Theileria sergenti and T. buffeli: polymerase chain reaction-based marker system for differentiating the parasite species from infected cattle blood and infected tick salivary gland.

Benign Theileria species in cattle. Theileria sergenti and T. buffeli, are morphologically indistinguishable. The polymerase chain reaction (PCR) was used to amplify the genes encoding the 33- and 34-kDa major piroplasm antigens (p33/34) of T. sergenti and T. buffeli from cattle blood infected with these parasites and tick salivary gland infected with T. sergenti. Following amplification, the p33 gene from T. sergenti and the p34 gene from T. buffeli were clearly differentiated using the restriction enzyme sites that were not shared between them. The oligonucleotide primer set, designed from t

Identification of Rickettsia spp., Anaplasma spp., and an Ehrlichia canis-like agent in Rhipicephalus microplus from Southwest and South-Central China.

Rhipicephalus microplus is considered to be the most important tick infesting cattle, buffalo, horse, goats as well as other animals. They transmit diseases between domestic animals and act as vectors of a variety of zoonotic pathogens. Although pathogens harbored by R. microplus have been extensively studied, the Rickettsiales pathogens vectored by R. microplus in some areas of China remained largely unexplored. From August to October 2020, a total of 291 R. microplus ticks were collected from goats and cattle in three Southern China provinces, Guangxi (n = 138), Sichuan (n = 120) and Hubei (

Molecular prevalence and diagnostic gene targets of Anaplasma, Babesia, Theileria, and Trypanosoma in cattle and buffaloes across Southeast Asia (2015-2025): A systematic review and meta-analysis.

BACKGROUND AND AIM: Hemoparasitic infections caused by Anaplasma, Babesia, Theileria, and Trypanosoma adversely affect cattle and buffalo health, resulting in economic losses and reduced livestock productivity in Southeast Asia. Although numerous molecular epidemiological studies have been conducted in individual countries, a comprehensive regional synthesis of polymerase chain reaction (PCR)-confirmed prevalence and molecular diagnostic targets has been lacking. This study aimed to estimate the pooled molecular prevalence of major hemoparasites in cattle and buffaloes across Southeast Asia, s

Serologic survey for selected arboviruses and other potential pathogens in wildlife from Mexico.

During 1988 and 1989, a serologic survey of wildlife was conducted in northeastern Mexico to determine the presence, prevalence, and distribution of arboviruses and other selected disease agents. Eighty mammal specimens were tested. Antibodies to vesicular stomatitis-Indiana, Venezuelan equine encephalitis-Mena II, Rio Grande virus, and vesicular stomatitis-New Jersey were detected predominantly in small mammals. Deer and mouflon (Ovis musimon) had antibodies to bluetongue and epizootic hemorrhagic disease. Two species had serologic evidence of recent exposure to Francisella tularensis. A whit

Demonstration of transplacental transmission of a human isolate of Anaplasma phagocytophilum in an experimentally infected sheep.

Anaplasma phagocytophilum, first identified as a pathogen of sheep in Europe, more recently has been recognized as an emerging tick-borne pathogen of humans in the U.S. and Europe. Transmission of A. phagocytophilum is reported to be by ticks, primarily of the genus Ixodes. While mechanical and transplacental transmission of the type genus organism, A. marginale, occur in addition to tick transmission, these modes of transmission have not been considered for A. phagocytophilum. Recently, we developed a sheep model for studying host-tick-pathogen interactions of the human NY-18 A. phagocytophil

Comparative moleculo-immunological analysis of swamp- and riverine-type water buffaloes responses.

This moleculo-epidemiological and immunological study through cytokine response assessment was done to know the dynamics of cytokines in the initiation, persistence and association to physiological changes of a particular pathogen in water buffaloes. This is important to understand the magnitude and behavior of disease progression. Water buffalo blood samples gathered from different places in the Philippines revealed a 9.4%, 27.6%, 10.3% and 4.4% prevalence of bovine viral diarrhea virus (BVDV), bovine leukemia virus (BLV), Anaplasma marginale and Babesia bigemina infection, respectively. Detection of Multiple Microorganisms in Ruminant Ticks in Senegal Using High-Throughput Microfluidic Real-Time PCR.

Ticks are major vectors of numerous pathogens affecting both livestock and humans. In Senegal, data on the diversity of tick-borne pathogens (TBPs) in ruminant-associated ticks remain limited. In total, 1703 ticks were collected from goats, sheep, and cattle across three ecological zones of Senegal (Sudanian, Sahelian, and Sudano-Sahelian). Tick species were identified morphologically, and 300 individuals were screened for 36 microorganisms using a high-throughput microfluidic real-time PCR system. DNA was successfully extracted and amplified from 289 ticks. The most abundant species were Rhip

Evaluation of the immunodiagnostic potential of recombinant Spm2 antigen using indirect ELISA for detecting Theileria annulata infection in cattle.

Bovine tropical theileriosis, caused by Theileria annulata, is an economically important disease that reduces livestock productivity through decreased milk yield, high morbidity, and mortality, particularly in exotic and cross-bred cattle. Timely diagnosis and treatment are crucial for effectively managing susceptible animals. This study aimed to develop a robust serological diagnostic tool targeting the sporozoite and macroschizont 2 antigen (Spm2), a protein expressed across multiple parasite stages including sporozoites, macroschizonts, and piroplasms. The recombinant Spm2 (rSpm2) protein w

Simultaneous detection of bovine Theileria and Babesia species by reverse line blot hybridization.

A reverse line blot (RLB) assay was developed for the identification of cattle carrying different species of Theileria and Babesia simultaneously. We included Theileria annulata, T. parva, T. mutans, T. taurotragi, and T. velifera in the assay, as well as parasites belonging to the T. sergenti-T. buffeli-T. orientalis group. The Babesia species included were Babesia bovis, B. bigemina, and B. divergens. The assay employs one set of primers for specific amplification of the rRNA gene V4 hypervariable regions of all Theileria and Babesia species. PCR products obtained from blood samples were hyb

Molecular characterization of the recombinant protein RmLTI-BmCG-LTB: Protective immunity against Rhipicephalus (Boophilus) microplus.

The bovine tick Rhipicephalus (Boophilus) microplus is found in several tropical and subtropical regions of the world. This parasite transmits pathogens that cause disease, such as babesiosis (Babesia bovis and B. bigemina) and anaplasmosis (Anaplasma marginale). Tick infestations cause enormous livestock losses, and controlling tick infestations and the transmission of tick-borne diseases remains a challenge for the livestock industry. Because the currently available commercial vaccines offer only partial protection against R. (B.) microplus, there is a need for more efficient vaccines. Sever

Detection of Babesia bovis carrier cattle by using polymerase chain reaction amplification of parasite DNA.

Carrier cattle infected with Babesia bovis are difficult to detect because of the low numbers of parasites that occur in peripheral blood. However, diagnosis of low-level infections with the parasite is important for evaluating the efficacies of vaccines and in transmission and epidemiological studies. We used the polymerase chain reaction (PCR) to amplify a portion of the apocytochrome b gene from the parasite and tested the ability of this method to detect carrier cattle. The target sequence is associated with a 7.4-kb DNA element in undigested B. bovis genomic DNA (as shown previously), and

Bovine ticks harbour a diverse array of microorganisms in Pakistan.

BACKGROUND: Ticks and tick-borne pathogens (TTBP) are a major constraint to livestock production in Pakistan; despite a high prevalence of TTBPs, knowledge on the capacity of Pakistani ticks to carry pathogens and endosymbionts is limited. Furthermore, mixed infections with multiple microorganisms further complicate and limit the detection potential of traditional diagnostic methods. The present study investigated the tick-borne microorganisms in bovine ticks in Pakistan, employing a high-throughput microfluidic real-time PCR based technique. METHODS: Ticks were collected from clinically heal
