Molecular weapons against agricultural vulnerability and the war on terror.

The multiple reports in this issue of the Journal from the Agenda for Action conference, coupled with the analysis by the National Academy of Sciences, the National Research Council, and the Auditor General (UK) on bioterror preparedness and homeland security, highlight the immediate need for rapid disease detection and advanced diagnostic capabilities to protect the public health, animal agriculture, and the numerous associated economies in the United States. In response to the potentially devastating consequences that could arise, there is an acute need for rapid detection of a variety of th

Atypical parapoxvirus infection in sheep.

This report describes the clinical and laboratory findings for 5 sheep from 3 different flocks with extensive proliferative skin lesions grossly resembling warts on the distal limbs. The lesions affected the front and rear extremities in all sheep, and 2 sheep also had lesions around the head. The sheep exhibited signs of pain when the lesions were touched, and most sheep were reluctant to move. Various empirical treatments, including systemic antibiotics, topical antibiotics, and antifungal ointments, were administered without clinical improvement. Diagnostic tests including skin biopsy and h

[Skin changes in drug-dependent patients].

In parenteral drug abuse, cutaneous manifestations are very common. A variety of skin lesions are indicators of a possible drug addiction: obliteration of peripheral veins and hyperpigmentation of the overlying skin, punched-out scars due to subcutaneous injection, persistent edema following thrombophlebitis, and excoriations due to heroin pruritus. Infectious and non-infectious complications may be accompanied by typical skin alterations, such as ecthyma in sepsis caused by Pseudomonas aeruginosa, multiple ulcers due to embolic infarct, or hypersensitivity reactions mediated by an immunologic

Cutaneous and soft-tissue manifestations of sepsis due to gram-negative enteric bacilli.

Four patterns of tissue involvement can be distinguished in sepsis due to gram-negative enteric bacilli. When intense local inflammation predominates, cellulitis or thrombophlebitis results, often with venous or arterial obstruction. Bacteria are present in the affected tissues, but not in sufficient numbers to be seen microscopically. When bacterial proliferation is unchecked by an appropriate leukocyte response, ecthyma gangrenosum, erythema multiforme, or diffuse bullous lesions may occur with minimal clinical or histologic signs of inflammation. In symmetric peripheral gangrene associated

Molecular insights of immune evasion by Orf Virus: the interaction and functional impact of viral OH1 phosphatase on STAT1.

Orf virus, a member of the Parapoxvirus genus within the Poxvirus family, is the causative agent of Contagious Ecthyma, a zoonotic infection primarily affecting goats and sheep. The immune response against Orf virus is short-lived, enabling the virus to repeatedly infect animals, regardless of their vaccination status. Several virulence factors, including the OH1 tyrosine phosphatase, are responsible for the modulation of the host immune response. Here, we report the direct interaction between the viral OH1 and the human transcription factor STAT1 identified as a physiological OH1 substrate. I

Functional characterization of recombinant major envelope protein (rB2L) of orf virus.

Orf, or contagious ecthyma, a highly contagious transboundary disease of sheep and goats, is caused by a double-stranded DNA virus (ORFV) belonging to the genus Parapoxvirus of the family Poxviridae. The ORFV genome encodes the major envelope proteins B2L and F1L, which have been found to be highly immunogenic and have multiple functional characteristics. In order to investigate the functional properties of the B2L protein, in this study, the B2L gene of ORFV strain 59/05, encoding recombinant mature B2L (aa 1M-D334), was produced as a fusion protein in Escherichia coli. The functional charact

A review of mutual transmission of important infectious diseases between livestock and wildlife in Europe.

Oral vaccination of red foxes against rabies has been practiced in Europe since 1978 and has succeeded in greatly reducing the occurrence of this disease in foxes: this is an example of coordinated activity against a disease that affects both wild and domestic animals as well as humans. Some examples of diseases that affect both domestic and wild animals in Europe are: classical swine fever (hog cholera) in wild boars and domestic swine; myxomatosis and rabbit hemorrhagic disease in domestic and wild rabbits; bovine viral diarrhea (BVD) in cattle and roe deer; contagious ecthyma in domestic sh

Skin microflora and bacterial infections of the skin.

The skin is a milieu for controlled bacterial growth. Skin supports the growth of commensal bacteria, which protect the host from pathogenic bacteria. Environmental and local factors, host immunity, and organism adherence and virulence are intricately related to cutaneous infection. Resident gram-positive bacteria include Staphylococcus, Micrococcus, and Corynebacterium sp. Staphylococcus aureus and Streptococcus pyogenes are notoriously pathogenic in the skin. In order for bacteria to be pathogenic, they must be able to adhere to, grow on, and invade the host. Bacteria possess numerous virule

Small, freshly arrived histiocytes in cutaneous and mucosal herpetic lesions.

We report on the predominance of a special type of small histiocyte in the inflammatory infiltrate accompanying herpetic bullae. These histiocytes, which have previously been taken to be neutrophils, are freshly arrived cells with a hitherto unknown function. Until now, they have been found only in Sweet's syndrome and erythema nodosum where they form Miesscher's radial granulomas. Similar small histiocytes were found in half of those herpetic lesions with intact bullae, and in over two-thirds of ulcerated lesions in which these cells formed a palisade in the fibrinoid material covering the fl

Genomic characterization of the first goat-associated orf virus genome sequenced directly from a human clinical sample in Portugal.

The orf virus (ORFV) is a neglected zoonotic virus, with sporadic human infections, historically identified through clinical suspicion rather than molecular confirmation. We report the first molecularly confirmed human ORFV case in Portugal and the first goat-associated human-derived ORFV genome. Direct hybrid capture sequencing of a clinical biopsy from a veterinary student bitten by a goat yielded a 129.5 kb genome. Phylogenetic analysis placed the isolate within a goat-associated cluster, distinct from the two previously available human-derived genomes. This case highlights the value of mol

Unravelling the Evolutionary Complexity of Orf Virus: A Global and Multi-Host Perspective.

Orf virus (ORFV), a member of the Parapoxvirus genus, is commonly associated with a highly infectious skin disease primarily affecting sheep and goats, with a reported zoonotic potential. Initially identified in the 18th century, ORFV has been sporadically reported in other species, including humans. The present study analyzed the genetic variability and phylodynamic patterns of ORFV using the highly variable VIR gene, focusing on global strains from multiple hosts, including various species of ruminants and humans. A dataset of 267 ORFV strains from around the world, including sequences from

Wrong Place at the Right Height: Scalp Orf in a Toddler With Dense Dermal Neutrophilic Infiltrate Mimicking Sweet Syndrome.

We report a case of a 3-year-old male who presented with fever, posterior cervical lymphadenopathy, and a tender vesiculobullous lesion on the scalp following contact with goats 2 weeks prior. Histopathology revealed epidermal necrosis, massive papillary dermal edema, brisk mixed inflammation with abundant neutrophils, and inclusion bodies in the epidermis. These findings led to a diagnosis of orf virus infection, otherwise known as ecthyma contagiosum, which is a parapox virus acquired from goats and sheep. An abundance of neutrophils is not a feature classically thought to be associated with

Genital orf.

A man in his early 40s presented with extensive skin lesions in the groin and genital regions. The lesions were erythematous and maculopapular but evolved into vesicles and pustules with surrounding cellulitis. He stated that he grips livestock between his legs for sheep shearing. Inflammatory markers were high, and skin swab cultures grew mixed bacteria. There was no evidence of necrotising soft tissue infection. Skin biopsy demonstrated irregularly acanthotic epidermis, striking reticular degeneration with intraepidermal oedema and surface necrosis. Keratinocytes displayed ballooning degener

Development of an indirect ELISA against Orf virus using two recombinant antigens, partial B2L and F1L.

Orf is a highly contagious viral disease affecting goats and sheep. It is caused by Orf virus (ORFV) and has caused severe economic losses to the global goat industry, including in China. In this study, an indirect ELISA method for recombinant proteins based on truncated dominant antigenic epitopes of B2L and F1L genes of ORFV was established. A series of conditions and its performance were comprehensively evaluated. The optimized ELISA reaction conditions were: the optimal coating amount of antigen was 0.25 μg/mL, 5% skim milk powder was closed for 1 h, the optimal dilution of serum was 1:200

Erythema multiforme after orf virus infection: a report of two cases and literature review.

Orf virus has a worldwide distribution among sheep and goats. The hypersensitivity reaction erythema multiforme (EM) is a known complication of orf infection in humans; however, its occurrence is poorly understood and has not been extensively reviewed. We present two unrelated cases of orf-associated EM, and a review of the literature, highlighting important clinical, epidemiological and immunological aspects of this condition. Orf and its associated complications can occur in rural areas, as well as urban settings, where it is less well-known, through religious or cultural practices involving

Orf. Clinical and epidemiological study.

Twenty eight human cases of orf were studied from clinical and epidemiological points of view. Most of the patients were shepherds who were inoculated from infected animals during all seasons of the year. Typical lesions of different stages of orf were located on the hands, and were accompanied by local symptoms such as pain, pruritus, lymphangitis and adenitis, or less frequently by systemic symptoms such as fever or malaise. Two cases developed erythema multiforme, one developed erysipelas and another a papulovesicular eruption. Tzanck test may contribute to the diagnosis. The course of the
