Lower Epigenetic Age Acceleration in Older Adults with Preserved Global Cognitive Performance.

Epigenetic clocks estimate biological age, yet evidence linking decelerated epigenetic aging to healthy cognitive performance in community-dwelling older adults is limited. Cross-sectional study within the population-based ALEXANDROS cohort (Santiago, Chile). Healthy Cognitive Agers (HCA) were participants aged ≥80 years who consistently maintained a MMSE score =30 across all cohort assessments. Controls were cognitively normal peers from the same cohort (MMSE 28.5 ± 2.9). Epigenetic age was estimated using the Horvath, GrimAge, and PhenoAge clocks. Epigenetic age acceleration (EAA) was calcul

Pedal acceleration time-Guided management of diabetic foot ulcers with borderline ischemia.

Background: The management of diabetic foot ulcers (DFUs) with borderline ischemia remains challenging due to the absence of reliable physiological markers to guide revascularization decisions. Pedal acceleration time (PAT) is a Doppler-derived measure of distal perfusion that may improve risk stratification. This study aimed to evaluate a PAT-guided management strategy in patients with DFUs and borderline perfusion by assessing its ability to guide treatment allocation, and its association with wound healing and amputation-free survival (AFS). Methods: In this prospective quasi-experimental s

Biological aging acceleration associated with electronic cigarette use in the United States.

BACKGROUND: Smoking combustible cigarettes accelerates biological aging and contributes to subsequent aging-related cardiovascular decline. However, the aging-related health consequences of electronic cigarette (e-cigarette) use (vaping) remain largely unknown. This study aims to investigate whether e-cigarette use is associated with biological aging acceleration using a nationally representative sample. METHODS: This cross-sectional study included 8,595 participants aged 20-79 from two cycles (2015-2016 and 2017-2018) of the National Health and Nutrition Examination Survey. Biological aging

Acceleration of autofocusing algorithms for fielding solid targets at high-repetition-rates.

High-energy-density science experiments utilizing high-power laser systems increasingly operate at high repetition rates (HRRs), demanding automated and rapid target focusing to maintain data throughput. We present a robust autofocusing framework and a direct experimental validation of its performance. The system utilizes a dichroic mirror to collinearly align a continuous-wave 532 nm probe laser with the main beam, focusing both via an off-axis parabola to a <10 μm spot. A focal spot monitor positioned along the optical path captures back-reflected images of the probe light as a target is sca

Healthy Eating Index, Epigenetic Age Acceleration and Mortality Risk in US Adults.

This study examined associations between diet quality, epigenetic age acceleration (EAA), and mortality in two U.S. cohorts: NHANES (n = 2158) and HRS (n = 1752), while accounting for demographic and socioeconomic (SES) determinants. Diet was evaluated as a potentially modifiable exposure within broader social and biological pathways. Participants were linked to the National Death Index. Cox proportional hazards models, additive Bayesian networks (ABN), generalized structural equation models (GSEM), and four-way decomposition were used to estimate direct, indirect, and interaction effects, wit

Biological Age Acceleration Predicts Functional Decline and Surgical Risk in Knee Arthroplasty.

BACKGROUND: Chronological age is widely used in perioperative risk assessment, but does not adequately capture interindividual differences in physiological reserve. Recently developed phenotypic biological aging measures quantify biological age using clinical biomarkers and may better reflect frailty and surgical vulnerability. This study compared the predictive value of biological and chronological age for functional and perioperative outcomes in patients undergoing total knee arthroplasty (TKA). METHODS: We retrospectively analyzed 173 consecutive adults who had osteoarthritis undergoing TK

Spatial extension of parafoveal processing predicts reading speed gain during reading acceleration.

Fast and efficient reading involves not only processing the currently fixated word but also simultaneous processing of the upcoming text. Although the role of extrafoveal processing in natural reading has been extensively documented, its contribution to reading acceleration benefits has remained unexplored. In the present study, we sought to investigate whether reading acceleration might act by influencing the efficiency and spatial extent of parafoveal processing. Participants read at either a natural or an accelerated pace while the availability of parafoveal information was independently ma

Predictors of pelvic acceleration during treadmill running across various stride frequency conditions.

Pelvic running injuries often require extensive rehabilitation and pelvic girdle pain is a barrier to running engagement in population sub-groups, such as perinatal women. However, exploration into how external pelvic loading may be altered during running is limited. This study assessed which biomechanical variables influence changes in external peak pelvic acceleration during treadmill running, across various stride frequency conditions. Twelve participants (7 female, 5 male) ran (9 km∙h-1) at their preferred stride frequency, and at ± 5% and ± 10% of their preferred stride frequency. Coordin

Internal sensory models allow for balance control using muscle spindle acceleration feedback.

Motor control requires sensory feedback, and the nature of this feedback has implications for the tasks of the central nervous system (CNS): for an approximately linear mechanical system (e.g., a freely standing person, a rider on a bicycle), if the sensory feedback does not contain the state variables (i.e., joint position and velocity), then optimal control actions are based on an internal dynamical system that estimates these states from the available incomplete sensory feedback. Such a computational system can be implemented as a recurrent neural network (RNN), and it uses a sensory model

L-Type Cav1.3 and HCN Channels Mediate Heart Rate Acceleration by Catecholamines.

BACKGROUND: The ionic mechanism by which catecholamines increase the heart rate is incompletely understood. In this study, we have assessed the roles of sinoatrial node L-type Cav1.3 (α1D) Ca2+ channels, phosphorylation of L-type channel regulatory partner protein Rad (Ras-related RGK GTP-binding protein), and cAMP-dependent regulation of hyperpolarization-activated HCN (hyperpolarization-activated cyclic nucleotide-gated) channels. METHODS: We studied β-adrenergic regulation of heart rate and sinoatrial pacemaker activity in mice lacking Cav1.3 channels and in mice expressing dihydropyridine

PIGK defects induce apoptosis in Purkinje cells and acceleration of neuroectodermal differentiation.

Biallelic mutations in PIGK cause GPI biosynthesis defect 22 (GPIBD22), characterized with developmental delay, hypotonia, and cerebellar atrophy. The understanding of the underlying causes is limited due to the lack of suitable disease models. To address this gap, we generated a mouse model with PIGK deficits, specifically in Purkinje cells (Pcp2-cko) and an induced pluripotent stem cell (iPSC) model using the c.87dupT mutant (KI) found in GPIBD22 patients. Pcp2-cko mice demonstrated cerebellar atrophy, ataxia and progressive Purkinje cells loss which were accompanied by increased apoptosis a
