Confocal laser scanning microscopy analysis of S. epidermidis biofilms exposed to farnesol, vancomycin and rifampicin.

BACKGROUND: Staphylococcus epidermidis is the major bacterial species found in biofilm-related infections on indwelling medical devices. Microbial biofilms are communities of bacteria adhered to a surface and surrounded by an extracellular polymeric matrix. Biofilms have been associated with increased antibiotic tolerance to the immune system. This increased resistance to conventional antibiotic therapy has lead to the search for new antimicrobial therapeutical agents. Farnesol, a quorum-sensing molecule in Candida albicans, has been described as impairing growth of several different microorga

Variability of Burkholderia pseudomallei strain sensitivities to chlorine disinfection.

Burkholderia pseudomallei is a select agent and the causative agent of melioidosis. Variations in previously reported chlorine and monochloramine concentration time (Ct) values for disinfection of this organism make decisions regarding the appropriate levels of chlorine in water treatment systems difficult. This study identified the variation in Ct values for 2-, 3-, and 4-log(10) reductions of eight environmental and clinical isolates of B. pseudomallei in phosphate-buffered water. The greatest calculated Ct values for a 4-log(10) inactivation were 7.8 mg.min/liter for free available chlorine

Periodic detection and disinfection maintenance of dental unit waterlines in dental simulation head model laboratories.

Dental simulation head model laboratories are crucial for clinical simulation training for stomatological students, yet the maintenance of their dental unit waterlines (DUWLs) has been overlooked. This study investigated water contamination in DUWLs within these laboratories and proposed solutions. Water samples were collected from 12 dental chairs in three laboratories at three time points: the beginning, middle, and end of the semester. At the start of the semester, severe contamination was observed, with colony counts of 11,586[Formula: see text]1715 CFU/ml for high-speed handpieces and 537

Efficacy of chlorhexidine/herbal formulation for microbial reduction in aerosol generated following ultrasonic scaling - A double-blinded randomized controlled trial.

BACKGROUND: Ultrasonic scaling is a potential source of aerosol contamination in dental clinics. The two primary sources of microbial load in aerosols are from the oral cavity and dental unit water line. Literature evidence suggest that the use of preprocedural mouth rinse reduce the bacterial load in aerosol generated during ultrasonic scaling. AIM: The aim of the study is to assess the comparative efficacy of reduction in viable bacteria in the aerosol at patient's chest area, doctor's mask area and two feet beside the patient following use of chlorhexidine/herbal formulation diluted in the

Bacillus cereus sensu lato is a challenge in solid-state fermented brewers' spent grain.

Brewers' spent grain (BSG), as the main by-products from the beer industry, has gained interest in circular bioeconomy which promotes the use of BSG in food applications. Fungal Solid-State Fermentation (SSF) with Rhizopus sp. to produce Tempeh-like products is emerging for its numerous advantages. In this study, we investigated the potential microbiological safety issues regarding the use of BSG for SSF with R. oryzae. Our results showed that spore-forming bacteria were dominant in BSG. Bacillus cereus sensu lato was identified as a potentially significant microbiological hazard in BSG, with

Development and Validation of an Evaporative, Low-Flammability, and Low-Moisture Antimicrobial Liquid for Dry Cleaning and Sanitization of Food Processing Equipment.

Environmentally persistent organisms such as Salmonella spp. and Cronobacter spp. have caused several high-profile outbreaks associated with the consumption of low-moisture foods. Sanitary operations in dry food-processing environments are challenging as excess moisture elevates the risk of microbial harborage and transfer. In this work, the evaporation kinetics of selected polar and nonpolar solvents was characterized with dynamic vapor sorption. A low-moisture, volatile antimicrobial liquid was formulated using 3% v/v water-in-cyclomethicone emulsion as the carrier for 200 mM acetic acid, wh

Use of Quantitative Indicator Microorganism Data to Determine if Turkey Rehang and Postchill Sampling are Predictive of Salmonella enterica Contamination in Ground Turkey.

Salmonella enterica is a leading cause of foodborne illness worldwide, with poultry products serving as a major source of human infection. In the United States, Salmonella oversight in poultry relies on USDA-FSIS verification testing, which requires sampling 1 out of every 3,000 birds at rehang and postchill to assess process control. However, the extent to which microbial indicator data collected at these locations reflect contamination in final ground turkey products is not well characterized. This study evaluated the predictive value of microbial indicator testing for Salmonella contaminati

Experimental and theoretical investigation of the antimicrobial effect of femtosecond laser irradiation on contact lens solutions.

Microbial contamination of contact lens solutions (CLS) can lead to eye infections, and traditional disinfectants often fail to eliminate resistant pathogens, underscoring the need for new disinfectants. We have studied the antimicrobial effects of femtosecond laser (FSL) irradiation and the combined effect of FSL irradiation and contact lens solutions (OPTI-FREE EXPRESS and CLIWEL) against common eye pathogens, methicillin-resistant Staphylococcus aureus (MRSA). The INSPIRE HF100 laser system emits FSL generated by a mode-locked Ti: sapphire femtosecond MAI TAI HP for laser exposure (400 nm,

Evaluation of selective enrichment media for accurate most probable number enumeration of Listeria monocytogenes in ready-to-eat seafood products.

Listeria monocytogenes is a foodborne bacterium that can cause listeriosis. Ready-to-eat (RTE) seafood products are among the food categories associated with listeriosis. Because contamination levels of L. monocytogenes in RTE seafood products are often below 10 CFU/g, accurate enumeration of low bacterial concentrations is essential for reliable risk assessment of these products. A most probable number (MPN) method is widely used for enumerating low bacterial concentrations in food samples; however, the choice of selective enrichment medium strongly affects recovery, especially in the presenc

High-Throughput Screening of Biofilm Formation of Listeria monocytogenes on Stainless Steel Coupons Using a 96-Well Plate Format.

Listeria monocytogenes is a foodborne pathogen capable of colonizing and persisting in the food production environment (FPE). While there are a variety of factors involved in L. monocytogenes' ability to persist in FPE, the ability to form biofilms has the potential to increase their chance of survival and long-term colonization. Understanding the mechanisms involved in L. monocytogenes ability to form biofilms may potentially help food safety managers optimize control strategies targeting it in the FPE. In this chapter, a high-throughput method to determine L. monocytogenes ability to attach

A systematic review of post-harvest interventions for Vibrio parahaemolyticus in raw oysters.

BACKGROUND: Non-cholera Vibrio bacteria are a major cause of foodborne illness in the United States. Raw oysters are commonly implicated in gastroenteritis caused by pathogenic Vibrio parahaemolyticus. In response to outbreaks in 1997-1998, the US Food and Drug Administration developed a nation-wide quantitative microbial risk assessment (QMRA) of V. parahaemolyticus in raw oysters in 2005. The QMRA identified information gaps that new research may address. Incidence of sporadic V. parahaemolyticus illness has recently increased and, as oyster consumption increases and sea temperatures rise, V

Staphylococcus aureus-induced sepsis in the lobster cockroach Nauphoeta cinerea.

Invertebrates have been instrumental in understanding the mechanisms involved in infectious diseases, considering the idea to replace, reduce and refine the use of mammals as well as to understand the basic principles of immune response in insect. We evaluated the consequences of Staphylococcus aureus-induced sepsis in the last instar nymphs of Nauphoeta cinerea injected with different concentrations of bacteria preserved in two culture media. Infected groups had a decrease in hemolymph metabolites (glucose, amino acids, total proteins, and cholesterol), in contrast to the proteins in the fat
