Community-acquired pneumonia is an infection of the lower respiratory tract caused by various viral and bacterial pathogens, including influenza A virus (IAV), Streptococcus pneumoniae, and Staphylococcus aureus. To understand the disease pathology, it is important to delineate host metabolic responses to an infection. In this study, metabolome profiling of mono- and coinfected human bronchial epithelial cells was performed. We show that IAV and S. aureus silently survive within the cells with almost negligible effects on the host metabolome. In contrast, S. pneumoniae significantly altered va

Interest has grown in the use of reactive minerals for natural and engineered transformation of ground water contaminants. This study investigated how the structural properties of 10 model compounds representing natural organic matter (NOM) influenced their adsorption to chloride green rust (GR-Cl), and how this adsorption affected rate constants for transformation of carbon tetrachloride (CT) by GR-Cl. The affinity of benzoic acid, phthalic acid, trimesic acid, pyromellitic acid, and mellitic acid for the GR-Cl surface generally increased in the order of increasing number of carboxylic acid f

The influence of some aliphatic alcohols on oxygen uptake, carbon dioxide production, acid formation and lactate and pyruvate concentrations of rat liver slices was studied. At the concentrations used, none of the alcohols was found to influence oxygen uptake. Of the alcohols that are not oxidized by liver alcohol dehydrogenase, methanol increased carbon dioxide production, propan-2-ol decreased it and 2-methylpropan-2-ol was without influence. All the alcohols that are oxidized by the enzyme strongly decreased carbon dioxide production. The alcohols that are not oxidized had no marked effect

Activation of group III metabotropic glutamate (mGlu) receptors has been recently highlighted as a potential approach in the treatment of Parkinson's disease (PD). This study evaluates the antiparkinsonian action of systemic administration of the broad-spectrum agonist of group III mGlu receptors, 1-aminocyclopentane-1,3,4-tricarboxylic acid (ACPT-I), and investigates its site of action within the basal ganglia circuitry. Acute injection of ACPT-I reverses haloperidol-induced catalepsy, an index of akinesia in rodents. In a rat model of early PD based on partial bilateral nigrostriatal lesions

In this study, the impact of salicylic acid (SA), a prevalent non-steroidal anti-inflammatory drug metabolite, on microalgal-bacterial granular sludge (MBGS) systems was systematically investigated. Results demonstrated that the MBGS system exhibited high pollutant removal efficiency in both synthetic and real wastewater containing 1 mg/L SA, achieving 81.6 % organics, 72.3 % nitrogen, and 77.1 % phosphorus removal. This performance was attributed to increased extracellular polymeric substance secretion and the proliferation of stress-tolerant Proteobacteria, Cyanobacteria and Bacteroidota, wh

Although numerous studies have reported the involvement of glutamate in plant abiotic stress, relatively little is known about the role of glutamate in plant defence against pathogens. To gain further knowledge, we investigated the effect of glutamate on Alternaria alternata in tomato fruit. A multidisciplinary approach was pursued, combining exogenous glutamate applications, enzymatic activity measurements and real-time quantitative PCR analysis. The results showed glutamate significantly reduced the disease incidence in tomato caused by A. alternata, by inducing resistance (Duncan's test, p<

SUCLA2 is one of several nuclear-encoded genes that can cause encephalomyopathy accompanied by mitochondrial DNA depletion. The disorder usually manifests in early childhood and leads to early death. The gene encodes one of the subunits of succinyl-CoA synthase, the enzyme that catalyzes the reversible conversion of substrates succinyl-CoA and ADP to products succinate and ATP in the tricarboxylic acid pathway. Thirty-two individuals harboring mutations in SUCLA2 have so far been reported, and five different mutations were observed among these individuals. Here we report identification of a no

Acylglycerol kinase (AGK) is a mitochondrial lipid kinase that catalyzes the phosphorylation of monoacylglycerol and diacylglycerol to lysophosphatidic acid and phosphatidic acid, respectively. Mutations in AGK cause Sengers syndrome, which is characterized by congenital cataracts, hypertrophic cardiomyopathy, skeletal myopathy, exercise intolerance, and lactic acidosis. Here we identified AGK as a subunit of the mitochondrial TIM22 protein import complex. We show that AGK functions in a kinase-independent manner to maintain the integrity of the TIM22 complex, where it facilitates the import a

Secondary-treated municipal wastewater (MWW) could supply a viable alternative water resource for cooling water systems. Inorganic salts in the concentrated cooling water pose a great challenge to corrosion control chemicals. In this study, the inhibition effect of 1-hydroxy ethylidene-1,1-diphosphonic acid (HEDP), trimethylene phosphonic acid (ATMP) and 2-phosphonobutane-1,2,4-tricarboxylic acid (PBTCA) on corrosion of carbon steel in secondary-treated MWW was investigated by the means of potentiodynamic polarization and electrochemical impedance spectroscopy. The inhibition effect increased

A 74-year-old woman with type 2 diabetes mellitus developed ketoacidosis within six days of adding metformin to imeglimin treatment. The patient was insulin-sensitive and showed preserved insulin secretion; therefore, insulin insufficiency alone was unlikely to contribute to the development of ketoacidosis. Both imeglimin and metformin partially inhibit complex I in the mitochondrial respiratory chain. Inhibition of mitochondrial respiration can lead to tricarboxylic acid (TCA) cycle suppression. Thus, the entry of acetyl-coenzyme A into TCA cycle is restricted, and it is eventually used in ke

The mechanisms responsible for heart failure in single-ventricle congenital heart disease are unknown. Using explanted heart tissue, we showed that failing single-ventricle hearts have dysregulated metabolic pathways, impaired mitochondrial function, decreased activity of carnitine palmitoyltransferase activity, and altered functioning of the tricarboxylic acid cycle. Interestingly, nonfailing single-ventricle hearts demonstrated an intermediate metabolic phenotype suggesting that they are vulnerable to development of heart failure in the future. Mitochondrial targeted therapies and treatments

A chemical, aurine tricarboxylic acid was used to identify aluminum in bone in a group of chronic renal failure patients with severe osteomalacia. Thirteen out of 49 (26%) patients had a positive stain at the bone-osteoid junction. The presence of aluminum at the staining site was confirmed by electron dispersion x-ray analysis. The patients' biopsies staining negative with this clinical, although having high bone aluminum levels, had more significant features of hyperparathyroidism, both biochemically and histologically than those having a positive stain. The usefulness of this technique to d

Inflammatory diseases are pathological conditions precipitated by inflammation, with the gut microbiota and its metabolites playing a pivotal role in the inflammatory process. Succinate, a key metabolite generated by gut microorganisms, is implicated not only in the energy metabolism of the tricarboxylic acid cycle but also in the modulation of inflammatory processes via the succinate receptor 1-mediated immune regulatory pathway. This review provides a unified elaboration on the metabolic pathways and biological characteristics of succinate, the detection techniques for succinate, and the pot

Hepatitis B virus (HBV) establishes chronic infection through strategic manipulation of host metabolic networks, driving a spectrum of hepatic pathologies ranging from hepatitis to cirrhosis and hepatocellular carcinoma. Mechanistically, HBV reprograms core metabolic pathways, including glycolysis, tricarboxylic acid (TCA) cycle, oxidative phosphorylation, and lipid homeostasis, to fuel its replication machinery and evade immune surveillance. This review systematically synthesizes current evidence on HBV-induced glucose/lipid metabolic rewiring, with particular emphasis on how viral-host cross

Arsenic is either notorious toxicant or miracle cure for acute promyelocytic leukemia and several other diseases. It interacts with mitochondria directly or indirectly, by interacting with mitochondrial enzymes, such as respiratory chain complexes and tricarboxylic acid cycle proteins, or affecting mitochondrial homeostasis via ROS or mitochondrial outer membrane permeabilization. Given the ubiquitous presence of mitochondria and indispensable role in cellular metabolism, arsenical-mitochondrial interactions may manifest clinical importance by revealing mechanism of disease curation, preventin

Palmitic acid-induced lipotoxicity contributes to the development of nonalcoholic fatty liver disease (NAFLD). Hesperetin has been reported to alleviate oxidative stress, inflammation, and cell death in NAFLD, while its potential to mitigate palmitic acid-induced lipotoxicity remains unexplored. This study investigates the protective effects of hesperetin on palmitic-acid-stimulated lipotoxicity and elucidates the underlying molecular mechanisms. Our results showed that hesperetin decreased palmitic acid-activated lipotoxicity through inhibition of the intrinsic apoptosis pathway and promotion

UNLABELLED: Mosquito larvae rely on a living microbiota for normal development because the microbiota supplies essential nutrients, particularly vitamins. Beyond vitamin provision, transcriptomic data suggest that the microbiota also supports other key nutritional processes. Here, we explored these roles by conducting a metabolomics analysis on Aedes aegypti third instar larvae following microbiota depletion. We sampled larvae and dissected guts 12 and 20 h post-decolonization, and analysed methanol-soluble metabolites using untargeted gas chromatography-mass spectrometry. Our findings reveal

Zygosaccharomyces bailii is the most tolerant yeast species to acetic acid-induced toxicity, being able to grow in the presence of concentrations of this food preservative close to the legal limits. For this reason, Z. bailii is the most important microbial contaminant of acidic food products but the mechanisms behind this intrinsic resistance to acetic acid are very poorly characterized. To gain insights into the adaptive response and tolerance to acetic acid in Z. bailii, we explored an expression proteomics approach, based on quantitative 2DE, to identify alterations occurring in the protei
