Efficacy of intravitreal bevacizumab for stage 3+ retinopathy of prematurity.

BACKGROUND: Retinopathy of prematurity is a leading cause of childhood blindness worldwide. Peripheral retinal ablation with conventional (confluent) laser therapy is destructive, causes complications, and does not prevent all vision loss, especially in cases of retinopathy of prematurity affecting zone I of the eye. Case series in which patients were treated with vascular endothelial growth factor inhibitors suggest that these agents may be useful in treating retinopathy of prematurity. METHODS: We conducted a prospective, controlled, randomized, stratified, multicenter trial to assess intra

Intravitreal bevacizumab in retinopathy of prematurity: an interventional case series.

BACKGROUND: To evaluate the results of intravitreal bevacizumab in terms of regression of retinopathy of prematurity (ROP). METHODS: In this interventional case series, 12 eyes from January 2007 to December 2008 were included. Two groups of patients were considered. Group 1 included those ROP patients who did not respond to conventional laser therapy within 3 weeks or had some complications (hyphema or vitreous hemorrhage) after conventional laser therapy; intravitreal injection (0.625 mg) was performed as an adjuvant therapy. Group 2 included those with aggressive posterior ROP who received

On safety, pharmacokinetics and dosage of bevacizumab in ROP treatment - a review.

UNLABELLED: Off-label intravitreal use of the vascular endothelial growth factor (VEGF) antibody bevacizumab for retinopathy of prematurity (ROP) increases despite lack of studies on safety, pharmacokinetics and dosage in developing individuals. Systemic absorption has been considered negligible. A literature search was performed with emphasis on potential adverse systemic effects in developing individuals. CONCLUSION: Intravitreal bevacizumab enters the general circulation, suppresses plasma VEGF levels and remains in the blood for more than 8 weeks in primates. Possible adverse effects on V

Intravitreal bevacizumab (avastin) as an adjuvant for the treatment of posterior scleritis.

We report a case of posterior scleritis effectively managed with intravitreal bevacizumab. A 71-year-old woman was diagnosed with posterior scleritis. Although she was initially treated with systemic steroids, her clinical presentation deteriorated. She was then treated with a single intravitreal injection of bevacizumab and aqueous humor collection. The aqueous level of vascular endothelial growth factor prior to the intravitreal injection was 880.51 pg/mL, greater than that in the healthy control group (p < 0.001). One month later, the scleritis was completely resolved, and the patient remai

Reversible posterior leukoencephalopathy syndrome in a child treated with bevacizumab.

Bevacizumab is a monoclonal antibody targeting vascular endothelial growth factor (VEGF). Hypertension is a well-recognized, common side effect of VEGF blocking agents. The reversible posterior leukoencephalopathy syndrome (RPLS) has been described as a rare but serious consequence of bevacizumab administration. We present a case of a 6-year-old child with refractory hepatoblastoma who developed hypertensive crisis, seizures and MRI changes consistent with RPLS while receiving bevacizumab with gemcitabine and oxaliplatin. Findings completely resolved without neurologic sequelae with stringent

Bevacizumab (Avastin) as a potential novel adjunct in the management of pterygia.

In the last two decades of angiogenesis research, a variety of angiogenic regulators have been identified. The discovery of several antiangiogenic factors has led to the development of novel ophthalmic therapies. Pterygia are characterized by the encroachment of a fleshy fibrovascular tissue from the bulbar conjunctiva onto the cornea. The pathogenesis of pterygia is presently uncertain and its treatment is quiet controversial. It has also been postulated that the development of pterygia depends on a changed angiogenic stimulator-to-inhibitor ratio. On of the most important known mediators of

Bevacizumab in the treatment of breast cancer: rationale and current data.

Vascular endothelial growth factor (VEGF) has emerged as a key target for the treatment of cancer. As the ligand to the VEGF receptor, it plays a central role in promoting tumor angiogenesis. Overexpression of VEGF leads to poor outcomes in patients with breast cancer and other tumors. Preclinical studies have shown that the humanized monoclonal antibody to VEGF, bevacizumab (Avastin; Genentech, Inc., South San Francisco, CA), can reduce tumor angiogenesis and inhibit the growth of solid tumors, either alone or in combination with chemotherapy. As a single agent or added to vinorelbine, bevaci

Returning to PARP Inhibition: Rechallenge with Bevacizumab in Ovarian Cancer.

The KGOG 3056/NIRVANA-R trial demonstrated that niraparib rechallenge combined with bevacizumab may potentially be a promising maintenance strategy in patients with platinum-sensitive recurrent ovarian cancer previously treated with poly(ADP-ribose) polymerase inhibitor (PARPi). With a 68% 6-month progression-free survival rate, this combination may synergistically be able to overcome acquired PARPi resistance, particularly in patients with favorable platinum sensitivity. Therapeutic Plasma Exchange for Removal of Bevacizumab: A Case Series.

Bevacizumab is a humanized monoclonal antibody against vascular endothelial growth factor A. Its long half-life and low volume of distribution allow circulating drug to persist in the body for weeks, which becomes clinically relevant when urgent or emergent surgery is needed. Because bevacizumab is largely intravascular, therapeutic plasma exchange (TPE) is attractive, though published experience is limited. We describe four adults who underwent TPE to facilitate surgery or postoperative wound healing after bevacizumab exposure: two with glioblastoma multiforme (GBM) before craniotomy, one wit
