Preserved functional organization of human auditory cortex in individuals missing one temporal lobe from infancy.

Human cortical responses to natural sounds, measured with fMRI, can be approximated as the weighted sum of a small number of canonical response patterns (components), each having interpretable functional and anatomical properties. Here, we asked whether this organization is preserved in cases where only one temporal lobe is available due to early brain damage by investigating a unique family: one sibling born without a left temporal lobe, another without a right temporal lobe, and a third anatomically neurotypical. We analyzed fMRI responses to diverse natural sounds within the intact hemisphe

Ipsilateral auditory cortex responses to the intact ear after unilateral noise trauma in juvenile rats.

BACKGROUND: While ear stimulation produces a robust response in the contralateral auditory cortex (AC), it produces only a weak response in the ipsilateral AC, known as interhemispheric asymmetry. Unilateral deafness can lead to AC plastic changes, resulting in reduced interhemispheric asymmetry and auditory perceptual consequences. However, the unilateral hearing loss-associated plastic changes are far from fully understood. The purpose of this study was to investigate AC responses to the ipsilateral unimpaired ear after noise injury to the contralateral ear in juvenile rats. METHODS: Rats (

Electrophysiological evaluation of high and low-frequency transcranial random noise stimulation over the auditory cortex.

BACKGROUND: Transcranial random noise stimulation (tRNS) is a non-invasive brain stimulation technique which uses electrical alternating currents applied at random frequencies. Besides the ability to alter cortical excitability, past research demonstrated that high-frequency tRNS over the auditory cortex can modulate both spontaneous and auditory evoked oscillatory brain activity. OBJECTIVES: The aim of the present study was to examine the effects of high- and low-frequency auditory tRNS on EEG power and evoked activity. METHODS: Low-frequency (0.1-100Hz), high-frequency (100-640Hz) and sham

Functional organization of mouse primary auditory cortex in adult C57BL/6 and F1 (CBAxC57) mice.

The primary auditory cortex (A1) plays a key role for sound perception since it represents one of the first cortical processing stations for sounds. Recent studies have shown that on the cellular level the frequency organization of A1 is more heterogeneous than previously appreciated. However, many of these studies were performed in mice on the C57BL/6 background which develop high frequency hearing loss with age making them a less optimal choice for auditory research. In contrast, mice on the CBA background retain better hearing sensitivity in old age. Since potential strain differences could

Go/No-Go task engagement enhances population representation of target stimuli in primary auditory cortex.

Primary sensory cortices are classically considered to extract and represent stimulus features, while association and higher-order areas are thought to carry information about stimulus meaning. Here we show that this information can in fact be found in the neuronal population code of the primary auditory cortex (A1). A1 activity was recorded in awake ferrets while they either passively listened or actively discriminated stimuli in a range of Go/No-Go paradigms, with different sounds and reinforcements. Population-level dimensionality reduction techniques reveal that task engagement induces a s

[Functional evaluation of auditory cortex by functional magnetic resonance before cochlear implantation: a pilot study].

OBJECTIVE: To investigate the functional evaluation of auditory cortex by functional magnetic resonance imaging (fMRI) before cochlear implantation (CI). METHODS: Twenty-one children with severe to profound sensorineural hearing loss (SNHL) (7 cases with normal inner ear structure, 3 cases with large vestibular aqueduct syndrome, 1 case with cochlear nerve hypoplasia) and 7 children with normal hearing were examined by fMRI under sedation. The subjects received acoustic stimuli at four random frequencies (500, 1 000, 2 000, 4 000 Hz) during fMRI examination. Tones were interleaved with silenc

Brief hearing loss disrupts binaural integration during two early critical periods of auditory cortex development.

Early binaural experience can recalibrate central auditory circuits that support spatial hearing. However, it is not known how binaural integration matures shortly after hearing onset or whether various developmental stages are differentially impacted by disruptions of normal binaural experience. Here we induce a brief, reversible unilateral conductive hearing loss (CHL) at several experimentally determined milestones in mouse primary auditory cortex (A1) development and characterize its effects ~1 week after normal hearing is restored. We find that CHL shapes A1 binaural selectivity during tw

Activation of human auditory cortex during speech perception: effects of monaural, binaural, and dichotic presentation.

We used a rapid event-related functional magnetic resonance imaging (fMRI) paradigm to compare cortical activation to speech tokens presented monaurally to each ear, binaurally, and dichotically. Two forms of dichotic conditions were examined: one presented consonant-vowel (CV) syllables simultaneously to each ear while the other paired a CV syllable with a non-speech stimulus (band-limited noise). Right-handed adults were asked to differentially respond to serially presented target and distractor CV syllables. Activations were localized with reference to anatomic segmentation algorithms that

Early neural dysfunction reflected in degraded auditory cortex responses in pre-regression heterozygous Mecp2 rats.

Rett syndrome, a genetic disorder caused by mutations in the X-linked Mecp2 gene, is characterized by typical early development followed by rapid developmental regression between 6 and 18 months of age. Affected individuals exhibit seizures, cognitive impairments, motor deficits, and difficulties in speech-language processing. Post-regression rodent models of Rett syndrome have been observed to follow similar regression, presenting sensory processing difficulties during auditory discrimination tasks, as well as degraded auditory cortical responses. However, little is known about the auditory p

Auditory cortex ensembles jointly encode sound and locomotion speed to support sound perception during movement.

The ability to process and act upon incoming sounds during locomotion is critical for survival and adaptive behavior. Despite the established role that the auditory cortex (AC) plays in behavior- and context-dependent sound processing, previous studies have found that auditory cortical activity is on average suppressed during locomotion as compared to immobility. While suppression of auditory cortical responses to self-generated sounds results from corollary discharge, which weakens responses to predictable sounds, the functional role of weaker responses to unpredictable external sounds during

Neonatal Hypoxia-Ischemia Causes Persistent Intracortical Circuit Changes in Layer 4 of Rat Auditory Cortex.

The connection between early brain injury and subsequent development of disorders is unknown. Neonatal hypoxia-ischemia (HI) alters circuits associated with subplate neurons (SPNs). SPNs are among the first maturing cortical neurons, project to thalamorecipient layer 4 (L4), and are required for the development of thalamocortical connections. Thus, early HI might influence L4 and such influence might persist. We investigated functional circuits to L4 neurons in neonatal rat HI models of different severities (mild and moderate) shortly after injury and at adolescence. We used laser-scanning pho

Different Excitation-Inhibition Correlations Between Spontaneous and Tone-evoked Activity in Primary Auditory Cortex Neurons.

Tone-evoked synaptic excitation and inhibition are highly correlated in many neurons with V-shaped tuning curves in the primary auditory cortex of pentobarbital-anesthetized rats. In contrast, there is less correlation between spontaneous excitation and inhibition in visual cortex neurons under the same anesthetic conditions. However, it was not known whether the primary auditory cortex resembles visual cortex in having spontaneous excitation and inhibition that is less correlated than tone-evoked excitation and inhibition. Here we report whole-cell voltage-clamp measurements of spontaneous ex

Modified synaptic dynamics predict neural activity patterns in an auditory field within the frontal cortex.

Frontal areas of the mammalian cortex are thought to be important for cognitive control and complex behaviour. These areas have been studied mostly in humans, non-human primates and rodents. In this article, we present a quantitative characterization of response properties of a frontal auditory area responsive to sound in the brain of Carollia perspicillata, the frontal auditory field (FAF). Bats are highly vocal animals, and they constitute an important experimental model for studying the auditory system. We combined electrophysiology experiments and computational simulations to compare the r

Memantine Attenuates Salicylate-induced Tinnitus Possibly by Reducing NR2B Expression in Auditory Cortex of Rat.

Memantine, a noncompetitive antagonist of the N-methyl-D-aspartate (NMDA) receptor, suppresses the release of excessive levels of glutamate that may induce neuronal excitation. Here we investigated the effects of memantine on salicylate-induced tinnitus model. The expressions of the activity-regulated cytoskeleton-associated protein (ARC) and tumor necrosis factor-alpha (TNFα) genes; as well as the NMDA receptor subunit 2B (NR2B) gene and protein, were examined in the SH-SY5Y cells and the animal model. We also used gap-prepulse inhibition of the acoustic startle reflex (GPIAS) and noise burst

Organization of rodent auditory cortex: anterograde transport of PHA-L from MGv to temporal neocortex.

In the present study we analyzed the organization of the thalamocortical projections of the specific auditory relay nucleus of the thalamus, the ventral division of the medial geniculate body (MGv), using the anterograde axonal tracer Phaseolus vulgaris leucoagglutinin. All injections of MGv produced dense labeling of axonal fibers in temporal cortex. In all cases, labeled axons were predominantly concentrated in cortical layers III and IV and, to a lesser extent, at the junction of layers V and VI. Injections confined to the medial regions of MGv, and specifically to the ovoid nucleus of MGv

Auditory thalamic reticular nucleus of the rat: anatomical nodes for modulation of auditory and cross-modal sensory processing in the loop connectivity between the cortex and thalamus.

The auditory sector of the thalamic reticular nucleus (TRN) plays a pivotal role in gain and/or gate control of auditory input relayed from the thalamus to cortex. The TRN is also likely involved in cross-modal sensory processing for attentional gating function. In the present study, we anatomically examined how cortical and thalamic afferents intersect in the auditory TRN with regard to these two functional pathways. Iontophoretic injections of biocytin into subregions of the auditory TRN, which were made with the guidance of electrophysiological recording of auditory response, resulted in re

Altered dynamic functional connectivity of auditory cortex and medial geniculate nucleus in first-episode, drug-naïve schizophrenia patients with and without auditory verbal hallucinations.

BACKGROUND AND OBJECTIVE: As a key feature of schizophrenia, auditory verbal hallucination (AVH) is causing concern. Altered dynamic functional connectivity (dFC) patterns involving in auditory related regions were rarely reported in schizophrenia patients with AVH. The goal of this research was to find out the dFC abnormalities of auditory related regions in first-episode, drug-naïve schizophrenia patients with and without AVH using resting state functional magnetic resonance imaging (rs-fMRI). METHODS: A total of 107 schizophrenia patients with AVH, 85 schizophrenia patients without AVH (NA

Cell-type-specific reorganization of VGSCs in auditory cortex and therapeutic potential of Nav1.6 blockade for tinnitus.

Neuronal hyperexcitability resulting from an inhibitory-excitatory imbalance in the primary auditory cortex (A1) is a key pathological feature of tinnitus. Voltage-gated sodium channels (VGSCs) are crucial in regulating neuronal excitability by facilitating action potential generation and propagation. However, the specific involvement of VGSC subtypes in tinnitus-related hyperexcitability within the A1 cortex remains poorly understood. Previous studies have shown that acute and chronic salicylate administration can induce stable tinnitus in rats. In this study, we investigated the distribution

Syngap1 regulates the synaptic drive and membrane excitability of Parvalbumin-positive interneurons in mouse auditory cortex.

SYNGAP1 haploinsufficiency-related intellectual disability (SYNGAP1-ID) is characterized by moderate to severe ID, generalized epilepsy, autism spectrum disorder, sensory processing dysfunction, and other behavioral abnormalities. While numerous studies have highlighted a role of Syngap1 in cortical excitatory neurons development, recent studies suggest that Syngap1 plays a role in GABAergic inhibitory neuron development as well. However, the molecular pathways by which Syngap1 acts on GABAergic neurons, and whether they are similar or different from the mechanisms underlying its effects in ex

Perinatal methimazole exposure impairs the distribution and function of layer 5 neurons in the mouse auditory cortex.

Methimazole (MMI) is an antithyroid drug often prescribed for hyperthyroid conditions. While perinatal MMI exposure is used to model hypothyroidism in rodents, resulting in auditory cortex malformation, mice are reportedly less effective in lowering serum thyroxine levels. This raises the question on MMI-induced hypothyroidism being the underlying cause of cortical malformation. Here we examined if and how perinatal MMI exposure in mice influenced the serum thyroxine level and the distribution and function of deep layer projection neurons in the auditory cortex. MMI exposure resulted in little
