Cerebral Perfusion and Sensory Testing Results Differ in Interstitial Cystitis/Bladder Pain Syndrome Patients with and without Fibromyalgia: A Site-Specific MAPP Network Study.

PURPOSE: Fibromyalgia is a common co-morbidity in patients with interstitial cystitis/bladder pain syndrome. Quantitative sensory testing measures and regional cerebral blood flow measures have been noted to differ from healthy controls in both subjects with fibromyalgia and those with interstitial cystitis when studied independently. The present study examined such measures in subjects with the diagnosis of interstitial cystitis both with and without the co-diagnosis of fibromyalgia to determine whether differences in these measures may be associated with co-morbidity. PATIENTS AND METHODS:

Multimodal Evaluation of Neurovascular Functionality in Early Parkinson's Disease.

Parkinson's disease (PD) is a multisystem neurological condition affecting different neurotransmitter pathways characterized by aberrant functional connectivity (FC) and perfusion alteration. Since the FC, measuring neuronal activity, and cerebral blood flow (CBF) are closely related through the neurovascular coupling (NVC) mechanism, we aim to assess whether FC changes found in PD mirror perfusion ones. A multimodal MRI study was implemented by acquiring resting state functional MRI (rsfMRI) and arterial spin labeling (ASL) datasets on a group of 26 early PD (66.8 ± 8 years, 22 males, median

Disrupted Regional Cerebral Blood Flow, Functional Activity and Connectivity in Alzheimer's Disease: A Combined ASL Perfusion and Resting State fMRI Study.

Recent studies have demonstrated a close relationship between regional cerebral blood flow (rCBF) and resting state functional connectivity changes in normal healthy people. However, little is known about the parameter changes in the most vulnerable regions in Alzheimer's disease (AD). Forty AD patients and 30 healthy controls participated in this study. The data of resting-state perfusion and functional magnetic resonance imaging (fMRI) was collected. By using voxel-wise arterial spin labeling (ASL) perfusion, we identified several regions of altered rCBF in AD patients. Then, by using restin

Cytosolic, but not mitochondrial, oxidative stress is a likely contributor to cardiac hypertrophy resulting from cardiac specific GLUT4 deletion in mice.

We hypothesized that oxidative stress may contribute to the development of hypertrophy observed in mice with cardiac specific ablation of the insulin sensitive glucose transporter 4 gene (GLUT4, G4H(-/-) ). Measurements of oxidized glutathione (GSSG) in isolated mitochondria and whole heart homogenates were increased resulting in a lower ratio of reduced glutathione (GSH) to GSSG. Membrane translocation of the p67(phox) subunit of cardiac NADPH oxidase 2 (NOX2) was markedly increased in G4H(-/-) mice, suggesting elevated activity. To determine if oxidative stress was contributing to cardiac hy

Synergism in hyperhomocysteinemia and diabetes: role of PPAR gamma and tempol.

BACKGROUND: Hyperhomocysteinemia (HHcy) and hyperglycemia cause diabetic cardiomyopathy by inducing oxidative stress and attenuating peroxisome proliferator- activated receptor (PPAR) gamma. However, their synergistic contribution is not clear. METHODS: Diabetic Akita (Ins2+/-) and hyperhomocysteinemic cystathionine beta synthase mutant (CBS+/-) were used for M-mode echocardiography at the age of four and twenty four weeks. The cardiac rings from WT, Akita and hybrid (Ins2+/-/CBS+/-) of Akita and CBS+/- were treated with different doses of acetylcholine (an endothelial dependent vasodilator).

Prenatal high-salt diet impaired vasodilatation with reprogrammed renin-angiotensin system in offspring rats.

AIMS: High-salt diet is linked to hypertension, and prenatal high-salt diet increases the risk of cardiovascular diseases in the offspring. The present study investigated whether and how prenatal high-salt diet influenced nitric oxide-mediated vasodilatation in the offspring. METHODS AND RESULTS: Pregnant rats were fed either normal-salt (1% sodium chloride) or high-salt (8% sodium chloride) diet during gestation. Experiments were conducted in 5-month-old male offspring. Sodium nitroprusside (SNP, nitric oxide donor)-induced hypotensive responses (in vivo) and vascular dilatation (in vitro) w

Podocyte as the target for aldosterone: roles of oxidative stress and Sgk1.

Accumulating evidence suggests that mineralocorticoid receptor blockade effectively reduces proteinuria in hypertensive patients. However, the mechanism of the antiproteinuric effect remains elusive. In this study, we investigated the effects of aldosterone on podocyte, a key player of the glomerular filtration barrier. Uninephrectomized rats were continuously infused with aldosterone and fed a high-salt diet. Aldosterone induced proteinuria progressively, associated with blood pressure elevation. Notably, gene expressions of podocyte-associated molecules nephrin and podocin were markedly decr

alpha-Phenyl-N-tert-butylnitrone provides protection from dextran sulfate sodium-induced colitis in mice.

Nuclear factor-kappaB (NF-kappaB)-dependent up-regulation of inflammatory cytokines and inducible nitric oxide (iNOS) occurs in inflammatory bowel disease. We investigated the effect of alpha-phenylN-tert-butylnitrone (PBN), a spin-trapping agent that inhibits NF-kappaB activity, on dextran sulfate sodium (DSS)-induced colonic mucosal injury and inflammation in mice. Acute colitis was induced with DSS in female BALB/c mice receiving 0, 0.3, 3, and 30 mg/kg i.p. PBN daily. Colonic mucosal inflammation was evaluated biochemically and histologically. Nitric oxide was evaluated as luminal nitrite/

Increased oxidative stress brought on by pro-inflammatory cytokines in neurodegenerative processes and the protective role of nitrone-based free radical traps.

Nitrone-based free radical traps (NFTs) have been shown to be protective in several neurodegenerative models. Our research has strongly implicated that: A) several neurodegenerative conditions exhibit increased levels of pro-inflammatory cytokines which consequently result in increased levels of oxidative stress and B) that NFTs act in part by suppressing oxidative stress through suppression of the action of the cytokine cascade. Acquired Immune Deficiency Syndrome (AIDS) dementia complex (ADC) is one of several conditions where the data collected helped to develop these concepts. Novel observ

Multimodal MRI insights into glymphatic clearance and blood-brain barrier integrity in healthy aging.

Aging is associated with impaired CSF clearance in preclinical models, but its impact on blood-brain barrier (BBB) health and glymphatic function in humans remains unclear. The BBB and glymphatic functions are complementary and may partially overlap, yet their interaction is yet to be fully elucidated. In this study, we aimed to compare BBB integrity, extracellular free water, and glymphatic function between healthy young and older adults, and to examine mechanistic relationships among these measures using multimodal MRI techniques. 30 healthy participants with no known neurological or psychia

Early kidney injury occurs in the absence of detectable perirenal adipose tissue remodelling in the Munich Wistar Frömter rat model of chronic kidney disease.

Adipose tissue surrounding the kidney, known as perirenal adipose tissue (PRAT), is a metabolically active depot that may influence renal function through inflammatory and fibrotic mediators. In the Munich Wistar Frömter (MWF) rat, a genetic model of spontaneous chronic kidney disease (CKD), PRAT dysfunction has been described at 22 weeks but whether it is present at earlier stages related to the initiation of renal injury remains unclear. This study aimed to assess the temporal relationship between PRAT phenotype and renal injury in MWF rats. Male MWF and control Wistar rats were evaluated at

Acute exercise reduces intranasal insulin-mediated elevations in blood pressure in aging adults with cardiometabolic risk.

Insulin contributes to blood pressure (BP) regulation through, in part, effects on the brain. Whether exercise impacts brain insulin action in parallel with alterations in BP is unclear. The purpose of this study is to examine whether an acute exercise lowers BP after intranasal insulin (INI) and increases cerebral blood flow (CBF) in aging adults with cardiometabolic risk. In a counterbalanced design, 15 cognitively unimpaired (MoCA: 28.2 ± 1.3 au) adults [56.7 ± 2.1 yr; (maximal oxygen consumption) V̇o2max: 23.9 ± 0.9 mL/kg/min] with obesity (BMI 31.8 ± 1.3 kg·m-2) were randomized to a rest

Study protocol for neuroimaging using 7 T MRI in the investigation of baricitinib for reduction of HIV in the CNS: a randomized placebo-controlled trial.

BACKGROUND: While antiretroviral therapy is successful in conferring peripheral human immunodeficiency virus type 1 (HIV-1) suppression, eradication of HIV-1 reservoirs in the central nervous system (CNS) is an immediate goal in the management of HIV-1 infection. The primary goal of this randomized and placebo-controlled Phase IIb trial is to evaluate the effects of baricitinib versus placebo on HIV-1 CNS persistence markers in a ten-week mechanistic study, with secondary endpoints evaluating the effects on peripheral blood, neuroimaging, and neuropsychological measures. Given the large scope

ASLNet: an explainable deep learning framework for glioma grading and survival prediction.

INTRODUCTION: Arterial spin labeling (ASL) MRI provides noninvasive quantitative perfusion information and may capture vascular heterogeneity associated with glioma aggressiveness and prognosis. Deep learning (DL) approaches applied directly to ASL volumes may therefore support imaging-based prediction of tumor grade and survival. This study aimed to develop and validate ASLNet, an interpretable three-dimensional residual network framework trained on ASL MRI to predict histopathologic grade and overall survival (OS) in patients with diffuse glioma. METHODS: This retrospective study included 4
