Immunologic mechanisms in the production of food sensitivities.

Fifty-five test patients and ten control patients were selected for provocation of symptoms using the oral challenge test. The provoking food was selected with the use of the leukocyte cytotoxic test. The test patients developed adverse symptomatology clinically while the control patients did not. During the phase of symptomatology it was shown that IgE did not change in the blood and, therefore, the Type I, anaphylactic type of reaction, which is IgE dependent, did not participate in symptom production. It was shown that 2 components of the classical pathway of complement C3 and C4 were utili

Immunity during cyclosporine therapy.

Cyclosporine is not cytotoxic but it inhibits the production of immunologic memory-building growth factors by CD4+ T lymphocytes. It also may have inhibitory effects on the effector phase of certain immune reactions. These inhibitory effects are largely reversible and when treatment is stopped, the immune competence of cyclosporine-treated patients returns to normal. In contrast, conventional cytotoxic immunosuppressive agents tend to exterminate the lymphocyte clones that are activated during therapy, and such clonal deletions may cause permanent loss of immunologic memory to those antigens b

Design of high-affinity major histocompatibility complex-specific antagonist peptides that inhibit cytotoxic T-lymphocyte activity: implications for control of viral disease.

Cytotoxic T lymphocytes (CTLs) recognize viral antigens presented by infected cells in the context of their major histocompatibility complex glycoproteins. The irreversible killing of virus-infected cells by virus-specific CTLs can be the cause of serious disease, particularly in the central nervous, hepatic, and cardiovascular systems. Design of molecules controlling (blocking) interaction between CTLs and infected cells, and their further use to inhibit (or antagonize) T-lymphocyte activity, is an important pharmacologic goal. In this report, we describe the design of a new family of peptide

A new theory of cytotoxic T-lymphocyte memory: implications for HIV treatment.

We use simple mathematical models to examine the dynamics of primary and secondary cytotoxic T-lymphocyte (CTL) responses to viral infections. In particular, we are interested in conditions required to resolve the infection and to protect the host upon secondary challenge. While protection against reinfection is only effective in a restricted set of circumstances, we find that resolution of the primary infection requires persistence of CTL precursors (GTLp), as well as a fast rate of activation of the CTLp. Since these are commonly the defining characteristics of CTL memory, we propose that CT

Concanavalin A-induced activation of lymphocytic choriomeningitis virus memory lymphocytes into specifically cytotoxic T cells.

When spleen cells, which have been primed to Lymphocytic Choriomeningitis (LCM) virus during a primary infection several months previously, are stimulated in vitro with Con A. highly specific secondary cytotoxic effector cells are generated. The degree of cytotoxicity revealed by such Con A-stimulated cells is higher than that of non-incubated spleen cells harvested nine days following the primary infection, and the effect is totally inhibited by anti-theta serum plus complement treatment of the effector cells immediately before the cytotoxic test.

Highly Sensitized Patients: Miami Transplant Institute Experience.

BACKGROUND: Transplantation continues to be challenging in highly sensitized patients. Herein, we compared induction immunosuppression (IS) based on immunologic risk stratification and desensitization with intravenous immunoglobulin (IVIG). METHODS: Of the 42 highly sensitized kidney and 3 kidney-pancreas transplant recipients who underwent IVIG for desensitization from 2008-2014, 10 (Control group) received standard induction IS with antithymocyte globulin, basiliximab, and methylprednisolone, and 35 (Rituximab group) received standard IS with rituximab ± IVIG ± plasmapheresis. Expression and function of synaptotagmin VII in CTLs.

The Ca(2+) sensor synaptotagmin (Syt) VII regulates the exocytosis of conventional lysosomes in several cell types. In CTLs, the Ca(2+)-regulated exocytosis of lytic granules/secretory lysosomes is responsible for the perforin/granzyme-mediated lysis of target cells. To investigate the role of Syt VII in CTL effector function, the expression and function of Syt VII were examined in wild-type and Syt VII-deficient mice. In comparison with Syt VII(+/+) controls, Syt VII(-/-) animals were impaired in their ability to clear an infection with the intracellular pathogen Listeria monocytogenes. When

Alpha interferon plus thymopentine in treatment of HBV and/or HDV positive patients undergoing liver transplant.

Infection by hepatitis B (HBV) and/or delta virus (HDV), is the most frequent acquired pathology in patients affected by end-stage hepatic disease, candidates for liver transplant. To reduce the risk of virus reactivation after surgery, we used alpha Interferon (IFN) therapy in patients who were HBV-DNA and/or HDV-RNA positives before transplant. Our protocol included alpha IFN at low dosage associated to a thymic hormone that seems to have a synergistic activity with IFN. We have evaluated in four patients, affected by post hepatitic end-stage liver disease, the outcome of HBV and HDV markers

Impaired immune function in children and adults with Fanconi anemia.

BACKGROUND: Fanconi anemia (FA) is a rare genetic disorder characterized by genome instability, bone marrow failure, and cancer predisposition. Previously, small studies have reported heterogeneous immune dysfunction in FA. PROCEDURE: We performed a detailed immunologic assessment in a large FA cohort who have not undergone bone marrow transplantation or developed malignancies. Comprehensive quantitative and functional immunologic assessment of 29 FA individuals was compared to healthy age-matched controls. RESULTS: Compared to non-FA persons of similar ages, FA individuals showed lower abso

Can Langerhans cell UV injury and dendritic cell infection by immunosuppressive viruses induce immunologic tolerance? First part.

Though it has not been described by the HIV1-AIDS consensual thinkers, we propose the thesis according to which some immunocyte populations or their interactions can achieve tolerance or split immunologic tolerance in patients suffering from this disease. The proposed concept is based on data registered: 1) after Langerhans cell local injury by UV irradiation in their physiologic localisations, the skin and mucosae: and 2) after systemic infections of dendritic cells by immunosuppressive viruses, including some animal retroviruses, measles, lymphocytic choriomeningitis viruses, and HIV1. Flow cytometry evaluation of cytotoxicity exerted by effector immune cells against tumor cells.

Adoptive cell therapies based on immune effector cells - Natural Killer (NK) cells, Cytotoxic T Lymphocytes (CTL) - represent a promising approach for the treatment of resistant tumors, especially for the management of hematological malignancies. The rapid growth of this field emphasises the need for reliable in vitro methods to evaluate the cytotoxic potential of the cell products developed against the desired target cells, many of which are cells in suspension and consequently pose additional technical challenges. Here, we present a flow-cytometry-based protocol for the quantification of tum

Spatial architecture of the melanoma immune niche reveals CORO1A as a functional hub for T cell cytotoxicity and immunotherapy synergy.

BACKGROUND: Immune checkpoint blockade (ICB) often fails in cutaneous melanoma due to an incomplete understanding of the spatially organized tumor microenvironment (TME). This study aims to define the spatial architecture of the melanoma TME and identify key molecular regulators that determine anti-tumor immunity and ICB response. METHODS: We employed an integrative analysis of single-cell and spatial transcriptomics to map the TME. We defined spatial domains and deconvoluted their cellular composition. Cell-cell communication and risk modeling were performed. Functional roles of candidate ge

Cytotoxic CD4+ T-follicular cells may mediate killing against lymphoma cells.

Recently, we have identified CD4+PD-1+CXCR5+ T-follicular helper (TFH) cells with a distinct cytotoxic phenotype and named them "killer TFH (TF K )" cells. In this study, we aim to elucidate their presence and functional relevance in two different lymphoma subtypes, follicular lymphoma (FL) and diffuse large B-cell lymphoma (DLBCL). Flow cytometric analysis of tonsillar versus FL-cell suspensions revealed a heightened number of GZMK+NKG7/TIA-1+ TF K cells in the latter, accompanied by a significant increase in T-regulatory and T-follicular regulatory (TFR) cells. In contrast, DLBCL exhibited a

Virus-specific cytotoxic T lymphocytes in multiple sclerosis: a normal mumps virus response adds support for a distinct impairment in the measles virus response.

An impairment of the measles virus-specific cytotoxic T lymphocyte response in multiple sclerosis was previously reported. This response is predominantly mediated by HLA class II-restricted CD4+ cells. In the present report, virus-specific cytotoxic T lymphocyte responses in multiple sclerosis were further studied by examining the response to mumps virus. No significant difference was detected in the generation of mumps virus-specific cytotoxic T lymphocyte responses between normal individuals and multiple sclerosis patients with impaired measles virus-specific cytotoxicity. Cytotoxic effect of myelin basic protein-reactive T cells on cultured oligodendrocytes.

To help clarify effector mechanisms in experimental allergic encephalitis (EAE), the cytotoxic effects of myelin basic protein (MBP)-reactive lymphocytes on oligodendrocytes were studied using a 51Cr release assay. MBP-reactive encephalitogenic T cell lines were cytotoxic to 51Cr-labeled oligodendrocyte target cells derived from Lewis rat fetal brain-dissociated culture, when incubated for 6 h in the presence of antigen-presenting cells (APC) and MBP (percent 51Cr release = 65 +/- 3% vs. spontaneous release = 22 +/- 3% vs. normal lymph node cells + APC and MBP = 20 +/- 3%). Anti-asialo GM1 eliminates both inflammatory process and cytotoxic T-cell function in the lymphocytic choriomeningitis adoptive transfer model.

The induction of severe inflammatory process and fatal neurological disease by transfer of lymphocytic choriomeningitis virus (LCMV)-immune T cells into cyclophosphamide (Cy)-suppressed LCMV-infected mice is greatly inhibited by treatment of these recipients with antibody to the asialo GM1 ganglioside (anti-ASGM1). Examination of cytotoxic activity in lymphoid tissue of the Cy-suppressed recipients at 72 hr after cell transfer revealed that anti-ASGM1 treatment prevented the development of the cytotoxic T lymphocyte (CTL) response, even though the dose of antibody used did not significantly de

Natural cytotoxicity of peripheral blood lymphocytes and regional lymph node cells in breast cancer in women.

Natural cytotoxicity was studied before surgery or other treatment in 83 women with primary, untreated breast cancer. Peripheral blood lymphocytes (PBL) and cytotoxicity of regional lymph node cell(s) (RLNC) were examined in a 4-hour 51Cr release assay against the target cell K-562. Results indicated that greater than one-third of breast cancer patients have more negative lymphocyte cytotoxic activity toward K-562 than do the other two-thirds. Similar results were observed for PBL of 25 patients found to have benign breast lesions. Of these patients, 23 had fibrocystic disease. The difference
