EGF receptor in organ development, tissue homeostasis and regeneration.

The epidermal growth factor receptor (EGFR) is a protein embedded in the outer membrane of epithelial and mesenchymal cells, bone cells, blood and immune cells, heart cells, glia and stem neural cells. It belongs to the ErbB family, which includes three other related proteins: HER2/ErbB2/c-neu, HER3/ErbB3, and HER4/ErbB4. EGFR binds to seven known signaling molecules, including epidermal growth factor (EGF) and transforming growth factor-alpha (TGF-α). This binding triggers the formation of receptor pairs (dimers), self-phosphorylation of EGFR, and the activation of several signaling pathways

LRP-1 and LRP-2 receptors function in the membrane neuron. Trafficking mechanisms and proteolytic processing in Alzheimer's disease.

Low density lipoprotein receptor-related protein (LRP) belongs to the low-density lipoprotein receptor family, generally recognized as cell surface endocytic receptors, which bind and internalize extracellular ligands for degradation in lysosomes. Neurons require cholesterol to function and keep the membrane rafts stable. Cholesterol uptake into the neuron is carried out by ApoE via LRPs receptors on the cell surface. In neurons the most important are LRP-1 and LRP-2, even it is thought that a causal factor in Alzheimer's disease (AD) is the malfunction of this process which cause impairment i

Islet cell neogenesis in the pancreas.

The beta cell population of the endocrine pancreas may expand by either of two processes, neogenesis or replication. While replication requires the existence of an already differentiated beta cell, neogenesis depends on the presence of active stem cells. Since the replicative activity of highly specialized cells such as beta cells seems to be limited, it is interesting to study the potential of the endocrine pancreas for beta cell neogenesis. In this article we review both the current state of knowledge of beta cell neogenesis under natural conditions and in response to stimulation, and the si

The endothelium-bone axis in development, homeostasis and bone and joint disease.

Blood vessels form a versatile transport network that is best known for its critical roles in processes such as tissue oxygenation, metabolism and immune surveillance. The vasculature also provides local, often organ-specific, molecular signals that control the behaviour of other cell types in their vicinity during development, homeostasis and regeneration, and also in disease processes. In the skeletal system, the local vasculature is actively involved in both bone formation and resorption. In addition, blood vessels participate in inflammatory processes and contribute to the pathogenesis of

Changes in extracellular matrix proteins and actin during corneal endothelial growth.

Basement membranes influence growth, shape and differentiation of cells and tissues. However, the role and influence of Descemet's membrane during corneal development is not understood. To address this question, the relationships between cell growth and fibronectin, laminin and actin distribution in the developing rat corneal endothelium in vivo has been examined. During fetal development, rat corneal endothelial cells undergo DNA synthesis and mitosis. However, at day 14 of gestation both processes begin to decline and neither can be detected in endothelium of 1-month-old animals. By this tim

Bacterial cell growth is arrested by violet and blue, but not yellow light excitation during fluorescence microscopy.

BACKGROUND: Fluorescence microscopy is a powerful tool in cell biology, especially for the study of dynamic processes. Intensive irradiation of bacteria with UV, blue and violet light has been shown to be able to kill cells, but very little information is available on the effect of blue or violet light during live-cell imaging. RESULTS: We show here that in the model bacterium Bacillus subtilis chromosome segregation and cell growth are rapidly halted by standard violet (405 nm) and blue light (CFP) (445-457 nm) excitation, whereas they are largely unaffected by green light (YFP). The stress

Mechanics of compression-driven morphogenesis.

Mechanical forces shape multiscale biological processes across many living systems, ranging from cell proliferation and apoptosis, to tissue folding and pattern formation. In recent years, tissue pressure has emerged as a fundamental regulator of morphogenesis across animal and plant kingdoms. However, a fundamental understanding of the definition and origin of tissue pressure remains lacking. In this Review, we synthesise how tissue pressure is generated from diverse sources, including tissue growth, actomyosin contractility, topological defects and fluid- or extracellular matrix-driven swell

Dual Functions of Polyamines in Shaping Host-Specific Pathogen Dynamics.

Polyamines such as putrescine, spermidine, and spermine play essential roles in most living organisms. They regulate fundamental processes, like cell proliferation, differentiation, growth, gene expression (DNA/RNA stability, transcription, and translation), and signal transduction. As important regulators, polyamines influence development, stress responses, and the progression of health and disease, including cancer and aging. These positively charged molecules have been extensively studied for decades. In humans, polyamines are often researched as potential therapeutic targets for diseases s

Roles and therapeutic targeting of SLC transporters in gastrointestinal malignancies.

The solute carrier (SLC) transporter family, serving as metabolic gatekeepers, transports a range of substrates (nutrients, metabolites, ions, drugs) and regulates key physiological processes (proliferation, migration, angiogenesis, chemoresistance, immunomodulation, energy metabolism). Gastrointestinal malignancies are a leading cause of cancer-related deaths globally, marked by high prevalence and aggressive growth. Metabolic reprogramming, as an emerging cancer hallmark, promotes uncontrolled tumour proliferation. It dynamically adjusts the distribution of nutrients and energy in the tumour

Expedited growth factor-mediated specification of human embryonic stem cells toward the hepatic lineage.

Human embryonic stem cells (hESCs) have the potential to be a promising source of liver cells, hepatocytes, for regenerative medicine given their unlimited proliferative and pluripotent differentiative capacity. However, the inefficient embryoid body process and limited understanding of molecular signals potentiating cell-specific differentiation plague the use of hESCs as a hepatic source. In this study, we describe an efficient growth factor-based process for directed differentiation of hESCs that bypasses embryoid body development. The system involves adherent hESC culture exposure to activ

Implications of LINC01094 for human malignancies.

Long non-coding RNAs (lncRNAs) constitute a large class of ribonucleic acids, participating in multiple biological events within tumor cells, especially the regulation of transcription. A growing body of literature has revealed that abnormalities of lncRNA expression could result in carcinogenesis and oncogenesis by exerting inhibition or oncogenic effects. LINC01094 is a recently identified lncRNA found to be dys-regulated in an assortment of cancer tissues and control multiple biological processes via competing endogenous RNA (ceRNA) mechanisms. Specifically, LINC01094 functions as a ceRNA t

Polycystic liver diseases.

Polycystic liver diseases (PCLDs) are genetic disorders with heterogeneous etiologies and a range of phenotypic presentations. PCLD exhibits both autosomal or recessive dominant pattern of inheritance and is characterized by the progressive development of multiple cysts, isolated or associated with polycystic kidney disease, that appear more extensive in women. Cholangiocytes have primary cilia, functionally important organelles (act as mechanosensors) that are involved in both normal developmental and pathological processes. The absence of polycystin-1, 2, and fibrocystin/polyductin, normally

H3K9me3 represses G6PD expression to suppress the pentose phosphate pathway and ROS production to promote human mesothelioma growth.

The role of glucose-6-phosphate dehydrogenase (G6PD) in human cancer is incompletely understood. In a metabolite screening, we observed that inhibition of H3K9 methylation suppressed aerobic glycolysis and enhances the PPP in human mesothelioma cells. Genome-wide screening identified G6PD as an H3K9me3 target gene whose expression is correlated with increased tumor cell apoptosis. Inhibition of aerobic glycolysis enzyme LDHA and G6PD had no significant effects on tumor cell survival. Ablation of G6PD had no significant effect on human mesothelioma and colon carcinoma xenograft growth in athymi

Dynamic behavior analysis of a fractal tumor-immune model with drug resistance.

Chemotherapy is a type of treatment that uses drugs to treat cancer and prevent tumor growth. Different chemotherapy protocols and procedures have been developed. Mathematical models have been proposed to analyze the dynamical processes related to cancerous cell proliferation. In this work, we propose a fractal model with healthy, cancerous cells, and chemotherapeutic agents, as well as effector immune cells interacting with cancerous cells. The model incorporates cancer drug resistance as a result of mutation processes triggered by chemotherapy. We demonstrate that the order of our fractal tu

A Cytosine Methyltransferase Modulates the Cell Envelope Stress Response in the Cholera Pathogen [corrected].

DNA methylation is a key epigenetic regulator in all domains of life, yet the effects of most bacterial DNA methyltransferases on cellular processes are largely undefined. Here, we used diverse techniques, including bisulfite sequencing, transcriptomics, and transposon insertion site sequencing to extensively characterize a 5-methylcytosine (5mC) methyltransferase, VchM, in the cholera pathogen, Vibrio cholerae. We have comprehensively defined VchM's DNA targets, its genetic interactions and the gene networks that it regulates. Although VchM is a relatively new component of the V. cholerae gen

Airborne exposure of Rhizobium leguminosarum strain E20-8 to volatile monoterpenes: Effects on cells challenged by cadmium.

Volatile organic compounds (VOCs) are produced by plants, fungi, bacteria and animals. These compounds are metabolites originated mainly in catabolic reactions and can be involved in biological processes. In this study, the airborne effects of five monoterpenes (α-pinene, limonene, eucalyptol, linalool, and menthol) on the growth and oxidative status of the rhizobial strain Rhizobium leguminosarum E20-8 were studied, testing the hypothesis that these VOCs could influence Rhizobium growth and tolerance to cadmium. The tested monoterpenes were reported to have diverse effects, such as antibacter

[The pathologically changed Descemet membrane. Cultivation of human corneal endothelium on transformed membrane].

Corneal endothelial diseases are connected with structural and biochemical changes of Descemet's membrane (DM). Little is known about the possible effects of these changed basement membranes. In the present study we investigated the influence of changed DM on endothelium. Human corneal endothelial cells were cultured on DM of healthy corneas as well as corneas with Fuchs' endothelial dystrophy or bullous keratopathy, and cell growth and morphology were compared. On healthy DM, cells formed a confluent monolayer within 4 days. Cells cultured on Fuchs' DM showed a similar pattern. The polygonal

A Simple Microfluidic Chip for Long-Term Growth and Imaging of Caenorhabditis elegans.

Caenorhabditis elegans (C. elegans) have proved to be a valuable model system for studying developmental and cell biological processes. Understanding these biological processes often requires long-term and repeated imaging of the same animal. Long recovery times associated with conventional immobilization methods done on agar pads have detrimental effects on animal health making it inappropriate to repeatedly image the same animal over long periods of time. This paper describes a microfluidic chip design, fabrication method, on-chip C. elegans culturing protocol, and three examples of long-ter

Targeting the Akt signaling pathway: Exploiting curcumin's anticancer potential.

Cancer is recognized as one of the leading causes of death worldwide. In recent years, advancements in early detection and expanding treatment options have contributed to a decrease in mortality rates. However, the emergence of drug-resistant cancers necessitates the exploration of innovative and more effective drugs. The Akt kinases play a central role in various signaling pathways that regulate crucial cellular processes, including cell growth, proliferation, survival, angiogenesis, and glucose metabolism. Due to frequent disruptions of the Akt signaling pathway in numerous human cancers and
