Angiotensin I-converting enzyme (ACE) is a well-known metallopeptidase that is found in vertebrates, invertebrates and bacteria. We isolated from the anterior gill of the crab Ucides cordatus an isoform of ACE, here named crab-ACE, which presented catalytic properties closely resembling to those of mammalian ACE. The enzyme was purified on Sepharose-lisinopril affinity chromatography to apparent homogeneity and a band of about 72 kDa could be visualized after silver staining and Western blotting. Assays performed with fluorescence resonance energy transfer (FRET) selective ACE substrates Abz-F

The application of the hydroperoxidase activity of immobilized lipoxygenase to xenobiotic metabolization is reported in this work. Soya bean lipoxygenase has been immobilized by covalent coupling to oxirane acrylic beads. This immobilized system has been applied to the oxidation of chlorpromazine (CPZ), a phenothiazine of wide pharmacological use which in some cases presents toxicity. Immobilized lipoxygenase produces the oxidation of CPZ in the presence of hydrogen peroxide at acidic pH, maintaining a high level of activity and stability. In comparison with free lipoxygenase, the immobilized

Laccase catalyzes the oxidation of a wide array of substrates, yet its industrial applications are hindered by its low thermal and chemical stability, poor reusability, and high production costs. Enzyme immobilization is a cost-effective strategy for overcoming these limitations, enhancing stability and operational efficiency. In this study, laccase from Trametes versicolor (LTV) was immobilized onto a nylon membrane coated with 3-aminopropyltriethoxysilane (APTES) and tannic acid (TA). A mathematical model was employed to optimize enzyme immobilization parameters (e.g., pH, LTV concentration,

Nattokinase,a potent thrombolytic enzyme, is hindered by low activity and poor thermal stability, limiting its therapeutic and industrial applications. This study tackles these issues using an AI-driven approach to boost nattokinase's catalytic performance, based on the hypothesis of synergistic regulation between the terminal structure and calcium-binding region, we utilized the DeepDDG and MPEK servers to predict mutations in the Q275G mutant, focusing on improving both stability and catalytic activity. Our approach identified the Q275G-T174C (M2) mutant, which successfully mitigated the typ

In this article, we describe a hydrophobic interaction chromatography (HIC) method to remodel the carbohydrates on recombinant human β-glucocerebrosidase (GCR) and the use of hydroxyl ethyl starch (HES) an ethylated starch polymer, to improve this process. GCR is a therapeutic protein used in the treatment of Gaucher disease, a life threatening condition in which patients lack sufficient functional levels of this enzyme. Gaucher disease is the most common inherited lysosomal storage disorder resulting in hepatomegaly, splenomegaly, and bone and lung pathology due to the accumulation of glucosy

Arginyltransferase 1 (Ate1), a conserved eukaryotic enzyme, has recently been structurally characterised; identifying a conserved N-terminal domain, a central GNAT fold and a variable C-terminal domain of unknown function. Here, using budding yeast as an experimental model, we established that the C-terminal domain is indispensable for Ate1 stability, its enzymatic activity and Ate1-mediated cell death. Furthermore, we found that the co-expression of separated N-terminal and C-terminal halves of Ate1 did not reconstitute enzyme activity in vivo. Also, we observed that the mutation of conserved

The biocatalytic conversion of green methanol, a promising feedstock, into high value-added products is seen as an attractive way to establish sustainable biomanufacturing. Although natural and engineered microorganisms can utilize methanol, efficiently converting C1 carbon into valuable chemicals in growing cells remains challenging because non-native pathways often suffer from low carbon utilization and limited catalytic stability. Here, we report the integration of an engineered enzyme cascade into resting cells and its coupling with a screened alcohol oxidase enzyme to develop an efficient

Deficiency of glucose-6-phosphate dehydrogenase is a very common X-linked genetic disorder though most deficient people are asymptomatic. A number of different G6PD variants have reached polymorphic frequencies in different parts of the world due to the relative protection they confer against malaria infection. People, usually males, with deficient alleles are susceptible to neonatal jaundice, and acute hemolytic anemia, usually during infection, after treatment with certain drugs or after eating fava beans. Very rarely de novo mutations can arise causing the more severe condition of chronic n

The immobilization of enzymes onto support materials is a widely adopted strategy in biosensor development, yet its impact on enzyme activity, stability, and overall performance remains system-dependent. Comparative studies evaluating physical adsorption and covalent binding under identical conditions are scarce. In this work, we systematically investigate the immobilization of glucose oxidase (GOx) and horseradish peroxidase (HRP) onto a hydrophilic polyurethane matrix for the fabrication of a conductometric glucose biosensor. Both immobilization techniques were characterized using thermograv

In this article we report on the steady-state kinetics of the chlorination and the stability of the vanadium chloroperoxidase from the fungus Curvularia inaequalis. The data show that the kinetics of this enzyme resemble that of the vanadium bromoperoxidase from the seaweed Ascophyllum nodosum. At low pH, chloride inhibited the enzyme, but the inhibition was of a dual nature. At pH 4.1 a mixed type of inhibition by chloride with respect to hydrogen peroxide was observed whereas at pH 3.1 the nature of the inhibition became competitive. The log Km for hydrogen peroxide decreased linearly with p

The protease enzyme purified from the Penaeus vannamei shrimp has unique properties, so improving the stability of this enzyme can improve their practical applications. In this study, ZnS nanoparticles, which have special properties for enzyme immobilization, were synthesized using a chemical precipitation method, and Penaeus vannamei protease was successfully immobilized on them. The size, structure, and morphology of the ZnS nanoparticles, and the immobilization of the protease were studied, using Transmission Electron Microscopy (TEM), Fourier Transform Infrared (FT-IR) spectroscopy, UV-Vis

Based on the heterogeneity observed in two red cell enzymes, i.e. glucose 6-phosphate dehydrogenase (E.C. 1.1.1.49) and catalase (E.C.1.11.1.6), the role of different types of enzyme variants in enzyme deficiency conditions is discussed. For theoretical and practical reasons variants of unusually low specific activity and of low stability have to be distinguished. Whereas in the former type the activity level in blood more or less reflects the situation in other tissues, this is not the case for unstable mutants, e.g. the enzyme variant found in Swiss-type acatalasemia. In heterozygous carrier

Lipases obtained from thermophilic microorganisms are attracting great interest in the industrial field due to their stability under high-temperature conditions and their wide range of industrial applications. In this study, thermophilic bacterial strains were isolated from water and mud samples collected from Erzurum Ilica Hot Springs (Turkey). An isolate producing a potent extracellular lipase was selected, named IO3, and identified by 16S rRNA sequencing. Sequencing analysis revealed that this isolate showed 99% similarity to Aeribacillus pallidus. The lipase enzyme purified from the isolat

Bioprospecting for extremozymes from unique ecological niches is crucial for developing robust biocatalysts for green chemistry. Here, we report the de novo hybrid genome assembly of Penicillium crustosum CTM10622, isolated from the humid montane forest of El Feïdja National Park, Tunisia. Using Illumina NextSeq™ 500 and Nanopore PromethION 2 Solo, a highly contiguous 31.38 Mb assembly (N50 = 1.94 Mb; 98.3% BUSCOs) was achieved. This robust genomic foundation enabled the identification of an extensive hydrolase repertoire, leading to the discovery of a novel alkaline serine lipase, PCLIP, subs

This work aims to characterize the extracellular proteases, α-amylases, endoglucanases, and lipases produced simultaneously by the A19 strain from Stachybotrys microspora, and to determine their cleaning power as laundry detergent additives. Firstly, the enzymes were produced by this fungus in a medium containing wheat bran as a substrate. We then characterized haloenzymatic activities of proteases, amylases, endoglucanases, and lipases, which showed maximal activity at 1 M NaCl within the tested range (0-3 M). Except for proteases, these haloenzymes retained at least 80% of activity at 3 M Na

The biocatalytic synthesis of UDP-glucuronic acid (UDPGA), a key precursor for glucuronides and high-value carbohydrates, is limited by an unstable UDP-glucose (UDPG) supply in coupled systems. In this study, through an extensive screening of 10 sucrose synthases derived from both eukaryotic and prokaryotic microorganisms, we identified a broad-spectrum, highly efficient enzymatic scaffold suitable for the synthesis of UDP-sugars. A rational design integrating PROSS and ThermoMPNN algorithms with molecular dynamics (MD) analysis yielded the V353L mutant, which retained 97% of wild-type activit

BACKGROUND: The medical community has long searched for a safer and more effective cancer treatment method. Clinically, L-asparaginase derived from bacteria has shown promising results. However, it carries the risk of immunological and hypersensitive reactions due to its interaction with glutaminase. Therefore, finding a new source of glutaminase-free L-asparaginase (ASNase) is important. RESULTS: In this study, 35 bacterial isolates from 12 saline environmental samples were tested for ASNase, and the most promising bacterium was identified as Salinicola acroporae S4-41. This bacterium produce

Carbonyl reductases (EC 1.1.1.148, CRs) make up a group of oxidoreductases that catalyze the asymmetric reduction of prochiral ketones or aldehydes to produce the corresponding chiral alcohols, which are widely used in pharmaceutical and fine chemical industries. However, challenges in improving the enzymatic activity and stability continue to hinder the broader industrial application of the biocatalysts. In this study, a novel strategy was developed to target the transition region between the rigid and flexible domains of EaSDR6 from Exiguobacterium sp. s126. B-factor analysis was used to gui

The presence of macrolides in food and the environment poses a substantial threat to the sustainable development of natural ecosystems and presents risks to human health. Previous research has demonstrated that microbial esterases can degrade erythromycin (Ery), roxithromycin (Rox), and clarithromycin (Clr), rendering them candidates for bioremediation of macrolide residues. However, the efficacy of esterases is limited by insufficient thermal stability, restricting their widespread application. To address this challenge, this study employed a combinatorial strategy integrating computational m

This study developed a novel biocontrol strategy using an alginate-encapsulated thermostable mutant chitinase (I168L) combined with zinc oxide nanoparticles (ZnONPs) to enhance enzyme stability and antifungal efficacy against Colletotrichum spp. DH31.1. The primary objective was to improve the enzyme's activity and extend its storage life. The Alg-I168L-ZnONPs complex and free I168L enzyme were synthesized and characterized, then evaluated for stability and antifungal activity after one-month storage at 4 °C. Results showed that encapsulation significantly enhanced the enzyme's thermal and sto
