Female rats are more vulnerable to binge drinking behavior in an operant self-administration paradigm: implication for transition to alcohol use disorders.

While binge drinking can significantly impact health negatively, it has become increasingly important to understand how sex differences contribute to this hazardous behavior, which may also serve as a risk factor for alcohol use disorder. We employed the binge drinking experimental model we developed previously to specifically analyze sex differences. Forty male and 40 female Long Evans rats were tested in the alcohol self-administration procedure, operationalized as alcohol responding in short daily session. We tested other parameters, including motivation, seeking, responses during cue omiss

Neuropeptide Y (NPY) in the central nucleus of the amygdala (CeA) does not affect ethanol-reinforced responding in binge-drinking, nondependent rats.

The central nucleus of the amygdala (CeA) has been implicated as having a significant role in mediating alcohol-drinking behavior. Neuropeptide Y (NPY) has been investigated as a potential pharmacotherapeutic due to its ability to attenuate ethanol intake, particularly when administered into the CeA. Previous research suggests, though the evidence is somewhat conflicting, that the efficacy of NPY is contingent upon genetic background and/or prior history of ethanol dependence in rats. However, studies looking at the effects of NPY in nonselected animals lacking a history of ethanol dependence

Nonoperative Management of a Spontaneous Perforation of Neobladder Secondary to Blackout From Solitary Binge Drinking: A Case Report and Current Literature Review.

The orthotopic ileal neobladder is becoming a popular technique of urinary diversion after radical cystectomy (RC) for localized muscle-invasive bladder cancer (MIBC), allowing patient continence, with a more desirable body image and good quality of life. Minimally invasive robot-assisted RC and neobladder have the potential to minimize physical and psychological trauma and are increasingly being adopted for patients with MIBC worldwide. Spontaneous perforation of orthotopic neobladder is uncommon;however, it represents serious complications. Solitary binge drinking can be dangerous in a patie

Adolescent binge drinking causes sex-specific deficits in behavioral reactivity, endocrine recovery from stress and altered expression of hypothalamic KCC2 in males.

RATIONALE: Binge drinking during adolescence is associated with a higher risk of developing Alcohol Use Disorders and impaired stress reactivity in adulthood. The hypothalamic-pituitary-adrenal axis matures during adolescence and is under strict GABAergic control. Yet, how early binge drinking alters GABAergic signaling in this region, and whether these changes contribute to later-life disruptions in stress reactivity, remain unknown. OBJECTIVES: Our goal with these set of experiments was to test the hypothesis that adolescent binge drinking would impair maturation of GABAergic signaling in t

Binge drinking as a potential cardiometabolic risk factor during adolescence in rats: novel protective antinitrosative mechanism of folic acid via caveolin-1.

Binge drinking (BD) is the predominant pattern of alcohol consumption among adolescents. It is clinically associated with cardiovascular and hepatic alterations due to its strong pro-oxidant effects. Folic acid (FA), a cardioprotective vitamin, is related to caveolins (Cav), proteins involved in cellular nitrosative balance. This study aims to evaluate BD as a possible cardiometabolic risk factor (CMRF) during adolescence by examining its effects on cardiac nitrosative equilibrium as well as the potential of FA supplementation to mitigate these effects. Four groups of adolescent rats were empl

Dynamic regulation of CeA gene expression during acute and protracted abstinence from chronic binge drinking of male and female C57BL/6J mice.

Binge alcohol consumption is a major risk factor for developing Alcohol Use Disorder (AUD) and is associated with alcohol-related problems like accidental injury, acute alcohol poisoning, and black-outs. While there are numerous brain regions that have been shown to play a role in this AUD in humans and animal models, the central nucleus of the amygdala (CeA) has emerged as a critically important locus mediating binge alcohol consumption. In this study, we sought to understand how relative gene expression of key signaling molecules in the CeA changes during different periods of abstinence foll

Binge drinking and mortality among older adults: Findings from the Canadian Community Health Survey linked to the Canadian Vital Statistics Death Database.

BACKGROUND: Binge drinking is a growing public health concern among older adults, yet research has disproportionately focused on younger adults. The relationship between binge drinking and mortality among older adults represents a critical research gap. OBJECTIVES: (1) To examine the association between binge drinking frequency and mortality among older adults; (2) To identify whether this relationship is attenuated by controlling for various sociodemographic and health factors. METHODS: Data were drawn from the 2005-2014 Canadian Community Health Survey linked to the Canadian Vital Statisti

Ethanol Self-Administration Reduces mGlu2/3 Protein Expression in the Nucleus Accumbens and mGlu2/3 Activation Suppresses Binge Drinking in Male Mice.

BACKGROUND: Alcohol use disorder (AUD) is associated with dysregulated glutamatergic signaling within mesocorticolimbic circuits that govern reinforcement and excessive ethanol intake. Group II metabotropic glutamate receptors (mGlu2/3) act primarily as presynaptic autoreceptors that regulate glutamate release. However, how voluntary alcohol intake alters mGlu2/3 expression within reward circuitry remains unclear. METHODS AND RESULTS: We examined mGlu2/3 protein expression following a history of voluntary ethanol exposure and operant self-administration and, in separate cohorts, assessed the

Biochemical changes precede affective and cognitive anomalies in aging adult C57BL/6J mice with a prior history of adolescent alcohol binge-drinking.

The early initiation of binge-drinking and biological sex are critical risk factors for the development of affective disturbances and cognitive decline, as well as neurodegenerative diseases including Alzheimer's disease. Further, a history of excessive alcohol consumption alters normal age-related changes in the pattern of protein expression in the brain, which may relate to an acceleration of cognitive decline. Here, we aimed to disentangle the interrelation between a history of binge-drinking during adolescence, biological sex and normal aging on the manifestation of negative affect, cognit

Epigenetic Repression of the Serotonergic Neuron Phenotype Following Adolescent Binge Drinking Is Restored through Inhibition of Proinflammatory Signaling by Exercise and Glycyrrhizic Acid.

The serotonergic system regulates diverse social and emotional processes, including formation of social networks that continue to mature throughout adolescence. In adolescent intermittent ethanol (AIE)-exposed male and female rats and postmortem human dorsal raphe nucleus (DRN) tissue from adolescent-onset alcohol use disorder (AUD) cases, we observed persistent reductions of serotonergic markers (TPH2+, 5HT+) within the DRN and its projections, accompanied by robust neuroimmune activation (HMGB1, TLR4, pNFκBp65). Alcohol motivations associated with frequency of alcohol use, binge drinking, and alcohol problems among active duty junior enlisted soldiers and non-commissioned officers.

Problematic alcohol use is a serious threat to the behavioral health of active-duty Service Members (ADSM), resulting in numerous calls from governmental agencies to better understand mechanistic factors contributing to alcohol misuse within the military. Alcohol use motives are reliable predictors of alcohol-related behaviors and are considered malleable targets for prevention and intervention efforts. However, empirical research indicates that drinking motives vary across contextually distinct populations. Although some research has been conducted among veteran and reservist populations, lim

Aspirin and N-acetylcysteine co-administration markedly inhibit chronic ethanol intake and block relapse binge drinking: Role of neuroinflammation-oxidative stress self-perpetuation.

Chronic alcohol intake leads to neuroinflammation and cell injury, proposed to result in alterations that perpetuate alcohol intake and cued relapse. Studies show that brain oxidative stress is consistently associated with alcohol-induced neuroinflammation, and literature implies that oxidative stress and neuroinflammation perpetuate each other. In line with a self-perpetuating mechanism, it is hypothesized that inhibition of either oxidative stress or neuroinflammation could reduce chronic alcohol intake and relapse. The present study conducted on alcohol-preferring rats shows that chronic et

Amitifadine, a triple monoamine uptake inhibitor, reduces binge drinking and negative affect in an animal model of co-occurring alcoholism and depression symptomatology.

The co-occurrence of alcoholism and depression is highly prevalent and difficult to treat. In an animal model of binge drinking that exhibits abstinence-induced behaviors reminiscent of negative affective states, the triple monoamine uptake inhibitor, amitifadine, produced a selective, dose dependent attenuation of binge drinking. Amitifadine also reversed abstinence-induced increases in the intracranial self-stimulation threshold, a model of anhedonia, and immobility in the forced swim test, reflecting behavioral despair. In view of the safety profile of amitifadine in humans, including low r

Adolescent binge drinking disrupts hepatic lipid homeostasis, leading to steatosis in rats: protective role of folic acid in cholesterol and fatty acid balance.

Alcohol liver damage (ALD) is increasing worldwide among adolescents, along with binge drinking (BD). BD is an acute alcohol consumption pattern, strongly pro-oxidant in the liver, and may be associated with steatosis, the first step in ALD. Folic acid (FA), an antioxidant crucial for liver function, shows compromised hepatic stores after BD. Therefore, this study aims to analyze the hepatic lipid changes associated with BD-induced steatosis during adolescence in rats and to evaluate the efficacy of FA supplementation in preventing these alterations. Four groups of adolescent rats were used: c
