzyme of xanthophyll cycle. It plays a critical role in abscisic acid (ABA) biosynthesis, growth and stress responses in plants. Although functions of several VDE genes have been characterized, it is largely unknown whether OsVDE regulates the ABA biosynthesis and salt stress tolerance in rice. In this study, we generated the OsVDE overexpressing and CRISPR-Cas9-mediated gene-editing transgenic lines, and

Physiological and Transcriptome Analysis on Diploid and Polyploid Populus ussuriensis Kom. under Salt Stress.

Populus ussuriensis Kom. is a valuable forest regeneration tree species in the eastern mountainous region of Northeast China. It is known that diploid P. ussuriensis (CK) performed barely satisfactorily under salt stress, but the salt stress tolerance of polyploid (i.e., triploid (T12) and tetraploid (F20)) P. ussuriensis is still unknown. In order to compare the salt stress tolerance and salt stress response mechanism between diploid and polyploid P. ussuriensis, phenotypic observation, biological and biochemistry index detections, and transcriptome sequencing (RNA-seq) were performed on CK,

Impairment of ribosome maturation or function confers salt resistance on Escherichia coli cells.

We found that loss of integrity of the ribosome by removal of a putative ribosome maturation factor or a ribosomal protein conferred salt tolerance on Escherichia coli cells. Some protein synthesis inhibitors including kasugamycin and chloramphenicol also had a similar effect, although kasugamycin affected neither 16S rRNA maturation nor subunit association into a 70S ribosome. Thus, salt tolerance is a common feature of cells in which maturation or function of the ribosome is impaired. In these cells, premature induction of an alternative sigma factor, σ(E), by salt stress was observed. These

Index for the assessment of heat tolerance.

Heat tolerance was assessed by magnitude of strain induced in the body as a whole by heat load. The strain was represented by a combination of relative water loss, relative rise in rectal temperature and relative salt loss, using those critical values of the three factors which cause heat stroke (40.6 degrees C), water depletion heat exhaustion (7% of body weight) and salt depletion heat exhaustion (0.75 g per kg of body weight). As this numerical heat tolerance index is defined as the degree of disturbance induced by heat exposure concerning thermal regulation, water and electrolyte metabolis

Synergistic effects of phytohormones and membrane transporters in plant salt stress mitigation.

Plants are frequently exposed to high salinity, negatively affecting their development and productivity. This review examined the complex roles of membrane transporters (MTs) and phytohormones in mediating salt stress. MTs are crucial in capturing sodium ions (Na+) and maintaining a delicate balance between sodium (Na+) and potassium (K+), essential for supporting cellular homeostasis and enhancing overall plant health. These MTs were instrumental in regulating ion balance and promoting the absorption and segregation of vital nutrients, thereby enhancing salt stress tolerance. Various plant ho

Haplotype and diversity analysis of indigenous rice for salinity tolerance in early-stage seedling using simple sequence repeat markers.

We evaluated the seedling-stage salt tolerance of a total of 50 indigenous rice genotypes from coastal Tamil Nadu. Using a hydroponic system, we studied the different agronomic characters 14 days after exposure to six different concentrations of saline solution. Shoot and root length as well as plant biomass at seedling stage decreased with increasing salinity. Genotypes showing significant interaction and differential response towards salinity were assessed at the molecular level using 20 simple sequence repeat (SSR) markers linked with salt-tolerance QTL. These genotypes were grouped into el

Salt sensitivity in young normotensive subjects is associated with a hyperinsulinemic response to oral glucose.

Insulin resistance associated with a hyperinsulinemic response to oral glucose intake has been found in patients with essential hypertension and is believed to play a role in inducing hypertension by causing renal sodium and water retention. We therefore examined whether salt-sensitive, young normotensives, assumed to be predisposed to essential hypertension, exhibit impaired glucose tolerance in a similar way. The plasma insulin and glucose response to oral glucose intake (75 g) was assessed in 23 healthy, lean, male volunteers ingesting either 20 mmol or 260 mmol NaCl/day for 6 days each in
