Action potential duration restitution kinetics in human atrial fibrillation.

OBJECTIVES: We undertook this study to determine whether human atrial fibrillation (AF) relates to steeply sloped action potential duration restitution (APDR) kinetics and whether the spatial nonuniformity of APDR promotes persistence of AF. BACKGROUND: A steeply sloped APDR curve is known to be an important determinant of the induction of more complex action potential duration (APD) dynamics and fibrillation. METHODS: Patients with chronic atrial fibrillation (CAF) (n = 18), paroxysmal atrial fibrillation (PAF) (n = 14) and normal control subjects (n = 9) were studied. The monophasic action

BAG-1M: a potential specificity determinant of corticosteroid receptor action.

BAG-1M is a eukaryotic cochaperone that associates with several proteins, including the glucocorticoid receptor (GR). It down-regulates GR-mediated transactivation by a mechanism that requires its prior recruitment by the liganded receptor from cytoplasm into the nucleus. In the nucleus, it uses a repeated sequence motif ([EEX4]8) at its NH2 terminus to inhibit DNA binding, as well as transactivation functions of the receptor. The mineralocorticoid receptor (MR), a structural and functional homologue of the GR, is unable to translocate BAG-1M into the nucleus, and its transactivation function

Potential sites of action of TNPA: a dopamine-2 receptor agonist.

The purpose of this study was to correlate potential mechanisms with site(s) of action for TNPA-induced ocular hypotension. In response to R(-)-2, 10, 11-trihydroxy-N-propyl-noraporphine hydrobromide (TNPA, 75 microg), a D2 dopamine receptor agonist, the intraocular pressure decreased by 4.5 and 8 mm Hg at 1 and 2 hr, respectively, as measured by pneumatonometry. The levels of norepinephrine in aqueous humor, as determined by high performance liquid chromatography with electrochemical detection, were reduced by 38% and 79% at 1 and 2 hr, respectively, following topical application of TNPA (75

Normethylclozapine potentiates the action of quinpirole in the 6-hydroxydopamine lesioned rat.

Systemic administration of the 5-HT2c receptor antagonist, normethylclozapine, to previously untreated unilaterally 6-hydroxydopamine-lesioned rats elicits rotations contraversive to the lesion when given with a subthreshold dose of the dopamine D2 receptor agonist, quinpirole. Normethylclozapine alone does not possess any anti-parkinsonian action. In animals which have previously received dopamine replacement therapy, i.e. primed, normethylclozapine potentiates the contraversive rotations induced by quinpirole. We speculate that these actions may result from reduced excitation of the output r

The effects of contralateral noise on masked compound action potential in humans.

The effects of contralateral noise on the masked compound action potential (CAP) were examined in human subjects. Small but significant enhancements of masked CAP amplitude (anti-masking effects) were observed in some patients with facial palsy, as well as in some subjects with healthy ears when noise was added to the contralateral ear. There were no significant differences in fractional changes of CAP by the addition of contralateral noise between subjects with healthy ears and patients with facial palsy in which acoustic reflexes of middle ear muscles (MEMs) had disappeared or were impaired.

Specificity of the anticollagenase action of tetracyclines: relevance to their anti-inflammatory potential.

The concentrations of doxycycline and 4-de-dimethylaminotetracycline required to inhibit 50% of collagenase activity were found to be 15 to 30 microM for human neutrophil and gingival crevicular fluid collagenases. Fibroblast collagenase was relatively resistant to inhibition by tetracyclines; the 50% inhibitory concentrations of doxycycline and 4-de-dimethylaminotetracycline were 280 and 510 microM, respectively.

FOXO family and neurodegenerative diseases: Mechanisms of action and therapeutic potential.

Neurodegenerative diseases (NDDs) lack effective disease-modifying therapies. The FOXO transcription factors serve as integrative hubs of cellular stress responses, operating through cell-autonomous homeostasis, intercellular coordination, intracellular quality control, and cell fate decisions four hierarchical tiers. This review systematically examines isoform-specific functions: FOXO1 governs metabolic reprogramming and mitochondrial biogenesis; FOXO3 acts as the principal oxidative stress sensor with context-dependent neuroprotective or pro-apoptotic outputs; FOXO4 regulates cellular senesc

[LSD revisited: Mechanisms of action and therapeutic potential in mental health].

Major depressive disorder (MDD) represents a colossal worldwide burden of health, affecting more than 350 million individuals. While traditional antidepressants with mechanisms of action at the serotonin, dopamine, or norepinephrine pathways are routinely prescribed, approximately one-third of patients fail to achieve adequate remission and develop treatment-resistant depression (TRD). This therapeutic shortfall underscores the necessity for novel approaches to treatment. Over the past two decades there has been a resurgence of scientific interest in psychedelics, such as lysergic acid diethyl

Exploiting autophagy-targeting natural compounds for potential antimicrobial actions.

Natural products are biologically active compounds used for therapeutic interventions for various diseases, particularly infections. Autophagy is an intracellular catabolic pathway involving lysosomal degradation and is closely associated with immunological pathways, effectively combating bacterial, viral, fungal, and parasitic infections. Accumulating evidence suggests that autophagy activation or inhibition by natural products promotes antimicrobial responses against various pathogens. Numerous natural products can modulate autophagy through diverse signaling pathways, suggesting their poten

Action potential: Impact of climate change on neurological disease in Ireland.

Anthropogenic climate change has led to a significant increase in global average temperatures and the frequency of extreme heat events. Rising temperatures have a broad range of health implications and have the potential to significantly impact neurological disorders and brain health. A growing body of evidence suggests that the incidence and severity of many common neurological conditions (stroke, epilepsy, headache, neuroinflammation, and neurodegenerative disorders) are affected by climate change. Ireland's average temperature is rising, and we are likely to experience extreme heat events m

Elamipretide: A Review of Its Structure, Mechanism of Action, and Therapeutic Potential.

Mitochondria serve an essential metabolic and energetic role in cellular activity, and their dysfunction has been implicated in a wide range of disorders, including cardiovascular conditions, neurodegenerative disorders, and metabolic syndromes. Mitochondria-targeted therapies, such as Elamipretide (SS-31, MTP-131, Bendavia), have consequently emerged as a topic of scientific and clinical interest. Elamipretide has a unique structure allowing for uptake in a variety of cell types and highly selective mitochondrial targeting. This mitochondria-targeting tetrapeptide selectively binds cardiolipi

Antifungal potential, mechanism of action, and toxicity of 1,4-naphthoquinone derivatives.

BACKGROUND: The rising prevalence of fungal infections and challenges such as adverse effects and resistance against existing antifungal agents have driven the exploration of new antifungal substances. METHODS: We specifically investigated naphthoquinones, known for their broad biological activities and promising antifungal capabilities. It specifically examined the effects of a particular naphthoquinone on the cellular components of Candida albicans ATCC 60193. The study also assessed cytotoxicity in MRC-5 cells, Artemia salina, and the seeds of tomatoes and arugula. RESULTS: Among four tes

Opioids and Immunosuppression: Clinical Evidence, Mechanisms of Action, and Potential Therapies.

BACKGROUND: In addition to the more well-known adverse effects of opioids, such as constipation, mounting evidence supports underlying immunosuppressive effects as well. METHODS: In this study, we provide a narrative review of preclinical and clinical evidence of opioid suppression of the immune system as well as possible considerations for therapies. RESULTS: In vitro and animal studies have shown clear effects of opioids on inflammatory cytokine expression, immune cell activity, and pathogen susceptibility. Observational data in humans have so far supported preclinical findings, with multi

Electrocardiographic marker of ventricular action potential triangulation (the simulation study).

BACKGROUND: The aim of this simulation was to examine the utility of a novel ECG-based index of cardiac action potential (AP) triangulation, the Tstart-to-Tpeak (TsTp) interval-to-JTstart (JTs) interval ratio, for assessment of changes in AP profile imposed through variations in the duration of the plateau phase and the phase 3 repolarization. METHODS: ECGs were simulated using a realistic rabbit model based on experimental data. The AP plateau was measured at APD30, and the phase 3 was assessed as APD90-to-APD30 difference (AP durations at 90 % and 30 % repolarization, respectively). AP tria

The mechanical properties of nerves, the size of the action potential, and consequences for the brain.

The action potential is widely regarded as a purely electrical phenomenon. However, one also finds mechanical and thermal changes that can be observed experimentally. In particular, nerve membranes become thicker and axons contract. The spatial length of the action potential can be quite large, ranging from millimeters to many centimeters. This suggests the use of macroscopic thermodynamics methods to understand its properties. The pulse length is several orders of magnitude larger than the synaptic gap, larger than the distance of the nodes of Ranvier and even larger than the size of many neu
