Thalamic damage in periventricular leukomalacia: novel pathologic observations relevant to cognitive deficits in survivors of prematurity.

Despite major advances in the long-term survival of premature infants, cognitive deficits occur in 30-50% of very preterm (<32 gestational weeks) survivors. Impaired working memory and attention despite average global intelligence are central to the academic difficulties of the survivors. Periventricular leukomalacia (PVL), characterized by periventricular necrosis and diffuse gliosis in the cerebral white matter, is the major brain pathology in preterm infants. We tested the novel hypothesis that pathology in thalamic nuclei critical for working memory and attention, i.e. mediodorsal nucleus

Crocodilian Forebrain: Evolution and Development.

Organization and development of the forebrain in crocodilians are reviewed. In juvenile Caiman crocodilus, the following features were examined: identification and classification of dorsal thalamic nuclei and their respective connections with the telencephalon, presence of local circuit neurons in the dorsal thalamic nuclei, telencephalic projections to the dorsal thalamus, and organization of the thalamic reticular nucleus. These results document many similarities between crocodilians and other reptiles and birds. While crocodilians, as well as other sauropsids, demonstrate several features o

Development of the dorsal thalamus in a reptile: identification of subdivisions and their associated nuclei.

How the dorsal thalamus of amniotes (reptiles, birds, and mammals) is organized remains an important but incompletely answered question. Identification of meaningful subdivisions would greatly aid in its understanding. Because the dorsal thalamus is more simply organized during development, studies have examined this structure during embryogenesis. Most reports using this approach have examined the developing dorsal thalamus in mammals and birds. Only rarely has the development of the dorsal thalamus been investigated in reptiles. Regardless, any approach to identify subdivisions, the presumed

The parasubthalamic nucleus: A novel eating center in the brain.

Eating behavior stands as a fundamental determinant of animal survival and growth, intricately regulated by an amalgamation of internal and external stimuli. Coordinated movements of facial muscles and the mandible orchestrate prey capture and food processing, propelled by the allure of taste and rewarding food properties. Conversely, satiation, pain, aversion, negative emotion or perceived threats can precipitate the cessation or avoidance of eating activities. In recent years, the parasubthalamic nucleus (PSTN), located in the lateral hypothalamic area, has emerged as a focal point in feedin

Enhancing precision in MRgFUS for tremor treatment: a systematic review of tractography-based VIM targeting approaches.

BACKGROUND: Magnetic resonance guided focused ultrasound (MRgFUS) thalamotomy has been an effective treatment option in the management of various tremor etiologies. The process of targeting the VIM thalamic location relies on indirect atlas/coordinate-based systems using anatomic reference points. With advancing imaging modalities, thalamic structures can be better visualized resulting in additional targeting techniques and improved accuracy. OBJECTIVES: To identify and evaluate MRgFUS thalamotomy targeting techniques for tremor beyond atlas/coordinate-based systems. METHODS: A systematic re

The Paraventricular Thalamic Nucleus and Its Projections in Regulating Reward and Context Associations.

The paraventricular thalamic nucleus (PVT) is a brain region that mediates aversive and reward-related behaviors as shown in animals exposed to fear conditioning, natural rewards, or drugs of abuse. However, it is unknown whether manipulations of the PVT, in the absence of external factors or stimuli (e.g., fear, natural rewards, or drugs of abuse), are sufficient to drive reward-related behaviors. Additionally, it is unknown whether drugs of abuse administered directly into the PVT are sufficient to drive reward-related behaviors. Here, using behavioral as well as pathway and cell-type specif

Thalamic Local Field Potentials and Closed-Loop Deep Brain Stimulation in Orthostatic Tremor.

BACKGROUND: Orthostatic tremor (OT) is a rare movement disorder characterized by a feeling of unsteadiness and a high-frequency tremor in the legs (13-18 Hz) relieved by sitting or walking. OBJECTIVES: The aims were to study the brain electrophysiology captured chronically in a person with medication-refractory OT while standing and walking and in the semi-recumbent position using bilateral ventral intermedius nucleus deep brain stimulation (DBS) (Medtronic Percept PC) and to describe the clinical use of closed-loop DBS. METHODS: A sensing survey was used to capture baseline local field pote

Suppression of absence seizures by using different stimulations in a reduced corticothalamic-basal ganglion-pedunculopontine nucleus model.

Coupled neural network models are playing an increasingly important part in the modulation of absence seizures today. However, it is currently unclear how basal ganglia, corticothalamic network and pedunculopontine nucleus can coordinate with each other to develop a whole coupling circuit, theoretically. In addition, it is still difficult to select effective parameters of electrical stimulation on the regulation of absence seizures in clinical trials. Therefore, to develop a coupled model and reduce computation cost, a new model constructed by a simplified basal ganglion, two corticothalamic c

Do crocodiles have a zona incerta?

In mammals, the zona incerta is thought to be involved in a number of behaviors: visceral activity, arousal, attention, and posture and locomotion. These diverse and complex features suggested that the zona incerta functions as a global or integrative node. Nevertheless, despite multiple investigations into its anatomy, physiology, and behavior in a variety of mammals, no specific character identifies the zona incerta besides its appearance in fiber-stained material and its relationship to surrounding structures. One such structure is the thalamic reticular nucleus whose caudal pole often cont

Cortical control of the unit activity in nucleus medialis dorsalis thalami of the cat.

Effects of electrical stimulation of OFC (gyrus proreus, gyrus orbitalis, gyrus sigmoideus anterior) on MD unitary activity were studied. Acute preparations, curarized and unanesthetized or anesthetized with chloralose or barbiturates, were used. Steel microelectrodes were used for recording. Single rectangular pulses (0.5 msec and 0.1-0.5 ma) or short duration trains of stimuli were delivered. 479 neurons were tested and 244 (50.9 percent) responsive cells were revealed. Responses patterns: Type I, excitatory in nature. The discharge was composed of one or more spikes (latencies below 100 mse

Multi-scale structural alterations of the thalamus and basal ganglia in focal epilepsy using 7T MRI.

Focal epilepsy is characterized by repeated spontaneous seizures that originate from cortical epileptogenic zone networks (EZN). Analysis of intracerebral recordings showed that subcortical structures, and in particular the thalamus, play an important role in seizure dynamics as well, supporting their structural alterations reported in the neuroimaging literature. Nonetheless, between-patient differences in EZN localization (e.g., temporal vs. non-temporal lobe epilepsy) as well as extension (i.e., number of epileptogenic regions) might impact the magnitude as well as spatial distribution of s

Magnetic resonance imaging of the thalamic mediodorsal nucleus and pulvinar in schizophrenia and schizotypal personality disorder.

BACKGROUND: The importance of neuronal interactions in development, the cortical dependence of many thalamic nuclei, and the phenomenon of transsynaptic degeneration suggest possible abnormalities in thalamic nuclei with connections to other brain regions implicated in schizophrenia. Because frontal and temporal lobe volumes are diminished in schizophrenia, volume loss could characterize their primary thalamic relay nuclei (mediodorsal nucleus [MDN] and pulvinar). METHODS: Tracers delineated the thalamus, MDN, and pulvinar on contiguous 1.2-mm magnetic resonance images in 12 schizophrenic pat

Functional impairment due to white matter ischemia after middle cerebral artery occlusion in cats.

We recorded regional cerebral blood flow, somatosensory evoked potentials, and auditory evoked potentials in the thalamic relay nuclei (ventral posterior lateral nucleus and medial geniculate body) and in the somatosensory and auditory cortices during and after 1 hour of transient left middle cerebral artery occlusion in nine cats. Regional cerebral blood flow was also measured in the thalamocortical tracts of five of these cats. Additionally, the integrity of thalamocortical connections was tested by retrograde labeling of the thalamic nuclei with horseradish peroxidase in eight cats (three o

Dissociable spatial and non-spatial attentional deficits after circumscribed thalamic stroke.

Thalamic nuclei act as sensory, motor and cognitive relays between multiple subcortical areas and the cerebral cortex. They play a crucial role in cognitive functions such as executive functioning, memory and attention. In the acute period after thalamic stroke attentional deficits are common. The precise functional relevance of specific nuclei or vascular sub regions of the thalamus for attentional sub functions is still unclear. The theory of visual attention (TVA) allows the measurement of four independent attentional parameters (visual short term memory storage capacity (VSTM), visual perc

Preserved repetition in thalamic afasia. A pathophysiological hypothesis.

The ability to repeat words is almost always preserved in thalamic aphasia. The pathophysiology of both thalamic aphasia and preservation of repetition are not fully understood. In a case of severe aphasia with preserved repetition after a left thalamic hemorrhage, MRI disclosed left thalamic lesion and loss of fractional anisotropy in the left centrum semiovale. FDG-PET showed severe hypometabolism in the left cerebral hemisphere, except for superior and transverse temporal gyri, calcarine fissure and frontopolar regions. Primary sensory function may be less functionally dependent on thalamic

Bicuculline injections into the rostral and caudal motor thalamus of the monkey induce different types of dystonia.

The pathophysiology of dystonia remains unclear in comparison with other movement disorders. Recent data suggest that there may exist in dystonia an increased thalamic drive to the mesial premotor cortex. To test this hypothesis, we induced overactivity of the motor thalamus by injecting a GABA-A (gamma-aminobutyric acid) antagonist (bicuculline) into the rostral (pallidal) and caudal (cerebellar) ventrolateral nuclei of the thalamus in both hemispheres of one monkey. Dystonic postures were observed in the contralateral limbs and axis. Electromyographic recordings revealed bursts of muscular a

Disrupted thalamic functional hierarchy on resting-state fMRI in idiopathic generalized epilepsy.

OBJECTIVE: Abnormal thalamo-cerebral and thalamo-cerebellar interactions are implicated in idiopathic generalized epilepsy (IGE), but their influence on thalamic functional hierarchy remains unclear. This study aimed to characterize thalamic functional hierarchy across varying connectivity strengths in IGE. MATERIALS AND METHODS: Resting-state functional magnetic resonance imaging (fMRI) data were collected for 144 IGE patients and 222 healthy controls. The voxel-wise thalamo-cerebral and thalamo-cerebellar functional connectivity (FC) was first computed. Connectome gradient mapping was appli
