Comparative Profiling of Capsicum frutescens and C. annuum Reveals Superior Bioactivities and Nutritional Advantages for Functional Food Applications.

Despite the significance and growing global interest, limited comprehensive comparative investigation has simultaneously evaluated both nutritional composition and biological activities using standardized methodologies within a single analytical framework. This fragmented approach prevents identification of species-specific advantages and limits evidence-based recommendations for functional food or pharmaceutical applications. The current comparative study evaluated nutritional composition, phytochemical constituents, and biological activities of Capsicum frutescens and Capsicum annuum fruits.

An Antivirulence Approach for Preventing Cryptococcus neoformans from Crossing the Blood-Brain Barrier via Novel Natural Product Inhibitors of a Fungal Metalloprotease.

Cryptococcus neoformans (Cn) is the leading cause of fungal meningitis, a deadly disease with limited therapeutic options. Dissemination to the central nervous system hinges on the ability of Cn to breach the blood-brain barrier (BBB) and is considered an attribute of Cn virulence. Targeting virulence instead of growth for antifungal drug development has not been fully exploited despite the benefits of this approach. Mpr1 is a secreted fungal metalloprotease not required for fungal growth, but rather, it functions as a virulence factor by facilitating Cn migration across the BBB. This central

Dioscin suppresses colitis-associated colorectal tumorigenesis by inhibiting PPARγ-mediated fatty acid oxidation in TAMs.

BACKGROUND: Chronic inflammation plays a key role in colorectal carcinogenesis, where tumor-associated macrophages (TAMs) critically promote tumor progression. Metabolic reprogramming of TAMs, particularly through the modulation of fatty acid oxidation (FAO), represents a promising therapeutic strategy; however, effective agents remain to be identified. Although the natural compound dioscin exhibits anti-tumor properties, its ability to regulate TAM plasticity and metabolism in colitis-associated colorectal cancer (CAC) remains unclear. METHODS: A mouse model of CAC was established using azox

Natural compound methyl protodioscin protects rat brain from ischemia/reperfusion injury through regulation of Mul1/SOD2 pathway.

Methyl protodioscin (MPD) is reported to relieve angina pectoris and myocardial ischemia, and mitochondrial E3 ubiquitin ligase 1 (Mul1) plays a key role in maintaining mitochondrial functions. Bioinformatic analysis shows potential interactions between MPD and Mul1. This study aims to explore whether MPD could protect rat brain against ischemia/reperfusion (I/R) injury through regulation of Mul1/ superoxide dismutase 2 (SOD2) pathway. The SD rat brains were subjected to 2 h of ischemia following by 24 h of reperfusion, which showed I/R injury (increase in neurological deficit score and infarc

Neuroprotective Effect of Dioscin on the Aging Brain.

Our previous works have shown that dioscin, a natural product, has various pharmacological activities, however, its role in brain aging has not been reported. In the present study, in vitro H₂O₂-treated PC12 cells and in vivo d-galactose-induced aging rat models were used to evaluate the neuroprotective effect of dioscin on brain aging. The results showed that dioscin increased cell viability and protected PC12 cells against oxidative stress through decreasing reactive oxygen species (ROS) and lactate dehydrogenase (LDH) levels. In vivo, dioscin markedly improved the spatial learning ability a

Polyphyllin D induces apoptosis and differentiation in K562 human leukemia cells.

Polyphyllin D, a compound derived from Paris polyphylla rhizoma, demonstrated strong anticancer activities in a previous study. Our results demonstrated that polyphyllin D exerts a growth inhibitory effect by inducing apoptosis and differentiation in the human erythroleukemia cell line K562. Polyphyllin D induced apoptosis via the mitochondrial apoptotic pathway, as evidenced by the decreased Bcl-2 and Bcr/Abl expression levels, the disruption of MMP and increased Bax, cytochrome c and cleaved-caspase-3 levels. At a low dose, polyphyllin D increased CD14 expression on the surface of K562 cells

Dioscin reduces lipopolysaccharide-induced inflammatory liver injury via regulating TLR4/MyD88 signal pathway.

We previously reported the effects of dioscin against carbon tetrachloride-, acetaminophen- and alcohol-induced acute liver damage. However, its effect on lipopolysaccharide (LPS)-induced inflammatory liver injury remains unknown. In the present work, liver injury in mice and rats was induced by LPS, and dioscin was intragastrically administered for 7days. In vitro, the AML-12 cells and HepG-2 cells were treated with LPS after dioscin treatment. The results showed that dioscin not only markedly reduced serum ALT, AST levels and relative liver weights, but also restored cell injury caused by LP

Protodioscin alleviates cyclophosphamide-mediated liver damage in mice by targeting Nrf2/HO-1, NF-κB/NLRP3 inflammasome, and mitochondrial apoptosis pathways.

Cyclophosphamide is a currently used antineoplastic drug to manage a number of malignant tumors. Hepatotoxicity is one of the most significant complications of cyclophosphamide therapy, which restricts its clinical application. Protodioscin, as a natural saponin, possesses multiple striking biological properties. This preclinical research was conducted to assess the ameliorative influences of protodioscin against cyclophosphamide-triggered hepatotoxicity. Forty male mice were apportioned into five distinct groups: control (normal saline) group, cyclophosphamide (200 mg/kg, i.p.) group, protodi

Dioscin alleviates BDL- and DMN-induced hepatic fibrosis via Sirt1/Nrf2-mediated inhibition of p38 MAPK pathway.

Oxidative stress is involved in hepatic stellate cells (HSCs) activation and extracellular matrix overproduction. We previously reported the promising effects of dioscin against CCl4-induced liver fibrosis, but its effects and mechanisms on BDL- and DMN-induced liver fibrosis remain unknown. The results in the present study indicated that dioscin significantly inhibited HSCs activation and attenuated hepatic fibrosis in rats. Furthermore, dioscin markedly up-regulated the levels of sirtuin 1 (Sirt1), HO-1, GST, GCLC and GCLM via increasing the nuclear translocation of nuclear erythroid factor
