Essential role of Cdc42 in cardiomyocyte proliferation and cell-cell adhesion during heart development.

Cdc42 is a member of the Rho GTPase family and functions as a molecular switch in regulating cell migration, proliferation, differentiation and survival. However, the role of Cdc42 in heart development remains largely unknown. To determine the function of Cdc42 in heart formation, we have generated a Cdc42 cardiomyocyte knockout (CCKO) mouse line by crossing Cdc42 flox mice with myosin light chain (MLC) 2a-Cre mice. The inactivation of Cdc42 in embryonic cardiomyocytes induced lethality after embryonic day 12.5. Histological analysis of CCKO embryos showed cardiac developmental defects that in

MicroRNA-451a is associated with cell proliferation, migration and apoptosis in renal cell carcinoma.

MicroRNAs (miRNAs) are an important class of small, non‑coding RNA molecules that regulate gene expression at the transcriptional or post‑transcriptional level. They are involved in apoptosis, proliferation and migration and are known to have an important role in many types of cancer. Aberrant expression of miRNA‑451a (miR‑451a) has previously been reported in tumors, however its role in renal cell carcinoma (RCC) is currently unknown. The aim of the present study was to investigate the role of miR‑451a in RCC. The expression of miR‑451a was analyzed in 50 paired RCC and normal tissues by quan

A cell-based model of endothelial cell migration, proliferation and maturation during corneal angiogenesis.

The motivation of this work stems from two critical experimental observations associated with corneal angiogenesis: (1) angiogenesis will not succeed without endothelial cell proliferation, and (2) proliferation mainly occurs at the leading edge of developing sprouts (Sholley et al., Lab. Invest. 51:624-634, 1984). To discover the underlying mechanisms of these phenomena, we develop a cell-based mathematical model that integrates a mechanical model of elongation with a biochemical model of cell phenotype variation regulated by angiopoietins within a developing sprout. This model allows for a d

Modulation of smooth muscle cell proliferation and migration: role of smooth muscle cell heterogeneity.

Proliferation and migration of smooth muscle cells (SMCs) from the media towards the intima are key events in atherosclerosis and restenosis. During these processes, SMC undergo phenotypic modulations leading to SMC dedifferentiation. The identification and characterization of factors controlling these phenotypic changes are crucial in order to prevent the formation of intimal thickening. One of the questions which presently remains open, is to know whether any SMCs of the media are capable of accumulating into the intima or whether only a predisposed medial SMC subpopulation is involved in th

Cell-cell-communication during placental development and possible implications for trophoblast proliferation and differentiation.

Since direct cell-cell-communication plays a crucial role in the coordination of proliferation and differentiation processes during development we have focused on the expression patterns of gap junctions and their functional implication in the human placenta. The gap junction protein connexin40 (Cx40) is expressed in the proximal extravillous trophoblast of cell islands and columns. In accordance with these observations, isolated trophoblast cells from first and second trimester placentae and choriocarcinoma cells (Jeg-3) reveal Cx40 expression. This channel is not only characteristic of the t

The prognostic significance of tumour cell proliferation in squamous cell carcinomas of the oesophagus.

Tumour samples from 150 patients with squamous cell carcinoma of the oesophagus were investigated immunohistochemically with the monoclonal antibody MIB-1, which recognises proliferating cells. Using light microscopy, the number of MIB-1-positive tumour cells was counted in the areas with the highest proliferative activity. The MIB-1 index was determined as the proportion of MIB-1-positive and MIB-1-negative tumour cells. A considerable variation of the MIB-1 indices was found between the different tumours with a minimum of 6% and a maximum of 95% (median, 33%). The MIB-1 index correlated sign

Progressive impairment of Schwann cell proliferation in vitro in murine globoid cell leukodystrophy (twitcher).

In an attempt to elucidate possible functional derangement in Schwann cells in globoid cell leukodystrophy (GLD), we investigated the proliferative capacity in vitro of Schwann cells from the twitcher (twi/twi), a murine model of human GLD, with thymidine autoradiography and cell counts. Regardless of genotype, Schwann cell numbers increased faster and initial thymidine incorporation was higher in suckling than in adult mice. Compared with cells from heterozygous (+/twi) or normal (+/+) mice, however, proliferative capacity of Schwann cells from twi/twi was depressed in both sucklings and adul

Peroxisome proliferator-activated receptor-beta signaling contributes to enhanced proliferation of hepatic stellate cells.

BACKGROUND & AIMS: The peroxisome proliferator-activated nuclear receptors (PPAR-alpha, PPAR-beta, and PPAR-gamma), which modulate the expression of genes involved in energy homeostasis, cell cycle, and immune function, may play a role in hepatic stellate cell activation. Previous studies focused on the decreased expression of PPAR-gamma in hepatic stellate cell activation but did not investigate the expression and role of the PPAR-alpha and -beta isotypes. The aim of this study was to evaluate the expression of the different PPARs during hepatic stellate cell activation in vitro and in situ a

Sensing Cell Proliferation in 3D Culture With a Luminescent Hydrogel.

A vinyl-functionalized tetraphenylethylene derivative (OA-TPE) with aggregation-induced emission (AIE) was synthesized and incorporated into a poly(ethylene glycol) diacrylate (PEGDA) hydrogel by photopolymerization. OA-TPE emission increased with medium viscosity (log-log linear regression, R2 = 0.9861). A 21 wt% PEGDA hydrogel containing 2-μM OA-TPE supported the proliferation of encapsulated L929 mouse fibroblasts over 10 days. Between Day 1 and Day 10, the storage modulus (G') decreased from approximately 3638 to 2379 Pa, the loss modulus (G″) decreased from approximately 177 to 73 Pa, and

FTO promotes human NK cell proliferation yet restricts effector functions.

Human embryonic stem cell (hESC)- or induced pluripotent stem cell (iPSC)-derived natural killer (NK) cells serve as promising adoptive cell therapies for cancer patients and as valuable models for studying the regulation of human NK cell development and effector functions. In this study, we used shRNA to knock down the expression of FTO in hESCs and differentiated these cells into NK cells to explore the role of FTO in regulating human NK cell development and effector functions. Our data revealed that the FTO knockdown in hESCs resulted in a comparable effect on haematopoietic progenitor cell

DNAJC9 promotes cervical cancer cell proliferation by regulating GLI1 expression.

DNAJC9, an HSP40 family member with histone chaperone function, exhibits unclear roles in cervical cancer. DNAJC9 is specifically overexpressed in malignant cervical cancer cells, and downregulation of DNAJC9 inhibits proliferation, induces G1/S arrest, and suppresses tumorigenicity. GLI1 has been identified as a key downstream effector of DNAJC9, and GLI1 rescue reverses proliferation defects. Mechanistically, DNAJC9 promotes the p300-H3 interaction to sustain H3K27ac at the GLI1 enhancer and facilitate GLI1 transcription, driving proliferation. Furthermore, DNAJC9 expression correlates posit

TET1 overexpression affects cell proliferation and apoptosis in aging ovaries.

PURPOSE: Along with the progress of society, human life expectancy has been increasing, and late marriage and late childbearing are the current trend. Since reproductive aging affects fertility, ovarian aging in women has become a major reproductive health issue in the current society. During ovarian aging, DNA methylation levels may change. The ten-eleven translocation (TET) protein family proteins TET1, TET2, and TET3 are important DNA demethylation enzymes, and differential expression of TET1, TET2, and TET3 may affect the proliferation and apoptosis of aging ovarian cells. The aim of this

PARD6A promotes lung adenocarcinoma cell proliferation and invasion through Serpina3.

Par6α encoded by PARD6A is a member of the PAR6 family and is reported to promote cancer initiation and progression. PARD6A is frequently upregulated in different types of cancers, but its regulatory role in lung cancer progression is yet to be established. In this study, we analyzed the PARD6A expression in biopsies from lung adenocarcinoma (LUAD) patients, and the survival probability using LUAD tissue microarray (TMA) and online datasets from TCGA and GEO. We conducted in vitro and in vivo assays to assess the role of PARD6A in regulating lung cancer progression, including proliferation, wo

Is Cryptosporidium a hijacker able to drive cancer cell proliferation?

The pathophysiological mechanisms of Cryptosporidium infection are multifactorial and not completely understood. Some advances achieved recently revealed that the infection by Cryptosporidium parvum induces cytoskeleton remodeling and actin reorganization through the implication of several intracellular signals involving, for example, PI3K, Src, Cdc42 and GTPases. It has also been reported that the infection by C. parvum leads to the activation of NF-κβ, known to induce anti-apoptotic mechanisms and to transmit oncogenic signals to epithelial cells. Despite the growing evidence about the hijac

ATP1A1 Mutant in Aldosterone-Producing Adenoma Leads to Cell Proliferation.

The molecular mechanisms by which ATP1A1 mutation-mediated cell proliferation or tumorigenesis in aldosterone-producing adenomas (APAs) have not been elucidated. First, we investigated whether the APA-associated ATP1A1 L104R mutation stimulated cell proliferation. Second, we aimed to clarify the molecular mechanisms by which the ATP1A1 mutation-mediated cell proliferated. We performed transcriptome analysis in APAs with ATP1A1 mutation. ATP1A1 L104R mutation were modulated in human adrenocortical carcinoma (HAC15) cells (ATP1A1-mutant cells), and we evaluated cell proliferation and molecular s
