Conversion of CO2 into valuable products: engineering the PirC-PGAM switch in cyanobacteria to direct carbon flux into desired products.

BACKGROUND: In response to rising CO2 emissions driving global warming, there is an urgent need for a transition toward a sustainable bioeconomy. Photo-biotechnological processes based on oxygenic photosynthesis hold high potential for achieving CO2 neutrality and in this regard, cyanobacteria have emerged as promising biocatalysts. Rational metabolic engineering of cyanobacteria depends on a thorough understanding of regulatory mechanisms governing primary metabolism, because native metabolic flux through specific pathways and, consequently, the formation of target products can be limited. Saline Permafrost and Cryopegs as Potentially Important Sources of CO2-Assessing Organic Carbon Mineralization Potentials on the Alaskan Coastal Plain.

Thermokarst lake and drained lake basin (DLB) dynamics are intensifying across the Alaskan Arctic Coastal Plain. Thawing, drainage, and erosion expose surface and deep sediments (> 1 m) to aerobic conditions, with saline deposits being particularly vulnerable due to freeze-point depression. As organic carbon mineralization remains poorly constrained, we determined potentials with an aerobic one-year long incubation at 10°C in permafrost upland, lake talik, lake cryopeg, and refrozen saline DLB sediments. We linked CO2 production to biochemical, hydrochemical, and microbial factors, and assesse

Rewiring photosynthetic carbon flow: Engineered cyanobacterial factories for sustainable carbohydrate production and carbon-negative biomanufacturing.

Cyanobacteria are promising platforms for light-driven carbon fixation and carbohydrate biosynthesis. However, optimization strategies that focus solely on carbon allocation are insufficient to achieve substantial improvements in yield and sustainability. Here, Synechococcus elongatus PCC 7942 was engineered to enhance sucrose production by simultaneously increasing total carbon input and reinforcing the artificial sink. The engineered strain secreted 5.821 g L-1 sucrose, which was 27.4 times higher than the wild-type. Transcriptomic analysis revealed upregulation of abundant genes involved in

Increased Belowground Carbon Allocation Reduces Soil Carbon Losses Under Long-Term Warming.

The response of the carbon cycle in forests to global warming could lead to a positive climate feedback if warming accelerates the mineralization of soil organic carbon (SOC), thereby causing net emissions of CO2 into the atmosphere. In Europe, carbon-rich alpine forest soils could be particularly affected by global warming, as a greater rise in temperature is expected in this region than the global average. Here we show that nearly two decades of experimental soil warming (+4°C during the snow-free seasons) in a mountain forest in the Northern Limestone Alps significantly (~13% per 1°C warmin

Unusual regulation of the CO2 concentrating mechanism of marine chemolithoautotroph Thiomicrospira pelophila.

Autotrophic organisms fix dissolved inorganic carbon (DIC: CO2, HCO3-, CO3-2) into biomass, introducing organic carbon into the global carbon cycle. Many lineages of autotrophs developed CO2 concentrating mechanisms (CCMs) to grapple with the catalytic constraints of the carboxylase of the Calvin-Benson-Bassham cycle, ribulose-1,5-bisphosphate carboxylase/oxygenase (RubisCO). Carboxysomes and DIC transporters act together to form CCMs, which facilitate DIC fixation by autotrophic bacteria when DIC is scarce. Autotroph Thiomicrospira pelophila has carboxysomes, with an unusual carbonic anhydras

Summer carbon budget simulation in a lagoon ecosystem with submerged aquatic vegetation.

As international efforts against global warming advance, blue carbon has gained attention as a CO₂ sink. This study quantitatively evaluated the carbon budget of an eelgrass (Zostera marina) ecosystem in Komuke Lagoon, a restricted coastal lagoon on the northeastern coast of Hokkaido, Japan. Based on outdoor experiments conducted in 2018 and 2023, a new DIC equation was constructed that incorporates photosynthetic responses to photon flux density and water temperature. Furthermore, a box model integrating river inflow, seawater exchange with the open ocean, air-water CO₂ flux, bottom dilution,

DOM-microbe interactions shape carbon storage strategies in rhizosphere and detritus-rich wetland soils.

Wetland macrophytes are associated with carbon storage through photosynthetic CO2 uptake and interactions with microbes. Rhizodeposition and detritus accumulation represent two key pathways examined in this study. However, how dissolved organic matter (DOM) interacts with microbial carbon storage remains incompletely understood. Here, we investigated links between DOM characteristics and microbial carbon transformation pathways in Wuchang Lake using FT-ICR-MS, metagenomics, and geochemical analyses. Macrophyte presence was associated with a higher relative abundance of genes related to microbi

Littoral and wetland vegetation decrease carbon emissions from dry inland waters.

Lakes are recognized as active components of the inland water carbon (C) cycle, as organic matter is processed by microbial respiration, inducing large carbon dioxide (CO2) and methane (CH4) emissions. In the context of long-lasting drought periods, large uncertainties remain about: (1) the influence of wet-dry cycle on CO2 and CH4 fluxes in littoral zones and lacustrine wetlands; and (2) the contribution of emergent vegetation to C fluxes in dry inland waters. At the water-land interface of two shallow lakes, this study focuses on CO2 and CH4 fluxes from vegetated and bare dry inland waters i

C1 photochemotrophy - rethinking one-carbon metabolism in phototrophs.

Excessive CO2 emissions, caused by an imbalance between carbon oxidation and reduction, drive climate change. To address this, we propose photochemotrophic metabolism as an alternative to both canonical photosynthesis and synthetic one-carbon (C1) metabolism in heterotrophs. In photochemotrophy, naturally phototrophic microorganisms such as cyanobacteria serve as the chassis to assimilate chemically reduced and soluble C1 compounds such as formate or methanol by using carbon fixation cycles that are more efficient than the native Calvin cycle. Key potential advantages of photochemotrophy inclu

Shifts in Microbial Thermal Traits Mitigate Heat-Induced Carbon Losses in Soils.

Global warming is expected to transfer carbon from soil organic matter to atmospheric CO2, with microbial communities playing a crucial role in regulating this exchange. While the immediate impact of temperature on microbial functions is well understood and causes soil carbon losses, the long-term response remains unclear, with losses stabilising over time, reducing the overall effect of chronic warming on soil organic carbon (SOC) stocks. Here, we examined the temperature dependence of microbial respiration and growth after 9 years of +5°C warming in a temperate forest. Using these temperatur

Carboxysomes: The next frontier in biotechnology and sustainable solutions.

Some bacteria possess microcompartments that function as protein-based organelles. Bacterial microcompartments (BMCs) sequester enzymes to optimize metabolic reactions. Several BMCs have been characterized to date, including carboxysomes and metabolosomes. Genomic analysis has identified novel BMCs and their loci, often including genes for signature enzymes critical to their function, but further characterization is needed to confirm their roles. Among the various BMCs, carboxysomes, which are found in cyanobacteria and some chemoautotrophic bacteria, and are most extensively investigated. The

Microbial drivers of biogeochemical cycles in deep sediments of the Kathiawar Peninsula Gulfs of India.

Deep marine sediments are rich in microbial diversity, which holds metabolic repertoire to modulate biogeochemical cycles on a global scale. We undertook the environmental microbiome inhabiting the Gulf of Kathiawar Peninsula as a model system to understand the potential involvement of the deep marine sediment microbial community and as a cohort in the carbon, nitrogen, and sulfur biogeochemical cycles. These gulfs are characterized by dynamic tidal variations, diverse sediment textures, and nutrient-rich waters, driven by coastal processes and the interaction between natural coastal dynamics

Unlocking the Carbon Mitigation Potential of Green Infrastructure: Evidence from China's Sponge City Program.

Carbon mitigation and nature-based green infrastructure (GI) have become central themes in global efforts to address climate and urban sustainability. The net carbon benefits of GI and the factors shaping them remain poorly understood due to data and methodological gaps. This study draws on 679 engineering cases from 15 demonstration Sponge Cities in China to conduct multiscale life-cycle carbon accounting of GI. Overall, sponge retrofitting shifted conventional urban surfaces from a net source to a sink. At the infrastructure scale, carbon benefits varied widely across different types of GI,

Systematic engineering of synthetic serine cycles in Pseudomonas putida uncovers emergent topologies for methanol assimilation.

The urgent need for a circular carbon economy has driven research into sustainable substrates, including one-carbon (C1) compounds. The non-pathogenic soil bacterium Pseudomonas putida is a promising host for exploring synthetic methylotrophy due to its versatile metabolism. In this research article we implemented synthetic serine cycle variants in P. putida for methanol assimilation, combining modular engineering and growth-coupled selection, whereby methanol assimilation supported biosynthesis of the essential amino acid serine. We adopted three synthetic variants (serine-threonine cycle, ho

Deletion of Re-citrate synthase allows for analysis of contributions of tricarboxylic acid cycle directionality to the growth of Heliomicrobium modesticaldum.

UNLABELLED: Heliomicrobium modesticaldum, a phototrophic member of the phylum Firmicutes and family Clostridiales, possesses most of the enzymes specific to the reductive tricarboxylic acid (rTCA) cycle, except for the key enzyme, ATP-citrate lyase. It is thought to utilize a split TCA cycle when growing on pyruvate as a carbon source, in which the oxidative TCA (oTCA) direction generates most of the 2-ketoglutarate, but some can be produced in the reductive direction. Although a typical Si-citrate synthase gene is not found in the genome, it was suggested that gene HM1_2993, annotated as homo

Dynamic photosynthetic labeling and carbon-positional mass spectrometry monitor in vivo RUBISCO carbon assimilation rates.

RIBULOSE-1,5-BISPHOSPHATE CARBOXYLASE/OXYGENASE (RUBISCO) is the most abundant enzyme and CO2 bio-sequestration system on Earth. Its in vivo activity is usually determined by 14CO2 incorporation into 3-phosphoglycerate (3PGA). However, the radiometric analysis of 3PGA does not distinguish carbon positions. Hence, RUBISCO activity that fixes carbon into the 1-C position of 3PGA and Calvin-Benson-Bassham (CBB) cycle activities that redistribute carbon into its 2-C and 3-C positions are not resolved. This study aims to develop technology that differentiates between these activities. In source fra

Sulfur-carbon loop enhanced efficient nitrogen removal mechanism from iron sulfide-mediated mixotrophic partial denitrification/anammox systems.

This study successfully established Iron Sulfide-Mediated mixotrophic Partial Denitrification/Anammox system, achieving nitrogen and phosphorus removal efficiency of 97.26% and 78.12%, respectively, with COD/NO3--N of 1.00. Isotopic experiments and X-ray Photoelectron Spectroscopy analysis confirmed that iron sulfide enhanced autotrophic Partial Denitrification performance. Meanwhile, various sulfur valence states functioned as electron buffers, reinforcing nitrogen and sulfur cycles. Microbial community analysis indicated reduced heterotrophic denitrifiers (OLB8, OLB13) under lower COD/NO3--N

Assessment of CO2 fluxes in the hinterland of the Gurbantunggut Desert and its response to climate change.

Desert ecosystems, as an important part of terrestrial ecosystems, are considered potential hidden carbon sinks in the global carbon cycle. The Gurbantunggut Desert, as China's largest fixed/semi-fixed desert, has received little research on its role in the global carbon cycle and future trends. This study utilizes continuous observational data from the Gurbantunggut Desert from 2018 to 2022 and integrates CMIP6 global climate model scenario data to study the evolution of carbon balance in the desert ecosystem, carbon source/sink functions, and future trends. The result showed that: 1) The CO2
