Flavor perception and biological activities of bitter compounds in food.

Bitter compounds in food produce a distinct bitter taste that significantly influences overall flavor and quality, while also possessing valuable biological activities. Therefore, a systematic review summarizing recent research advances on bitter compounds is necessary for a better understanding of them. This review discusses the sources of bitter substances in food, the mechanism of bitterness perception, their biological activities and key issues for future research. Bitter compounds in food mainly include polyphenols, alkaloids, terpenoids, bitter peptides and Maillard reaction products. Bi

Oleanolic acid improves diet-induced obesity by modulating fat preference and inflammation in mice.

Obesity, triggered by high-fat diet (HFD), is associated to altered gustatory perception of dietary lipids. Oleanolic acid (OLA), a triterpene, has been reported to exert anti-obesity effects in animal models. Hence, we investigated the role of OLA in the modulation of oro-sensory perception of lipids in control and HFD-induced obese mice. As expected, OLA-treated obese mice exhibited a decrease in body, liver, and visceral adipose tissue weights. OLA treatment improved glucose tolerance, insulin level, plasma lipopolysaccharide (LPS), and hepatic cholesterol and triglyceride concentrations. O

Changes in gustatory perceptions of patients with major depression treated with vagus nerve stimulation (VNS).

BACKGROUND: Olfactory and gustatory functions were investigated before and during vagus nerve stimulation (VNS) in a group of 9 patients with therapy-resistant depression, implanted with a VNS system. METHODS: Gustation and olfaction were tested using standard sniffing tests. Subjects participated in 2 sessions with the vagal stimulator switched on and off, respectively. RESULTS: Under conditions of stimulation of the VNS, there were statistically significant differences of the threshold of perception, with an intensification of the taste "sweet" (Z = -2.0; p = 0.048) and "bitter" (Z = - 2.5

How Does Food Taste in Anorexia and Bulimia Nervosa? A Protocol for a Quasi-Experimental, Cross-Sectional Design to Investigate Taste Aversion or Increased Hedonic Valence of Food in Eating Disorders.

Background: Despite on-going efforts to better understand dysregulated eating, the olfactory-gustatory deficits and food preferences in eating disorders (ED), and the mechanisms underlying the perception of and responses to food properties in anorexia nervosa (AN) and bulimia nervosa (BN) remain largely unknown; both during the course of the illness and compared to healthy populations. It is, therefore, necessary to systematically investigate the gustatory perception and hedonics of taste in patients with AN and BN. To this end, we will examine whether aversions to the taste of high-calorie fo

Sweets and fats tasting in patients with anorexia nervosa: the role of the thought-shape fusion cognitive distortion.

INTRODUCTION: It has been found that the olfactorygustatory function is altered in patients with eating disorders, with an impairment affecting the perception of olfactory and gustatory stimuli. OBJECTIVE: The aim was to explore the subjective reactivity after the exposure and tasting of foods with different gradient of sweetness and different fats textures. In addition, changes in the thought-shape fusion (TSF) cognitive distortion were assessed after tasting those different presentations as well as the correlations between the initial scores on TSF-Questionnaire (TSF-Q) and the different re

Losing the sense of smell does not disrupt processing of odor words.

Whether language is grounded in action and perception has been a key question in cognitive science, yet little attention has been given to the sense of smell. We directly test whether smell is necessary for comprehension of odor language, by comparing language processing in a group of participants with no sense of smell (anosmics) to a group of control participants. We found no evidence for a difference in online comprehension of odor and taste language between anosmics and controls using a lexical decision task and a semantic similarity judgment task, suggesting olfaction is not critical to t

Association between physical activity and taste-The advantage of increased intensity for some but not all individuals.

INTRODUCTION: Taste is a key sensory modulator of energy intake, and while the effects of acute physical activity (PA) on taste have been recently confirmed, its chronic effects remain poorly documented. METHODS: Data were extracted from the NHANES database, including salty and bitter taste tests, moderate PA (MPA) and vigorous PA (VPA) levels, and anthropometric data. Binary logistic regressions were conducted, and odds ratios (ORs) represent the association between physical activity level and successful taste tests. RESULTS: 3,114 participants (51.5% women, mean age 58.5±11.9 years, mean B

Correlation Between the TAS2R38 Genetic Polymorphism, PROP Perception and Eating Habits: A Cross-Sectional Molecular Epidemiological Study.

Background/Objectives: Genetic variation in bitter taste receptors, particularly TAS2R38, influences sensitivity to bitter compounds such as 6-n-propylthiouracil (PROP). However, its influence on food preferences and dietary habits remains unclear. This study investigated the relationship between TAS2R38 rs713598 polymorphism, a variant in strong linkage disequilibrium with other common variants and therefore served as a proxy for TAS2R38 bitter taste-related genetic variation, PROP responsiveness, food preferences, and adherence to the Mediterranean diet. Methods: A cross-sectional molecular

Sensitivity to Sweet and Salty Tastes in Children and Adolescents with Type 1 Diabetes.

Taste function impairment is observed in people with type 1 diabetes (T1D). It is most often related to sweet taste. It is associated with such factors as diabetic neuropathy, smoking, age, duration of the disease and a rigorous diet that eliminates easily digestible carbohydrates. The aim of the study was to compare sensitivity to sweet and salty tastes between healthy children and adolescents and children and adolescents with T1D. The study group consisted of children with T1D (n = 35), with at least 5 years of disease history, while the group of healthy children included 46 individuals sele

Sensation of dietary nutrients by gut taste receptors and its mechanisms.

Nutrients sensing is crucial for fundamental metabolism and physiological functions, and it is also an essential component for maintaining body homeostasis. Traditionally, basic taste receptors exist in oral cavity to sense sour, sweet, bitter, umami, salty and et al. Recent studies indicate that gut can sense the composition of nutrients by activating relevant taste receptors, thereby exerting specific direct or indirect effects. Gut taste receptors, also named as intestinal nutrition receptors, including at least bitter, sweet and umami receptors, have been considered to be activated by cert

The taste of neuroinflammation: Molecular mechanisms linking taste sensing to neuroinflammatory responses.

Evidence indicates a critical role of neuroinflammatory response as an underlying pathophysiological process in several central nervous system disorders, including neurodegenerative diseases. However, the molecular mechanisms that trigger neuroinflammatory processes are not fully known. The discovery of bitter taste receptors in regions other than the oral cavity substantially increased research interests on their functional roles in extra-oral tissues. It is now widely accepted that bitter taste receptors, for instance, in the respiratory, intestinal, reproductive and urinary tracts, are cruc

Discovery of a Novel P2X2/3 Allosteric Antagonist as an Efficient Bitter Taste Blocker.

Masking the taste of unpalatable medicines is a key approach to improving treatment compliance, especially with children, where this issue is exacerbated. The P2X2/3 purinergic receptor is a ligand-gated ion channel activated by ATP that relays information from taste cells to the brain. Inhibition of this receptor has been demonstrated to block taste perception in humans and mice, suggesting that the application of a potent and short-lived antagonist directly on the tongue could provide a solution to improve treatment compliance. The previously reported dual P2X3 and P2X2/3 antagonist AF-353 (

Exploring the characteristics and mechanisms of novel salt taste-enhancing peptides from porcine bone collagen via virtual screening, sensory evaluation, and molecular dynamics simulations.

This study aimed to identify salt taste-enhancing peptides from porcine bone collagen. In silico hydrolysis coupled with virtual screening identified eight potential salt taste-enhancing peptides. Further validation using sensory evaluation and electronic tongue analysis demonstrated that peptides DGGR, GAPGDR, VGPRGERGF, PGQRGERGF, and EMATQLAFMR enhanced the saltiness perception of a 0.3 % NaCl solution. Among these, DGGR exhibited the strongest salt taste-enhancing effect. DGGR, GAPGDR, VGPRGERGF, and EMATQLAFMR exhibited synergistic effects with NaCl, while PGQRGERGF showed an additive eff

Discovery and taste mechanisms of novel umami peptides from edible grasshopper (Oxya chinensis sinuosa) based on machine learning, molecular docking and dynamics simulation.

This study aimed to reveal umami peptides from grasshopper, and then explored their umami perception mechanism and umami enhancement. Four potential umami peptides (GDRFL, NGWGDY, NVTY and VVNY) were isolated, characterized and screened from grasshopper protein hydrolysate through machine learning and in silico approaches. Subsequently, sensory evaluation and electronic tongue confirmed their umami taste with thresholds of 0.08-0.25 mg/mL, which were lower than that of monosodium glutamate (0.3 mg/mL). The potent umami taste of GDRFL and NGWGDY was mainly driven by their umami amino acid compo
