Cyclooxygenase products mediate the cutaneous vasodilation induced by endothelin-1 in humans.

Intradermal injection of endothelin-1 (ET-1) causes vasoconstriction (pallor) at the injection site, surrounded by a larger area of vasodilation (flare) in humans. Some of the vasomotor responses to ET-1 are thought to be mediated by prostaglandins. In the present study, we investigated the involvement of cyclooxygenase-derived products of arachidonic acid metabolism on the cutaneous vasomotor responses to ET-1. Ten normal subjects (25-44 years) were studied after treatment with either indomethacin (50 mg t.i.d.) or placebo according to a double blind cross-over design. Five doses of ET-1 (5 x

Prolonged impairment of coronary vasodilation after reversible ischemia. Evidence for microvascular "stunning".

Reperfusion after brief, reversible myocardial ischemia is associated with prolonged depression of contractile function (myocardial "stunning"); however, the effect on coronary vascular function has not been defined. Thus, open-chest dogs (n = 14) underwent a 15-minute left anterior descending coronary artery (LAD) occlusion followed by reflow. Four hours after reperfusion, regional myocardial blood flow (microspheres) was significantly (p less than 0.01) lower and coronary vascular resistance significantly (p less than 0.01) higher in the postischemic as compared with the nonischemic endocard

Neurogenic cerebral vasodilation from electrical stimulation of the cerebellum in the monkey.

The effect of stimulation of fastigial nucleus (FN) on cerebral blood flow (CBF) was examined in anesthetized, paralyzed monkeys. CBF was measured with a flow transducer chemically implanted on the left common carotid artery. The left external carotid artery and its branches were ligated. Electrical stimulation of the fastigal nucleus produced rapid (within two seconds) increases in arterial pressure (AP) and heart rate. Mean carotid blood flow (MCBF) rose 77% (+/- 40% SEM) above control and followed the increases in mean arterial pressure (MAP). Calculated cerebrovascular resistance decreased

Increased frequency of angina attacks caused by switching a brand-name vasodilator to a generic vasodilator in patients with vasospastic angina: Two case reports.

It is well known that calcium channel blockers (CCBs) are the first line of therapy for vasospastic angina (VSA). Here, we report two cases of VSA with an increase in the frequency of angina attacks after switching from a brand-name to a generic CCB. In both cases, angina recurred upon switching from a brand-name CCB to a generic CCB during follow-up. The patients' condition improved upon switching back to the original CCB. Both cases involved a high severity of VSA, based on the results of spasm provocation testing. These findings suggest that, in some patients with severe VSA, the frequency

Beyond vasodilation: sotatercept and the remodeling hypothesis in hfpef-associated pulmonary hypertension.

Pulmonary hypertension complicating heart failure with preserved ejection fraction (HFpEF), particularly the combined post- and pre-capillary form (CpcPH), has lacked any mechanism-specific therapy, as repeated trials of pulmonary vasodilators have failed or caused harm. This mini-review highlights findings from the Phase 2 CADENCE study evaluating sotatercept, an activin signaling inhibitor that targets vascular remodeling rather than vascular tone. Sotatercept met its primary endpoint by reducing pulmonary vascular resistance at 24 weeks and produced concordant improvements in mean pulmonary

The Evolving Landscape of Pulmonary Hypertension Treatment: From Vasodilation to Disease Modification.

Pulmonary hypertension (PH) is a complex disorder associated with significant morbidity and mortality. PH is characterized by an elevated pulmonary vascular load, leading to right ventricular dysfunction and remodeling, with right heart failure and premature death if left untreated. PH is classified into five groups (WHO Groups 1-5) based on etiology and pathophysiology. Group 1 PH, known as pulmonary arterial hypertension (PAH), has seen recent therapeutic advances with life-extending treatments, including a new therapeutic option that targets the underlying vascular abnormalities. PAH treatm

Diphenhydramine inhibits acetylcholine-induced, nitric oxide-mediated vasodilation in isolated rat aortas.

Nitric oxide (NO), stimulated by the muscarinic receptor agonist acetylcholine (ACh) released from the endothelium via mitochondrial acetyl-CoA production, appears to contribute to physiological flow-mediated vasodilation. Diphenhydramine (DPH), a muscarinic receptor antagonist, may interfere with ACh-induced NO-mediated vasodilation. However, whether DPH affects ACh-induced, NO-mediated vasodilation remains unknown. This study aimed to examine the effect of DPH on ACh-induced, NO-mediated vasodilation and its underlying mechanisms, including muscarinic receptor subtypes. The effects of diphen

Synthetic Biocatalysis in Erythrocyte Microreactors for Gasotransmitter H2S Production and Vessel Vasodilation.

Designing therapeutic microreactors that emulate natural cells to execute vital metabolic processes and interact with biological systems is a significant challenge in synthetic biology and bioengineering. In this study, erythrocyte microreactors capable of prolonged hydrogen sulfide (H₂S) production for therapeutic vasodilation are developed. The microreactors are fabricated by encapsulating a pyridoxal 5'-phosphate-polylysine (PLP-PLys) complex into erythrocytes using a hypotonic swelling method. The optimized process achieved a loading efficiency of 61.5%, ensuring the uniform distribution o

New device for assessment of endothelial function: plethysmographic flow-mediated vasodilation (pFMD).

Measurement of flow-mediated vasodilation (FMD) in the brachial artery by using ultrasound is a well-established technique for evaluating endothelial function. To make the measurement quicker and simpler than the measurements of conventional ultrasound FMD (uFMD), we have developed a new noninvasive method, plethysmographic FMD (pFMD), to assess vascular response to reactive hyperemia in the brachial artery. The aim of this study was to determine the accuracy of measurement of pFMD in comparison to that of measurement of conventional uFMD. This study was a multi-center, cross-sectional study.

Hybrid Photoacoustic Ultrasound Imaging System for Cold-Induced Vasoconstriction and Vasodilation Monitoring.

Lewis hunting reaction refers to the alternating cold-induced vasoconstriction and dilation in extremities, whose underlying mechanism is complex. While numerous studies reported this intriguing phenomenon by measuring cutaneous temperature fluctuation under cold exposure, few of them tracked peripheral vascular responses in real-time, lacking a non-invasive and quantitative imaging tool. To better monitor hunting reaction and diagnose relevant diseases, we developed a hybrid photoacoustic ultrasound (PAUS) tomography system to monitor finger vessels' dynamic response to cold, together with si

Impact of Vasodilation on Oxygen-Enhanced Functional Lung MRI at 0.55 T.

BACKGROUND: Oxygen-enhanced magnetic resonance imaging (OE-MRI) can be used to assess regional lung function without ionizing radiation. Inhaled oxygen acts as a T1-shortening contrast agent to increase signal in T1-weighted (T1w) images. However, increase in proton density from pulmonary hyperoxic vasodilation may also contribute to the measured signal enhancement. Our aim was to quantify the relative contributions of the T1-shortening and vasodilatory effects of oxygen to signal enhancement in OE-MRI in both swine and healthy volunteers. METHODS: We imaged 14 anesthetized female swine (47 ±

Short stature is associated with low flow-mediated vasodilation in Japanese men.

An inverse association between height and the risk of cardiovascular disease has been reported. The objective of this study was to examine the association between height and endothelial function assessed by flow-mediated vasodilation (FMD). We evaluated cross-sectional associations of height with FMD in 7682 Japanese men. All participants were divided into four groups based on height: <155.0 cm, 155.0-164.9 cm, 165.0-174.9 cm, and ≥175.0 cm. Subjects in a lower quartile of FMD were defined as subjects having low FMD values. Univariate regression analysis revealed that height was significantly

Quantitative myocardial first-pass perfusion imaging of CO2 -induced vasodilation in rats.

Inducible hypercapnia is an alternative for increasing the coronary blood flow necessary to facilitate the quantification of myocardial blood flow during hyperemia. The current study aimed to quantify the pharmacokinetic effect of a CO2 gas challenge on myocardial perfusion in rats using high-resolution, first-pass perfusion CMR and compared it with pharmacologically induced hyperemia using regadenoson. A dual-contrast, saturation-recovery, gradient-echo sequence with a Cartesian readout was used on a small-animal 9.4-T scanner; additional cine images during hyperemia/rest were recorded with a

Increased cerebral endothelium-dependent vasodilation in rats in the postcardiac arrest period.

Cardiovascular lability is common after cardiac arrest. We investigated whether altered endothelial function is present in cerebral and mesenteric arteries 2 and 4 h after resuscitation. Male Sprague-Dawley rats were anesthetized, intubated, ventilated, and intravascularly catheterized whereupon rats were randomized into four groups. Following 7 min of asphyxial cardiac arrest and subsequent resuscitation, cardiac arrest and sham rats were observed for either 2 or 4 h. Neuron-specific enolase levels were measured in blood samples. Middle cerebral artery segments and small mesenteric arteries w

Puerarin induces mouse mesenteric vasodilation and ameliorates hypertension involving endothelial TRPV4 channels.

Puerarin (Pue) is an isoflavone derived from the root of Pueraria lobata, which has been widely used as food and a herb for treating cardiovascular and cerebrovascular diseases. Transient receptor potential vanilloid 4 (TRPV4), a Ca2+-permeable channel with multiple modes of activation, plays an important role in vascular endothelial function and vasodilation. However, no reports have shown the effects of Pue on TRPV4 channels and mouse small mesenteric arteries. In the present study, we performed a molecular docking assay by using Discovery Studio 3.5 software to predict the binding of Pue to

Quantification of Myocardial Perfusion With Vasodilation Using Arterial Spin Labeling at 1.5T.

BACKGROUND: Myocardial perfusion is evaluated in first-pass MRI using a gadolinium-based contrast agent, which limits its repeatability and restricts its use in patients with abnormal kidney function. Arterial spin labeling (ASL) is a promising technique for measuring myocardial perfusion without contrast injection. The ratio of stress to rest perfusion, termed myocardial perfusion reserve (MPR), is an indicator of the severity of stenosis in patients with coronary artery disease (CAD). PURPOSE: To quantify perfusion increases with pharmacological vasodilation, explore MPR differences between

Comparison Between Two Pharmacologic Strategies to Alleviate Rewarming Shock: Vasodilation vs. Inodilation.

Rewarming from hypothermia is often challenged by coexisting cardiac dysfunction, depressed organ blood flow (OBF), and increased systemic vascular resistance. Previous research shows cardiovascular inotropic support and vasodilation during rewarming to elevate cardiac output (CO). The present study aims to compare the effects of inodilatation by levosimendan (LS) and vasodilation by nitroprusside (SNP) on OBF and global oxygen transport during rewarming from hypothermia. We used an in vivo experimental rat model of 4 h 15°C hypothermia and rewarming. A stable isotope-labeled microsphere techn

Royal jelly causes hypotension and vasodilation induced by increasing nitric oxide production.

Among royal jelly's (RJ) various biological activities, its possible antihypertension and vasorelaxation effects deserve particular attention, but the underlying mechanisms of action remain unclear. Therefore, this study used the spontaneously hypertensive rats (SHR) hypertension model and the isolated rabbit thoracic aorta rings model to explore the mechanisms underlying the hypotension and vasorelaxation effects of RJ. Rats were divided into the following groups (n = 6): WKY-control group, SHR-control group, and SHR-RJ group. SHR-RJ group was received 1 g/kg of RJ via oral administration dai
