De Novo Acute Myeloid Leukemia with Combined CBFB-MYH11 and BCR-ABL1 Gene Rearrangements: A Case Report and Review of Literature.

Acute myeloid leukemia (AML) with inv(16)(p13.1q22) resulting in CBFB-MYH11 fusion is associated with a favorable prognosis. The presence of a KIT mutation modifies it to an intermediate prognosis. Additionally, inv(16) can cooperate with other genetic aberrations to further increase cell proliferation. Coexistence of inv(16) and t(9;22) is extremely rare (20 cases). We present a case of a 55-year-old male with elevated white blood cell count. Bone marrow evaluation and flow cytometry analysis were compatible with AML with monocytic features. Cytogenetic studies revealed two-related clones, a

Fungal infections of the central nervous system and paranasal sinuses in onco-haematologic patients. Epidemiological study reporting the diagnostic-therapeutic approach and outcome in 89 cases.

Invasive fungal infections (IFI) of the Central Nervous System (IFI-CNS) and Paranasal Sinuses (IFI-PS) are rare, life-threatening infections in haematologic patients, and their management remains a challenge despite the availability of new diagnostic techniques and novel antifungal agents. In addition, analyses of large cohorts of patients focusing on these rare IFI are still lacking. Between January 2010 and December 2016, 89 consecutive cases of Proven (53) or Probable (36) IFI-CNS (71/89) and IFI-PS (18/89) were collected in 34 haematological centres. The median age was 40 years (range 5-7

Deep cutaneous fungal infections in immunocompromised children.

BACKGROUND: Life-threatening infections from ubiquitous fungi are becoming more prevalent in adults and children because of the increased use of immunosuppressive agents and broad-spectrum anti-infective drugs. Extremely low birth weight premature neonates and patients with a disrupted epidermal barrier are also at increased risk. Lethality is high, particularly with delayed diagnosis. As cutaneous lesions are often the first manifestation of such infections, early recognition of suspicious lesions is crucial to decrease associated morbidity and mortality. The clinical features of deep cutaneo

Treatment of fungal keratitis by penetrating keratoplasty.

AIM: To study the use of penetrating keratoplasty (PKP) for the treatment of severe fungal keratitis that could not be cured by antifungal medication. METHODS: A retrospective analysis of 108 cases of severe fungal keratitis in which PKP was performed served as the basis for this study. Fungal keratitis was diagnosed by KOH staining of corneal scrapings or by confocal microscopic imaging of the cornea. All patients received a combination of topical and oral antifungal medicines without steroids as the first course of therapy. Patients whose corneal infection was not cured or in whom the infec

Plasma pre-treatment and peroxynitric acid soaking as a novel oxidative seed disinfection strategy against seed-borne fungal pathogens in maize.

INTRODUCTION: Seed-borne fungal contamination is an important challenge in maize production and may contribute to grain deterioration and mycotoxin contamination. This study evaluated peroxynitric acid (PNA), alone and in combination with nonequilibrium gaseous plasma pretreatment, as a rapid oxidative strategy for disinfecting maize seeds contaminated with Aspergillus flavus and Fusarium fujikuroi. METHODS: Maize seeds were artificially inoculated with approximately 10⁶ spores of either A. flavus or F. fujikuroi. For plasma pretreatment, seeds were sequentially exposed to hydrogen (0.2 s) an

Optimization of fermentation conditions for Metarhizium robertsii and its effect on rice growth.

UNLABELLED: Metarhizium robertsii ARSEF 23 is a significant insect-pathogenic fungus and plant-associated endophyte, with dual roles as an insect-control agent and plant growth-enhancing agent that have fueled its extensive use in agricultural practices. However, the growth-promoting impacts of M. robertsii (referred to as MAA) on rice, and their underlying mechanisms are still not fully clarified. This research verified the growth-enhancing effects of both MAA itself and its fermented supernatant on rice plants. By means of single-factor tests and response surface methodology, the fermentatio

Identification of pathogenic fungi causing ocular infections using full rRNA operon sequencing with Oxford Nanopore Technologies.

Although fungal eye infections are a major cause of visual impairment worldwide, standard clinical laboratory methods remain slow, insensitive, and limited in their taxonomic resolution. Sequencing of the full ribosomal RNA (rRNA) operon provides a comprehensive marker for fungal identification. In this study, twenty fungal isolates associated with ocular infections were obtained from Srinagarind Hospital, Thailand, and characterized using four identification approaches. Initial hospital-based routine identification relied on conventional morphological methods and matrix-assisted laser desorpt

Invasive fungal infections in critically ill children: epidemiology, risk factors and antifungal drugs.

BACKGROUND: Invasive fungal infections (IFIs) are important infectious complications amongst critically ill children. The most common fungal infections are due to Candida species. Aspergillus, Zygomycetes and Fusarium are also emerging because of the empirical use of antifungal drugs. This updated review discusses the epidemiology of IFIs as well as antifungal drugs, dosing and potential adverse effects in critically ill children. METHODS: A PubMed search was conducted with Clinical Queries using the key terms "antifungal", "children", "critical care" AND "paediatric intensive care unit" OR "

Debaryomyces hansenii Strains Isolated From Danish Cheese Brines Act as Biocontrol Agents to Inhibit Germination and Growth of Contaminating Molds.

The antagonistic activities of native Debaryomyces hansenii strains isolated from Danish cheese brines were evaluated against contaminating molds in the dairy industry. Determination of chromosome polymorphism by use of pulsed-field gel electrophoresis (PFGE) revealed a huge genetic heterogeneity among the D. hansenii strains, which was reflected in intra-species variation at the phenotypic level. 11 D. hansenii strains were tested for their ability to inhibit germination and growth of contaminating molds, frequently occurring at Danish dairies, i.e., Cladosporium inversicolor, Cladosporium si

Topical application of acidified nitrite to the nail renders it antifungal and causes nitrosation of cysteine groups in the nail plate.

BACKGROUND: Topical treatment of nail diseases is hampered by the nail plate barrier, consisting of dense cross-linked keratin fibres held together by cysteine-rich proteins and disulphide bonds, which prevents penetration of antifungal agents to the focus of fungal infection. Acidified nitrite is an effective treatment for tinea pedis. It releases nitric oxide (NO) and other NO-related species. NO can react with thiol (-SH) groups to form nitrosothiols (-SNO). OBJECTIVES: To determine whether acidified nitrite can penetrate the nail barrier and cure onychomycosis, and to determine whether ni

Repression of deoxynivalenol-triggered cytotoxicity and apoptosis by mannan/β-glucans from yeast cell wall: Involvement of autophagy and PI3K-AKT-mTOR signaling pathway.

Deoxynivalenol (DON) is the most common trichothecene distributed in food and feed. So far, much work has focused on investigating the cytotoxicity of DON, while there is few researches aimed at intervening in the toxic impacts on humans and livestock posed by DON. The objective of this study is to investigate the underlying mechanism of biomacromolecules mannan/β-glucans from yeast cell wall (BYCW) for their potency to impede the cytotoxicity and apoptosis caused by DON with porcine jejunum epithelial cell lines (IPEC-J2) used as a cell injury model. We analyzed the cell morphology, cell acti

Fabrication of a drug delivery system that enhances antifungal drug corneal penetration.

Fungal keratitis (FK) remains a severe eye disease, and effective therapies are limited by drug shortages and critical ocular barriers. Despite the high antifungal potency and broad spectrum of econazole, its strong irritant and insolubility in water hinder its ocular application. We designed and fabricated a new drug delivery system based on a polymeric vector for the ocular antifungal application of econazole. This novel system integrates the advantages of its constituent units and exhibits superior comprehensive performance. Using the new system, drug content was significantly increased mor

Effects of different amendment treatments on survival rate and soil microbial community of continuous cropping Atractylodes macrocephala.

Prolonged continuous cropping of Atractylodes macrocephala usually cause soil-borne diseases such as root rot, resulting in significant yield losses even total crop failure. Burning, fumigation, and microbial inoculant application are commonly used to suppress soil-borne diseases in agricultural systems. However, their effects on plant performance and soil microbial communities in continuously cropped A. macrocephala systems remain poorly understood. We conducted a field experiment with high-throughput amplicon sequencing to examine the effects of burning, soil fumigation, microbial inoculant

Microbial keratitis at a single tertiary eye care in Andhra Pradesh: A 12-year analysis of microbiological profile and in vitro susceptibility of bacterial isolates.

CONTEXT: Microbial keratitis is a potentially vision-threatening emergency that demands prompt diagnosis and treatment to ensure better vision recovery. Empirical use of broad-spectrum antimicrobial therapies is often the primary treatment modality, which is then tailored according to clinical response, cultured organisms, and their sensitivities. Since the spectrum of microbial agents associated with corneal ulcers is wide and varies from one geographical location to another, it is imperative that local microbial trends and susceptibility patterns are analyzed to enable better care in the man
