Novel Strategy to Assess the Neurotoxicity of Organic Solvents Such as Glycol Ethers: Protocol for Combining In Vitro and In Silico Methods With Human-Controlled Exposure Experiments.

BACKGROUND: Chemicals are not required to be tested systematically for their neurotoxic potency, although they may contribute to the development of several neurological diseases. The absence of systematic testing may be partially explained by the current Organisation for Economic Co-operation and Development (OECD) Test Guidelines, which rely on animal experiments that are expensive, laborious, and ethically debatable. Therefore, it is important to understand the risks to exposed workers and the general population exposed to domestic products. In this study, we propose a strategy to test the n

[Specific drug therapy of acute poisoning].

Antidote therapy is an important component of urgent treatment of acute poisonings. However, due to high specificity of the antidote action they can be effectively and safely used only under certain conditions. According to Ye. A. Luzhnikov's classification, 4 main groups of antidotes are distinguished: toxicotropic, toxicokinetic, symptomatic, and antitoxic immunopreparations. The use of individual antidotes, mainly symptomatic and some toxicotropic and toxicokinetic, is determined by the presence of typical clinical signs of poisoning and the anamnesis. In the rest cases the diagnosis is to

Comparative insights into the dermal absorption, distribution, and elimination of conventional and nano formulations of tebuconazole in rats.

BACKGROUND: Nanopesticides have been developed to improve the efficacy and utilization efficiency of conventional formulations; however, information on their toxicokinetics behavior following dermal exposure remains limited. Tebuconazole is a widely used triazole fungicide for which dermal exposure represents an important occupational route. This study aimed to compare the absorption, distribution, and elimination of tebuconazole in rats after dermal exposure to nano-formulations (nanoemulsion and nano-capsules) and conventional formulations (suspension concentrate and micro-capsules), providi

Development of a Physiologically Based Pharmacokinetic Model for Inorganic Mercury Salts in Rodents and Humans and Its Application in the Derivation of Health-Based Toxicity Metrics.

Inorganic mercury salts (iHg) are mercury compounds that lack carbon-hydrogen bonds. Exposure to iHg is associated with toxic effects, primarily shown in animal studies, yet the lack of cross-species toxicokinetic models has limited our ability to assess human health risks. This study aimed to develop and validate the first physiologically based pharmacokinetic (PBPK) model for iHg, incorporating species-specific physiological and chemical parameters to better characterize iHg kinetics in mice, rats, and humans. We included a hierarchical Markov chain Monte Carlo (MCMC) approach to address the

Hepatotoxic assessment in a microphysiological system: Simulation of the drug absorption and toxic process after an overdosed acetaminophen on intestinal-liver-on-chip.

To compensate the limitation of animal models, new models were proposed for drug safety evaluation to refine and reduce existing models. To mimic drug absorption and metabolism and predict toxicokinetic and toxic effects in an in vitro intestinal-liver microphysiological system (MPS), we constructed an intestinal-liver-on-chip and detected the acute liver injury process after an overdose of acetaminophen (APAP). Caco-2 and HT29-MTX-E12 cell lines were utilized to establish intestinal equivalents, along with HepG2, HUVEC-T1, and THP-1 induced by PMA and human hepatic stellate cell to establish

Acute trichloroethylene poisoning by ingestion: clinical and pharmacokinetic aspects.

Five hours after ingestion of an unknown amount of trichloroethylene (TCE) a 32-year-old woman was admitted to hospital in deep coma. The neurological condition remained unchanged for 3 days, after which there was an improvement of the central nervous system function. The concentrations of TCE in the blood, which were measured during 7 days after the solvent ingestion, gave us the opportunity to study some toxicokinetic parameters of TCE. Using a physiologically based pharmacokinetic model, the toxicokinetic parameters and blood concentrations of TCE are discussed in relation to the neurologic

Clinical and anatomic pathology effects of serial blood sampling in rat toxicology studies, using conventional or microsampling methods.

UNLABELLED: As a general practice in rodent toxicology studies, satellite animals are used for toxicokinetic determinations, because of the potential impact of serial blood sampling on toxicological endpoints. Besides toxicological and toxicokinetic determinations, blood samples obtained longitudinally from a same animal may be used for the assessment of additional parameters (e.g., metabolism, pharmacodynamics, safety biomarkers) to maximize information that can be deduced from rodents. We investigated whether removal of up to 6 × 200 μL of blood over 24h can be applied in GLP rat toxicology

Dietary compounds as modulators of metals and metalloids toxicity.

A large part of the population is exposed to metals and metalloids through the diet. Most of the in vivo studies on its toxicokinetics and toxicity are conducted by means of exposure through drinking water or by intragastric or intraperitoneal administration of aqueous standards, and therefore they do not consider the effect of the food matrix on the exposure. Numerous studies show that some components of the diet can modulate the toxicity of these food contaminants, reducing their effect on a systemic level. Part of this protective role may be due to a reduction of intestinal absorption and s

Toxicological aspects of metallothionein.

Metallothionein (MT) is expressed to a certain extent in almost all mammalian tissues. The biological significance of MT is related to its various forms MT-1, MT-2, MT-3 and MT4. For MT-1 several isoforms of the protein exist and it is likely that these isoforms are related to various functions involved in developmental processes occurring at various stages of gestation. Toxicokinetics and biochemistry of essential and toxic metals such as cadmium, zinc, mercury and copper in organs e.g. kidney, CNS, are often related to metallothionein. It is debated whether there is a relation or not for oth

Juvenile rat and pediatric trazodone studies: how to gain extra sensitivity to overcome bioanalytical challenges.

AIM: Trazodone (TZD) is used for the treatment of depression in adults and, off-label, as a sleep medication in adult and pediatric populations. The off-label use is well documented, however further clinical studies are needed to confirm its efficacy and safety for the treatment of sleep disorders. In this scenario, we developed a bioanalytical method to quantify low TZD concentrations in samples collected by capillary microsampling (CMS) to support dose finding, Good Laboratory Practice juvenile rat toxicokinetic and upcoming pediatric studies. METHODOLOGY: A method using only 8 μl of plasma

Environmental pollutants and bad bugs work hand in glove.

'Bad bacteria' could alter the toxicokinetics of environmental pollutants, thereby exacerbating chemically induced tumorigenesis. Recently, Roje et al. reported that specific gut microbiota can metabolize nitrosamine compounds to a toxic oxidation product, aggravating bladder cancer development and progression. These findings have important implications for tumor intervention through the gut microbiota.

Effect of blood microsampling (50 μL) on toxicological assessment in rats treated with tacrine, a drug known to have adverse effects that increase neutrophils.

To promote the 3Rs in toxicological assessment, the recommendation for blood microsampling in toxicokinetic evaluation is noted in the ICH Harmonized Guideline S3A Q&As. However, there are only a few articles reporting the practical application of microsampling in the toxicological assessment with toxic drugs. In this study, we investigate the effect of microsampling on toxicological assessment in rats treated with tacrine, which is known to have toxic effects that induce an increase in neutrophils and behavioral abnormalities. Thirty female Sprague-Dawley rats were divided into microsampling

Separating the Role of Gut Enzymatic Transformation in Modulating Internal Exposure to Three Major Phthalates.

The gastrointestinal tract (GIT) is crucial in the absorption and metabolism of xenobiotics, including phthalates─widespread environmental contaminants associated with various health risks. Estimating human exposure to phthalates via biomonitoring is challenging due to their complex metabolic pathways, resulting in a mass-balance gap between internal and external exposure. The relative contributions of the GIT and liver to phthalate metabolism remain underexplored. This study investigated the metabolism of three representative phthalate diesters─dibutyl phthalate (DBP), di(2-ethylhexyl)phthala

[Interactions between alcohol and work exposure to chemical substances].

BACKGROUND AND OBJECTIVES: The quite diffused habit of a significant assumption of alcohol drinks, can interfere with the professional exposure to chemical substances. The interaction may result in increasing their toxicity and/or modifying the parameters of the biological monitoring. It may also act as a confounding factor, not only in epidemiologic researches but also at individual level when the assessment of the occupational exposure and/or the diagnosis of an occupational diseases, is under consideration. We review available references in the literature summarizing major scientific eviden

Integrating variability in half-lives and dietary intakes to predict mercury concentration in hair.

Methylmercury (MeHg) is a bioaccumulative environmental toxin that exerts its effect on fetal and infant neurodevelopment. Mercury concentration in hair is a good biomarker of MeHg accumulation in the body, with seafood being the main source of MeHg in humans. Therefore, modeling the link between food intake and mercury concentration in hair is a key step in assessing the risk of MeHg exposure. Using repeated measurements of diet and mercury concentration in hair, we studied 125 French pregnant women who consumed seafood (e.g., fish, mollusks and crustaceans) and compared their individual esti

From exposure models to multi-organ toxicity of 6PPD-quinone in mice: A mini-review.

Emerging evidence positions 6PPD-quinone (6PPD-Q) as a global environmental contaminant, with confirmed toxicity to various species and widespread detection in human biological samples. Despite these observations, toxicological understanding of 6PPD-Q in mammals remains fragmented and inconsistent. Therefore, this review integrates current evidence from murine models within an "exposure-toxicokinetics-organ toxicity" framework, revealing exposure route-dependent toxicokinetics and toxicological outcomes. Moreover, 6PPD-Q perturbs lipid metabolism, mitochondrial function, and immune homeostasis

Incorporation of the glutathione conjugation pathway in an updated physiologically-based pharmacokinetic model for perchloroethylene in mice.

BACKGROUND: Perchloroethylene (perc) induced target organ toxicity has been associated with tissue-specific metabolic pathways. Previous physiologically-based pharmacokinetic (PBPK) modeling of perc accurately predicted oxidative metabolites but suggested the need to better characterize glutathione (GSH) conjugation as well as toxicokinetic uncertainty and variability. OBJECTIVES: We updated the previously published "harmonized" perc PBPK model in mice to better characterize GSH conjugation metabolism as well as the uncertainty and variability of perc toxicokinetics. METHODS: The updated PBP
