Surfen, a proteoglycan binding agent, reduces inflammation but inhibits remyelination in murine models of Multiple Sclerosis.

Proteoglycans are promising therapeutic targets in Multiple Sclerosis (MS), because they regulate many aspects of the immune response. This was studied using surfen, an agent that binds both heparan sulphate proteoglycans (HSPGs) and chondroitin sulphate proteoglycans (CSPGs). Initial cell culture work on bone marrow derived macrophages (BMDMs) found that surfen reduced concentrations of the chemokines CCL2, CCL4 and CCL5, with reduced messenger (m)RNA expression for Tumor Necrosis Factor, IL-6, IL-1β and inducible nitric oxide synthase. These data were further explored using Experimental Auto

The Effect of Coronary Artery Bypass Surgery on Interleukin-18 Concentration and Biomarkers Related to Vascular Endothelial Glycocalyx Degradation.

Surgical myocardial revascularization, regardless of the technique used, causes ischemia-reperfusion injury (IRI) in the myocardium mediated by inflammation and degradation of the endothelial glycocalyx (EG). We investigated the difference between on-pump and off-pump techniques in terms of the concentration of proinflammatory interleukin (IL)-18 and the EG degradation products syndecan-1 and hyaluronic acid measured by ELISA in the peripheral and cardiac circulation during open heart surgery and in the early postoperative period. The concentration of IL-18, C-reactive protein (CRP), and cardi

Coagulopathy in Dabie bandavirus infection is related to endogenous heparinoids release and mast cells activation.

BACKGROUND: The prolonged activated partial thromboplastin time (APTT) and thrombin time in patients infected with Dabie bandavirus (DBV) have been recognized to be poor prognostic indicators and attributed to a heparin-like effect rather than a decrease in coagulation factor levels. OBJECTIVES: To clarify the correlation between endogenous heparinoids and coagulopathy in patients infected with DBV, and their possible sources. METHODS: One hundred and twenty-one consecutive patients with confirmed DBV infection were enrolled in this prospective, single-center, observational study. Routine co

14-3-3η protein ameliorates glomerular endothelial cell injury in lupus nephritis by regulating mTOR pathway-mediated autophagy.

BACKGROUND: Lupus nephritis (LN), a severe complication of systemic lupus erythematosus (SLE), is characterized by glomerular endothelial cell injury, inflammation, and immune complex deposition. The 14-3-3η protein, a multifunctional regulator, plays roles in various cellular processes, however, its specific role in LN and the underlying mechanisms remain unclear. This study investigates the role of 14-3-3η in LN by modulating mTOR pathway-mediated autophagy. METHODS: Human renal glomerular endothelial cells (HRGECs) overexpressing 14-3-3η were treated with plasma from healthy donors and LN

Hyaluronan and N-ERC/mesothelin as key biomarkers in a specific two-step model to predict pleural malignant mesothelioma.

PURPOSE: Diagnosis of malignant mesothelioma is challenging. The first available diagnostic material is often an effusion and biochemical analysis of soluble markers may provide additional diagnostic information. This study aimed to establish a predictive model using biomarkers from pleural effusions, to allow early and accurate diagnosis. PATIENTS AND METHODS: Effusions were collected prospectively from 190 consecutive patients at a regional referral centre. Hyaluronan, N-ERC/mesothelin, C-ERC/mesothelin, osteopontin, syndecan-1, syndecan-2, and thioredoxin were measured using ELISA and HPLC

Fibrinogen based resuscitation mitigates lung injury in mice with bacterial pneumonia after hemorrhagic shock.

While pneumonia is a common complication following severe trauma, the optimal treatment for this complication remains unclear. Our previous studies have shown that fibrinogen and fresh frozen plasma (FFP) administration have beneficial effects in mitigating lung dysfunction in mice after trauma and hemorrhagic shock (HS), due to the restoration of endothelial syndecan-1. The objective of the current study was to test these two therapeutics in a combined model of HS and pneumonia. We hypothesized they would be equally protective. C57BL/6 mice underwent HS [mean arterial pressure (MAP) of 40-45

Comparative diagnostic value of NGAL and multibiomarker models in pediatric sepsis-associated acute kidney injury in Indonesia: a preliminary study.

PURPOSE: Sepsis is a leading cause of pediatric critical illness and a major contributor to acute kidney injury (AKI). Sepsis-associated AKI (SA-AKI) worsens outcomes, yet current diagnostic criteria based on serum creatinine and urine output lack sensitivity for early detection. This study evaluated the diagnostic performance of neutrophil gelatinase-associated lipocalin (NGAL), angiopoietin-2, and syndecan-1 for early identification of SA-AKI in children with sepsis. METHODS: A prospective nested case-control study was conducted in children aged 1-18 years with sepsis. Participants were cla

Distinct B-cell populations are present in hepatic and intestinal Schistosoma mansoni granulomas.

Although it is generally accepted that schistosomal granuloma formation results from a T-cell dependent host response towards the parasite egg, attention has recently been focused on the involvement of B-cells in the induction of schistosome-induced pathology. In this study we investigated the involvement of two functionally different B-cell populations in the formation of the Schistosoma mansoni granuloma: naive and antigen-stimulated B-cells. In liver granulomas two distinct B-cell populations were found, namely unstimulated B-cells at the periphery of the granuloma and antigen-stimulated, s

Neutrophil elastase and syndecan shedding contribute to antithrombin depletion in murine anthrax.

Bacillus anthracis infection is associated with severe hemostatic disturbances but their roles and contribution to fatality remain incompletely characterized. We undertook analyses of circulating antithrombin levels during the course of infection using a comparison of lethal and nonlethal murine anthrax models. Plasma samples were obtained from DBA/2 mice challenged intraperitoneally with the spores of either toxigenic B. anthracis Sterne strain or nontoxigenic, avirulent delta Sterne strain. We found that plasma antithrombin levels were rapidly depleted in Sterne spore-challenged mice, concom

Degradation of endothelial glycocalyx in Tanzanian children with falciparum malaria.

A layer of glycocalyx covers the vascular endothelium serving important protective and homeostatic functions. The objective of this study was to determine if breakdown of the endothelial glycocalyx (eGC) occurs during malaria infection in children. Measures of eGC integrity, endothelial activation, and microvascular reactivity were prospectively evaluated in 146 children: 44 with moderately severe malaria (MSM), 42 with severe malaria (SM), and 60 healthy controls (HC). Biochemical measures of eGC integrity included plasma syndecan-1 and total urinary glycosaminoglycans (GAG). Side-stream dark

Endothelium Protective Effect of 5% Human Albumin-Based Fluid Therapy in Perforation Peritonitis Patients Undergoing Emergency Laparotomy: A Randomized Controlled Trial.

INTRODUCTION: Preclinical and retrospective data indicated that albumin therapy may be associated with endothelial protection and lower net fluid balance in sepsis. However, endothelial protective effect of albumin has never been evaluated in a clinical trial. We therefore hypothesized that an infective, inflammatory condition like perforation peritonitis, which requires substantial fluid replacement, may benefit from albumin therapy. METHODS: Adult patients undergoing emergent/urgent abdominal surgery for perforation peritonitis were randomized to either group A or group P receiving 5% human

Early Endothelial Injury in Pancreas Transplantation: Insights from a Prospective Cohort Largely Composed of Simultaneous Pancreas-Kidney Recipients.

BACKGROUND/OBJECTIVES: Ischemia-reperfusion injury (IRI) contributes to graft dysfunction in solid organ transplantation, with the pancreas vulnerable due to its fragile vasculature. Endothelial glycocalyx (eGCX) disruption is central to this process. This study prospectively examined perioperative endothelial injury in pancreas transplantation. METHODS: Fifty-two recipients were included, of whom 47 underwent simultaneous pancreas-kidney (SPK) transplantation and 5 pancreas retransplantation. Biomarkers of eGCX degradation (syndecan-1, heparan sulfate (HS) and hyaluronan) and endothelial inj

Kawasaki disease and increased cardiovascular risk: Is there a link to circulating glycocalyx biomarkers?

AIMS: Kawasaki disease (KD) is an acute systemic vasculitis with possible long-term impact of general cardio-vascular health. An endothelial glycocalyx disorder during the disease's acute phase might predispose to long-term vascular anomalies leading to endothelial dysfunction and atherosclerosis. To investigate any association between increased cardiovascular risk and endothelial glycocalyx, we assessed circulating glycocalyx components in patients with a KD history, and analysed their association with acute-phase clinical features and more importantly, with patients' current cardiovascular r

Impairment of platelet NO signalling in coronary artery spasm: role of hydrogen sulphide.

BACKGROUND AND PURPOSE: The pathophysiology of coronary artery spasm (CAS), with its associated ischaemic crises, is currently poorly understood and treatment is frequently ineffective. In view of increasing evidence that platelet-based defects may occur in CAS patients, we investigated platelet reactivity in CAS patients and whether symptomatic crises reflect activation of platelet-endothelial interactions. EXPERIMENTAL APPROACH: CAS patients were evaluated during acute and/or chronic symptomatic phases and compared with healthy control subjects. Inhibition of ADP-induced platelet aggregatio

Nerve injury induces the expression of EXT2, a glycosyltransferase required for heparan sulfate synthesis.

Heparan sulfate proteoglycans, which bear long chains of heparan sulfate glycosaminoglycan, play significant roles during embryogenesis, including the formation of the CNS. However, their involvement in nerve regeneration has not yet been clarified. Here, we found that the mRNA expression of EXT2, one of the crucial enzymes for heparan sulfate-glycosaminoglycan synthesis, was markedly up-regulated in injured hypoglossal motor neurons after axotomy. In addition, immunohistochemical staining with an antibody specific for heparan sulfate-glycosaminoglycan chains demonstrated increased expression

Plasma growth differentiation factor-15 is associated with cardiovascular events in patients hospitalized for acute exacerbation of COPD.

Patients with acute exacerbation of chronic obstructive pulmonary disease (AECOPD) are at increased risk of major adverse cardiovascular events (MACE) and death. Growth differentiation factor-15 (GDF-15), a marker of cellular stress and inflammation, and Syndecan-1, a marker of endothelial dysfunction, have been suggested as prognostic biomarkers in plasma for MACE. We aimed to assess their association with a combined outcome of MACE or all-cause mortality over a 5-year period. This sub-study was embedded within the randomized controlled trial CORTICOsteroid reduction in COPD (CORTICO-COP), wh

Monocyte IL-1β predicts adverse cardiovascular events and associates with coronary microvascular dysfunction in kidney transplant recipients.

AIMS: Cardiovascular disease remains the leading cause of death after kidney transplantation. Coronary microvascular dysfunction (CMD) is common in kidney transplant recipients (KTRs), prognostically informative for cardiovascular events, and tightly related to inflammation. We aimed to test whether pretransplant monocytic expression of pyroptosis-related genes (IL-1β, GSDMD, Caspase-1, and NLRP3) independently predicts long-term mortality and major adverse cardiovascular events (MACE) in KTRs, and to evaluate its associations with CMD. METHODS: We enrolled 305 KTRs. Monocytes were isolated p
