Influence of Polycyclic Aromatic Compounds and Oxidation States of Soot Organics on the Metabolome of Human-Lung Cells (A549): Implications for Vehicle Fuel Selection.

Decades of research have established the toxicity of soot particles resulting from incomplete combustion. However, the unique chemical compounds responsible for adverse health effects have remained uncertain. This study utilized mass spectrometry to analyze the chemical composition of extracted soot organics at three oxidation states, aiming to establish quantitative relationships between potentially toxic chemicals and their impact on human alveolar basal epithelial cells (A549) through metabolomics-based evaluations. Targeted analysis using MS/MS indicated that particles with a medium oxidat

The Epithelial-to-Mesenchymal Transition-Like Process Induced by TGF-β1 Enhances Rubella Virus Binding and Infection in A549 Cells via the Smad Pathway.

Virus-host cell interactions in rubella virus (RuV) are of great interest in current research in the field, as their mechanism is not yet well understood. By hypothesizing that the epithelial-to-mesenchymal transition (EMT) may play a role in RuV infection, this study aimed to investigate the influence of TGF-β1-induced EMT of human lung epithelial A549 cells on the infectivity of RuV. A549 cells were cultured and treated with TGF-β1 for 1 to 2 days prior to virus infection (with a clinical strain). Viral infectivity was determined by flow cytometry analysis of cells harvested at 24 and 48 h p

Synergetic and Antagonistic Molecular Effects Mediated by the Feedback Loop of p53 and JNK between Saikosaponin D and SP600125 on Lung Cancer A549 Cells.

We jointly analyzed the changes in cell cycle arrest and distribution, the accumulation of subphase cells, apoptosis, and proliferation in A549 cells treated with Saikosaponin D (Ssd) and JNK inhibitor SP600125 alone or in combination. Our results indicated that cell cycle arrest at G0/G1, S, and G2/M phases was coupled with the accumulation of subG1, subS, and subG2 cells, corresponding to early apoptosis, DNA endoreplication, and later inhibitory proliferation, respectively. Analyzing the expression of 18 cell cycle regulatory genes and JNK and phosphorylated JNK (pJNK) levels revealed an en

The Radiobiological Effect of Flattening Filter Free (FFF) Beam Delivery Parameters on A549 and MCF-7 Cell Survival: An In Vitro Study.

BACKGROUND: Recent improvements, such as flattening filter-free (FFF) beams, enable the delivery of treatments at higher dose rates and in shorter times. However, the biological influence still remains uncertain. Here, we examined how dose rate and dose per pulse influence responses to FFF beams compared with flattened beams in cancer cells with different radiosensitivities. METHODS: A549 (lung, high α/β) and MCF-7 (breast, low α/β) cells were irradiated using VMAT (6 MV FF, 6 and 10 MV FFF; 600-2400 MU/min; 5-20 Gy). Cell viability was assessed by the WST-1 assay, and reproductive survival b

Acylhydrazone-based ferrocene-iridium(III) heteronuclear metal complexes: In vivo/in vitro anticancer evaluation towards A549 cell lines.

Iridium(III) complexes and ferrocenyl derivatives have recently emerged as promising alternatives to platinum-based anticancer agents, owing to their distinctive anticancer mechanisms. This study presented the creation of six acylhydrazone-based ferrocene‑iridium(III) heteronuclear complexes, each featuring polymetallic core, despite the inherent challenges in the design and synthesis of polynuclear metal complexes. Among these, IrFP6, which integrating two iridium(III) centers with a ferrocene unit, demonstrated superior bioactivity, surpassing that of cisplatin, and showing efficacy against

An A549 Cell-Based Approach Using Repeated Fluorescence Readouts for Assessing Reactive Oxygen Species Activity of Atmospheric Particulate Matter.

Exposure to atmospheric particulate matter (PM) is a major public-health concern, in part because PM can perturb cellular redox homeostasis. This study evaluates an in vitro A549/DCFH-DA approach using repeated fluorescence readouts to assess PM2.5-induced oxidative activity. Untreated and assay-specific controls were combined with zymosan and NIST Standard Reference Material® 2584 suspended in PBS, and fluorescence was monitored at multiple readout times over a 15 min-6 h window. Method performance was characterized using the coefficient of variation (CV) and signal-to-noise ratio (SNR). A de

Process development for high-titer production of adenovirus devoid of replication-competent particles in suspension-adapted complementing A549 cell culture.

Adenovirus is one of the most attractive viral vectors for therapeutic vaccines and gene therapy with the caveat that replication-competent adenoviruses (RCA) can be produced. To remediate this problem, engineered A-549 adenoviral vector complementing cells (SF-BMAdR cells) were previously generated by our organization for the production of E1-deleted adenoviral vectors without RCA. However, the production process remained to be improved for high titer production and scalability, as cost-effective and scalable biomanufacturing processes are critical for commercializing adenovirus-based vaccine

Ability of plasma-activated acetated Ringer's solution to induce A549 cell injury is enhanced by a pre-treatment with histone deacetylase inhibitors.

Non-thermal plasma (NTP) is applicable to living cells and has emerged as a novel technology for cancer therapy. NTP affect cells not only by direct irradiation, but also by an indirect treatment with previously prepared plasma-activated liquid. Histone deacetylase (HDAC) inhibitors have the potential to enhance susceptibility to anticancer drugs and radiation because these reagents decondense the compact chromatin structure by neutralizing the positive charge of the histone tail. The aim of the present study was to demonstrate the advantage of the combined application of plasma-activated acet
