Impaired Insulin Action Is Associated With Increased Glucagon Concentrations in Nondiabetic Humans.

CONTEXT: Abnormal glucagon concentrations contribute to hyperglycemia, but the mechanisms of α-cell dysfunction in prediabetes are unclear. OBJECTIVE: We sought to determine the relative contributions of insulin secretion and action to α-cell dysfunction in nondiabetic participants across the spectrum of glucose tolerance. DESIGN: This was a cross-sectional study. A subset of participants (n = 120) was studied in the presence and absence of free fatty acid (FFA) elevation, achieved by infusion of Intralipid (Baxter Healthcare, Deerfield, IL) plus heparin, to cause insulin resistance. Characterization of a Bacteriocin-like Substance from Loigolactobacillus coryniformis WYQ-2 and Its Antibacterial Activity Against Escherichia coli.

Escherichia coli is a common porcine diarrheal pathogen transmitted via the fecal-oral route through contaminated environments and feed. This study evaluated the antimicrobial potential of a bacteriocin-like substance produced by Loigolactobacillus coryniformis WYQ-2 isolated from pickled vegetables. Its physicochemical stability, molecular mass, and antimicrobial spectrum were characterized, and the mode of action against Escherichia coli was investigated through intracellular ATP quantification, live/dead staining, and scanning electron microscopy (SEM). The substance exhibited broad-spectru

Paradoxical action of zolpidem: interplay between dysregulation of the synergetic actions of γ-aminobutyric acid type A receptors and neuronal cotransporters (KCC2/NKCC1).

Zolpidem, or commercially known as Ambien or Stilnox, is a sedative-hypnotic agent, which is usually prescribed to manage sleeping difficulties in individuals with insomnia. The site of its sedative-hypnotic action is the γ-aminobutyric acid type A receptor, which it shares with benzodiazepines. However, this substance has been consistently associated to awaken patients with brain injuries such as trauma, stroke, disorders of consciousness and has also been expanded to the recovery of brain function in the event of hypoxic damage, cerebrovascular ischemic injury, infection of the central nervo

The neuroscience of action semantics in neurodegenerative brain diseases.

PURPOSE OF REVIEW: Within the last decade, the neurobiology of action processing has moved from an obscure topic of specialist interest to one of the most popular themes in modern neuroscience. However, the wealth of literature and the diversity of approaches and theoretical models can make the field complex and, at times, bewildering. This review presents the main currents of research, examining their theoretical underpinnings in an interdisciplinary context. RECENT FINDINGS: The presence of specific deficits in verb and action processing has been documented in a wide range of neurodegenerat

Mechanism of tetracycline phototoxicity.

Studies were made to determine factors important in the phototoxicity mechanism of 7 clinically used tetracyclines (TC). The clinical phototoxicity, the rates of photochemical degradation, and the in vitro phototoxicity of the TCs were qualitatively but not quantitatively correlated. Phototoxicity in vitro was partially oxygen-dependent and possibly singlet oxygen is involved. The contribution of photoproducts to the phototoxic process may be the basis for the reported differences between the in vivo action spectrum and the absorption spectrum of demethylchlorotetracycline. A mechanistic model

Sulfonated (1 → 6)-β-d-Glucan (Lasiodiplodan): A Promising Candidate against the Acyclovir-Resistant Herpes Simplex Virus Type 1 (HSV-1) Strain.

Herpes simplex virus type 1 (HSV-1) is a persistent human pathogen, and the emergence of strains resistant to Acyclovir (ACV, reference drug) shows the urgency to develop new treatments. We report the antiherpetic mechanism of the action of lasiodiplodan (LAS-N, (1 → 6)-β-d-glucan) and its sulfonated derivative (LAS-S3) in vitro and in vivo. LAS-S3 showed anti-HSV-1 action with high selectivity indices for HSV-1 KOS (88.1) and AR (189.2), sensitive and resistant to ACV, respectively. LAS-S3 inhibited >80% of HSV-1 infection in different treatment protocols (virucidal, adsorption inhibition,

Pharmacological and neurochemical properties of 1,4-diazepines with two annelated heterocycles ('hetrazepines').

1,4-Diazepines with two annelated heterocycles ('hetrazepines') such as brotizolam (WE 941), WE 973 and WE 1008 bind with high affinities to benzodiazepine receptors in the central nervous system. Brotizolam has a pharmacologic spectrum of action similar to clinically useful benzodiazepines, while the closely related derivatives WE 973 and WE 1008 appear to lack hypnotic action. Unlike other benzodiazepine receptor ligands which share common pharmacologic properties with the benzodiazepines, the apparent affinities of WE 973 and WE 1008 are not increased significantly in the presence of GABA,

Antimicrobial Peptides Derived from Bacteria: Classification, Sources, and Mechanism of Action against Multidrug-Resistant Bacteria.

Antimicrobial peptides (AMPs) are short, usually cationic peptides with an amphiphilic structure, which allows them to easily bind and interact with the cellular membranes of viruses, bacteria, fungi, and other pathogens. Bacterial AMPs, or bacteriocins, can be produced from Gram-negative and Gram-positive bacteria via ribosomal synthesis to eliminate competing organisms. Bacterial AMPs are vital in addressing the increasing antibiotic resistance of various pathogens, potentially serving as an alternative to ineffective antibiotics. Bacteriocins have a narrow spectrum of action, making them hi

ISG20: an enigmatic antiviral RNase targeting multiple viruses.

Interferon-stimulated gene 20 kDa protein (ISG20) is a relatively understudied antiviral protein capable of inhibiting a broad spectrum of viruses. ISG20 exhibits strong RNase properties, and it belongs to the large family of DEDD exonucleases, present in both prokaryotes and eukaryotes. ISG20 was initially characterized as having strong RNase activity in vitro, suggesting that its inhibitory effects are mediated via direct degradation of viral RNAs. This mechanism of action has since been further elucidated and additional antiviral activities of ISG20 highlighted, including direct degradation

Fungicidal activity of a phospholipase-A2-derived synthetic peptide variant against Candida albicans.

pEM-2, a 13-mer synthetic peptide variant derived from myotoxin II, a phospholipase A(2) homologue present in Bothrops asper snake venom, has shown potent bactericidal activity in previous studies due to the combination of cationic and hydrophobic amino acids, including three tryptophan-substituted residues in its sequence. This study reports that pEM-2 also exerts potent fungicidal activity against a variety of clinically relevant Candida species, killing 100% of yeasts at concentrations near 10 mg/l (5 microM), as indicated by plate counting assays. Thus, this peptide displays a broad-spectr

Combination chemotherapy of dental plaque infections.

Chemotherapy against dental plague micro-organisms has proved effective in combatting both caries and periodontal disease. Chlorhexidine is the drug of choice when a broad-spectrum antiplaque agent is called for. However, because its mechanism of action is unspecific and the high concentrations needed for maximum effect may cause side effects, combination chemotherapy has been suggested. Chlorhexidine has been successfully combined with fluoride and, more recently, with xylitol. The combination allows the use of less concentrated solutions than if the agents were administered alone. Further, t

Molecular mechanisms of action of ribavirin.

Ribavirin (1-beta-D-ribofuranosyl-1,2,4-triazole-3-carboxamide) is a broad-spectrum antiviral agent whose molecular mode of action remains remarkably controversial. This antiviral agent was approved by the U.S. Food and Drug Administration in 1986 for use as an aerosol for infants with serious infections due to respiratory syncytial virus. Ribavirin is and has been under clinical investigation for activity against a variety of viral illnesses, including those due to influenza virus, Lassa fever virus, Hantaan virus, and human immunodeficiency virus (HIV). There has been a great deal of clinica

Paraneoplastic Autoimmune Multiorgan Syndrome: Diagnostic Lessons for Pediatricians.

PURPOSE: Persistence of skin and wound infections is nowadays accepted being linked to bacterial biofilms, which are highly recalcitrant to treatments and contribute to maintain a constant inflammation state and prevent a correct healing. Topical antimicrobials are the most common first-line self-medications; however, treatment failure is not uncommon and emerging resistance to antibiotics is alarming. Chlorquinaldol is an antimicrobial with a wide spectrum of activity and desirable characteristics for topical application. Aim of this study was to evaluate the efficacy of chlorquinaldol to pre

Search for effective bacteriolytic enzymes and their combinations for the treatment of polymicrobial biofilm-associated infections.

Polymicrobial biofilms resistant to antibiotic therapy are often associated with different severe human diseases, including skin and soft tissue injuries, local mucosal and dental infections, post-traumatic and chronic osteomyelitis, and implant-associated infections. A possible treatment option for these challenging infections may be the use of enzybiotics containing bacteriolytic enzymes (or lysins). However, elimination of polymicrobial infections requires an effective combination of enzymes to expand the narrow activity spectrum of individual lysins and to ensure successful treatment. We s

The epidemiology of UV induced skin cancer.

There is persuasive evidence that each of the three main types of skin cancer, basal cell carcinoma (BCC), squamous cell carcinoma (SCC) and melanoma, is caused by sun exposure. The incidence rate of each is higher in fairer skinned, sun-sensitive rather than darker skinned, less sun-sensitive people; risk increases with increasing ambient solar radiation; the highest densities are on the most sun exposed parts of the body and the lowest on the least exposed; and they are associated in individuals with total (mainly SCC), occupational (mainly SCC) and non-occupational or recreational sun expos

Development of Novel Membrane Disrupting Lipoguanidine Compounds Sensitizing Gram-Negative Bacteria to Antibiotics.

A new class of amphiphilic molecules, the lipoguanidines, designed as hybrids of guanidine and fatty acid compounds, has been synthesized and developed. The new molecules present both a guanidine polar head and a lipophilic tail that allow them to disrupt bacterial membranes and to sensitize Gram-negative bacteria to the action of the narrow-spectrum antibiotics rifampicin and novobiocin. The lipoguanidine 5g sensitizes Klebsiella pneumonia, Acinetobacter baumannii, Pseudomonas aeruginosa, and Escherichia coli to rifampicin, thereby reducing the antibiotic minimum inhibitory concentrations (MI

[Antifungal agents in the treatment of systemic infections: Relevance of mechanism of action, activity profile and resistances].

The availability of different therapeutic alternatives has modified the treatment of systemic fungal infections. There commendations of antifungal therapy vary according to species which causes the mycosis and its susceptibility. Consequently, the knowledge of action mechanism, activity profile and resistances to antifungal agents are essential for the clinical practice. Amphotericin B is the antifungal agent exhibiting the broadest spectrum of activity, it is a fungicidal drug and resistances have been hardly ever described. The triazoles compounds also have a broad spectrum, but their massiv
