Helicobacter pylori (H. pylori), one of the most common infectious pathogens in the world, can cause gastritis, digestive ulcers, and even gastric cancer. H. pylori urease (HPU) is a distinctive virulence factor of H. pylori that allows it to be distinguished from other pathogens. Dried ginger is a famous edible and medicinal herb that is commonly used to prevent and treat gastrointestinal tract-related diseases. In this study, phytochemical analysis of the aqueous extract of dried ginger (DGE) and the inhibition of DGE on H. pylori was investigated. Subsequently, we evaluated the inhibitory a

Helicobacter pylori (H. pylori) is a gram-negative aerobic pathogen that primarily colonizes the gastric mucosa. Peptic ulcer disease, atrophic gastritis, gastric cancer, and mucosal-associated lymphoid tissue lymphoma have all been linked to chronic H. pylori infection. Hence, it is critical to diagnose and treat it as early as possible. There are both invasive and noninvasive tests available to detect it. In this review, the diagnostic abilities of two invasive tests - histology and the rapid urease test (RUT) - are compared in a variety of clinical situations. This systematic review was car

The nickel-dependent urease enzyme is responsible for the hydrolysis of urea to ammonia and carbon dioxide. A number of bacteria produce urease (ureolytic bacteria) and are associated with various infectious diseases and ammonia emissions from agriculture. We report the first comprehensive comparison of the inhibition of urease activity by compounds analysed under the same conditions. Thus, 71 commercially available compounds were screened for their anti-ureolytic properties against both the ureolytic bacterium Klebsiella pneumoniae and purified jack bean urease. Of the tested compounds, 30 sh

The arthropod-borne transmission route of Yersinia pestis, the bacterial agent of plague, is a recent evolutionary adaptation. Yersinia pseudotuberculosis, the closely related food-and water-borne enteric species from which Y. pestis diverged less than 6,400 y ago, exhibits significant oral toxicity to the flea vectors of plague, whereas Y. pestis does not. In this study, we identify the Yersinia urease enzyme as the responsible oral toxin. All Y. pestis strains, including those phylogenetically closest to the Y. pseudotuberculosis progenitor, contain a mutated ureD allele that eliminated urea

BACKGROUND: Ureases are multifunctional enzymes that display biological activities independent of their enzymatic function, including exocytosis induction and insecticidal effects. The hemipteran Rhodnius prolixus is one of the known susceptible models for this toxicity. It has been shown that Jack bean urease (JBU) has deleterious effects on R. prolixus, and these effects are modulated by eicosanoids, which are synthesized in a cascade involving phospholipase A2 (PLA2) enzymes.
METHODS: R. prolixus genome was screened for putative PLA2s and matching transcripts were cloned. Predicted amino a

We compared immunohistochemical and silver stains of pediatric gastric biopsy sections for the identification of Helicobacter pylori infection with chronic inflammation and a negative urease screening test. Thirty-seven patients (age range 10 months to 21 years) whose gastric antral biopsies were negative for the rapid urease test (CLO(R)) but positive for lymphocytic infiltration were selected for a retrospective study. Specimens had been subjected to a rapid urease test (CLO(R)) and hematoxylin and eosin staining, and Dieterle silver staining and immunohistochemical staining specific for H.

Yersinia pestis, the plague agent, is a naturally nonureolytic microorganism, while all other Yersinia species display a potent urease activity. In this report we demonstrate that Y. pestis harbors a complete urease locus composed of three structural (ureABC) and four accessory (ureEFGD) genes. Absence of ureolytic activity is due to the presence of one additional G residue in a poly(G) stretch, which introduces a premature stop codon in ureD. The presence of the same additional G in eight other Y. pestis isolates indicates that this mutation is species specific. Spontaneous excision of the ex

CLO-tests: A statistical analysis for the evaluation of the presence of Campylobacter pylori in antral mucosal biopsies. Antral biopsies taken in a sample of 475 endoscopy patients were positive for CLO on 21% of 77 controls, compared with 39% of non-ulcer dyspepsia (NUD) patients without and 69% with a history of former peptic ulcer. Epigastric pain was associated with a positive CLO-test more frequently than belching or heart burn. Patients with NUD of Turkish or Yugoslavian nationality had a significantly higher rate of positive CLO-reactions compared with Germans or Italians with the same

Thirty-nine single antral biopsies (phase 1) and 99 sets of six antral biopsies (phase 2) were collected from 132 patients, and 87 (63%) yielded positive cultures for Campylobacter pylori. Of several primary media tested in phase 1, tryptic soy agar and Skirrow agar, each supplemented with 10% whole sheep blood, supported relatively good growth of C. pylori. In phase 2, four of the six biopsies in each set were tested with different urease systems. Selective urea agar for rapid identification was the most sensitive (39 of 63 [62%] at 1 h) and specific (100%); however, the difference between th

Three new phenolic compounds, curaromin A-C (1-3), were isolated from the rhizomes of Curcuma aromatica Salisb. through chromatographic technique. All chemical structures were determined from HRESIMS and 1D-, 2D-NMR data. The absolute configuration of curaromin C (3) was established by ECD spectra. All isolated compounds were evaluated for inhibitory activity against α-glucosidase and urease. Compounds 1-3 exhibited strong α-glucosidase inhibition, with curaromin A (1) being the most potent (IC50 = 9.8 µM), followed by curaromin B (2; IC50 = 26.4 µM) and curaromin C (3; IC50 = 42.0 µM), and al

The well-known inhibitory strength of 3d metal Schiff base complexes against urease enzymes has long been acknowledged, but their untapped potential to act as ureolytic mimics of active metallobiosites remained unexplored. To break the new ground, we present pyrrolidine-based mononuclear Ni(II)-azide complex 1 {[NiL(HL)(N3)]·1.5(H2O)} using the N,N,O donor ligand, namely (E)-4-bromo-2-(((2-(pyrrolidin-1-yl)ethyl)imino)methyl)phenol. The initial spectrophotometric analysis showed catalytic inefficiency in methanol since it undergoes unexpected ligand dissociation to generate a new octahedral ni

The global rise in antibiotic resistance has posed significant challenges to the effective management of Helicobacter pylori (H. pylori), a gastric pathogen linked to chronic gastritis, peptic ulcers, and gastric cancer. Conventional antibiotic therapies, while effective, face significant challenges, such as increasing antibiotic resistance, high recurrence rates, and adverse effects such as gut microflora dysbiosis. These limitations have driven the exploration of alternative antibiotic-free therapies, including the use of plant-based compounds, probiotics, nanoparticles, phage therapy, antim

Designing and developing small organic molecules for use as urease inhibitors is challenging due to the need for ecosystem sustainability and the requirement to prevent health risks related to the human stomach and urinary tract. Moreover, imaging analysis is widely utilized for tracking infections in intracellular and in vivo systems, which requires drug molecules with emissive potential, specifically in the low-energy region. This study comprises the synthesis of a Schiff base ligand and its selected transition metals to evaluate their UV/fluorescence properties, inhibitory activity against

Urease plays a major role in the pathogenesis of peptic and gastric ulcer and also causes acute pyelonephritis and development of infection-induced reactive arthritis. Carbonic anhydrases (CA) cause pathological disorders such as epilepsy (CA I), glaucoma, gastritis, renal, pancreatic carcinomas, and malignant brain tumors (CA II). Although various synthetic urease and carbonic anhydrase inhibitors are known, these have many side effects. Hence, present studies were undertaken on ethyl acetate extract of Aspergillus nidulans, an endophytic fungus separated from the leaves of Nyctanthes arbor-t

The problem of catheter encrustation stems from infection by urease-producing bacteria. These organisms generate ammonia from urea, elevate the pH of urine and cause crystals of calcium and magnesium phosphates to form in the urine and the biofilm that develops on the catheter. In this study, a laboratory model was used to compare the ability of 12 urease-positive species of urinary tract pathogens to encrust and block catheters. Proteus mirabilis, Proteus vulgaris and Providencia rettgeri were able to raise the urinary pH above 8.3 and produce catheter-blocking crystalline biofilms within 40

Urease, an essential virulence factor in pathogens such as Proteus mirabilis, requires nickel incorporation into its active site for maturation. This process is mediated by accessory proteins, with a key role of UreE-UreG interaction representing an early step in nickel delivery. Disruption of this interaction impairs enzyme maturation, reduces urease secretion, and limits microbial survival under acidic conditions. In this study, peptide inhibitors were designed and evaluated to target the protein-protein interaction within the UreE-UreG complex. Protein structures were predicted using AlphaF

The eu3-a mutant exhibited greater tolerance to salt stress but increased sensitivity to copper stress, with distinct impacts on nitrogen metabolism, photosynthesis, and antioxidant responses. The eu3-a soybean mutant is urease-null, lacking all urease activity responsible for catalyzing the hydrolysis of urea into ammonia and carbon dioxide. In this study, the urease-null eu3-a soybean mutant was used to assess the saline and copper stresses on nitrogen metabolism. Seeds of eu3-a/eu3-a and the corresponding dominant homozygous Eu3 precursor line were collectively referred to as near-isogenic

Prevalence of gastric diseases caused by Helicobacter Pylori bacteria is very common especially in developing countries. H. pylori is not only responsible for initiating gastric complaints like gastritis and peptic ulcer but may also lead to gastric cancer. The aim of this research study is to explore natural flora that exhibit anti urease potential. For this purpose, fifteen edible and medicinal herbs (Silybium marianum, Fagonia arabica, Nigella sativa, Curcuma longa, Ocimum sanctum, Salvia rosmainus, Hyssopus officinalis, Antrirrhinum majus, Salvia splendens, Tropaeolum majus, Dalbergia siss

This study reports the design, synthesis, and biological evaluation of a new series of indole-thiadiazole-sulfonamide hybrids as potential dual inhibitors of α-glucosidase and urease enzymes. Eighteen derivatives were synthesized and structurally characterized using standard spectroscopic techniques. Several compounds exhibited significant inhibitory activity, with IC₅₀ values ranging from 5.68 ± 1.28 to 45.42 ± 2.51 μM against α-glucosidase and from 12.99 ± 3.80 to 54.63 ± 3.26 μM against urease, compared to the reference inhibitors acarbose (18.73 ± 3.53 μM) and thiourea (21.70 ± 3.80 μM). L-Asparaginase (L-asparagine aminohydrolase, E.C. 3.5.1.1) has been proven to be competent in treating Acute Lymphoblastic Leukaemia (ALL), which is widely observed in paediatric and adult groups. Currently, clinical L-Asparaginase formulations are derived from bacterial sources such as Escherichia coli and Erwinia chrysanthemi. These formulations when administered to ALL patients lead to several immunological and hypersensitive reactions. Hence, additional purification steps are required to remove toxicity induced by the amalgamation of other enzymes like glutaminase and urease. Production of
