Could β-glucans enhance the immune response to SARS-CoV-2 vaccine by inducing trained immunity and boosting neutralizing antibody production?

β-Glucans stimulate the immune system, training it to recognize and respond to antigens, which bolsters immunity, including to vaccines. The present study evaluated the capacity of β-glucans to stimulate immunity subsequent to primary vaccination for SARS-CoV-2 with ChAdOx1. Thirty-four SARS-CoV-2-non-immune men (18-49 years) were split into two groups: one receiving 500 mg of oral insoluble yeast β-glucans daily (the glucan group) and the other a placebo (the control group). The supplementation period lasted fourteen days in total, including seven days before and seven days after the initial

Trained immunity as a systemic bridge: the liver-gut-immune-oral axis in the comorbidity of chronic liver disease and periodontitis.

Trained immunity (TI) reveals that innate immune cells acquire long-term functional memory through metabolic and epigenetic reprogramming. This review examines TI in chronic liver diseases and periodontitis, proposing the "Liver-Gut-Immune-Oral Axis" as a framework where TI bridges these comorbidities. The forward pathway, currently inferred from mechanistic and associative studies, proposes that liver-gut dysfunction induces bone marrow training, generating hyper-reactive monocytes that amplify periodontal inflammation. The reverse pathway, similarly conceptual, proposes that periodontal path

Evaluation of trained immunity as a potential strategy for a universal Streptococcus suis vaccine using immunogenic proteins in a murine model.

BACKGROUND: Universal vaccine against Streptococcus suis (S. suis) remains a challenge due to the high number of serotypes/strains, the absence of cross-protection among them and the existence of different immune escape strategies. In this study, the immunomodulatory potential of live and inactivated porcine derived Bacillus Calmette-Guérin (dpB) in combination with specific immunogenic proteins S. suis is evaluated in murine model. RESULTS: Results revealed different immune responses depending on dpB formulation. Live-dpB administered intravenously induced consistent results with trained imm

Spatiotemporal immune gradients in gout: immune response-driven activation of the NLRP3-IL-1β axis and its transition to trained immunity.

Gout is a crystal-associated autoinflammatory disease triggered by monosodium urate (MSU) crystals, clinically characterized by recurrent transitions between acute inflammatory flares dominated by innate immunity and a state of "trained immunity" during the remission phase. However, previous studies have mostly focused on single time points or local lesions. Such approaches fail to systematically explain the recurrent nature of acute gout flares and the mechanisms underlying multi-system involvement. By integrating evidence from single-cell and spatial transcriptomics as well as mechanistic in

An Rv1471-expressing chimpanzee adenovirus vaccine confers protection against tuberculosis by inducing alveolar macrophage trained immunity and polyfunctional T-cell responses.

The limited protection afforded by Bacille Calmette-Guérin (BCG) against pulmonary tuberculosis (TB) underscores the critical need for novel vaccine strategies. Alveolar macrophages (AMs), as the primary sentinel cells encountering inhaled Mycobacterium tuberculosis (Mtb), play a decisive role in early infection outcomes, yet their potential as a direct vaccine target remains largely untapped. Here, we developed a chimpanzee adenovirus vaccine expressing the Mtb antigen Rv1471 (rAd-Rv1471), which we previously identified for its unique capacity to induce innate immune memory. In murine models,

"Trained Immunity" from Mycobacterium spp. exposure (BCG vaccination and environmental) may have an impact on the incidence of early childhood leukemia.

Preventive variables for childhood leukemia incidence (LI) remain unknown. Past assertions that childhood vaccinations, especially BCG, may be potentially protective have remained disputed for over five decades because of the lack of a unifying framework to explain variable outcomes in different studies. An examination of the early childhood LI for 2020 in European Region countries with supposedly similar underlying confounders but differential childhood vaccination coverage displays negative covariation with prevailing Mycobacterium spp. exposure in BCG-vaccinated children. The childhood LI i

A Combination of Polybacterial MV140 and Candida albicans V132 as a Potential Novel Trained Immunity-Based Vaccine for Genitourinary Tract Infections.

Recurrent urinary tract infections (RUTIs) and recurrent vulvovaginal candidiasis (RVVCs) represent major healthcare problems with high socio-economic impact worldwide. Antibiotic and antifungal prophylaxis remain the gold standard treatments for RUTIs and RVVCs, contributing to the massive rise of antimicrobial resistance, microbiota alterations and co-infections. Therefore, the development of novel vaccine strategies for these infections are sorely needed. The sublingual heat-inactivated polyvalent bacterial vaccine MV140 shows clinical efficacy for the prevention of RUTIs and promotes Th1/T

Trained immunity as a novel approach against COVID-19 with a focus on Bacillus Calmette-Guérin vaccine: mechanisms, challenges and perspectives.

COVID-19 is a severe health problem in many countries and has altered day-to-day life in the whole world. This infection is caused by the SARS-CoV-2 virus, and depending on age, sex and health status of the patient, it can present with variety of clinical symptoms such as mild infection, a very severe form or even asymptomatic course of the disease. Similarly to other viruses, innate immune response plays a vital role in protection against COVID-19. However, dysregulation of innate immunity could have a significant influence on the severity of the disease. Despite various efforts, there is no

Innate immunity and training to subvert original antigenic sin by the humoral immune response.

Originally defined in the context of influenza vaccines by Thomas Francis Jr. in the late 1950s, original antigenic sin (OAS) refers to the tendency of the immune system to preferentially recall B cell memory against primary antigen after secondary exposure to different but related antigen. This competes with the elicitation of de novo antibodies by lowering the frequency of antigen reception by the naïve B cell lymphocyte pool residing within secondary lymphoid organs. Consequently, OAS imposes a 'primary addiction' that modulates the target epitope specificity of the secondary antibody respo

A proactive immune training strategy to generate anticipatory immunity against viral evasion.

INTRODUCTION: RNA viruses rapidly evolve to evade host immunity, posing a significant challenge to conventional vaccines. Natural infection, however, provides a blueprint for broad immunity by training the immune system against a diverse antigenic landscape. This study addresses the problem of creating a vaccine that mimics this complex training process. OBJECTIVES: We aimed to develop and validate a novel vaccination paradigm, "Proactive Immune Training" (PIT), designed to enhance cross-reactive neutralization breadth against variants within the predicted evolutionary space. METHODS: Using

Trained immunity-based mucosal immunotherapies for preventing respiratory infections: a translational, evidence-graded appraisal of platforms, populations, and the protection-immunopathology balance.

Conventional respiratory vaccines protect through antigen-specific immunity, leaving broad gaps against antigenically diverse and emerging pathogens and inducing little protection at the airway surface itself. Trained immunity, the durable epigenetic and metabolic reprogramming of innate immune cells and their bone marrow progenitors, offers antigen-agnostic, heterologous protection and can be induced directly at the respiratory mucosa, providing a mechanistic rationale for a new class of prophylactic immunotherapies. Here we appraise this field from a clinical-translational, evidence-graded s

Influenza vaccination post-COVID-19 expands vaccine-specific effector CD4 T-cells and Tregs under positive influence of host trained innate immunity.

SARS-CoV-2 immunity and innate immune training may influence influenza vaccine immunogenicity. We investigated this in India. Adult volunteers with hybrid SARS-CoV-2 immunity were administered FluarixTM Tetra (GlaxoSmithKlein) 2022/2023 NH Vaccine in 2022. Significant induction of hemagglutinin inhibition-specific antibodies and polyfunctional central memory CD4+ T-cells (TCM) were observed 1-week post-vaccination with variable induction of CD8+T-cell and innate effectors. Vaccination also expanded Flu-specific regulatory T-cells (Treg), which negatively correlated with CD4 responses, highligh

Outcomes and Lessons Learned From Immunization in Practice Training.

Background The Kurdistan Region of Iraq (KRI) faces persistent challenges in immunization service delivery due to workforce shortages, limited training opportunities, and unequal resource distribution. Most vaccinators lack formal training and rely on informal mentorship, often resulting in knowledge and skill gaps. In response, the Kurdistan Region Ministry of Health (KMOH), with support from the United Nations Children's Fund (UNICEF), implemented training sessions based on the World Health Organization (WHO) Immunization in Practice (IIP) guide to enhance vaccinators' competencies. Objectiv

Changes in Immune Function during Initial Military Training.

PURPOSE: Initial military training (IMT) is a transitionary period wherein immune function may be suppressed and infection risk heightened due to physical and psychological stress, communal living, and sleep deprivation. This study characterized changes in biomarkers of innate and adaptive immune function, and potential modulators of those changes, in military recruits during IMT. METHODS: Peripheral leukocyte distribution and mitogen-stimulated cytokine profiles were measured in fasted blood samples, Epstein-Barr (EBV), varicella zoster (VZV), and herpes simplex 1 (HSV1) DNA was measured in

Bacillus Calmette-Guérin vaccination induces a trained innate immunity phenotype in adults over 50 years of age: A randomized trial in Guinea-Bissau.

BACKGROUND: The beneficial effects of Bacillus Calmette-Guérin (BCG) as an intervention against non-mycobacterial infections have been extensively studied in randomized trials. These non-specific effects have been linked to a heterologous increase of pro-inflammatory cytokine production by innate immune cells. It is unknown if BCG induces such responses in older individuals from TB-endemic countries. METHODS: In a single-blinded trial in Guinea-Bissau, 40 adults over 50 years of age were randomized 1:1 in a block of 40 to intradermal injection of BCG-Japan (intervention) or solvent (placebo).

[BCG, muramylpeptides, trained immunity (part II): a low molecular weight alternative to multicomponent bacterial immunostimulants for prevention of respiratory infections during a pandemic].

During a pandemic, nonspecific immunoprophylaxis of SARS-CoV-2 infection and other acute respiratory infections (ARI), which can worsen the course of COVID-19, is increasingly in demand in addition to specific immunization. BCG vaccine appears to be one of the candidate immunostimulants in this regard. At the same time, other microbe-derived preparations capable of inducing a state of trained immunity deserve attention. BCG and other bacterial immunostimulatory agents containing a large number of biologically active subunits have long been considered as objects of search for promising pharmaco

Trained Innate Immunity Induced by Vaccination with Low-Virulence Candida Species Mediates Protection against Several Forms of Fungal Sepsis via Ly6G+ Gr-1+ Leukocytes.

We recently discovered a novel form of trained innate immunity (TII) induced by low-virulence Candida species (i.e., Candida dubliniensis) that protects against lethal fungal/bacterial infection. Mice vaccinated by intraperitoneal (i.p.) inoculation are protected against lethal sepsis following Candida albicans/Staphylococcus aureus (Ca/Sa) intra-abdominal infection (IAI) or Ca bloodstream infection (BSI). The protection against IAI is mediated by long-lived Gr-1+ leukocytes as putative myeloid-derived suppressor cells (MDSCs) and not by prototypical trained macrophages. This study aimed to de

Heat-killed Mycobacterium tuberculosis-induced trained immunity elicits antitumor activity in PBMCs from colon cancer patients with unresponsiveness to anti-PD-1 in vitro.

INTRODUCTION: Trained immunity (TI) is a form of innate immune memory characterized by boosted innate immune responses to secondary heterologous challenges following an initial stimulus. Several inducers, including fungal β-glucan and the Bacillus Calmette-Guérin (BCG) vaccine, induce TI in human peripheral blood mononuclear cells (PBMCs). Although the potential of TI in colon cancer has been explored in murine models, its relevance in patients remains poorly understood. METHODS: Building on our previous demonstration that heat-killed Mycobacterium tuberculosis (HKMtb) induces TI, we applied

The promoting effect of high-fat diet-induced trained immunity on ulcerative colitis and the mechanism of the therapeutic effect of Dahuang Mudan Decoction.

BACKGROUND: Ulcerative colitis (UC) is a chronic inflammatory disease driven by intestinal immune imbalance. In addition, its pathogenesis includes environment, genetics, microbiota, genes, and diet. Epidemiological and related studies have shown that a high-fat diet habit can promote the progression of UC. The trained immunity induced by a high-fat diet is a key factor in the occurrence and development of chronic inflammatory diseases. As a chronic inflammatory disease, the research on how the trained immunity induced by a high-fat diet promotes the progression of UC is still unclear. Previou
