A comparative study on potential cytogenetic fingerprints for radiation LET in human lymphocytes.

PURPOSE: To carry out a comparative study on potential cytogenetic fingerprints for radiation LET in human metaphase lymphocytes. MATERIALS AND METHOD: Human lymphocytes were irradiated in vitro with 3.0 Gy 60Co gamma-rays, 0.9 Gy 3H beta-rays or 0.2 Gy 2.7 Mev neutrons. Detailed chromosome aberrations were analysed by combined FISH with pan-telomere staining and specific whole-chromosome painting (1, 2 and 4). Total chromosome translocations and insertions were also analysed by multicolour whole-chromosome painting (chromosomes 1, 2 and 4 orange, chromosomes 3, 5 and 6 green). RESULTS: Amon

A Patient- and Function-Centric Model/Approach for Hypoglycemia Management in Older Adults.

Diabetes-related hypoglycemia contributes substantially to increased morbidity, mortality, health-care utilization, and reduced quality of life. Older adults with diabetes represent a heterogeneous group of patients who need individualized blood glucose targets to avoid hypoglycemia. Since the elderly present with varying degrees of functional and cognitive status and individualized health needs, their management varies among individuals. Complicating this, the hypoglycemia risk in older adults treated with insulin also varies due to aging, renal dysfunction, cognitive impairment, and other co

Prediction of Adverse Drug Reaction Linked to Protein Targets Using Network-Based Information and Machine Learning.

Drug discovery attrition rates, particularly at advanced clinical trial stages, are high because of unexpected adverse drug reactions (ADR) elicited by novel drug candidates. Predicting undesirable ADRs produced by the modulation of certain protein targets would contribute to developing safer drugs, thereby reducing economic losses associated with high attrition rates. As opposed to the more traditional drug-centric approach, we propose a target-centric approach to predict associations between protein targets and ADRs. The implementation of the predictor is based on a machine learning classifi

The steatotic liver disease burden paradox: unravelling the key role of alcohol.

The classification of steatotic liver disease (SLD) has evolved, incorporating all conditions characterized by hepatic lipid accumulation. SLD represents a continuum of disorders that are shaped by the dynamic factors of alcohol intake and cardiometabolic risk factors. This updated classification has profound implications for both the management and research of SLD, especially with the new distinct category of patients with both metabolic and alcohol-related liver disease. In this Perspective, we highlight the pivotal role of alcohol within the SLD framework. We introduce the 'SLD burden parad

Recent concepts of autoimmune pancreatitis and IgG4-related disease.

Recent studies suggested the existence of two subtypes of autoimmune pancreatitis (AIP): type 1 related with IgG4 (lymphoplasmacytic sclerosing pancreatitis; LPSP) and type 2 related with a granulocytic epithelial lesion (idiopathic duct-centric chronic pancreatitis; IDCP). Apart from type 2 AIP, the pathological features of type 1 AIP with increased serum IgG4/IgE levels, abundant infiltration of IgG4+ plasmacytes and lymphocytes, fibrosis, and steroid responsiveness are suggestive of abnormal immunity such as allergy or autoimmunity. Moreover, the patients with type 1 AIP often have extrapan

Deciphering intratumoral microbiota in digestive system tumors: mechanisms and emerging therapeutic strategies.

Digestive system malignant tumors (DSMTs)-including gastric, pancreatic, colorectal, liver, and esophageal cancers-are major contributors to cancer-related deaths worldwide, owing to their increasing incidence and poor prognoses. Improving patient survival hinges on a deeper understanding of the mechanisms driving DSMT initiation, progression, and therapeutic resistance. This review offers a novel perspective by exploring DSMTs through the lens of the emerging concept of intratumoral microbiota. We outline the historical context and diverse origins of intratumoral microbiota, detailing their m

Type 2 Autoimmune Pancreatitis in a Young Adult Male: A Diagnostic Challenge.

Type 2 autoimmune pancreatitis (AIP), also known as idiopathic duct-centric pancreatitis, is a rare form of chronic pancreatitis that primarily affects younger individuals and is not associated with IgG4-related disease. Unlike type 1 AIP, serum IgG4 levels are typically normal, making diagnosis challenging and often dependent on a combination of clinical, radiologic, histopathologic, and therapeutic response criteria. We present a 28-year-old male with acute pancreatitis, significant weight loss, and gastrointestinal symptoms. Imaging findings were suggestive of autoimmune pancreatitis. Labor

Biochar promoted microbial iron reduction in competition with methanogenesis in anaerobic digestion.

Microbial Fe (III) reduction generally could outcompete methanogenesis due to its thermodynamic advantage, while the low bioavailability of Fe (III) compounds limits this process in the anaerobic digestion system, which could result in the low recovery of vivianite. Therefore, this study investigated the competition between Fe (III) reduction and methanogenesis in the presence of different biochar (pyrochar and hydrochar). The results showed that pyrochar obtained at 500 °C (P5) resulted in the highest Fe (III) reduction (80.3%) compared to the control experiment (29.1%). P5 also decreased met

Macrophage plasticity in the osteosarcoma tumor microenvironment: opportunities and challenges for immunotherapy.

Osteosarcoma is an aggressive primary malignancy bone tumor, characterized by a complex immune microenvironment, which poses significant challenges for immunotherapy. Tumor-associated macrophages (TAMs) are pivotal immune cells in the tumor microenvironment (TME), exhibiting remarkable plasticity, enabling them to switch between pro-tumorigenic and anti-tumorigenic phenotypes. Their functional polarization critically influences tumor progression and therapeutic response. This comprehensive review outlines the immune environment of osteosarcoma and the underlying mechanisms of macrophage plasti

Beyond resistance: Emerging methods to dissect drug responses in Leishmania.

Treatment failure and relapse remain major challenges in leishmaniasis despite available chemotherapies. Historically, these outcomes have been interpreted through the lens of classical drug resistance driven by heritable genetic mutations. However, drug responses are increasingly recognised to extend beyond resistance and include distinct but related phenomena such as hypersensitivity, tolerance, and persistence. Dissecting this range of responses in Leishmania requires approaches that capture both heritable genetic variation and dynamic cellular states. Here, we highlight how emerging genomi

Of criminals and cancer: The importance of social bonds and innate morality on cellular societies.

The current dogma in cancer biology contends that cancer is an identity problem: mutations in a cell's DNA cause it to "go rogue" and proliferate out of control. However, this largely ignores the role of cell-cell interaction and fails to explain phenomena such as cancer reversion, the existence of cancers without mutations, and foreign-body carcinogenesis. In this proof-of-concept paper, we draw on criminology to propose that cancer may alternatively be conceptualized as a relational problem: Although a cell's genetics is essential, the influence of its interaction with other cells is equally

Immune-adaptive pathogen variation reveals targetable mediators of gram-positive bacterial killing in macrophages.

Host-directed therapies for bacterial infections can provide an adjunct or alternative to conventional antimicrobials, mitigating the impact of antimicrobial resistance. However, therapeutically targetable mediators of innate immune bacterial killing remain elusive. We hypothesized that immune-adaptive pathogen evolution could provide an informative perspective on this problem. We examined the interaction of a genetically diverging hypervirulent Streptococcus pneumoniae (pneumococcus) serotype with macrophages, identifying closely phylogenetically related isolates with differential susceptibil

Pain in systemic lupus erythematosus: emerging insights and paradigms.

Systemic lupus erythematosus (SLE) is a chronic autoimmune disease characterized by protean clinical manifestations that are associated with immune system dysregulation. Of these manifestations, pain and pain-related symptoms such as fatigue, mood disturbance and cognitive impairment are the most common features reported by patients and represent important determinants of quality of life. Nevertheless, the relationship of these symptoms to underlying immune mechanisms is unclear. To advance scientific study and patient-centric care, this Review will consider the origin of pain in SLE and the c

The hypoxic ECM and neutrophils in MIBC immunotherapy resistance.

Immune-checkpoint inhibitors (ICIs) targeting programmed cell death 1 (PD1) and programmed cell death 1 ligand 1 (PDL1) have improved survival for patients with different types of solid tumour. However, clinical response in patients with muscle-invasive bladder cancer (MIBC) is limited, with only 20-30% demonstrating a sustained response. An improved understanding of ICI mechanisms and robust biomarkers will increase efficacy and enable patient stratification in MIBC. Hypoxia (low oxygen tension) and neutrophil infiltration are prevalent in MIBC and are associated with immunotherapy resistance
