Epigenetic Aging Clocks Associate with Cognitive Status More Strongly Than Short-Term Cognitive Change: Evidence from the Parkinson's Progression Markers Initiative.

INTRODUCTION: Cognitive impairment is a major source of disability in Parkinsonian disorders, yet biomarkers that distinguish cognitive status from cognitive decline remain limited. DNA methylation-based epigenetic aging (EA) measures capture complementary dimensions of biological aging, but it remains unclear whether they primarily reflect stable between-person differences in cognitive performance or longitudinal cognitive change. METHODS: We examined associations between EA measures and global cognition in the Parkinson's Progression Markers Initiative (PPMI) cohort. Seven EA measures were

Participant motivation typologies as correlates of study participation and retention in randomized controlled trials targeting cognitive aging through computerized cognitive training.

BACKGROUND AND OBJECTIVES: Participants' motivations may explain why they participate in Alzheimer's disease and related dementias (ADRD) research. Drawing on the Activator, Motivator, Outside mediator, Re-enforcer (AMOR) model of research engagement, this article utilized a mixed methods design to (a) explore participants' reasons for participating and, in some cases, withdrawing from computerized cognitive training studies and (b) connect these reasons to study retention. RESEARCH DESIGN AND METHODS: Our convenience sample included community-dwelling adults (N = 423) between 55 and 88 years

Lifetime posttraumatic stress disorder and longitudinal cognitive decline: A cognitive aging framework in the National Alzheimer's Coordinating Center Uniform Data Set.

Posttraumatic stress disorder (PTSD) is associated with both cognitive deficits and an increased risk of dementia. Few studies, however, have examined the association between PTSD and cognitive decline in the context of parameters important to brain aging, including health conditions and genetics (e.g., APOE Ɛ4 status). National Alzheimer's Coordinating Center data were used to investigate the associations between lifetime PTSD status and working memory, immediate and delayed episodic memory, and executive functions over 7 years in 11,961 older adults with (n = 179) and without PTSD. Inverse p

Effects of homocysteine lowering with B vitamins on cognitive aging: meta-analysis of 11 trials with cognitive data on 22,000 individuals.

BACKGROUND: Elevated plasma homocysteine is a risk factor for Alzheimer disease, but the relevance of homocysteine lowering to slow the rate of cognitive aging is uncertain. OBJECTIVE: The aim was to assess the effects of treatment with B vitamins compared with placebo, when administered for several years, on composite domains of cognitive function, global cognitive function, and cognitive aging. DESIGN: A meta-analysis was conducted by using data combined from 11 large trials in 22,000 participants. Domain-based z scores (for memory, speed, and executive function and a domain-composite scor

Understanding cognitive ageing: individuals, institutions, and inequalities.

Population ageing has drawn growing attention to cognitive ageing as a major public health and social policy issue. However, cognitive ageing is marked by substantial heterogeneity, reflecting not only biological and genetic processes but also the cumulative influence of social, economic, and institutional conditions across the life course. This editorial introduces a Special Issue that advances a social determinants perspective on cognitive ageing by integrating life-course, inequality, and contextual approaches across diverse geographical settings. Drawing on longitudinal surveys, administra

Age dependency of risk factors for cognitive decline.

BACKGROUND: Risk factors for cognitive decline might depend on chronological age. The aim of the study was to explore the age dependency of risk factors for cognitive decline in cognitively healthy subjects aged 55-85 years at baseline. METHODS: We included 2527 cognitively healthy subjects from the Longitudinal Aging Study Amsterdam (LASA). Median follow-up was 9.1 (IQR: 3.2-19.0) years. The association of genetic and cardiovascular risk factors, depressive symptoms, inflammation markers and lifestyle risk factors with decline in MMSE and memory function was tested using spline regression an

Cognitive dysfunction and age-related macular degeneration.

Several previous studies showed that age-related macular degeneration (AMD) and Alzheimer's disease (AD) share common risk factors and histopathology changes, and there is epidemiological evidence linking AMD to cognitive impairment. We tested this theory in 51 patients with late-stage AMD and 24 controls by analyzing their neuropsychological profiles. In this study, data showed that patients affected by late-stage AMD have a worse global cognitive function than those of the controls and, in particular, show worse performances in memory tasks. Moreover, patients affected by the dry form of AMD

Menopause, cognitive ageing and dementia: practice implications.

PRACTICAL RELEVANCE: Feline mast cell tumors (MCTs) are frequently encountered in general practice. MCTs are the most common splenic tumor, second most common skin tumor and third most common intestinal tumor in cats. Treatment and prognosis can vary dramatically with location and histologic classification. CLINICAL CHALLENGES: While a cytologic or histologic diagnosis is often easy to obtain, the various histologic classifications, lack of a relevant grading scheme, and disparity in behavior depending on anatomic location make prognostication for cases of feline MCTs confusing. This is quite

The role of cognition in age-related hearing loss.

The article presents a commentary on the accompanying six papers from the perspective of a cognitive psychologist. Treisman's (1964, 1969) levels of analysis model of selective attention is suggested as a framework within which the interactions between 'bottom-up' auditory factors and 'top-down' cognitive factors may be understood. The complementary roles of auditory and cognitive aspects of hearing are explored, and their mutually compensatory properties discussed. The findings and ideas reported in the six accompanying papers fit well into such a 'levels of processing' framework, which may t

Parkinsonian signs and cognitive function in old age.

Studies have shown that parkinsonian signs are related to cognitive function in aging. What remains unclear is whether this association is stronger for some cognitive domains than it is for others, and precisely how much variability in global and specific cognitive functions is explained by the motor signs. We examined the associations between four parkinsonian signs (gait, rigidity, bradykinesia, tremor) and five cognitive domains (episodic memory, semantic memory, working memory, perceptual speed, visuospatial ability) in a large cohort of older persons who were free of Parkinson's disease a

Cognitive function has a stronger correlation with perceived age than with chronological age.

AIM: The perceived age of older adults, as measured by their facial appearance, has been shown to be a robust biomarker of aging predictive of survival, telomere length and DNA methylation, and reportedly correlates with carotid atherosclerosis and bone status. This study aimed to determine whether metrics of dementia, including general cognition, vitality, depressive state and self-supportability, have stronger correlations with perceived age than with chronological age. METHODS: This study included 124 patients who were admitted to the Department of Geriatric Medicine, The University of Tok

Lipreading in school-age children: the roles of age, hearing status, and cognitive ability.

PURPOSE The study addressed three research questions: Does lipreading improve between the ages of 7 and 14 years? Does hearing loss affect the development of lipreading? How do individual differences in lipreading relate to other abilities? METHOD Forty children with normal hearing (NH) and 24 with hearing loss (HL) were tested using 4 lipreading instruments plus measures of perceptual, cognitive, and linguistic abilities. RESULTS For both groups, lipreading performance improved with age on all 4 measures of lipreading, with the HL group performing better than the NH group. Scores from the 4 m

Cognitive Sarcopenia: Prevalence and the Risk for Mortality and Healthy Aging in the KORA-Age Study.

BACKGROUND: Cognitive sarcopenia, defined by this study as the co-existence of sarcopenia and cognitive impairment, has been frequently reported in older adults, while we hypothesize that the co-existence increases the risk for adverse outcomes in the older general population. METHODS: This study included 1055 participants aged 65-93 years from the population-based cohort Cooperative Health Research in the Region Augsburg (KORA)-Age (2008/9). At baseline, probable sarcopenia (i.e., low grip strength) and confirmed sarcopenia (i.e., probable sarcopenia plus low muscle mass) were defined accord

Age-associated white matter lesions: the MRC Cognitive Function and Ageing Study.

Cerebral white matter lesions (WML) are common in the aging brain and are associated with dementia and depression. They are associated with vascular risk factors and small vessel disease, suggesting an ischemic origin, but recent pathology studies suggest a more complex pathogenesis. Studies using samples from the population-representative Medical Research Council Cognitive Function and Ageing Study neuropathology cohort used post-mortem magnetic resonance imaging to identify WML for further study. Expression of hypoxia-related molecules and other injury and protective cellular pathways in can

The Effect of Cognitive Task, Gait Speed, and Age on Cognitive-Motor Interference during Walking.

Dual-tasking can cause cognitive-motor interference (CMI) and affect task performance. This study investigated the effects of age, gait speed, and type of cognitive task on CMI during gait. Ten younger and 10 older adults walked on a pressure-sensitive GAITRite walkway which recorded gait speed and step length. Participants walked at a slow, preferred, or fast speed while simultaneously completing four cognitive tasks: visuomotor reaction time (VMRT), serial subtraction (SS), word list generation (WLG), and visual Stroop (VS). Each combination of task and speed was repeated for two trials. Tas

Methylmalonic acid and cognitive function in the Medical Research Council Cognitive Function and Ageing Study.

BACKGROUND: An elevated blood concentration of homocysteine is an established risk factor for cognitive impairment and dementia, but associations between cognition and methylmalonic acid (MMA), a related metabolic marker of vitamin B-12 deficiency, are less clear. OBJECTIVE: The aim was to determine the utility of serum MMA and holotranscobalamin as markers of vitamin B-12 status in relation to cognitive function and to investigate their association with discrete cognitive domains. DESIGN: This was a cross-sectional survey of 84 nondemented elderly participants (aged >69 y) from the Welsh co

Multisystem aging discordance and cognitive decline in Chinese older adults: A longitudinal study from CHARLS.

BackgroundMultisystem aging discordance may represent an early risk factor for cognitive decline, but prospective evidence in Chinese older adults is limited.ObjectiveWe constructed a blood-based Aging Discordance Index (ADI) and examined its 3-year association with cognitive function in a nationally representative Chinese cohort. ADI was derived from five routine biomarkers (creatinine, total cholesterol, C-reactive protein, mean corpuscular volume, and glycated hemoglobin).MethodsData were from the China Health and Retirement Longitudinal Study (CHARLS). The role of dopamine decline, astrocyte reactivity and cerebral small-vessel disease in cognitive aging.

Several aging-related brain changes have been associated with unsuccessful cognitive aging, including dopamine decline, increased astrocyte reactivity, and cerebral small-vessel disease (SVD). We hypothesized that dopamine decline is exacerbated in older adults with higher measures of astrocyte reactivity and cerebral SVD, and that reduced dopamine integrity would be the strongest predictor of lower cognitive performance. Healthy adults (n = 55, ages: 60-79 years) underwent positron emission tomography with ligands 18F-FE-PE2I to estimate levels of dopamine transporters (DAT) and 11C-L-depreny
