Process optimization and hybrid nanoflower synthesis of 5- hydroxymethylfurfural oxidase for efficient oxidation of HMF and its derivatives to 2,5- furandicarboxylic acid.

2,5-furandicarboxylic acid (FDCA) is a precursor of the biopolymer Polyethylene furandicarboxylate (PEF). 5-hydroxymethylfurfural oxidase (5-HMFO) oxidizes 5- hydroxymethylfurfural (HMF) to FDCA. One such promising biocatalyst is 5-HMFO from Pseudomonas poae (PSPO). The major limitations of free enzymes include high cost, low thermal stability, limited storage stability and lack of reusability. To mitigate these limitations, the protein-inorganic hybrid nanoflowers of PSPO and Mn2+ (PSPO@MnNF) was developed. PSPO@MnNF exhibited a flower-like morphology, as observed under scanning electron micr

Host serine protease ACOT2 assists DENV proliferation by hydrolyzing viral polyproteins.

Dengue fever is a mosquito-borne tropical disease caused by the dengue virus (DENV). The replication of DENV relies on the processing of its genome-encoded polyprotein by both viral protease NS3 (NS3pro) and host proteases. However, the impact of host proteases on DENV proliferation is not well understood. In this study, we utilized fluorophosphonate-based probes (FPs) to investigate the up-regulation of host serine proteases during DENV infection in detail. Among the identified proteases, acyl-CoA thioesterase 2 (ACOT2), an enzyme that hydrolyzes acyl-CoA molecules to generate fatty acids and

Ribitol enhances matriglycan of α-dystroglycan in breast cancer cells without affecting cell growth.

The laminin-binding glycan (matriglycan) on α-dystroglycan (α-DG) enables diverse roles, from neuronal development to muscle integrity. Reduction or loss of matriglycan has also been implicated in cancer development and metastasis, and specifically associated with high-grade tumors and poor prognoses in breast cancers. Hyperglycosylation of α-DG with LARGE overexpression is shown to inhibit cancer cell growth and tumorigenicity. We recently demonstrated that ribitol, considered to be a metabolic end-product, enhances matriglycan expression in dystrophic muscles in vivo. In the current study, w

Na-Ion Battery Anodes: Materials and Electrochemistry.

The intermittent nature of renewable energy sources, such as solar and wind, calls for sustainable electrical energy storage (EES) technologies for stationary applications. Li will be simply too rare for Li-ion batteries (LIBs) to be used for large-scale storage purposes. In contrast, Na-ion batteries (NIBs) are highly promising to meet the demand of grid-level storage because Na is truly earth abundant and ubiquitous around the globe. Furthermore, NIBs share a similar rocking-chair operation mechanism with LIBs, which potentially provides high reversibility and long cycling life. It would be

Arylsulphatase A activity and sulphatide concentration in the female rabbit oviduct are under physiological hormonal influence.

Oviduct samples of female rabbits in different phases of the reproductive cycle were analysed to establish the role of sex steroid hormones in the regulation of sulphatide concentration and arylsulphatase A activity. In addition to biochemical procedures, histochemical techniques were used to localize both enzyme activity and the natural substrate. The plasma concentrations of progesterone and 17 beta-oestradiol were determined by radioimmunoassay (RIA). The findings show that the parameters examined undergo considerable changes during the different phases of the reproductive cycle. Oestrogens

Phosphorylation of Synaptojanin Differentially Regulates Endocytosis of Functionally Distinct Synaptic Vesicle Pools.

UNLABELLED: The rapid replenishment of synaptic vesicles through endocytosis is crucial for sustaining synaptic transmission during intense neuronal activity. Synaptojanin (Synj), a phosphoinositide phosphatase, is known to play an important role in vesicle recycling by promoting the uncoating of clathrin following synaptic vesicle uptake. Synj has been shown to be a substrate of the minibrain (Mnb) kinase, a fly homolog of the dual-specificity tyrosine phosphorylation-regulated kinase 1A (DYRK1A); however, the functional impacts of Synj phosphorylation by Mnb are not well understood. Here we

Changes in sensorimotor inhibition across the menstrual cycle: implications for neuropsychiatric disorders.

The startle reflex is inhibited when the starting noise is preceded 30-500 msec by a weak prepulse. "Prepulse inhibition" (PPI) is reduced in specific neuropsychiatric disorders characterized clinically by impaired inhibition of sensory, motor, or cognitive information. PPI is sexually dimorphic, with men exhibiting significantly more PPI than women. We examined possible neuroendocrine substrates for this sex difference in PPI. The startle reflex, and a measure of visuospatial priming, were measured in 10 men, and in 46 normal women at different points in their menstrual cycle. In women, PPI w

A multifaceted role for ATM in genome maintenance.

The pleiotropic nature of the clinical phenotypes of patients with ataxia-telangiectasia (A-T)--which encompass cerebellar degeneration (leading to ataxia), gonadal atrophy, and cancer predisposition--suggests multiple functions of the gene responsible for the disease. The ataxia-telangiectasia mutated gene product (ATM), whose loss of function is responsible for ataxia-telangiectasia, is a protein kinase that interacts with several substrates and is implicated in mitogenic signal transduction, chromosome condensation, meiotic recombination, cell-cycle control and telomere maintenance. Area-Selective Atomic Layer Deposition of Metal Oxides on Noble Metals through Catalytic Oxygen Activation.

Area-selective atomic layer deposition (ALD) is envisioned to play a key role in next-generation semiconductor processing and can also provide new opportunities in the field of catalysis. In this work, we developed an approach for the area-selective deposition of metal oxides on noble metals. Using O2 gas as co-reactant, area-selective ALD has been achieved by relying on the catalytic dissociation of the oxygen molecules on the noble metal surface, while no deposition takes place on inert surfaces that do not dissociate oxygen (i.e., SiO2, Al2O3, Au). The process is demonstrated for selective

Structural characterization of the reaction pathway in phosphoserine phosphatase: crystallographic "snapshots" of intermediate states.

Phosphoserine phosphatase (PSP) is a member of a large class of enzymes that catalyze phosphoester hydrolysis using a phosphoaspartate-enzyme intermediate. PSP is a likely regulator of the steady-state d-serine level in the brain, which is a critical co-agonist of the N-methyl-d-aspartate type of glutamate receptors. Here, we present high-resolution (1.5-1.9 A) structures of PSP from Methanococcus jannaschii, which define the open state prior to substrate binding, the complex with phosphoserine substrate bound (with a D to N mutation in the active site), and the complex with AlF3, a transition

Phospholipid/calcium-dependent protein kinase (protein kinase C) system: a major site of bioregulation.

The substrate specificity determinants of protein kinase C are probed using synthetic peptides encompassing the major phosphorylation site serine 115 in bovine MBP. The results indicate that basic residues arginine 107 and 113 N-terminal to the phosphorylation site are essential for the substrate activity of the peptides. Substitutions of these basic residues by alanine cause a marked decrease in their substrate activity and the resulting peptide analogs become specific and rather potent inhibitors of protein kinase C. Leukemic cells are particularly abundant in protein kinase C and its substr

Celiac disease plasma cells recognize the active form of transglutaminase 2 with and without bound substrate.

Autoantibodies to the enzyme transglutaminase 2 (TG2) are characteristic of celiac disease, and TG2-specifc plasma cells are abundant in the celiac gut lesion. The production of antibodies by effector B cells (i.e. plasma cells) are believed to result from collaboration between TG2-specific B cells and gluten-specific CD4+ T cells - a collaboration facilitated by formation of TG2-gluten enzyme-substrate complexes. Since binding of substrate is associated with conformational changes in TG2, we sought to address whether the conformation of TG2 influences plasma cell reactivity. Using effector mo

Inhibition of falcilysin from Plasmodium falciparum by interference with its closed-to-open dynamic transition.

In the absence of an efficacious vaccine, chemotherapy remains crucial to prevent and treat malaria. Given its key role in haemoglobin degradation, falcilysin constitutes an attractive target. Here, we reveal the mechanism of enzymatic inhibition of falcilysin by MK-4815, an investigational new drug with potent antimalarial activity. Using X-ray crystallography, we determine two binary complexes of falcilysin in a closed state, bound with peptide substrates from the haemoglobin α and β chains respectively. An antiparallel β-sheet is formed between the substrate and enzyme, accounting for seque

Activity, specificity, and probe design for the smallpox virus protease K7L.

The K7L gene product of the smallpox virus is a protease implicated in the maturation of viral proteins. K7L belongs to protease Clan CE, which includes distantly related cysteine proteases from eukaryotes, pathogenic bacteria, and viruses. Here, we describe its recombinant high level expression, biochemical mechanism, substrate preference, and regulation. Earlier studies inferred that the orthologous I7L vaccinia protease cleaves at an AG-X motif in six viral proteins. Our data for K7L suggest that the AG-X motif is necessary but not sufficient for optimal cleavage activity. Thus, K7L require

Correlative Electron-Ion Beam Analysis of the Effect of Lithiophilic Interlayers on Regulating the Li+ Flux Distribution in Anode Free Solid-State Batteries.

Anode-free solid-state batteries (AFSSBs) are promising candidates for next-generation high-energy-density batteries, but their widespread use is impeded by nonuniform lithium plating on the anode-side current collector (CC), which compromises stability and cycle life. Although the use of lithiophilic interlayers exhibits uniform Li plating, the underlying mechanisms by which they regulate lithium deposition are not yet fully understood. Here, a correlative focused ion beam-scanning electron microscopy-secondary ion mass spectrometry (FIB-SEM-SIMS) imaging approach combined with lithium isotop

Impact of high-fat, low-carbohydrate diet on myocardial substrate oxidation, insulin sensitivity, and cardiac function after ischemia-reperfusion.

High-fat, low-carbohydrate Diet (HFLCD) impairs the myocardial response to ischemia-reperfusion, but the underlying mechanisms remain elusive. We sought to determine the magnitude of diet-induced alterations in intrinsic properties of the myocardium (including insulin sensitivity and substrate oxidation) and circulating substrate and insulin differences resulting from diet, leading to this impaired response. Rats were fed HFLCD (60% kcal from fat/30% protein/10% carbohydrate) or control diet (CONT) (16%/19%/65%) for 2 wk. Isolated hearts underwent global low-flow ischemia followed by reperfusi

CD8 T lymphocytes deploy embryonic cell cycle control mechanisms for rapid cell proliferation.

Rapid proliferation of CD8 T cells is crucial for adaptive immunity against viral infection. CD8 T cells can complete division cycles in less than 6 h, representing a physiological extreme for somatic mammalian cells. Embryonic stem cells utilize specialized cell cycle control mechanisms, including subdued periodic expression, for rapid cell division cycles. CD8 T cell cycle control remains poorly understood. Here, we test whether CD8 T cells utilize embryonic mechanisms to promote rapid cell cycles. We comprehensively measure protein abundances in G1, S, and G2&M phases in three murine cell t

Interlocked Interface Enhances Mechanical Integrity for Thermal-Cycling-Stable Perovskite Solar Cells With 26.82% Certified Efficiency.

Perovskite solar cells (PSCs) experience mechanical damage and failure (i.e. degradation and fracture) induced by temperature changes under thermal cycling. However, few studies have been able to simultaneously suppress interface delamination and delay chemical degradation to ensure the mechanical integrity of perovskite film under thermal shock, making it challenging to improve the thermal cycling stability of PSCs. We report a universal interlocking strategy via the modification of polymethyl(hydro)/polymethylvinylsilazane (PHVS), which achieves interfacial interlocking through the condensat

Quantifying the underestimated plant potential for phosphorus removal in constructed wetlands: Revealing the mediating mechanism of radial oxygen loss.

Substrate adsorption is considered the primary phosphorus removal pathway in subsurface flow constructed wetlands (CWs), but the role of plants in this process remains underestimated. To clarify this issue, this study conducted root restriction by decreasing substrate depth from 60 cm to 10 and 20 cm to intensify the radial oxygen loss (ROL). On this basis, the resultant enhancement to soluble total phosphorus (TP) removal from late spring to early winter was quantified. Further, the underlying enhancing mechanisms were revealed by analyzing the P mass balance and substrate surface interfacial
