Reduced basal macrovascular and microvascular cerebral blood flow in young adults with metabolic syndrome: potential mechanisms.

Ninety-million Americans suffer metabolic syndrome (MetSyn), increasing the risk of diabetes and poor brain outcomes, including neuropathology linked to lower cerebral blood flow (CBF), predominantly in anterior regions. We tested the hypothesis that total and regional CBF is lower in MetSyn more so in the anterior brain and explored three potential mechanisms. Thirty-four controls (25 ± 5 yr) and 19 MetSyn (30 ± 9 yr), with no history of cardiovascular disease/medications, underwent four-dimensional flow magnetic resonance imaging (MRI) to quantify macrovascular CBF, whereas arterial spin lab

ACU193, a Monoclonal Antibody that Selectively Binds Soluble Aß Oligomers: Development Rationale, Phase 1 Trial Design, and Clinical Development Plan.

BACKGROUND: Alzheimer's disease is a large and growing unmet medical need. Clinical trial designs need to assess disease-related outcomes earlier to accelerate the development of better treatments for Alzheimer's disease. ACU193 is a monoclonal antibody that selectively targets amyloid β oligomers, thought to be the most toxic species of Aβ that accumulates early in AD and contributes to downstream pathological effects. Nonclinical data indicate that ACU193 can reduce the toxic effects of amyloid β oligomers. ACU193 is currently being investigated in a phase 1 clinical trial designed with the

Mixed nut consumption improves brain insulin sensitivity: a randomized, single-blinded, controlled, crossover trial in older adults with overweight or obesity.

BACKGROUND: Improving brain insulin sensitivity, which can be assessed by measuring regional cerebral blood flow (CBF) responses to intranasal insulin, may prevent age-related metabolic and cognitive diseases. OBJECTIVES: This study aimed to investigate longer-term effects of mixed nuts on brain insulin sensitivity in older individuals with overweight/obesity. METHODS: In a randomized, single-blinded, controlled, crossover trial, 28 healthy adults (mean ± standard deviation: 65 ± 3 years; body mass index: 27.9 ± 2.3 kg/m2) received either daily 60-g mixed nuts (15 g of walnuts, pistachio, ca

Effects of alfaxalone, propofol and isoflurane on cerebral blood flow and cerebrovascular reactivity to carbon dioxide in dogs: A pilot study.

Propofol total intravenous anesthesia is a common choice to anesthetize patients with increased intracranial pressure, reducing cerebral blood flow while maintaining cerebrovascular reactivity to CO2. Propofol and alfaxalone are commonly used for total intravenous anesthesia in dogs, but the effects of alfaxalone on cerebral blood flow and cerebrovascular reactivity to CO2 are unknown. Our hypothesis was that alfaxalone would not be significantly different to propofol, while isoflurane would increase cerebral blood flow and decrease cerebrovascular reactivity to CO2. Six healthy hound dogs wer

Brain Differences in Adolescents Living With Perinatally Acquired HIV Compared With Adoption Status Matched Controls: A Cross-sectional Study.

BACKGROUND AND OBJECTIVES: Despite effective combination antiretroviral therapy (cART), adolescents with perinatally acquired HIV (PHIV) exhibit cognitive impairment, of which structural changes could be the underlying pathophysiologic mechanism. Prior MRI studies found lower brain volumes, higher white matter (WM) hyperintensity (WMH) volume, lower WM integrity, and differences in cerebral blood flow (CBF). However, these findings may be confounded by adoption status, as a large portion of adolescents with PHIV have been adopted. Adoption has been associated with malnutrition and neglect, whi

The influence of body temperature on tissue stiffness, blood perfusion, and water diffusion in the mouse brain.

While hypothermia of the brain is used to reduce neuronal damage in patients with conditions such as traumatic brain injury or stroke, little is known about how temperature affects the biophysical properties of in vivo brain tissue. Therefore, we measured shear wave speed (SWS), apparent diffusion coefficient (ADC), and cerebral blood flow (CBF) in the mouse brain at different body temperatures to investigate the relationship between temperature and tissue stiffness, water diffusion, and blood perfusion in the living brain. Multifrequency magnetic resonance elastography (MRE), diffusion-weight

Separating neural and vascular effects of caffeine using simultaneous EEG-FMRI: differential effects of caffeine on cognitive and sensorimotor brain responses.

The effects of caffeine are mediated through its non-selective antagonistic effects on adenosine A(1) and A(2A) adenosine receptors resulting in increased neuronal activity but also vasoconstriction in the brain. Caffeine, therefore, can modify BOLD FMRI signal responses through both its neural and its vascular effects depending on receptor distributions in different brain regions. In this study we aim to distinguish neural and vascular influences of a single dose of caffeine in measurements of task-related brain activity using simultaneous EEG-FMRI. We chose to compare low-level visual and mo

Slice-Wise Quality Assessment of High b Value Breast DWI via Deep Learning-Based Artifact Detection.

BACKGROUND: Diffusion-weighted imaging (DWI) can support lesion detection and characterization in breast MRI. Therefore, DWI is increasingly being incorporated into breast MRI protocols to address some of the shortcomings of routine clinical breast MRI. However, especially high b value DWI can be prone to intensity artifacts that can affect diagnostic image assessment. PURPOSE: To detect hyper- and hypointense artifacts on high b value DWI (b = 1500 s/mm2) using deep learning, employing either a binary classification (artifact presence) or a multiclass classification (artifact intensity) appr

Cerebral hypoperfusion in pulmonary arterial hypertension: Association with right ventricular dysfunction and risk stratification.

BackgroundPulmonary arterial hypertension (PAH) is a progressive disease that affects the central nervous system. The cardiopulmonary-brain axis hypothesis links right ventricular dysfunction to cerebral hypoperfusion, yet the association between regional cerebral blood flow (CBF) alterations, right heart dysfunction, and the clinical utility of CBF for risk stratification remains poorly understood.ObjectivesTo investigate the association between CBF and right heart dysfunction in patients with PAH, and to evaluate the potential of CBF as a non-invasive biomarker for risk stratification.Design

Regional Cerebral Blood Flow Patterns on ASL in Subacute Sclerosing Panencephalitis: Quantitative Analysis and Clinical Correlation.

BACKGROUND AND PURPOSE: Subacute sclerosing panencephalitis (SSPE) is a progressive, uniformly fatal neuroinflammatory disease caused by persistent measles virus encephalitis, characterised by chronic T-cell-mediated inflammation, progressive demyelination, and cortico-striatal-thalamic network dysfunction. Conventional MRI characterises established structural disease but does not capture early haemodynamic alterations. We aimed to characterise regional perfusion patterns using cerebellar-normalised arterial spin labeling (ASL) ratios, define qualitative perfusion phenotypes, and examine their

Altered coupling of cerebral blood perfusion and neuronal activity in children with MRI-negative drug refractory epilepsy.

BACKGROUND: Drug refractory focal epilepsy (DRFE) often gives rise to structural and functional damage in the brain accompanied by neuronal activity and cerebral vascular hemodynamics changes, which may result in neurovascular decoupling. However, neuroimaging evidence on neurovascular decoupling remains scarce. This study aimed to assess the manifestation of neurovascular coupling (NVC) in childhood DRFE using resting-state functional magnetic resonance imaging (rs-fMRI) and arterial spin labeling imaging (ASL). METHODS: The rs-fMRI and ASL imaging data were obtained from 24 children with DR
