Preliminary Structural Characterization and Bioactivity of Exopolysaccharides Produced by Sanghuangporus vaninii (Agaricomycetes).

Sanghuangporus vaninii produces bioactive polysaccharides with therapeutic potential, but the structural features and anti-obesity mechanisms of its exopolysaccharides (EPS) remain unclear. Two polysaccharide fractions (Fr-I and Fr-II) have been purified from the crude EPS of S. vaninii via gel filtration chromatography. The structural characterization of the two fractions was comprehensively characterized at multiple levels, employing SEC/MALLS, GC and FT-IR. SEC/MALLS indicated their average molecular weights (Mw) have been 3.162 ȕ 106 (Fr-I) and 2.613 ȕ 106 (Fr-II). Both fractions exhibited

Deciphering the Molecular Interplay Between Wheat and Puccinia striiformis f. sp. Tritici: Mechanisms of Interaction and Resistance Strategies.

Wheat stripe rust, caused by the obligate biotroph Puccinia striiformis f. sp. tritici (Pst), remains one of the most destructive fungal diseases threatening global wheat production. Beyond traditional control through chemical application and resistant cultivar breeding, rapid advances in molecular biology have substantially reshaped our understanding of the intricate molecular dialogue between wheat and Pst. In recent years, research has increasingly focused on Pst pathogenicity factors, wheat immune perception, and the multilayered signaling networks that orchestrate defense responses. Parti

Two ancient nuclear lineages and divergent reproductive strategies drive global success of the wheat stripe rust pathogen.

Plant pathogen evolution is driven by complex biological and ecological processes with major consequences for food security. Understanding how genetic diversity arises and is maintained is critical for predicting disease emergence. The wheat stripe rust fungus Puccinia striiformis f. sp. tritici is among the world's most destructive crop pathogens. As a dikaryotic fungus harboring two distinct haploid nuclei, Puccinia striiformis f. sp. tritici offers an excellent model to investigate nuclear-level evolution. Here, using population genomics of 507 global isolates and haplotype-phased genomes,

Modulation in physicochemical properties and digestibility of potato starch by ethanol-fractionated polysaccharides from Phlebopus portentosus.

In this study, the effects of Phlebopus portentosus polysaccharides on the physicochemical properties and digestibility of potato starch (PS) were systematically evaluated. Alkali-extracted polysaccharides from Phlebopus portentosus (APBP) were fractionated into three subfractions (APBP-30, APBP-70, and APBP-80) through gradient ethanol precipitation. These subfractions differed in monosaccharide composition and molecular weight. Further, PS-APBP subfraction composites were prepared and investigated for their physicochemical and structural properties, as well as in vitro digestibility. Pasting

BN-PAGE of Ustilago maydis Respiratory Complexes.

Isolated mitochondria are valuable for the study of the respiratory chain, protein complex assembly, and the activities of the Krebs cycle enzymes, among others. Here, we present a protocol for the isolation of functional U. maydis mitochondria, which involves mechanically disrupting the cell wall with glass beads and performing two centrifugations. The first step is carried out at low speed to remove intact cells and cell debris, followed by a high-speed centrifugation to obtain mitochondria. For the separation of respiratory complexes and supercomplexes, we describe the solubilization of pro

Mycotoxins in Basidiomycete Mycelium and Fruiting Bodies: Implications for Eu Novel Food Assessment and Selected International Regulatory Frameworks.

Advances in technology have enabled the efficient and sustainable production of basidiomycete vegetative mycelium, particularly from Pleurotus species, as a novel food source. However, mycelium has yet to receive regulatory approval in many global markets, requiring a better understanding of factors affecting its safety. Yet, various studies on different species and mycotoxins suggest that mycelium may be safer than its fruiting body. This review explores the occurrence and distribution of basidiomycete mycotoxins (BMTs) across different species and fungal structures, with a particular focus o

Wheat Germ Agglutinin-Alexa Fuor488/Propidium Iodine Costaining and Fluorescence Microscopic Imaging of Ustilago maydis Infected Maize Anthers.

Ustilago maydis infects all aerial maize tissues, forming tumors and altering host development. However, most published methods focus on seedling leaf infections in order to understand the interaction between maize and U. maydis. Cytological analysis, particularly fluorescence microscopy, is vital not only for examining fungal infection progression, but also for understanding the host's response. Thus, a detailed protocol is presented here for dissecting U. maydis-infected maize anthers, followed by costaining with wheat germ agglutinin-Alexa Fluor 488 (WGA-AF488) and propidium iodide (PI). Immunodetection of Cell-Surface-Associated Proteins in Ustilago maydis and Infected Maize Leaves.

Immunolocalization relies on the sequential binding of a specific primary antibody to its target antigen, followed by detection with a fluorophore-conjugated secondary antibody to visualize the subcellular localization of proteins. This technique enables precise and high-resolution mapping of protein localization and distribution patterns after secretion. This chapter presents a detailed protocol for the immunolocalization of secreted proteins from Ustilago maydis, both in vitro and in infected maize leaves.

Antifungal activity and function mechanisms of chitooligosaccharide against Gymnosporangium pleoporum, the pathogen causing rust in Juniperus przewalskii.

Juniperus przewalskii, a keystone species in China's ecologically fragile Sanjiangyuan region, faces severe threats from Gymnosporangium pleoporum rust. This study establishes chitooligosaccharides (COS) as potent antifungal agents against G. pleoporum, dose-dependently suppressing teliospore germination and viability. Integrated physiological analyses revealed COS-induced severe membrane damage evidenced by electrolyte leakage, soluble protein efflux, SEM-confirmed structural deformities, and elevated malondialdehyde indicating lipid peroxidation. Concurrently, COS triggered oxidative catastr

Constituents from the Polar Extract of Pisolithus arhizus and Their Anti-inflammatory Activity.

The phytochemical study of the Pisolithus arhizus fruiting body methanol extract led to the isolation of six new triterpenoids (1-6) and one new naphthalenoid pulvinic acid derivative (7), together with five known compounds, including norbadione A (8). Their structure was established from 1D and 2D NMR spectroscopy and HRESIMS analyses. The absolute configuration of the triterpenoids was determined by circular dichroism. The two pulvinic acid derivatives 7 and 8, showing the highest activity in modulating IL-6 secretion, were tested for their effect on COX-2, STAT3, and p-STAT3 proteins; both

Piriformospora indica colonization enhances remediation of cadmium and chromium co-contaminated soils by king grass through plant growth promotion and rhizosphere microecological regulation.

Poor plant growth and low pollutant bioavailability in contaminated soils limit phytoremediation efficiency. Pot experiments were conducted to investigate the effects and mechanisms of Piriformospora indica inoculation on the phytoremediation of Cd-Cr co-contaminated soils from farmland using king grass. P. indica colonization increased plant biomass by 20.4-24.6% and enhanced Cd/Cr accumulation in root, stem and leave tissues. Root vascular cylinder and cortex were the major structures for Cd/Cr transportation in plants. The amounts of Cd and Cr extracted by king grass considerably increased

Imaging of long-distance alpha-aminoisobutyric acid translocation dynamics during resource capture by Serpula lacrymans.

alpha-Aminoisobutyric acid (AIB) is a nonmetabolized amino acid analogue of alanine, which at low (muM) concentrations acts as a tracer for amino acid movements. At high concentrations (mM), it competitively inhibits membrane transport and metabolism of protein amino acids and acts as a systemic translocated inhibitor of mycelial extension in fungi. AIB can control mycelial spread of the basidiomycete Serpula lacrymans, the cause of brown rot of wood in buildings. However, it is not known how effectively the inhibitor is distributed throughout the mycelium. Realistically heterogeneous microcos

Zinc phosphate transformations by the Paxillus involutus/pine ectomycorrhizal association.

In this research, we investigate zinc phosphate transformations by Paxillus involutus/pine ectomycorrhizas using zinc-resistant and zinc-sensitive strains of the ectomycorrhizal fungus under high- and low-phosphorus conditions to further understand fungal roles in the transformation of toxic metal minerals in the mycorrhizosphere. Mesocosm experiments with ectomycorrhizas were performed under sterile conditions with zinc phosphate localized in cellophane bags: zinc and phosphorus mobilization and uptake by the ectomycorrhizal biomass were analyzed. In the presence of a phosphorus source, an ec

Effects of available water on growth and competition of southern pine beetle associated fungi.

Competitive, interactions among bark beetle associated fungi are potentially influenced by abiotic factors. Water potential, in particular, undergoes marked changes over the course of beetle colonization of tree hosts. To investigate the impact of water potential on competition among three southern pine beetle associated fungi, Ophiostoma minus, Entomocorticium sp. A and Ceratocystiopsis ranaculosus, we utilized artificial media with water potentials of 0, -5, -10, and -20 MPa. Growth of all three fungi, when grown alone, decreased on media with lower water potentials. Growth rates of all thre

Listeria monocytogenes septicemia associated with consumption of salted mushrooms.

Listeria monocytogenes septicemia in an 80-year-old man is described. On the day before clinical symptoms appeared the patient had eaten homemade salted mushrooms, rufous milkcap (Lactarius rufus Fr.). L. monocytogenes serotype 4b was isolated in blood cultures. The mushrooms which had been stored in cold for 5 months before consumption contained the same listeria serotype at a level of 10(6) CFU/g. Salt content (NaCl) of the mushrooms was 7.5%. Fever and diarrhea disappeared with penicillin therapy and the patient was discharged after 4 weeks in the hospital.
