[Pseudo-targeted metabolomics study of immune stress-mediated idiosyncratic liver injury induced by synergistic effects of bavachin and epimedin B].

Chinese patent medicine preparations containing Epimedii Folium and Psoraleae Fructus have been associated with the occurrence of idiosyncratic drug-induced liver injury(IDILI). However, the specific toxic biomarkers and mechanisms underlying these effects remain unclear. This study aimed to comprehensively assess the impact of bavachin and epimedin B, two principal consti-tuents found in Psoraleae Fructus and Epimedii Folium, on an IDILI model induced by tumor necrosis factor-α(TNF-α) treatment, both in vitro and in vivo. To evaluate the extent of liver injury, various parameters were assesse

Deep phenotyping of the lipidomic response in COVID-19 and non-COVID-19 sepsis.

BACKGROUND: Lipids may influence cellular penetrance by viral pathogens and the immune response that they evoke. We deeply phenotyped the lipidomic response to SARs-CoV-2 and compared that with infection with other pathogens in patients admitted with acute respiratory distress syndrome to an intensive care unit (ICU). METHODS: Mass spectrometry was used to characterise lipids and relate them to proteins, peripheral cell immunotypes and disease severity. RESULTS: Circulating phospholipases (sPLA2, cPLA2 (PLA2G4A) and PLA2G2D) were elevated on admission in all ICU groups. Cyclooxygenase, lipox

The Effect of Neoglandin on the Activity of N-Acetyl-β-D-Hexosaminidase in the Serum and Urine of Alcohol-Dependent Men.

UNLABELLED: Dietary supplementation of gamma-linolenic acid (GLA) in the form of a commercial drug neoglandin (containing GLA and vitamin E), in people following alcohol abuse allows bypassing of the ineffective delta-6-desaturase system involved in the transformation of linoleic acid into GLA. Determination of the activity of N-acetyl-β-D-hexosaminidase (HEX) in the serum and urine reflects neoglandin action on the catabolism of glycoconjugates and the functioning of liver and kidneys in people following alcohol abuse. MATERIAL AND METHODS: The serum and urine were collected from men with al

Calcium salts of fatty acids with varying fatty acid profiles in diets of feedlot-finished Bos indicus bulls: impacts on intake, digestibility, performance, and carcass and meat characteristics.

We hypothesized that the inclusion of calcium salts of fatty acid (CSFA) into the diets and the fatty acid (FA) profile of the supplements would impact performance and meat characteristics of Bos indicus bulls. Hence, the objective was to evaluate the effects of CSFA profiles on intake, body weight (BW), carcass, and meat characteristics of feedlot-finished B indicus bulls. Fifty-three Nellore bulls [initial BW 315 ± 5.9 kg and 20 ± 2 mo] were used. At the beginning, 6 bulls were randomly chosen and slaughtered for determination of their BW composition, and the remaining 47 bulls were evaluate

Metabolism of lipoidal derivatives of estradiol-17-beta in human mammary cancer tissue and cell lines.

Estradiol-17 beta (E2) is converted exclusively to intracellular metabolites, termed lipoidal estrogens [long chain fatty acid 17 beta-esters (E2-L)], by human mammary cancer tissue and cell lines. In order to further evaluate the biological role of lipoidal estrogens, rates of saturation of the estrogen receptor (ER) along with formation of [3H]E2-L have been measured in human mammary cancer cells exposed to 5 nM [3H]E2. Extensive specific binding of E2 to ER in MCF-7 cells (approximately 37%) and ZR-75-1 cells (approximately 62%) occurred before appreciable synthesis of E2-L was evident and

Crohn's disease: a review of treatment options and current research.

Crohn's disease is an autoimmune disorder that affects nearly 1.4 million Americans. The etiology of Crohn's disease is not completely understood, however, research has suggested a genetic link. There is currently no known cure for Crohn's disease and, as a result, most government-funded research is being conducted to increase the quality of life of afflicted patients (i.e. reducing chronic inflammation and alleviating growth impairment in pediatric patients). A number of treatment options are available including an alpha-4 integrin inhibitor and several TNF-alpha inhibitors. Furthermore, rese

Deficiency of tetralinoleoyl-cardiolipin in Barth syndrome.

Barth syndrome is an X-linked cardiac and skeletal mitochondrial myopathy. Barth syndrome may be due to lipid alterations because the product of the mutated gene is homologous to phospholipid acyltransferases. Here we document that a single mitochondrial phospholipid species, tetralinoleoyl-cardiolipin, was lacking in the skeletal muscle (n = 2), right ventricle (n = 2), left ventricle (n = 2), and platelets (n = 6) of 8 children with Barth syndrome. Tetralinoleoyl-cardiolipin is specifically enriched in normal skeletal muscle and the normal heart. These findings support the notion that Barth

Metabolomic Atlas of Cardiovascular Diseases: Mapping Shared and Specific Signatures.

BACKGROUND: Cardiovascular disease (CVD) remains the leading global cause of death. While metabolic dysregulation is central to CVD pathogenesis, the extent to which distinct clinical subtypes exhibit unique or shared metabolic signatures remains unclear. OBJECTIVES: The purpose of this study was to systematically characterize metabolomic patterns across a broad spectrum of CVD subtypes and to delineate both shared and subtype-specific metabolic features. METHODS: We analyzed nuclear magnetic resonance-based metabolomic data (325 metabolites) from 244,567 UK Biobank participants, including 2

Antioxidant and Antimicrobial Activities of Essential Oil Extracted from Leaves of African Boxthorn (Lycium ferocissimum): In-Vitro and In-Silico Approaches.

The African boxthorn (Lycium ferocissimum) is a medicinal plant belonging to the Solanaceae family that is used in traditional pharmacopeia. Here, its biological potential was assessed by analyzing the essential oil from its leaves (EOLF), extracted by hydro-distillation and major terpenes characterized by gas chromatography coupled to mass spectrometry (GC-MS). GC-MS analysis of EOLF identified fourteen compounds representing 99.95% of the total mass, which was dominated by carvacrol (35.96%) and sabinene (17.06%). EOLF exhibited antioxidant activity as characterized by several assays, includ

Exploring the Regulatory Effect of LPJZ-658 on Copper Deficiency Combined with Sugar-Induced MASLD in Middle-Aged Mice Based on Multi-Omics Analysis.

Globally, metabolic dysfunction-associated steatotic liver disease (MASLD), previously termed nonalcoholic fatty liver disease (NAFLD), is one of the most common liver disorders and is strongly associated with copper deficiency. To explore the potential effects and mechanisms of Lactiplantibacillus plantarum LPJZ-658, copper deficiency combined with a high-sugar diet-induced MASLD mouse model was utilized in this study. We fed 40-week-old (middle-aged) male C57BL/6 mice a copper-deficient and high-sugar diet for 16 weeks (CuDS), with supplementary LPJZ-658 for the last 6 weeks (CuDS + LPJZ-658

Alcohol and Prostate Cancer: Time to Draw Conclusions.

It has been a long-standing debate in the research and medical societies whether alcohol consumption is linked to the risk of prostate cancer (PCa). Many comprehensive studies from different geographical areas and nationalities have shown that moderate and heavy drinking is positively correlated with the development of PCa. Nevertheless, some observations could not confirm that such a correlation exists; some even suggest that wine consumption could prevent or slow prostate tumor growth. Here, we have rigorously analyzed the evidence both for and against the role of alcohol in PCa development.

Larvicidal activity of Ricinus communis extract against mosquitoes.

BACKGROUND & OBJECTIVES: Vector control strategies play significant role in reducing the transmission of malaria, dengue and other vector-borne diseases. The control of vector population using synthetic insecticides has resulted in development of insecticide resistance and negative effects on humans and environment. The present investigation evaluated the larvicidal potential of methanol, dichloromethane and hexane extracts of leaves and seeds of Ricinus communis (castor) plant against the early IV instar larvae of the dengue vector, Aedes aegypti, and malaria vector, Anopheles culicifacies.

Quantification of fatty acid ethyl esters (FAEE) and ethyl glucuronide (EtG) in meconium from newborns for detection of alcohol abuse in a maternal health evaluation study.

Fatty acid ethyl esters (FAEE) and ethyl glucuronide (EtG) were determined in 602 meconium samples in a maternal health evaluation study for detection of gestational alcohol consumption. A validated headspace solid phase microextraction method in combination with GC-MS was used for FAEE and the cumulative concentration of ethyl palmitate, ethyl linoleate, ethyl oleate, and ethyl stearate with a cut-off of 500 ng/g was applied for interpretation. A new and simple method was developed and validated for quantification of EtG from 10-20 mg meconium with D(5)-EtG as internal standard consisting of

The effects of glucagon-like peptide-1 receptor agonists on chemosensory function and ingestive behavior.

Glucagon-like peptide-1 receptor agonists (GLP-1RAs) reduce energy intake and produce clinically meaningful weight loss, but the chemosensory and behavioral contributions to these effects remain poorly characterized. This study examined whether GLP-1RA use was associated with differences in chemosensory function and ingestive behavior in adults with obesity. In this cross-sectional study, 79 adults with obesity were categorized as "pre-use": individuals eligible for GLP-1RA treatment but not yet using one (n = 27), 1-month GLP-1RA users (n = 25), or 6-month GLP-1RA users (n = 27). Investigating changes in serum metabolome and urinary endocrine disrupting chemicals in cats with hyperthyroidism.

Domestic cats share indoor environments with humans and are exposed to endocrine-disrupting chemicals (EDCs) from both household sources and cat-specific products capable of disrupting thyroid hormone signaling. The prevalence of feline hyperthyroidism (FHT) continues to rise, and while some EDCs have been implicated in its etiopathogenesis, the metabolic consequences of FHT are unknown. Here, we tested whether hyperthyroid cats exhibit altered systemic metabolomic signatures that are associated with phthalate and paraben urinary levels, compared with healthy controls. Thirty-five pet cats wer

Mendelian randomization study: Metabolites as mediators of inflammatory factors in ulcerative colitis.

Ulcerative colitis (UC) involves dysregulated immune responses and metabolic reprogramming, yet the causal mechanisms linking inflammatory mediators to UC via metabolic intermediates remain elusive. This study employs integrated Mendelian randomization (MR) and mediation analysis to dissect the immune-metabolic axis, a novel conceptual framework for UC pathogenesis, where inflammatory factors exert causal effects through metabolite-mediated pathways. Using European genetic data (5931 UC cases; 405,386 controls), we performed bidirectional 2-sample MR to assess causal relationships between 91 i
