The "Gredouno" Cross Target: a new tool adapted to control Glossina palpalis gambiensis in the mangrove forests of Guinea.

BACKGROUND: In the mangroves of Guinea, where the most active foci of human African trypanosomiasis in West Africa are located, vector control against tsetse flies using insecticide-impregnated Tiny Targets was first introduced in 2012. While annual deployments of Tiny Targets have resulted in an overall 90% reduction in tsetse fly densities in control areas, managing tsetse densities in specific biotopes such as mangrove channels, which are susceptible to significant climatic disturbances, presents greater challenges. Thus, a new three-dimensional model called the Cross Target was designed to

Insights into trypanosomiasis transmission: Age, infection rates, and bloodmeal analysis of Glossina fuscipes fuscipes in N.W. Uganda.

BACKGROUND: Tsetse flies (Glossina) transmit species of Trypanosoma which cause human African trypanosomiasis (HAT) and animal African trypanosomiasis (AAT). Understanding the epidemiology of this disease and controlling the vector rationally requires analysis of the abundance, age structure, infection rates and feeding patterns of tsetse populations. METHODS: We analysed a population of G. fuscipes fuscipes in the Koboko district of Uganda. Seasonal variation in the abundance of tsetse was assessed from the numbers of tsetse caught in pyramidal traps. The age structure of the population was

Insecticide and Repellent Mixture Pour-On Protects Cattle against Animal Trypanosomosis.

BACKGROUND: African animal trypanosomosis (AAT), transmitted by tsetse flies and tick-borne diseases are the main constraints to livestock production in sub-Saharan Africa. Vector control methods such as pour-on offer individual protection against ticks but not against tsetse so far, for which protection has always been communal, through a reduction of their density. The latter requires the treatment of a large part of the herd in a given landscape and is not instantaneous. METHODOLOGY/PRINCIPAL FINDINGS: Two prospective surveys were conducted to evaluate the efficacy and persistence of a pou

Creating complete life histories of individual female tsetse (Glossina spp) to study the effects of meteorological conditions on fly size in Zimbabwe.

Using novel methodologies and ovarian dissection, we constructed life histories for ~90,000 female Glossina pallidipes and G. m. morsitans collected in Zimbabwe (1988-1999). Temperature-dependent development rates were used to estimate, for each fly, the dates spent as an oocyte, egg, larva, pupa - and adult in each ovarian category. This enabled analysis of relationships between wing and egg lengths of field-captured adults - and mean environmental conditions prevailing when these lengths were being determined, during oogenesis for eggs, and oogenesis plus pregnancy for wings. Lengths all osc

The population genetics of Trypanosoma brucei and the origin of human infectivity.

The African trypanosome, Trypanosoma brucei, is a zoonotic parasite transmitted by tsetse flies. Two of the three subspecies, T. brucei gambiense and T.b. rhodesiense, cause sleeping sickness in humans whereas the third subspecies, T.b. brucei, is not infective to humans. We propose that the key to understanding genetic relationships within this species is the analysis of gene flow to determine the importance of genetic exchange within populations and the relatedness of populations. T.brucei parasites undergo genetic exchange when present in infections of mixed genotypes in tsetse flies in the

Quantifying Heterogeneity in Host-Vector Contact: Tsetse (Glossina swynnertoni and G. pallidipes) Host Choice in Serengeti National Park, Tanzania.

BACKGROUND: Identifying hosts of blood-feeding insect vectors is crucial in understanding their role in disease transmission. Rhodesian human African trypanosomiasis (rHAT), also known as acute sleeping sickness is caused by Trypanosoma brucei rhodesiense and transmitted by tsetse flies. The disease is commonly associated with wilderness areas of east and southern Africa. Such areas hold a diverse range of species which form communities of hosts for disease maintenance. The relative importance of different wildlife hosts remains unclear. This study quantified tsetse feeding preferences in a wi

Trypanosoma brucei gambiense group 2 experimental in vivo life cycle: from procyclic to bloodstream form.

Trypanosoma brucei gambiense (Tbg) group 2 is a subgroup of trypanosomes able to infect humans and is found in West and Central Africa. Unlike other agents causing sleeping sickness, such as Tbg group 1 and Trypanosoma brucei rhodesiense, Tbg2 lacks the typical molecular markers associated with resistance to human serum. Only 36 strains of Tbg2 have been documented, and therefore, very limited research has been conducted despite their zoonotic nature. Some of these strains are only available in their procyclic form, which hinders human serum resistance assays and mechanistic studies. Furthermo

Epidemiological investigation of trypanosomosis in livestock and distribution of vector in Dabo Hana district, Southwest Oromia, Ethiopia.

The trypanosomosis remains unresolved due to its impact on various hosts, leading to production losses in Ethiopia. In the Southwest of Oromia, multiple livestock species share grazing land in tsetse-infested areas. Thus, a cross-sectional study was conducted from December 2020 to December 2021 to determine the prevalence and associated risk factors of trypanosomosis in bovines, small ruminants, and equines, as well as the distribution of the vector in the Dabo Hana district of Southwest Oromia, Ethiopia. A vector survey was carried out using 60 monoconical traps placed at intervals ranging fr

Prevalence of bovine trypanosomiasis in Gongola State of Northern Nigeria.

A study of bovine trypanosomosis was carried out in Gongola State (Northern Nigeria), a supposedly tsetse-free region during the months of April-July, 1990. Blood samples were collected from 1,065 cattle and examined by buffy coat and stained smear methods. Packed red cell volume (PCV) was determined simultaneously. Fourty-two (3.9%) of the animals examined were infected with trypanosomes. T. vivax was more frequently encountered than other species. Infection rate was high in the tsetse flies as 27 (22.7%) of Glossina tachinoides caught, dissected and examined were positive for T. vivax and T.

Infection rates in Glossina morsitans morsitans fed on waterbuck and Boran cattle infected with Trypanosoma congolense.

Teneral Glossina morsitans morsitans were fed on waterbuck (Kobus defassa) and Boran cattle (Bos indicus) infected experimentally with Trypanosoma congolense clone IL2895. Infection rates in tsetse varied from 9 to 31% when fed on cattle, and from 2 to 59% when fed on waterbuck. In waterbuck, infections were often detected through the development of parasites in tsetse at times when parasitaemia could not be detected through microscopic examination of blood. Male and female, and 1- and 2-day-old flies were equally susceptible to infection on both hosts. Infection in tsetse was associated with

Spatial analysis of G.f.fuscipes abundance in Uganda using Poisson and Zero-Inflated Poisson regression models.

BACKGROUND: Tsetse flies are the major vectors of human trypanosomiasis of the form Trypanosoma brucei rhodesiense and T.b.gambiense. They are widely spread across the sub-Saharan Africa and rendering a lot of challenges to both human and animal health. This stresses effective agricultural production and productivity in Africa. Delimiting the extent and magnitude of tsetse coverage has been a challenge over decades due to limited resources and unsatisfactory technology. In a bid to overcome these limitations, this study attempted to explore modelling skills that can be applied to spatially est

Observations on the prevalence of trypanosomosis in small ruminants, equines and cattle, in relation to tsetse challenge, in The Gambia.

The prevalence of trypanosome infections in Djallonké sheep and West African Dwarf goats at different sites in The Gambia showed a significant, positive correlation with contemporary assessments of tsetse challenge. A similar correlation was observed in village N'Dama cattle which showed comparable prevalence values in the same areas. Trypanosome prevalences also tended to be higher in horses and donkeys in areas with high tsetse challenge compared with sites with relatively few flies. A ranking of the numbers of tsetse blood-meals from cattle, small ruminants and equines (1:0.06: > 0.03) corr

[Ecology of Glossina palpalis VANDERPLANK, 1949 (Diptera: Glossinidae) in mangrove area of Guinea: influence of tides on tsetse densities].

The mangrove area on the Guinea littoral constitutes a favourable habitat for transmission of Trypanosoma brucei gambiens, the parasite causing sleeping sickness also called Human African Trypanosmosis (HAT), due the simultaneous presence of the vector (tsetse flies) and the human hosts. In order to assess the influence of the sea tides on the densities of Glossina palpalis gambiensis (Gpg), major vector of HAT in the mangrove, entomological surveys were performed using two transects, according to tides coefficient (great and small) and tide daily fluctuations (high and low). On each transect,

Microbiome frequency and their association with trypanosome infection in male Glossina morsitans centralis of Western Zambia.

Tsetse flies (Diptera: Glossinidae) are considered primary cyclical vectors that transmit pathogenic trypanosomes in Africa. They harbour a variety of microbes including Wolbachia, Sodalis and the salivary gland hypertrophy virus (SGHV) which are all vertically transmitted. Knowledge on tsetse microbiome and their interactions may identify novel strategies for tsetse fly and trypanosomiasis control. Area-wide application of such strategies requires an understanding of the natural microbiome frequency in the different species and subspecies of Glossina in their geographical populations. Consequ

Studies on Trypanosoma (nannomonas) congolense II. Observations on the cyclical transmission of three field isolates by Glossina morsitans morsitans.

Teneral flies of Glossina morsitans morsitans were fed on mice infected with cloned and uncloned derivatives of three recent field isolates of Trypanosoma (Nannomonas) congolense. Flies with mature infections were identified by the warm-slide probe method and phase-contrast microscopy. High infection rates were achieved when such flies were fed on mice at peak parasitaemia. The infection rates were low when flies were fed on mice prior to or late after peak parasitaemia. The duration of the developmental cycle of T. congolense in the tsetse fly varied from 7 to 40 days: in 45% of the infective

Tsetse EP protein protects the fly midgut from trypanosome establishment.

African trypanosomes undergo a complex developmental process in their tsetse fly vector before transmission back to a vertebrate host. Typically, 90% of fly infections fail, most during initial establishment of the parasite in the fly midgut. The specific mechanism(s) underpinning this failure are unknown. We have previously shown that a Glossina-specific, immunoresponsive molecule, tsetse EP protein, is up regulated by the fly in response to gram-negative microbial challenge. Here we show by knockdown using RNA interference that this tsetse EP protein acts as a powerful antagonist of establis

Variation in sensitivity of Trypanosoma congolense to diminazene during the early phase of tsetse-transmitted infection in goats.

Twenty-five goats were randomly allocated to five groups of five animals each and infected with Trypanosoma congolense IL 3274 via the bites of infected Glossina morsitans centralis. At intervals of 1, 4, 8, 12 or 19 days following infection, each group of five animals was treated intramuscularly with diminazene aceturate at a dose of 7.0 mg kg-1 body weight (b.w.). While treatment on Day 1 eliminated infections in all five goats, treatment on Day 19 did not cure any of the animals; in groups treated 4, 8 or 12 days following infection, two of five goats in each group were cured. Seasonal prevalence of bovine trypanosomosis in a tsetse-infested zone and a tsetse-free zone of the Amhara Region, north-west Ethiopia.

During a period of four consecutive years, trypanosomosis surveys were conducted in a tsetse-infested and tsetse-free area of the Amhara Region of north-west Ethiopia. In each study area randomly selected communal cattle were sampled and their blood was investigated using parasitological diagnostic methods. At the same time the population of biting flies was sampled. The monthly average prevalence of trypanosome infections in cattle did not differ significantly between study areas. In both study areas, the prevalence of trypanosome infections was highest during the long rainy season. Trypanoso

Vector competence re-evaluation of reared Glossina palpalis gambiensis for transmission of Trypanosoma congolense and Trypanosoma brucei brucei isolates for an experimental event.

BACKGROUND: Tsetse flies (Diptera: Glossinidae) are vectors of human and animal trypanosomes. The Glossina palpalis gambiensis Burkina Faso (BKF) colony, established in 1972 and rejuvenated once in 1981, is a long-standing closed colony used extensively for research and vector control. While its performance and competitiveness for sterile insect technique (SIT) programs are regularly monitored, its vector competence (VC) data are outdated. This study aimed to update the VC data of this 47-year-old colony (from the onset of experiment in 2019) against Trypanosoma congolense and T. brucei brucei
