Lipidomic changes in the liver of beagle dogs associated with Toxocara canis infection.

A global lipidomic analysis using liquid chromatography-tandem mass spectrometry was performed on the liver of beagle dogs infected with Toxocara canis to profile hepatic lipid species at 12 h post-infection (hpi), 24 hpi, and 36 days post-infection (dpi). This analysis identified six categories and 42 subclasses of lipids, including 173, 64, and 116 differentially abundant lipid species at 12 hpi, 24 hpi, and 36 dpi, respectively. Many of the identified lysophospholipids, such as lysophosphatidylglycerol, lysophosphatidylserine, and lysophosphatidylcholine, may contribute to the migration and

Socioeconomic factors associated with infection by Toxoplasma gondii and Toxocara canis in children.

The intense contact of children with domestic animals or environments contaminated with faeces of these animals, together with habits related to lack of hygiene, can facilitate infection by zoonoses. The study evaluated the seroprevalence and risk factors associated with toxoplasmosis and toxocariasis in schoolchildren in the city of Jataizinho, Paraná. Of the 412 children aged 4-15 years, 56.8% (234/412) presented antibodies reactive to Toxoplasma gondii, 42.5% (175/412) presented antibodies reactive to Toxocara canis, and 27.4% (113/412) were reactive for the two species. The analysis of ris

Saccharomyces boulardii reduces the vertical transmission of Toxocara canis larvae in mice.

Probiotics have been shown to reduce the intensity of Toxocara canis infection in mice. However, larval transmission of this nematode also occurs via transplacental and transmammary routes. Thus, the aim of this study was to evaluate the effect of the Saccharomyces boulardii probiotic on the vertical transmission of T. canis in Swiss mice. The mice received 107S. boulardii colony-forming units per gram of food. The supplementation began 15 days before mating and was maintained throughout pregnancy and lactation. The animals were inoculated with 300 T. canis embryonated eggs on the 14th day of

Seropositivity and risk factors of Toxocara canis infection in adult asthmatic patients.

This cross-sectional study involving 86 adult asthmatic patients aimed to determine the relationship between Toxocara seropositivity and severity of asthma in adult asthmatics and investigate the risk factors for Toxocara infection. In all cases, T. canis IgG level was measured using an anti-Toxocara IgG enzyme-linked immunosorbent assay kit. Total serum IgE and eosinophil count were also determined. The anti-Toxocara IgG seropositivity was 68.6% among asthmatic patients. There were no statistically significant associations between Toxocara seroprevalence and other risk factors, clinical sympt

Antigen detection: Insights into Toxocara and other ascarid infections in dogs and cats.

Detection of ascarid excreted or secreted (E/S) molecules is an alternative approach to the identification of infection by egg flotation. E/S molecules serve as direct markers for the ascarid nematode commonly found in cats and dogs (Toxocara spp., Toxascaris leonina and Baylisascaris procyonis). The nematode derived E/S material mixes with the intestinal contents of the host animal and is available for detection as a coproantigen in the host's faeces. Antigen capture immunological techniques allow sensitive coproantigen detection. Different patterns of antigen to egg agreement are demonstrate

Proteomic alterations in the plasma of Beagle dogs induced by Toxocara canis infection.

Toxocara canis causes ocular larva migrans and visceral larva migrans in humans. Knowledge about the molecular mechanism of T. canis-hosts interaction is limited. The proteomic alterations in the plasma of Beagle dogs induced by T. canis infection were studied by the quantitative mass spectrometry-based data-independent acquisition (DIA). 418, 414 and 411 plasma proteins were identified at 24 h post-infection (hpi), 96 hpi and 36 days post-infection (dpi), including 6, 5 and 23 proteins with differential abundance, respectively. At 24 hpi, the altered proteins, retinoic acid receptor responder

Imaging of Toxocara canis larvae labelled by CFSE in BALB/c mice.

Mice are used most often as a model for human toxocariasis caused by Toxocara canis larvae. Variety of symptoms developing during the infection reflects behaviour of the larvae, which are able to escape from the intestine and further invade and damage various host organs. In order to find an approach enabling observation on parasite behaviour in mouse in vivo, we used an epifluorescence method and a small animal imaging system (SAIS). Larvae of T. canis were labelled by carboxyfluorescein succinimidyl ester (CFSE) which incorporated on the parasite gastrointestinal tract. Following infection o

Effects of disinfectants on Toxocara canis embryogenesis and larval establishment in mice tissues.

The objectives of this study were (i) to evaluate the effects of several different disinfectant solutions on embryonic development of Toxocara canis eggs and (ii) to investigate the potential infectivity of exposed eggs by assessing larval establishment in various tissues in a murine model. All the disinfectants tested were products routinely used in veterinary clinics, kennels, animal shelters and laboratories. Ova were obtained from gravid female T. canis uteri. Thirty samples containing 10,000 eggs were divided into five groups of six identical sample tubes per group. The treatments for the

In vivo evaluation of albendazole microspheres for the treatment of Toxocara canis larva migrans.

Albendazole is a benzimidazole derivative with proven efficacy against many parasites such as intestinal helminths. Toxocariasis is one of the important parasitic diseases in humans and animals caused by Toxocara canis. It is well known that T. canis larvae migrate in paratenic hosts, including humans where it may cause visceral larva migrans. Thus, the present research was carried out using in vivo experiments with the aim of finding whether novel albendazole microparticles would be active against migrating larvae of the parasite. Albendazole-chitosan microparticles were prepared by ionotropi

A simple and inexpensive in vitro method for retrieving fertilized Toxocara canis eggs.

The erratic migration of nematode larvae is potentially deleterious for humans, especially those produced by Toxocara roundworms. Diagnosis of human toxocariosis: visceral larva migrans depends on antibody detection against excretions-secretions (E/S) from Toxocara larvae by enzyme-linked immunosorbent assay. In the present work, we wish to propose a simple and cheap culture procedure for keeping T. canis female worms alive. We removed living worms from the small intestine of euthanized puppies at the Canine Control Centre "Culhuacan" in the Southern part of Mexico City. To ascertain the utili

The high prevalence of asymptomatic Toxocara infection among schoolchildren in Manado, Indonesia.

We performed a serological survey of Toxocara canis infection in junior high school students from three districts in northern Sulawesi. Almost all of the 117 subjects from two rural districts near Manado allowed dogs in their houses, and there was an 84.6% prevalence of T. canis infection in this group. Fifty-three subjects (45.3%) had serum samples with a high titer of specific anti-Toxocara antibody. By contrast, 41 students tested in one urban district showed a 12.2% prevalence. To confirm the clinical symptoms of visceral larva migrans (VML) and ocular larva migrans (OLM) caused by Toxocar

Prevalence of Toxocara spp. eggs in the soil of public parks in Ankara, Turkey.

One hundred and seventy soil samples from forty-six public parks in Ankara area were examined to determine the level of contamination with Toxocara spp. eggs. 30.6% of the 170 soil samples were contaminated with Toxocara spp. eggs. The number of eggs in the positive soil samples varied from 1 to 10. A high proportion of the eggs was fully embryonated. The findings demonstrate the common occurrence of these eggs in locations likely to be important in the transmission of visceral larva migrans. Of 19 fecal collected, 5 (26.3%) contained Toxocara spp. eggs. Eggs of Ancylostomidae, Toxascaris leon

Eosinophil peroxidase levels in hearts and lungs of mice infected with Toxocara canis.

Toxocara canis infection of abnormal hosts results in a condition in which infective larvae migrate through the soft tissues of the body, exclusive of the skin. This condition is known as visceral larva migrans (VLM) and causes a syndrome characterized by hepatosplenomegaly, hyperglobulinemia, hypereosinophilia, and transient pulmonary infiltrates. Because of the known association between hypereosinophilia and eosinophilic heart disease, we have been studying the hearts of mice infected with T. canis for evidence of myocardial damage and have previously described a severe eosinophilic myocardi

Pathological and haematological responses of cats experimentally infected with Toxocara canis larvae.

Responses of eight adult cats to one or two infections with larvae of Toxocara canis were studied up to 39 days post infection (DPI). Clinically, all cats remained normal throughout the study. The major necropsy finding was multifocal, white to grey nodules mainly within the liver, lungs and kidneys; live larvae were found in liver nodules. Histologically, the nodules were eosinophilic granulomas. Granulomas containing a larval section were observed mainly within the liver. All infected cats had variably severe, eosinophilic arteritis and bronchiolitis and medial hypertrophy and hyperplasia of

The extraparasitic life cycle of Toxocara vitulorum in the village environment of Sri Lanka.

The extraparasitic life cycle of Toxocara vitulorum of buffalo in the villages of Sri Lanka is related to observations on buffalo behaviour, experimental studies on the development and persistence of the eggs in soil and in wallows and the presence of eggs in village locations. Calf faeces on soil were rapidly incorporated by insect activity and the eggs developed only slightly slower than in the laboratory. Some infective eggs persisted 3-4 cm deep for 17 months, finally dying during a prolonged hot, dry period. Eggs in a wallow developed intermittently over 16 months as it was flushed with r
