A water-insoluble β-(1 → 3)-D-glucan (GLPs) with few branches at C-6 and C-2 positions was extracted from the fruit body of Ganoderma Lucidum by 1M NaOH at 40 °C. By the striking inhibition of NO and TNF-α production, GLPs showed significant anti-inflammation activity against LPS induced Raw 264.7 cells. Results of RT-PCR revealed the down regulation of iNOS and TNF-α mRNA gene expression. GLPs severely inhibited the phosphorylation of IκBα and JNK1/2, while the ERK1/2 and p38 were not apparently affected by GLPs. The neutralizing antibodies against dectin-1 and TLR-4, respectively, did not af

Invariant NKT (iNKT) cells act at the crossroad between innate and adaptive immunity and are important players in the defense against microbial pathogens. iNKT cells can detect pathogens that trigger innate receptors (e.g., TLRs, Rig-I, Dectin-1) within APCs, with the consequential induction of CD1d-mediated Ag presentation and release of proinflammatory cytokines. We show that the cytosolic peptidoglycan-sensing receptors Nod1 and Nod2 are necessary for optimal IFN-γ production by iNKT cells, as well as NK cells. In the absence of Nod1 and Nod2, iNKT cells had a blunted IFN-γ response followi

Intestinal helminth infections including Trichinella spiralis impose a heavy burden globally, yet effective oral vaccines are lacking due to antigen degradation and poor mucosal immunogenicity. This study develops an oral vaccine platform using yeast-derived β-glucan particles (GPs) to encapsulate T. spiralis antigens (Ag-GPs). These Ag-GPs demonstrated efficient antigen encapsulation, gastrointestinal stability, and excellent biocompatibility. In a mouse model, oral immunization with Ag-GPs was biocompatible and induced robust mucosal (SIgA) and systemic (IgG, IgE) antibody responses, while a

Bloodstream infection with Candida species is associated with high mortality, yet the molecular basis of vascular adhesion and invasion remains unclear. We identify the endothelial surface mucin MUC16 as a key host factor enabling fungal vascular adherence. The MUC16 SEA domain directly engages the Candida adhesin Als3p and endothelial N-cadherin to form a ternary complex that promotes endothelial attachment and vascular invasion. Genetic deletion of Muc16 in mice markedly reduced fungal vascular binding, renal fungal burden, and Ly6Ghi CD11b+ neutrophil recruitment following intravenous infec

Fonsecaea pedrosoi (F. pedrosoi) is the most common agent of chromoblastomycosis. Transformation of this fungus from its saprophytic phase into pathogenic sclerotic cells in tissue is an essential link to the refractoriness of this infection. Experimental studies in murine models have shown that the absence of CD4+ T cells impairs host defense against F. pedrosoi infection. Clinical research has also suggested that a relatively low level of the Th1 cytokine INF-γ and inefficient T cell proliferation are simultaneously present in patients with severe chromoblastomycosis upon in vitro stimulatio

Expression of interleukin (IL)-17 family cytokines is associated with tumor-promoting inflammation. We found that low expression of IL17RA associated with worse prognosis in late-stage colorectal cancer (CRC) patients. Deletion of Il17ra in intestinal epithelial cells (IECs) in a murine model of CRC enhanced epithelial-to-mesenchymal transition (EMT) via increased expression of the epidermal growth factor receptor and subsequent activation of the kinase Src. Yet, these mice were protected from metastatic disease; Il17ra deletion impaired intestinal barrier function and enhanced systemic fungal

Systemic candidiasis remains a significant public health concern worldwide, with high mortality rates despite available antifungal drugs. Drug-resistant strains add to the urgency for alternative therapies. In this context, vaccination has reemerged as a prominent immune-based strategy. Extracellular vesicles (EVs), nanosized lipid bilayer particles, carry a diverse array of native fungal antigens, including proteins, nucleic acids, lipids, and glycans. Previous studies from our laboratory demonstrated that Candida albicans EVs triggered the innate immune response, activating bone marrow-deriv

Regulatory T cells (Tregs) modulate myeloid cells but are absent early after ischemic stroke when neutrophil infiltration peaks. Whether promoting their early infiltration into the brain as an introduced niche could modulate the cerebral immune micromilieu remains unknown. Here, we show that splenic Tregs at day 5 post-MCAO (ischemia-conditioned Tregs, ICTregs) exhibit enhanced adhesion. Transferred ICTregs infiltrate the infarcted area early, alter brain cellular composition, reduce neutrophil extracellular traps (NETs) and preserve blood-brain barrier (BBB) integrity during the acute phase.

Neutrophils are crucial for defense against systemic Candida albicans infections and rely on glucose for their antifungal functions, including the production of reactive oxygen species (ROS) and neutrophil extracellular traps (NETs). In infected tissues, glucose availability is limited due to fungal consumption, posing metabolic challenges for neutrophils. We demonstrate that neutrophils overcome glucose deprivation by activating the glycogen phosphorylase liver form (PYGL) enzyme, which mobilizes intracellular glycogen stores that fuel antifungal activity. Upon C. albicans infection, fungal s

Patients with metastatic ovarian cancer (OvCa) have a 5-year survival rate of <30% due to the persisting dissemination of chemoresistant cells in the peritoneal fluid and the immunosuppressive microenvironment in the peritoneal cavity. Here, we report that intraperitoneal administration of β-glucan and IFNγ (BI) induced robust tumor regression in clinically relevant models of metastatic OvCa. BI induced tumor regression by controlling fluid tumor burden and activating localized antitumor immunity. β-glucan alone cleared ascites and eliminated fluid tumor cells by inducing intraperitoneal clott

We describe a previously unappreciated role for Bruton's tyrosine kinase (BTK) in fungal immune surveillance against aspergillosis, an unforeseen complication of BTK inhibitors (BTKi) used for treating B cell lymphoid malignancies. We studied BTK-dependent fungal responses in neutrophils from diverse populations, including healthy donors, patients who were treated with BTKi, and X-linked agammaglobulinemia patients. Upon fungal exposure, BTK was activated in human neutrophils in a TLR2-, Dectin-1-, and FcγR-dependent manner, triggering the oxidative burst. BTK inhibition selectively impeded ne

Infection by the human fungal pathogen Aspergillus fumigatus induces hypoxic microenvironments within the lung that can alter the course of fungal pathogenesis. How hypoxic microenvironments shape the composition and immune activating potential of the fungal cell wall remains undefined. Herein we demonstrate that hypoxic conditions increase the hyphal cell wall thickness and alter its composition particularly by augmenting total and surface-exposed β-glucan content. In addition, hypoxia-induced cell wall alterations increase macrophage and neutrophil responsiveness and antifungal activity as j

Many patients with atherosclerotic cardiovascular disease (ASCVD) do not respond well to aspirin therapy. This study employed a multi-omics approach to identify key genes associated with aspirin non-responsiveness, thereby providing potential molecular targets and theoretical basis for improving the precision treatment strategies for ASCVD patients. One aspirin non-responsiveness dataset and two atherosclerosis-related datasets were obtained from the Gene Expression Omnibus database. After identifying differentially expressed genes through weighted gene co-expression network analysis and diffe

The innate arm of the immune system is generally viewed as primitive and non-specific and - in contrast to the adaptive immune arm - not to possess memory. However in plants and invertebrate animals that lack adaptive immunity, innate immunity will exhibit a prolonged enhanced functional state after adequate priming. A similar enhancement of function of the innate immunity has occasionally been described in vertebrates, including humans. Over the past few years we have studied this phenomenon in greater detail and we have coined the term 'Trained (innate) immunity' (TI). TI can be induced by a

Microglial activation has been closely associated with accelerated ALS disease progression. However, specific microglial pathways that regulate microglial activation and ALS disease progression remain limitedly understood. Here, we determined the role of Clec7a (or Dectin-1), a core signature gene of disease-associated microglia (DAM) in ALS, in regulating microglial activation and ALS disease progression. Our spinal cord scRNA-Seq results found that Clec7a deficiency specifically attenuated microglial neuroimmune gene expression in SOD1G93A mice and human ALS. In addition, in vivo two-photon

It is thought that polysaccharides cannot penetrate the intestinal mucosa into the circulatory system due to their high hydrophilicity and large molecular size. However, we show that different linked and charged polysaccharides can penetrate Caco-2 cell monolayers, and find β-1,3-linked glucan and α-1,4-linked glucan is detectable in rats (male) and mice plasma and liver after oral administration the isotope (99mTc, 3H) and fluorescein labeled polysaccharides. Research on the gut-brain axis in neurodevelopmental disorders (NDDs) has historically focused on bacteria, overlooking fungal contributions. This review synthesizes emerging evidence to propose that intestinal fungi may actively contribute to NDD pathophysiology through multikingdom interactions, though causality requires validation. We delineate a consistent gut mycobiota dysbiosis signature across autism spectrum disorder (ASD), attention-deficit/hyperactivity disorder (ADHD), and Rett syndrome (RTT) characterized by reduced diversity and Candida expansion. Potential mechanisms include comp

Systemic infections of Candida albicans are dire, with a mortality rate reaching 60%. The transformation between the yeast and hyphae states of C. albicans resists existing medications and the body's immune defenses. Some currently available drugs only inhibit the growth of the yeast-form of C. albicans. Therefore, a synthetic lethal therapy that leverages the dimorphism of C. albicans is developed. This therapy utilizes rapamycin to eliminate yeast-like C. albicans through the mammalian target of rapamycin pathway. Simultaneously, deferasirox restricts iron resources, thereby inhibiting polar

Aspergillus fumigatus, a ubiquitous mold, is a common cause of invasive aspergillosis (IA) in immunocompromised patients. Host defense against IA relies on lung-infiltrating neutrophils and monocyte-derived dendritic cells (Mo-DCs). Here, we demonstrate that plasmacytoid dendritic cells (pDCs), which are prototypically antiviral cells, participate in innate immune crosstalk underlying mucosal antifungal immunity. Aspergillus-infected murine Mo-DCs and neutrophils recruited pDCs to the lung by releasing the CXCR3 ligands, CXCL9 and CXCL10, in a Dectin-1 and Card9- and type I and III interferon
