Basilar Artery Lateral Displacement May Be Associated with Migraine with Aura.

OBJECTIVE: The objective of this study is to determine whether structural features of the vertebrobasilar arterial system are related to migraine. BACKGROUND: Alterations in cerebral vascular structure and function have been associated with migraine, possibly mediated by hypoperfusion and/or endothelial dysfunction triggering cortical spreading depression. Vessel tortuosity, in particular, has been associated with both altered hemodynamics and endothelial function. Symptoms of migraine with aura (MWA) often localize to the occipital cortex, and evidence supports the localization of a migraine

Secondary prevention of Alzheimer's dementia: neuroimaging contributions.

BACKGROUND: In Alzheimer's disease (AD), pathological changes may arise up to 20 years before the onset of dementia. This pre-dementia window provides a unique opportunity for secondary prevention. However, exposing non-demented subjects to putative therapies requires reliable biomarkers for subject selection, stratification, and monitoring of treatment. Neuroimaging allows the detection of early pathological changes, and longitudinal imaging can assess the effect of interventions on markers of molecular pathology and rates of neurodegeneration. This is of particular importance in pre-dementia

Clinical evaluation of a quantitative real time polymerase chain reaction assay for diagnosis of primary Epstein-Barr virus infection in children.

BACKGROUND: Epstein-Barr virus (EBV) infectious mononucleosis is often diagnosed based on characteristic clinical features and either a positive heterophil antibody test or serology, both of which can be unreliable in young children. Real time quantitative PCR assays that measure EBV DNA load in serum or plasma are highly sensitive in young children, but serum and plasma contain inhibitors of PCR which must be removed by DNA extraction techniques. A real time TaqMan PCR assay was designed and evaluated for simultaneously measuring EBV DNA load and validating the removal of PCR inhibitors from

Inhibition of natriuretic factors increases blood pressure in rats.

Renal dopamine and nitric oxide contribute to natriuresis during high-salt intake which maintains sodium and blood pressure homeostasis. We wanted to determine whether concurrent inhibition of these natriuretic factors increases blood pressure during high-sodium intake. Male Sprague-Dawley rats were divided into the following groups: 1) vehicle (V)-tap water, 2) NaCl-1% NaCl drinking water, 3) 30 mM l-buthionine sulfoximine (BSO), an oxidant, 4) BSO plus NaCl, and 5) BSO plus NaCl with 1 mM tempol (antioxidant). Compared with V, NaCl intake for 10 days doubled sodium intake and increased urina

Methylmercury-mediated inhibition of 3H-D-aspartate transport in cultured astrocytes is reversed by the antioxidant catalase.

Astrocytes are essential for removal of glutamate from the extracellular space in the central nervous system. The neurotoxic heavy metal methylmercury potently and specifically inhibits the transport of glutamate in cultured astrocytes by an unknown mechanism. Glutamate transport in astrocytes is also inhibited by reactive oxygen species. A glutamate-induced transporter current is inhibited both by reactive oxygen species and thiol oxidizing agents. These observations suggest that oxidation of the transporter might mediate methylmercury-induced inhibition of glutamate transport. In the present

Texture Analysis of Kidney MRI Using Diffusion-Weighted Imaging and Intravoxel Incoherent Motion for Classifying the Severity of Chronic Kidney Diseases.

Introduction Chronic kidney disease (CKD) is a significant global health concern, and noninvasive imaging biomarkers for chronic kidney disease have been investigated using magnetic resonance imaging (MRI), including diffusion‑weighted imaging (DWI), blood oxygenation level-dependent (BOLD) imaging, arterial spin labeling (ASL), and T1 mapping; however, there are currently no widely accepted imaging biomarkers for the non-invasive assessment of renal dysfunction severity. DWI and intravoxel incoherent motion (IVIM), when combined with texture analysis (TA), provide a promising approach for the

Integrating Metabolic, Perfusion, and Microstructural Parameters for Quantitative Neuroimaging in Rare Neurodegenerative Diseases: A Hybrid PET/MRI Approach.

Background/Objectives: The use of quantitative neuroimaging to establish objective biomarkers in neurodegenerative diseases (NDD) has attracted increasing interest over the last decade. Advanced magnetic resonance imaging (MRI) such as arterial spin labeling (ASL) and diffusion tensor imaging (DTI), as well as [18F]fluorodeoxyglucose ([18F]FDG) positron emission tomography (PET), could provide clinically meaningful biomarkers and may support differential diagnosis. The aim of this investigator-initiated, single-center, retrospective comparative study was to implement a framework for multimodal

Early neurodevelopmental brain perfusion abnormalities and functional connectivity findings in infants with Prader-Willi syndrome.

BACKGROUND: Prader-Willi syndrome (PWS) is a neurodevelopmental disorder characterized by distinct eating behaviors that evolve from early feeding difficulties to later hyperphagia. Early brain abnormalities remain poorly understood, with no data on brain perfusion during infancy. This study investigated early brain perfusion and functional connectivity in infants with PWS and their associations with feeding and social functioning. METHOD: Twenty-seven infants (mean age 3 months) were included in this prospective study. Thirteen genetically confirmed PWS infants (mean age 2.7 ± 1.2 months) und

Convergent functional, white-matter microstructural, and cerebral perfusion abnormalities in overweight and obese adults: a multimodal MRI study.

BACKGROUND: Obesity affects the brain, but most neuroimaging studies examine a single modality, leaving it unclear whether obesity-related functional, microstructural, and perfusion abnormalities co-localize. We combined three modalities across the normal-weight-overweight-obese spectrum. METHODS: Of 80 adults enrolled, 10 were excluded (sleep, incomplete acquisition, artifacts), leaving 70 (normal-weight, n = 27; overweight, n = 21; obese, n = 22), who did not differ in age, sex, or education. All underwent resting-state functional MRI, diffusion tensor imaging, and 3D arterial spin labeling

Hyperoxic cerebral vasoconstriction in healthy humans: global, lobe, and regions of interest.

Hyperoxia is used for hypoxia/ischemia-related conditions, but is associated with poorer clinical outcomes, potentially related to reductions in cerebral blood flow (CBF). Global CBF hyperoxia responses are variable; limited evidence suggests heightened sensitivity in anterior brain regions, leaving important gaps in understanding spatial resolution. Using 3-Tesla magnetic resonance imaging (MRI), we tested the hypotheses that 1) isocapnic hyperoxia would reduce CBF at the whole brain, gray matter (GM), and white matter (WM), and 2) this reduction would differ across all lobes. CBF was quantif

The efficacy and cerebral hemodynamics mechanisms of acupuncture on the posterior circulation ischemic stroke with vertigo: study protocol for a multicenter, randomized, controlled trial.

INTRODUCTION: Posterior circulation ischemic stroke (PCIS) with vertigo is a common central vertigo disease that significantly hinders patients' motivation for rehabilitation and increases the recurrence rate and mortality among stroke patients due to recurrent episodes of vertigo. Acupuncture has shown promising therapeutic effects in the treatment of PCIS with vertigo, but its underlying mechanisms remain unclear. This study is designed to investigate the impact of acupuncture on cerebral hemodynamics and brain structure in PCIS patients vertigo, and to evaluate its clinical effectiveness in

Integrated imaging and molecular profiling reveals APOE4-associated neurovascular and glial disruptions in young adult mice.

BACKGROUND: The Apolipoprotein-E ε4 (APOE4) allele is the strongest genetic risk factor for late-onset Alzheimer's disease (LOAD) and may contribute to neurodegeneration through a multi-hit hypothesis, in which vascular dysfunction, glial activation, and impaired lipid metabolism play central roles. Alterations in neurovascular unit (NVU) have emerged as an early APOE4-related phenotype, independent of amyloid and tau pathology. Astrocytes, as the primary source of APOE in the brain and key regulators of NVU homeostasis, may play a central role in these processes. This study investigates APOE4

The Acute Effects of Non-concussive Head Impacts on Brain Microstructure, Chemistry and Function in Male Soccer Players: A Pilot Randomised Controlled Trial.

BACKGROUND: Head impacts, particularly, non-concussive impacts, are common in sport. Yet, their effects on the brain remain poorly understood. Here, we investigated the acute effects of non-concussive impacts on brain microstructure, chemistry, and function using magnetic resonance imaging (MRI) and other techniques. RESULTS: Fifteen healthy male soccer players participated in a randomised, controlled, crossover pilot trial. The intervention was a non-concussive soccer heading task ('Heading') and the control was an equivalent 'Kicking' task. Participants underwent MRI scans ~ 45 min post-tas

Added prognostic value of histogram features from preoperative multi-modal diffusion MRI in predicting Ki-67 proliferation for adult-type diffuse gliomas.

BACKGROUND: Ki-67 labelling index (LI), a critical marker of tumor proliferation, is vital for grading adult-type diffuse gliomas and predicting patient survival. However, its accurate assessment currently relies on invasive biopsy or surgical resection. This makes it challenging to non-invasively predict Ki-67 LI and subsequent prognosis. Therefore, this study aimed to investigate whether histogram analysis of multi-parametric diffusion model metrics-specifically diffusion tensor imaging (DTI), diffusion kurtosis imaging (DKI), and neurite orientation dispersion and density imaging (NODDI)-co

Multiparametric cardiac magnetic resonance identifies macrophage nitric oxide synthase 2-mediated benefits of preventive sodium-glucose cotransporter 2 inhibition in a mouse model of metabolic heart disease.

BACKGROUND: Sodium-glucose cotransporter 2 (SGLT2) inhibitors improve metabolic and cardiovascular outcomes, but the mechanisms remain incompletely understood. We utilized cardiovascular magnetic resonance (CMR) and complementary methods to investigate whether preventive SGLT2 inhibitor administration attenuates the development of metabolic heart disease in a high-fat, high-sucrose diet (HFHSD) mouse model. METHODS: Male wild-type (WT) C57BL/6 J mice were fed an HFHSD for 18 weeks to induce obesity, coronary microvascular disease, and diastolic dysfunction. WT mice treated preventively with a

Alterations in cerebral perfusion and substrate metabolism in type 2 diabetes: interactions with APOE-ε4.

AIMS/HYPOTHESIS: Epidemiological studies indicate that type 2 diabetes increases the risk for Alzheimer's disease. Alterations in cerebral metabolism have been proposed as a potential mechanism underlying this association. A better understanding of these metabolic changes may elucidate potential pathways linking type 2 diabetes to Alzheimer's disease. The aim of the current exploratory study was to investigate whether cerebral metabolism, including glucose and fatty acid uptake as well as cerebral blood flow, is altered in individuals with type 2 diabetes compared with both overweight individu
