Moscona, in the early sixties [A.A. Moscona, Recombination of dissociated cells and the development of cell aggregates, in: B.M. Willmer (Ed.), Cells and Tissues in Culture, Academic Press, New York, 1965, pp. 489-529.] [16], discovered that aggregation of dissociated cells is a property of embryonal cells. Several features of the aggregate culture system are particularly attractive for the conduct of biochemical and molecular studies on the human fetal brain. (i) All the pertinent procedural parameters can be readily controlled and standardized, resulting in a consistently reproducible system

Embryonal carcinoma cells were aggregated with cleavage stage mouse embryos, cultured briefly, and transferred as morulae to the uteri of pseudopregnant females. When midgestation foetuses were examined, many were morphologically abnormal. The severity of this abnormal development was correlated with the extent of contribution by embryonal carcinoma cells to the foetuses as indicated by GPI (glucose phosphate isomerase) analysis. This was true for all three of the cell lines studied, NG-2, PSA-1, and LT1-2D. The clear correlation between increasingly abnormal development and more extensive par

Humoral immunity is a central component of host defense in teleost fish, yet the dynamics of IgM+ B cells during infection remain poorly understood. In this study, we generated and characterized a monoclonal antibody (mAb) against native IgM protein purified from grass carp (Ctenopharyngodon idella) sera. This mAb specifically recognized the recombinant grass carp IgM heavy chain expressed in the HEK293 cells and showed cross-reactivity with IgM molecules from multiple cyprinid species, including rare minnow, zebrafish, black carp, and common carp, but not with IgM from largemouth bass. An early and continual event in Alzheimer's disease (AD) is upregulation of the immune response concomitant with accumulation of aggregated amyloid-β peptide (Aβ). The mechanisms by which Aβ can trigger an immune response involve stimulation of cell surface receptors and intracellular pathways, as well as internalization of Aβ into the cell cytosol. The HEK293T cell line has been a valuable model system for exploring a variety of biological processes, including internalization of protein aggregates such as Aβ. There have been some questions regarding the role of Aβ conformation in the HEK293T

BACKGROUND: Rapid and stable regeneration of bone defects remains a pressing clinical challenge. We previously fabricated stem cell aggregates (CA) by mimicking developmental condensation and demonstrated their efficacy in promoting bone defect repair. Endogenous Gli1+ skeletal stromal/progenitor cells (SSPCs) are a pivotal SSPC subtype known to maintain bone homeostasis and enhance bone regeneration; however, the functional properties and translational potential of CA derived from these cells (Gli1+ CA) remain largely elusive.
METHODS: Single-cell RNA sequencing was performed to characterize

Tau aggregation is a central pathological feature of Alzheimer's disease (AD) and other tauopathies. Although oxidative stress has been implicated in AD pathogenesis, the effects of intracellular redox perturbation on tau-seeded aggregation remain incompletely understood. In this study, we investigated the role of thiol redox homeostasis in tau-seeded aggregation using HEK293 TauRD fluorescence resonance energy transfer biosensor cells. Intracellular redox conditions were manipulated using the glutathione synthesis inhibitor l-buthionine-(S,R)-sulfoximine (BSO) and through overexpression or kn

Aggregation and deposition of amyloid-β protein (Aβ42) in extracellular spaces are key neuropathological features of Alzheimer's disease (AD). Extracellular Aβ42 aggregates not only impair the function of the extracellular matrix but also directly damage brain cells through binding interactions. Chemically, tea polyphenols are amphiphilic, similar to the Aβ42 molecule. This study found that catechin (CA) and epigallocatechin gallate (EGCG) effectively bound to Aβ42 monomers (Aβ42M) and oligomers (Aβ42O), preventing Aβ42 aggregation; in contrast, EGCG also showed some binding to protofibrils (A

Parkinson's disease (PD) is a severe pathology caused by a progressive degeneration of neurons in the substantia nigra pars compacta, hypothalamus, and thalamus. Although etiology of PD remains unclear, accumulating evidence indicates that neurodegenerative effects are triggered by the abrupt aggregation of α-synuclein (α-Syn), a small membrane protein that is responsible for cell vesicle trafficking. α-Syn aggregates are highly toxic to neurons and immune cells present in the brain, including macrophages, microglia, and dendritic cells. Transition metal dichalcogenide nanoflowers (TMD NFs) ar

Intracellular protein aggregation is a hallmark of aging and contributes to pathology in some age-associated diseases. In hereditary adult-onset neuromuscular diseases (NMDs), protein aggregates play a key role in disease onset and progression. The wild-type Poly(A) binding protein nuclear 1 (PABPN1) forms benign nuclear aggregates, whereas a short trinucleotide expansion leads to the formation of pathogenic aggregates, a hallmark of Oculopharyngeal Muscular Dystrophy (OPMD). In OPMD, the mutant PABPN1 causes skeletal muscle weakness. So far, the structural differences between benign and patho

Mesenchymal stromal cell (MSC) transplantation has achieved significant clinical benefits for many diseases, such as systemic lupus erythematosus (SLE) and inflammatory diseases. However, the detailed therapeutic mechanism of MSCs is not fully understood. Here, in the SLE treatment, we show that MSC transplantation triggers recipient bone marrow neutrophil aggregation to generate an endogenous extracellular vesicle (EV) storm in the circulation via the TNFα/ICAM-1/Rab11b axis. Interestingly, blockade of the EV storm abolishes the MSC-mediated therapeutic effect for SLE. The level of EV storm i

Amyotrophic lateral sclerosis (ALS) is a progressive neurodegenerative disorder that currently lacks effective therapy. Given the heterogeneity of clinical and molecular profiles of ALS patients, personalized diagnostics and pathological characterization represent a powerful strategy to optimize patient stratification, thereby enabling personalized treatment. Immortalized lymphocytes from sporadic and genetic ALS patients recapitulate some pathological hallmarks of the disease, facilitating the fundamental task of drug screening. However, the molecular aggregation of ALS has not been character

In this work, we develop a nanoprobe for the detection of hypochlorous acid (HOCl), a key reactive oxygen species, utilizing dark-field microscopy (DFM). The nanoprobe is constructed by covalently attaching PEG-SH and an HOCl-sensitive molecule, FD, to gold nanoparticles (GNPs-FD-PEG). This probe detects HOCl by observing a color shift from green to red due to the aggregation of GNPs, which is triggered by HOCl-induced the cleavage of FD molecule. This aggregation occurs primarily due to electrostatic interactions between the differently charged particles following the cleavage. By recording t

Hair regeneration using hair follicle stem cells (HFSCs) and dermal papilla cells is a promising approach for the treatment of alopecia. One of the challenges faced in this approach is the quantitative expansion of HFSCs while maintaining their hair induction capacity. In this study, HFSC expansion was achieved through the formation of uniform-diameter cell aggregates that were subsequently encapsulated in Matrigel. We designed a microwell array device, wherein mouse HFSCs were seeded, allowed to form loosely packed aggregates for an hour, and then embedded in Matrigel. Quantitative analysis r

Accumulation and aggregation of α-Synuclein (α-Syn) are the hallmarks of the incidence of α-Synucleinopathies, which comprises dementia with Lewy bodies (LBs). Aggregation inhibitors are anticipated to reduce α-Syn toxicity and serve as therapeutic agents. As a result, α-Syn is regarded as the potential and priority target for drug development. Here, we report inhibition of α-Syn aggregation by a certain lysophospholipids (LPLs) species. LPLs are small bioactive lipid molecules characterized by a single carbon chain and polar head group. The LPLs used here were extracted from porcine liver dec

OBJECTIVE: Therapies targeting B cells have been used in the clinic for multiple sclerosis (MS). In patients with relapsing MS, anti-CD20 therapy often suppresses relapse activity; yet, their effect on disease progression has been disappointing. Most anti-CD20 therapeutic antibodies are type I, but within the unique microenvironment of the brain, type II antibodies may be more beneficial, as type II antibodies exhibit reduced complement-dependent cytotoxicity and they have an increased capacity to induce direct cell death that is independent of the host immune response.
METHODS: We compared t

Polyglutamine (polyQ) diseases are a group of neurodegenerative disorders, involving the deposition of aggregation-prone proteins with long polyQ expansions. However, the cytotoxic roles of these aggregates remain highly controversial, largely due to a lack of proper tools for quantitative and nonperturbative interrogations. Common methods including in vitro biochemical, spectroscopic assays, and live-cell fluorescence imaging all suffer from certain limitations. Here, we propose coupling stimulated Raman scattering microscopy with deuterium-labeled glutamine for live-cell imaging, quantificat

Protective MHC class I-dependent immune responses require an overlap between repertoires of proteins directly presented on target cells and cross-presented by professional APC, specifically dendritic cells. How stable proteins that rely on defective ribosomal proteins for direct presentation are captured for cell-to-cell transfer remains enigmatic. In this study, we address this issue using a combination of in vitro (C57BL/6-derived mouse cell lines) and in vivo (C57BL/6 mouse strains) approaches involving stable and unstable versions of OVA model Ags displaying defective ribosomal protein-dep

The eukaryotic cytosolic proteome is vulnerable to changes in proteostatic and redox balance caused by temperature, pH, oxidants, and xenobiotics. Cysteine-containing proteins are especially at risk, as the thiol side chain is subject to oxidation, adduction, and chelation by thiol-reactive compounds. The thiol-chelating heavy metal cadmium is a highly toxic environmental pollutant demonstrated to induce the heat shock response and recruit protein chaperones to sites of presumed protein aggregation in the budding yeast Saccharomyces cerevisiae. However, endogenous targets of cadmium toxicity r

BACKGROUND: Mesenchymal stromal cells (MSCs) are promising cell therapy candidates. Clinical application is considered safe. However, minor side effects have included thromboembolism and instant blood-mediated inflammatory reactions suggesting an effect of MSC infusion on hemostasis. Previous studies focusing on plasmatic coagulation as a secondary hemostasis step detected both procoagulatory and anticoagulatory activities of MSCs. We now focus on primary hemostasis and analyzed whether MSCs can promote or inhibit platelet activation.
METHODS: Effects of MSCs and MSC supernatant on platelet a

Formaldehyde (FA), as a reactive carbonyl species and signaling molecule, plays an important role in living systems. Here, an FA-responsive probe with fast response and great selectivity is designed based on aggregation-induced emission. The probe is prepared by functionalizing tetraphenylethene (TPE) with two amine groups. FA is detected based on the solubility differences between the amine-functionalized TPE and the corresponding Schiff bases after reaction with FA. The probe exhibits a limit of detection of 40 nM and a response time of ∼90 s. Furthermore, its ability to detect both endogeno
