Spatial Memory Disturbance Following Transient Brain Ischemia is Associated with Vascular Remodeling in Hippocampus.

A number of studies have investigated the effects of ischemic injury on functional and cellular characteristics of hippocampus. There is only a limited study on vascular remodeling of it. The present study aimed at examining vascular remodeling in hippocampus and spatial memory disturbances after transient brain ischemia. Male Wistar rats were randomly divided into four groups, i.e. sham operated (SHAM), transient brain ischemia with 1 day reperfusion (IR1), 3 day reperfusion (IR3), and 10 days reperfusion (IR10) groups. Transient brain ischemia was induced by bilateral common carotid artery o

Temporary conductive hearing loss in early life impairs spatial memory of rats in adulthood.

INTRODUCTION: It is known that an interruption of acoustic input in early life will result in abnormal development of the auditory system. Here, we further show that this negative impact actually spans beyond the auditory system to the hippocampus, a system critical for spatial memory. METHODS: We induced a temporary conductive hearing loss (TCHL) in P14 rats by perforating the eardrum and allowing it to heal. The Morris water maze and Y-maze tests were deployed to evaluate spatial memory of the rats. Electrophysiological recordings and anatomical analysis were made to evaluate functional and

Enriched Physical Environment Attenuates Spatial and Social Memory Impairments of Aged Socially Isolated Mice.

BACKGROUND: Social isolation in the elderly is one of the principal health risks in an aging society. Physical environmental enrichment is shown to improve sensory, cognitive, and motor functions, but it is unknown whether environmental enrichment can protect against brain impairments caused by social isolation. METHODS: Eighteen-month-old mice were housed, either grouped or isolated, in a standard or enriched environment for 2 months, respectively. Behavioral tests were performed to evaluate cognitive functional and social interaction ability. Synaptic protein levels, myelination, neuroinfla

Multiple bout rTMS on spatial working memory: a comparison study of two cortical areas.

It has been established that acute (within-session) repetitive transcranial magnetic stimulation (rTMS) improves spatial working memory (SWM). However, questions remain regarding the safety and effectiveness of multiple bouts of rTMS and the optimal cortical area to stimulate. This preliminary study investigated, in healthy participants, multiple bouts of rTMS over the dorsolateral pre-frontal cortex (DLPFC), or posterior parietal cortex (PPC) on SWM. Twenty participants (10m, 10f), all naïve to rTMS, where randomized into a DLPFC or PPC group, receiving six sessions of rTMS (5Hz at 80% of mot

Enhancing Spatial Memory: Anxiolytic and Antidepressant Effects of Tapinanthus dodoneifolius (DC) Danser in Mice.

We evaluated the anxiolytic and antidepressant effects of the aqueous extract of the bark of Tapinanthus dodoneifolius (TAE) (Danser) (25, 50, and 100 mg/kg), using open field, elevated plus maze, and forced swimming tests. Effect of TAE was compared to standard drugs diazepam (2 mg/kg) and imipramine (10 mg/kg). Additionally, the same doses of TAE were evaluated on rat's memory using Y-maze task. Results showed a significant (P < 0.05; 100 mg/kg) increase in the percentage of open arm entry and the time spent in the open arms in the elevated plus maze, suggesting an anxiolytic activity of the

DL-/PO-phosphatidylcholine restores restraint stress-induced depression-related behaviors and spatial memory impairment.

The present study investigated the effects of 1,2-dilinoleoyl-sn-glycero-3-phosphocholine (DL-PC) and 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (PO-PC) on depression-related behaviors and spatial memory impairment in mice subjected to restraint stress. The immobility time in forced-swim and tail-suspension tests for mice subjected to restraint stress was significantly longer than that for nonstressed control mice, and oral coadministration of DL-PC and PO-PC (DL-/PO-PC; DL-PC : PO-PC=1 : 1) shortened the prolonged immobility time in a dose (0.1-5 mg/kg)-dependent manner. Phenylbutyric acid protects against spatial memory deficits in a model of repeated electroconvulsive therapy.

Repeated electroconvulsive therapy (rECT) is widely applied in the treatment of refractory depression. Among the side effects of rECT, memory impairment is noticeable and needs effective protection. In this study, by employing a recognized repeated electroconvulsive shock (rECS) rat model, we found that rECS induced the significant spatial memory retention deficits with the simultaneous decreases in long-term potential (LTP), enhanced excitable postsynaptic potentials (EPSP), population spike (PS) and input/output curve in perforant pathway-dentate gyrus (PP-DG), but had no obvious neuron loss

Dorso- and ventro-lateral prefrontal volume and spatial working memory in schizotypal personality disorder.

Schizotypal personality disorder (SPD) individuals and borderline personality disorder (BPD) individuals have been reported to show neuropsychological impairments and abnormalities in brain structure. However, relationships between neuropsychological function and brain structure in these groups are not well understood. This study compared visual-spatial working memory (SWM) and its associations with dorsolateral prefrontal cortex (DLPFC) and ventrolateral prefrontal cortex (VLPFC) gray matter volume in 18 unmedicated SPD patients with no BPD traits, 18 unmedicated BPD patients with no SPD trai

Serotonin transporter genotype and depressive symptoms moderate effects of nicotine on spatial working memory.

Smokers may use nicotine to self-medicate for situation-specific or person-specific cognitive or affective deficits. Although evidence suggests that nicotine replacement therapy (NRT), relative to placebo, enhances spatial working memory (SWM) in smoking-abstinent smokers with schizophrenia, the extent to which NRT may be helpful in attenuating abstinence-related SWM in other groups with deficits in SWM is unknown. Depressive symptoms are associated with both tobacco smoking and deficits in SWM. Previous studies have found that smoking abstinence increases depressive affect and depression-rela

The neural correlates of non-spatial working memory in velocardiofacial syndrome (22q11.2 deletion syndrome).

Velocardiofacial syndrome (VCFS), also known as 22q11.2 deletion syndrome, is a neurogenetic disorder that is associated with both learning disabilities and a consistent neuropsychological phenotype, including deficits in executive function, visuospatial perception, and working memory. Anatomic imaging studies have identified significant volumetric reductions in the parietal lobe of individuals with VCFS, but several studies have reported that the frontal lobe is relatively preserved. We used functional magnetic resonance imaging to investigate the neural correlates of non-spatial working memo

Alpha-2 adrenoceptor activation inhibits phencyclidine-induced deficits of spatial working memory in rats.

N-methyl-D-aspartate (NMDA)/glutamate receptor antagonists, such as phencyclidine (PCP), induce behavioral abnormalities (locomotor hyperactivity, sensorimotor gating deficits, impairments of cognition) in animals that are thought to model aspects of schizophrenia. The administration of PCP increases noradrenaline transmission in the rat prefrontal cortex, a brain structure required for normal cognitive processes. Noradrenaline, in turn, works through a set of receptors that have themselves been implicated directly in NMDA antagonist-induced deficits; we recently reported that the alpha-2 agon

Impaired remote spatial memory after hippocampal lesions despite extensive training beginning early in life.

Damage to the hippocampus typically produces temporally graded retrograde amnesia, whereby memories acquired recently are impaired more than memories acquired remotely. This phenomenon has been demonstrated repeatedly in a variety of species and tasks, and it has figured prominently in theoretical treatments of memory and hippocampal function. A striking exception to the finding of temporally graded retrograde amnesia comes from studies with rodents using spatial tasks like the water maze. In these studies, recent and remote memory were similarly impaired following hippocampal lesions. In cont

Deficits of spatial and non-spatial memory and of auditory fear conditioning following anterior thalamic lesions in mice: comparison with chronic alcohol consumption.

This study was aimed at determining (i) whether or not bilateral subtotal lesions of the anterior thalamic nuclei (ATH) in rodents produced memory deficits for spatial and/or non-spatial information and of auditory fear conditioning, and (ii) if these eventual deficits resemble those produced by chronic alcohol consumption (CAC). Working memory was assessed using both spatial (spontaneous alternation) and non-spatial (temporal alternation) delayed response tasks. Results showed that ATH lesions induced delay-dependent memory impairments in both spatial and non-spatial alternation tasks, as wel

Gamma transcranial alternating current stimulation prevents scopolamine-induced damage to EEG activity and spatial memory.

BackgroundDementia is a serious public health concern, primarily caused by Alzheimer's disease (AD). Transcranial alternating current stimulation (tACS) can improve cognitive function by modulating neural oscillations. While tACS shows therapeutic potential for dementia, its preventive effects remain unknown. Because tACS has long-lasting modulatory effects, we propose that these offline effects may help prevent dementia.ObjectiveThis study aims to determine whether pre-administered 40 Hz tACS prevents scopolamine-induced impairment of electroencephalogram (EEG) activity and spatial cognition.

Spatial statistical learning of task relevance, rather than stimulus prevalence, improves visual working memory recall.

Our visual environment provides us with more information than we can process. Processing efficiency may be enhanced by leveraging inherent regularities in the environment, such as prioritizing likely target objects over unlikely ones. Although there is evidence that statistical learning improves visual working memory performance, it is unclear whether this improvement occurs through stimulus prevalence (how likely we are to encounter stimuli) or task relevance (how likely we are to use these stimuli). To distinguish between these two hypotheses, we examined whether frequent appearance, or freq

Spatial reorganization of object representations in high-level visual cortex distinguishes working memory from perception.

The visual system balances veridical object perception with flexible visual working memory (VWM), both engaging high-level visual regions. However, how these competing demands shape object-selective cortex (OSC) representations remains unclear. Here, we ask whether VWM inherits the spatial constraints observed in the OSC during perception or reorganizes representations to meet mnemonic demands. Using a matched task design and functional magnetic resonance imaging-based decoding, we systematically compared the localization of object-identity representations during perception and VWM. A robust d

Protective role of hemokinin-1 against age-dependent spatial memory and attention decline in mice.

Age-related cognitive decline is a major social and medical challenge, but its mechanisms remain poorly understood; prevention and therapy are unsatisfactory. The tachykinin hemokinin-1 (HK-1), encoded by the Tac4 gene, is expressed in brain regions involved in learning and memory and has been implicated in neuroimmune regulation, but its contribution to age-dependent cognitive impairment remains unclear. Here, we investigated whether endogenous HK-1 modulates cognitive performance during aging. Young (3-5 months), middle-aged (12-15 months), and old (18-20 months) male and female C57BL/6 J wi

Deletion of RGS5 attenuates high-fat diet-induced cerebrovascular pathology and associated spatial memory deficits.

Diet-induced metabolic dysregulation is associated with cerebral microvascular pathology contributing to cognitive decline and vascular dementia. These alterations include blood-brain barrier (BBB) leakage, pericyte dysfunction, aberrant angiogenesis and perivascular neuroinflammation. Restoration of BBB integrity has been achieved in other conditions by targeting pericytes, for example through knockout of Regulator of G-protein Signaling 5 (RGS5), a sensor protein for hypoxia and oxidative stress highly enriched in these cells. It remains unclear whether deletion of RGS5 can provide vascular

Remote spatial memory deficits in mouse models of neuropsychiatric disorders with immature dentate gyrus phenotype.

BACKGROUND: The hippocampal dentate gyrus (DG) is a critical region that contributes to recent and remote memory. Granule cells within this region, in which adult neurogenesis occurs, undergo dynamic and reversible maturation via genetic and environmental factors during adulthood. A pseudo-immature state of DG granule cells, called immature DG (iDG), has been observed in the adult mice of certain mutant strains, which are considered animal models of neuropsychiatric and neurodegenerative disorders, such as intellectual disability, schizophrenia, autism, and Alzheimer's disease. However, the as

EEG slow oscillations and overnight spatial navigational memory performance in CPAP-treated obstructive sleep apnea.

Obstructive sleep apnea (OSA) exerts pathogenic effects through a combination of sleep fragmentation (SF) and intermittent hypoxia (IH). The mechanisms through which sleep disruption impacts memory might arise by investigating disruption of specific sleep stages and, when such disruption occurs through OSA, by evaluating the individual contributions of SF and IH. Given that region-specific EEG slow activity during non-REM sleep has been associated with overnight declarative, motor, and spatial memory formation, we investigated the effects of disrupting slow wave sleep (SWS) on a virtual maze n
