INTRODUCTION: Gut microbiota play a critical role in aquaculture by enhancing nutrient metabolism, digestion, immune response, and reproductive performance in fish. Labeo catla, one of the most commercially important Indian major carps, demands better reproductive management; however, insights into its gut microbiome composition and functional dynamics, particularly during the crucial pre-spawning phase, remain limited.
METHODS: In this study, we investigated the structural and functional attributes of gut microbial communities in male and female L. catla reared in ICAR-CIFA ponds using high-

Although antibiotics are the main therapy for bacterial infections, the reports showed that the overuse (or misuse) of antibiotics will results in several problems such as the development of antibiotic-resistant strains, persistence of drug residues, and numerous environmental concerns. Therefore, finding antibiotic alternatives is considered of vital importance. Investigation of the antimicrobial properties of several plant substances and extracts is of great value to replace antibiotics. With this objective, this study aimed to evaluate the antimicrobial activities of an ethanolic extract pr

UNLABELLED: The escalating antimicrobial resistance crisis demands innovative strategies for antibiotic discovery. Conventional approaches for identifying antibiotic-producing microorganisms from environmental samples are often laborious and low-throughput, requiring prior isolation and purification of individual strains. Here, we developed a high-throughput screening platform integrating a microbial whole-cell biosensor into a double-layer plate assay, enabling rapid identification of bacteria producing cell wall-targeting antibiotics from environmental samples. The biosensor is based on the

Green synthesis of metal nanoparticles (NPs) using biological systems provides a sustainable alternative to conventional chemical fabrication methods. This protocol presents reproducible experimental and analytical procedures for the synthesis of gold (Au) and silver (Ag) NPs using bacterial cultures, fungal filtrates, and plant leaf extracts as reducing and stabilizing agents. AgNPs were synthesized using extracts from Psidium guayaquilensis, Acanthophora spicifera, and Earliella sp., exhibiting characteristic ultraviolet-visible (UV-Vis) absorption bands between 405 and 425 nm that confirmed

Bioreduction is considered to be one of the most effective methods for the bioremediation of uranium-contaminated soil or water, in which the low extracellular electron transfer (EET) rate of microorganisms seriously affects the reduction and fixation of uranium through microorganisms. Here, we develop an in-situ self-regenerable Shewanella putrefaciens (S. putrefaciens)-FeS biohybrid system to enhance the efficiency of uranium reduction. The dense layer of bio-FeS on the cell surface of S. putrefaciens provides excellent uranium tolerance and activity for bio-FeS-mediated uranium reduction. B

Sustainable recycling strategies are urgently needed to address the rapidly growing accumulation of spent lithium-ion batteries. Current strategies for the first step in recycling, the solubilization of lithium and other metals, are energy intensive and/or generate toxic waste. This study critically evaluated the option of 'microbial electro-extraction' (MEE), a process in which electroactive microbes supply electrons to reduce the nickel, cobalt, and manganese in spent lithium-ion battery powder, releasing the soluble reduced ions of these metals along with soluble Li+. We introduce a near-real-time optical imaging method that works via the detection of the intrinsic fluorescence of life forms upon excitation by deep-UV (DUV) illumination. A DUV (<250-nm) source enables the detection of microbes in their native state on natural materials, avoiding background autofluorescence and without the need for fluorescent dyes or tags. We demonstrate that DUV-laser-induced native fluorescence can detect bacteria on opaque surfaces at spatial scales ranging from tens of centimeters to micrometers and from communities to single cells. Given exposure times of 100 μs and lo

Homologs of the polar landmark proteins HubP and FimV are widespread among bacterial species. They all share several common features, including a periplasmic LysM-like domain, a transmembrane region, an extensive cytoplasmic domain enriched in acidic amino acids, and a C-terminal tetrapeptid-repeat (TPR) domain referred to as the FimV domain. Apart from these conserved general features, however, the proteins exhibit little homology across different bacterial genera. Here, we characterized Pseudomonas putida FimV (PpFimV) with respect to cellular processes involving FimV or HubP in other specie

There is an urgent need to develop novel antibiotics since antibiotic resistance is an increasingly serious threat to global public health. Whole-cell biosensors are one of the promising strategies for new antibiotic discovery. The peptidoglycan (PG) of the bacterial cell wall is one of the most important targets for antibiotics. However, the biosensors for the detection of PG-targeting antibiotics in Gram-negative bacteria have not been developed, mainly because of the lack of the regulatory systems that sense and respond to PG stress. Recently, we identified a novel two-component signal tran

In order to explore the impact of antibiotics on the bacterial metabolic cycling of nitrate within contaminated soil and groundwater environments, we compared the effects of polymyxin B (PMB) and ciprofloxacin (CIP) concentration gradients on the distribution and activity of a wild type (WT) and a flagella deficient mutant (Δflag) of Shewanella oneidensis MR-1 in a microfluidic gradient chamber (MGC). Complementary batch experiments were performed to measure bacteriostatic versus bactericidal concentrations of the two antibiotics, as well as their effect on nitrate reduction. Prior work demons

Flow-through aquifer columns packed with a middle layer of granular iron (Fe0) were used to study the applicability and limitations of bio-enhanced Fe0 barriers for the treatment of contaminant mixtures in groundwater. Concentration profiles along the columns showed extensive degradation of hexavalent chromium Cr(VI), nitrate, sulfate, and trichloroethene (TCE), mainly in the Fe0 layer. One column was bioaugmented with Shevanella algae BRY, an iron-reducing bacterium that could enhance Fe0 reactivity by reductive dissolution of passivating iron oxides. This strain did not enhance Cr(VI), which

The quinol form (AHDS) of 9,10-anthraquinone-2,6-disulfonate (AQDS) was used as a titrant to determine bioavailability of Fe(III) in pure iron minerals and several soils. AHDS oxidation to AQDS was coupled to Fe(III) reduction to Fe(ll) in biological media consisting of trace salts and vitamins, providing estimates of bioavailability consistentwith the biogeochemical mechanisms and conditions that control Fe(III) availability to iron-reducing bacteria. Iron(III) oxide sources were synthetic oxides (amorphous and crystalline) and three soils separated into two size fractions each (0-500 and 500

Enzymatic toxins targeting nucleic acids are a widespread strategy for resolving biological conflicts. Ubiquitous HEPN domain-containing proteins harbor a conserved RNase motif and are found in both prokaryotes and eukaryotes. In prokaryotes, HEPN domain toxins frequently pair with adjacent MNT domain antitoxins to form the prevalent type VII HepT/MntA toxin-antitoxin (TA) system. Here, we identified a group of fused MNT-HEPN proteins (renamed FhepTA) carried by mobile genetic elements in the Shewanella genus; these proteins show high similarity to the HepT/MntA TA systems. When encoded as sep

Calcium ions may adversely affect struvite biomineralization, but the exact role remains elusive and the way to eliminate its adverse effect needs to be explored. Herein, Shewanella oneidensis MR-1 was selected to induce struvite biomineralization. The increase of Ca/Mg ratios from 0 to 2:1 resulted in a decrease of struvite yield from 98.6 mg to 0 mg. Calcium ions inhibited struvite crystallization by forming amorphous calcium phosphate (ACP) at the early stage of biomineralization. The increase of disodium ethylenediaminetetraacetate (EDTA) concentrations from 1 mM to 3 mM led to a rise of s

Chlorinated organic compounds are widely used as solvents, but they are pollutants that can have adverse effects on the environment and human health. Dissimilatory iron-reducing bacteria (DIRB) such as Shewanella and Geobacter have been applied to treat a wide range of halogenated organic compounds due to their specific biological properties. Until now, there has been no systematic review on the mechanisms of direct or indirect degradation of halogenated organic compounds by DIRB. This work summarizes the discussion of DIRB's ability to enhance the dechlorination of reaction systems through di

Dissimilatory iron-reducing bacteria (DIRB) with extracellular electron transfer (EET) capabilities have shown significant potential for bioremediating halogenated hydrocarbon contaminated sites rich in iron and humic substances. However, the role and microbial molecular mechanisms of iron-humic acid (Fe-HA) complexes in the reductive dehalogenation process of DIRB remains inadequately elucidated. In this study, we developed a sustainable carbon cycling approach using Fe-HA complexes to modulate the electron flux from sawdust (SD), enabling almost complete reductive dechlorination by most DIRB

BACKGROUND: Shewanella spp. are gram-negative bacteria, saprophytes, and rarely pathogenic. Saccharomyces cerevisiae is the well-known yeast used for fermentation in industry and molecular biology for research. In humans, it is a very rare pathogen which colonizes the digestive tract, and its utility has been linked to the treatment and prevention of diarrhea associated with Clostridium difficile.
CASE REPORT: A 27-year-old male, victim of aggressive, blunt trauma with a 4-day history of symptoms was admitted to our surgery unit. Abdominal sonography revealed peritoneal fluid in all spaces wi

(99m)Tc-DTPA has been used as a conservative tracer to quantify water transport through porous media. However, more information on the reactivity of this (99m)Tc compound under varying geochemical conditions is desirable to better understand its potential uses. We measured the speciation of Tc following amendment of (99m)Tc-DTPA to batch systems spanning a range of controlled biogeochemical conditions. Our results suggest that (99m)Tc-DTPA is stable under the reducing conditions tested. However, freshly precipitated Al-ferrihydrite may displace Tc(IV) from DTPA in the absence of Fe(III)-reduci

Skin and soft tissue infections (SSTIs) are one of the most common infection syndromes and may be caused by a large number of microorganisms. Some principles of aquatic injuries are different than those of land-based trauma. Wounds sustained in marine environment are exposed to a milieu of bacteria rarely encountered in different settings. These include Vibrio spp., Aeromonas spp., Shewanella spp., Erysipelothrix rhusiopathiae, Mycobacterium marinum, Streptococcus iniae, and other microbes. Failure to recognize and treat these uncommon pathogens in a timely manner may result in significant mor

The nanomachine referred to as the type III secretion system (T3SS) is used by many Gram-negative pathogens or symbionts to inject their effector proteins into host cells to promote their infections or symbioses. Among the genera possessing T3SS is Vibrio, which consists of diverse species of Gammaproteobacteria including human pathogenic species and inhabits aquatic environments. We describe the genetic overview of the T3SS gene clusters in Vibrio through a phylogenetic analysis from 48 bacterial strains and a gene order analysis of the two previously known categories in Vibrio (T3SS1 and T3S
